Study on the Community Structure of Meiofauna in Marian Cove, King George Island, Antarctica (남극 King George Islands, Marian Cove의 중형저서생물 군집 구조에 관한 연구)

  • The community structure, vertical distribution and harpacticoids composition of the meiofauna community were observed from five stations in Marian Cove, King George Island and one station on the northeastern side of Nelson Island. Sample was taken by a free-fall corer in December 2002. Generally, 11 taxa of meiofauna were found, and meiofauna abundance ranged from 322 to 1575 indiv. $10cm^{-2}$ (mean 781 indiv. $10cm^{-2}$). Nematodes were the most dominant group, making up $89\%$ of total meiofauna, followed by harpacticoids $(6.8\%)$. Benthic harpacticoids appeared 19 species of nine families at all the stations, and most various taxa appeared at station B (13 species of seven families). For vertical distribution, more than $70\%$ of meiofauna was concentrated in the upper $0\~2cm$ sediment layers, and the density abruptly decreased with depth in all the stations. Total biomass of meiofauna varied between 41 and $360{\mu}gC\;10cm^{-2}$, and overall mean biomass was $205{\mu}gC\;10cm^{-2}$. Also nematodes had the highest percentage of total maiofauna biomass $(62.4\%)$. The analysis results of Canonical Correspondence Analysis between meiofauna community and sediment grain size showed that polychaets, oligochaets and cumaceans were influenced by silt&cray, and sand, granule and pebble had a influence on harpacticoids, kinorhynchs and ostracods respectively. But nematodes were not affected by sediment grain size.

Curcumin Effect on MMPs and TIMPs Genes in a Breast Cancer Cell Line

  • Curcumin (CM) possesses anti-cancer activity against a variety of tumors. Matrix metalloproteinases (MMPs) play an important role in remodeling the extracellular matrix and their activities are regulated by tissue inhibitor of metalloproteinases (TIMPs) family. Control of MMP and TIMP activity are now of great significance. In this study, the effect of CM is investigated on metastatic MMPs and anti-metastatic TIMPs genes on MDA breast cancer cells cultured in a mixture of DMEM and Ham's F12 medium and treated with different concentrations of CM (10, 20 and $40{\mu}M$ for various lengths of time. Reverse transcription followed by quantitative real time PCR was used to detect the gene expression levels of MMPs and TIMPs in CM-treated versus untreated cases and the data were analyzed by one-way ANOVA. At high concentrations of curcumin, TIMP-1, -2, -3 and -4 genes were up-regulated after 48 hours of treatment, their over-expression being accompanied by down-regulation of MMP-2 and MMP-9 gene expression levels in a concentration- and time-dependent manner. These results suggest that curcumin plays a role in regulating cell metastasis by inhibiting MMP-2 and MMP-9 and up-regulating TIMP1 and TIMP4 gene expression in breast cancer cells.

Aloe-Emodin Protects RIN-5F (Pancreatic β-cell) Cell from Glucotoxicity via Regulation of Pro-Inflammatory Cytokine and Downregulation of Bax and Caspase 3

  • To determine the protective effect of aloe-emodin (AE) from high glucose induced toxicity in RIN-5F (pancreatic ${\beta}$-cell) cell and restoration of its function was analyzed. RIN-5F cells have been cultured in high glucose (25 mM glucose) condition, with and without AE treatment. RIN-5F cells cultured in high glucose decreased cell viability and increased ROS levels after 48 hr compared with standard medium (5.5 mM glucose). Glucotoxicity was confirmed by significantly increased ROS production, increased pro-inflammatory (IFN-${\gamma}$, IL-$1{\beta}$,) & decreased anti-inflammatory (IL-6&IL-10) cytokine levels, increased DNA fragmentation. In addition, we found increased Bax, caspase 3, Fadd, and Fas and significantly reduced Bcl-2 expression after 48 hr. RIN-5F treated with both high glucose and AE ($20{\mu}M$) decreased ROS generation and prevent RIN-5F cell from glucotoxicity. In addition, AE treated cells cultured in high glucose were transferred to standard medium, normal responsiveness to glucose was restored within 8hr and normal basal insulin release within 24 hr was achieved when compared to high glucose.

Determination of Metal Levels in Shamma (Smokeless Tobacco) with Inductively Coupled Plasma Mass Spectrometry (ICP-MS) in Najran, Saudi Arabia

  • Objective: The use of Shamma (smokeless tobacco) by certain groups is giving rise to health problems, including cancer, in parts of Saudi Arabia. Our objective was to determine metals levels in Shamma using inductively coupled plasma mass spectrometry (ICP-MS). Methods: Thirty-three samples of Shamma (smokeless tobacco) were collected, comprising four types: brown Shamma (n = 14.0), red Shamma (n = 9.0), white Shamma (n = 4.0), and yellow Shamma (n = 6.0). All samples were collected randomly from Shamma users in the city of Najran. Levels of 11 elements (Al, As, Cd, Co, Cr, Cu, Li, Mn, Ni, Pb, and Zn) were determined by ICP-MS. Results: A mixed standard (20 ppb) of all elements was used for quality control, and average recoveries ranged from 74.7% to 112.2%. The highest average concentrations were found in the following order: Al ($598.8-812.2{\mu}g/g$), Mn ($51.0-80.6{\mu}g/g$), and Ni ($23.2-53.3{\mu}g/g$) in all four Shamma types. The lowest concentrations were for As ($0.7-1.0{\mu}g/g$) and Cd ($0.0-0.06{\mu}g/g$). Conclusions: The colour of each Shamma type reflects additives mixed into the tobacco. Cr and Cu were showed significant differences (P < 0.05) among Shamma types. Moreover, Pb levels are higher in red and yellow Shamma, which could be due to use (PbCrO4) as yellow colouring agent and lead tetroxide, Pb3O4 as a red colouring agent. The findings from this study can be used to raise public awareness about the safety and health effects of Shamma, which is clearly a source of oral exposure to metals.

