Interleukin-2/antibody complex expanding Foxp3+ regulatory T cells exacerbates Th2-mediated allergic airway inflammation

  • $Foxp3^+$ regulatory $CD4^+$ T (Treg) cells play an essential role in preventing overt immune responses against self and innocuous foreign antigens. Selective expansion of endogenous Treg cells in response to the administration of interleukin (IL)-2/antibody complex, such as the IL-2/JES6-1 complex (IL-2C) in mice, is considered an attractive therapeutic approach to various immune disorders. Here, we investigated the therapeutic potential of IL-2C in allergic airway inflammation models. IL-2C treatment ameliorated Th17-mediated airway inflammation; however, unexpectedly, IL-2C treatment exacerbated Th2-mediated allergic airway inflammation by inducing the selective expansion of Th2 cells and type-2 innate lymphoid cells. We also found that IL-2 signaling is required for the expansion of Th2 cells in lymphoproliferative disease caused by Treg cell depletion. Our data suggest that IL-2C is selectively applicable to the treatment of allergic airway diseases depending on the characteristics of airway inflammation.

Anti-arthritic Effects of Buthus martensi Karsch Herbal Acupuncture, Inhibiting Interleukin-1-induced Expression of Nitric Oxide Synthase and Production of Nitric Oxide in Human Chondrocytes (전갈 약침액의 인체연골세포에서 nitric oxide synthase의 interleukin-1 유도 유전형질 발현과 nitric oxide의 생산의 억제에 관한 연구)

  • 목적 : 면역억제 작용을 지닌 것으로 알려진 전갈약침(BMK)의 IL-1으로 야기된 1차성 골관절염 인체 연골 세포에 대한 항염증 효과 골 기능 효과에 대해 연구하였다. 방법 : 골관절염 연골에서 채취된 인체 연골세포는 ID-1(2ng/ml)에 의해 처리되어졌으며, IL-1과 BMK($10{\mu}g/ml$)를 함께 처리한 연골세포와 비교하였다. 결과 : IL-1 단독처리된 연골세포에 비해 BMK가 함께 처리된 연골세포에서 연골세포의 손실과 퇴화의 중요한 요소인 NO의 생산량이 의미있게 저하되었다. IL-1단독으로 처리된 연골세포보다 IL-1과 BMK가 함께 처리된 연골세포에서 iNOS mRNA의 단백질 합성이 의미있게 감소하였다. 또한, 전사인자로서의 NF-B의 활성화가 IL-1 단독으로 처리된 연골세포에 비하여 BMK가 함께 처리된 군에서 상대적으로 의미있게 억제되었다. 결론: 이상의 결과를 종합하면 BMK가 인제 골관절염 연골에 있어서 NF-B 활성화에 의존한 IL-1 유도염증의 치료상에 효과적인 반응억제제임을 시사하며, 골 세포의 골 재흡수 활동에 효과적임을 시사한다.

Effects of Cultivated Wild Ginseng Pharmacopuncture on Surgically Induced Endometriosis in Rats (산삼약침이 자궁내막증 유발 흰쥐에 미치는 영향)

  • Purpose : This study was carried out to identified the effects of Cultivated Wild Ginseng Pharmacopuncture(CWG Pharmacopuncture) to the rats with surgically induced endometriosis. Methods : Endometrial tissue was implanted in the serosal wall of the small intestine in rats. The rats were divided randomly into an experimental and control group. The experimental group was treated with CWG Pharmacopuncture intravenous injection to the vein of tail and the control group was given same dose of normal saline for 4 weeks. And the size of the ectopic uterine tissue was estimated, and the serum progesterone, estradiol and cytokine($TNF-{\alpha}$, IL-2, IL-4, IL-6, IL-10) concentrations were analyzed. Results : 1. The size of the ectopic uterine implants in the experimental group was much smaller than that in the control group. 2. In analysis of serum hormone, the concentration of progesterone showed higher value than that of control group and estradiol showed lower value. 3. In analysis of cytokine, $TNF-{\alpha}$, IL-2 and IL-6 concentrations showed lower value than that of control group. 4. In analysis of cytokine, IL-4 concentration showed no significant difference and IL-10 concentrations showed higher value than that of control group. Conclusion : These results indicated that CWG Pharmacopuncture is useful treatment for endometriosis.