Tolerance of Alternaria mali to Various Fungicides (사과 반점낙엽병균(斑點落葉病菌)의 각종(名種) 살균제(殺菌劑)에 대(對)한 내성(耐性))

  • Alternaria mali isolated from infected apples in storages and orchards of the four main apple growing areas of Taegu, Yesan, Jungup and Chungju in Korea was tested with various fungicides added in PSA medium. The fungal tolerance varied with little difference by orchard area and with great difference by king and concentration of fungicide. Minimum inhibitory concentration (MIC) of chlorothalonil and captafol was $100,238{\mu}g\;and\;81,000{\mu}g/ml$, respectively, showing low effects with high fungal tolerance; of iprodione and folpet was $3,285{\mu}g\;and\;3,000{\mu}g/ml$, respectively, showing moderate effects with moderate fungal tolerance; and of oxidong, polyoxin and polydong was $1,000{\mu}g,\;900{\mu}g\;and\;500{\mu}g/ml$, respectively, showing high effects with low fungal tolerance. Mycelial growth, conidia formation and conidia germination of the fungus showed a similar trend of response to the seven fungicides.

Historical Geography of Mu-Sim River and Nam-Suk Bridge in Chongju City (청주 무심천과 남석교에 관한 역사지리적 고찰)

  • It is recorded that the orignal Nam-Suk Bridge is constructed at the first year of Oh-Bong(B.C. 57). But I believe that it can be back to the 19 year of Jin-Hung King(AD. 558) by discovered bibliographies. The Nam-Suk Bridge is improved and remodelled from past to the late of Zosun dynasty by the development of walled city of Chongju. By the late of Gojong Empire, the Nam-Suk Bridge has stone post and monument of Nam-Suk Bridge. Because the alternations of Mu-Sim River channel, heavy floods, extension of built, up area and Japanese unconcern, the stone post and monument of Nam-Suk Bridge is lost approximately by 1910s and Nam-Suk Bridge is completely buried by the early 1930s. The Mu-Sim Hiver has negative aspects from past to the Japanese rule but has increasingly the positive aspects after the liberation of Korea. We must reconstruct these buried cultural resources to highlight the cultural identity of Chongju City.

Kanahia Laniflora Methanolic Extract Suppressed Proliferation of Human Non-Small Cell Lung Cancer A549 Cells

  • Introduction: Lung cancer is one of the most common cancers worldwide. In certain countries such as United States of America, it is the leading cause of related cancer mortality among both men and women. Natural products play an important role in overcoming the limitations of chemotherapy and radiotherapy. Objectives: In this study, we investigated the antiproliferative and apoptotic activities of Kanahia laniflora methanolic extract against human non-small cell lung cancer cells (A549). Methods: Sulforhodamine B colorimetric assays were used to determine the inhibitory effects of a leaf methanolic extract against A549 cells. Results: The extract showed strong cytotoxic activity against A549 cells with an $IC_{50}$ value of $0.13{\mu}g/ml$ compared to $0.21{\mu}g/ml$ for doxorubicin. The extract also significantly increased the percentage of apoptotic cells to 49.7% as compared to 1.4% and 47.4% for control and doxorubicin respectively. Conclusion: These results showed, for the first time, that a methanolic extract of Kanahia laniflora leaves can inhibit the proliferation of human non-small cell lung cancer cells (A549). Further attention to its potential as a new effective anticancer agent is warranted.

Study on Auto Focusing System of Laser Beam by Using Fiber Confocal Method (파이버 공초점법을 이용한 레이저 빔 자동 초점 제어 장치에 관한 연구)

  • Auto focusing system to find optimized focal position of laser beam used for material process has been investigated by using fiber confocal method. Wavelength of laser diode (LD) and diameter of single-mode fiber are 780nm and $5.3{\mu}m$, respectively. Intensity distributions of beam reflected from the surface of mirror and silicon bare wafer have been observed in a gaussian form. Experimental results show that focal position obtained by LD is shifted from one observed from surface scribed by laser about $80{\mu}m$. It is due to the difference of wavelength and each divergence of between LD and laser used for material process. It is confirmed that auto focusing control system through position calibration has operated steadily.

Production, Purification and Antifungal Activity of Antibiotic Substances Produced by Pseudomonas aeruginosa Strain B5

  • Pseudomonas aeruginosa strain B5 with antagonistic activity against Phytophthora capsici and Magnaporthe grisea, was isolated from pepper-growing soil. From the culture of P. aeruginosa strain B5 grown on King's medium B, antibiotic substances were purified using XAD-2 column chromatography. XAD-2 eluates inhibited not only the mycelial growth of P. capsid and M. grisea, but also the development of Phytophthora blight on pepper plants. The crude antibiotic substances were further purified by using silica gel column chromatography, Sephadex LH-20 column chromatography, thin layer chromatography on silica gel plates, and high performance liquid chromatography. Silica gel column chromatogrphy gave good separation of the four antibiotic substances. The pure antibiotics P1, P2, and P3 finally purified by preparative HPLC inhibited the mycelial growth of P. capsici, at concentrations from 7 to 10 $\mu g/ml$. Only P1 and P2 had antifungal activity against M. grisea at 8 $\mu g/ml$. P1 and P3 were highly inhibitory to the mycelial growth of Botryosphaeria dothidea and Botrytis cinerea at relatively low concentrations. However, the three antibiotics had no antifungal activity against Rhizoctonia solani. The chemical structures of these antibiotics are being identified.