Cytokine Release from Organotypic Human Oral Tissue Following Exposure to Oral Care Product Chemicals

  • Purpose: Measuring viability of a three-dimensional in vitro organotypic human oral tissue model has been suggested as an alternative test method to the oral mucosa irritation test of oral care products. The aim of this study was to investigate the production of two different cytokines using organotypic human oral tissue model following exposure to chemicals that are commonly used in oral care products. Materials and Methods: The organotypic human oral tissues were exposed to ethanol, sodium lauryl sulphate or hydrogen peroxide for 90 minutes. Following exposure, interleukin (IL)-1α and IL-8 productions were assessed and correlated with cell viability testing as well as histology of the organotypic human oral tissues. Result: High levels of IL-8 were released from organotypic human oral tissues in all of the test and control groups without any significant differences between them. In contrast, differences were found in IL-1α release between the test and control groups. Additionally, the trend of IL-1α release corresponded to the phenotypes observed in histological analysis while different trend existed between IL-1α release and cell viability. Conclusion: The study concluded the non-specific release of IL-8 for the assessment of oral care product chemicals' toxicity, while potential of measuring IL-1α cytokine level as the possible alternative test method.

  • This paper deals with a novel approach to unknown inputs observer(UIO) design for linear time-invariant dynamical systems using a fast Walsh transform and Walsh function's differential operation. Generally, UIO has a derivation of system outputs which is not available from the measurement directly. And it is an obstacle to estimate the unknown inputs properly when unexpected measurement noises are presented. Therefore, this paper propose an algebraic approach to eliminate such problems by using a Walsh function's differential operation.

Association of an Anti-inflammatory Cytokine Gene IL4 Polymorphism with the Risk of Type 2 Diabetes Mellitus in Korean Populations

  • Chronic inflammation has been implicated as one of the important etiological factors in insulin resistance and type 2 diabetes mellitus (T2DM). To investigate the role of anti-inflammatory cytokines in the development of T2DM, we conducted a case-control study to assess the association between IL4/IL4R polymorphisms and disease risk. We firstly identified single nucleotide poly-morphisms (SNP) at IL4 and IL4RA loci by sequencing the loci in Korean participants. Case-control studies were conducted by genotyping the SNPs in 474 T2DM cases and 470 non-diabetic controls recruited from community-based cohorts. Replication of the associated signals was performed in 1,216 cases and 1,352 controls. We assessed effect of IL4 -IL4RA interaction on T2DM using logistic regression method. The functional relevance of the SNP associated with disease risk was determined using a reporter expression assay. We identified a strong association between the IL4 promoter variant rs2243250 and T2DM risk (OR=0.77; 95% CI, 0.67~0.88; p=$1.65{\times}10^{-4}$ in the meta-analysis). The reporter gene expression assay demonstrated that the presence of rs2243250 might affect the gene expression level with ~1.5-fold allele difference. Our findings contribute to the identification of IL4 as a T2D susceptibility locus, further supporting the role of anti-inflammatory cytokines in T2DM disease development.

Effects of Bifidobacterium Strains Treated with Gastrointestinal Enzymes on Cytokine Induction in RAW 264.7 Macrophage Cells

  • The objective of the current study was to compare the abilities of undigested and enzymatically digested bifidobacteria to induce nitric oxide and cytokine release in RAW 264.7 macrophage cells. Nine different Bifidobacterium strains derived from herbivorous animals were digested with pepsin and then pancreatin, and the precipitates and supernatants were acquired via centrifugation. The RAW 264.7 cells were incubated with whole cells, the precipitate, or the supernatant, and the macrophage culture supernatants were analyzed with respect to the induction of nitric oxide and cytokines. Pronounced increases in the production of nitric oxide, interleukin (IL)-$1{\beta}$, IL-6, IL-12, and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) were observed when cultured with whole cells and the precipitates. It is noteworthy that the precipitates in most of the Bifidobacterium strains evidenced a trend toward superior IL-12 release compared with whole cells. The results showed that both whole cells and digested Bifidobacterium sp. are effective at stimulating RAW 264.7 cells to induce the production of nitric oxide and cytokines. The pepsin-pancreatin system used in the current study may be useful in unraveling the mechanism by which ingested lactic acid bacteria modulate the induction of macrophage mediators at the cellular level.

  • 유체의 전자기력에 의한 거동특성 파악을 위해 전자기 체적력에 의한 접근법을 이용하였다. 수정된 Kelvin force을 유체정역학 방정식에 도입하고 유체표면에서의 압력이 일정해야한다는 원리를 이용하여 유체 표면의 형상을 얻어낸다. 액적에 인가하는 전압의 크기와 전극을 꽂는 높이의 변화에 따른 액적의 Electrowetting 영향을 수치해석한다.

  • This paper describes axial magnetic field characteristics of multipolar axial magnetic field type electrodes for vacuum interrupter used in vacuum circuit breaker. It was distinguished that axial magnetic field distribution in consideration of eddy current effect between electrodes by finite element method. Throughout experiment of magnetic field measurement, confirmed validity of numericla analysis.
