Utilization of Substrate for the In vitro Lipid Synthesis in the Adipose Tissue of Hanwoo Steers

  • An ability to utilize the substrates (acetate, glucose and lactate) in the lipid synthesis was measured in vitro with the adipose tissues of 4 locations (subcutaneous, SUBC; intramuscular, INTR; tail and kidney, KIDN) in 12 Hanwoo (Korean native cattle) steers (26 and 28 months of ages, mean body weight 638.6 kg). The rates of lipid synthesis from acetate were higher than those from glucose in SUBC and ITRA adipose tissues, respectively. In contrast, the rates of lipid synthesis from glucose were higher than those from acetate in the adipose tissues of tail and KIDN, respectively. Lactate utilization was lowest in all the locations while that of acetate or glucose had the different trends of utilization in the lipogenesis. The rate of lipid synthesis from acetate was highest in the SUBC adipose tissue but was lowest in the KIDN while that from glucose was also higher in the SUBC adipose tissue than in the other tissue locations. The rate of lipid synthesis from lactate, however, was highest in the tail adipose tissue among the locations.

  • Hepatocyte growth factor (HGF), originally discovered and cloned as a powerful mitogen for hepatocytes, is a four kringle-containing growth factor which specifically binds to membrane-spanning tyrosine kinase, c-Met/HGF receptor. HGF has mitogenic, motogenic (enhancement of cell movement), morphogenic (e.g., induction of branching tubulogenesis), and anti-apoptotic activities for a wide variety of cells. During embryogenesis, HGF supports organogenesis and morphogenesis of various tissues, including liver, kidney, lung, gut, mammary gland, and tooth. In adult tissues HGF elicits an organotrophic function which supports regeneration of organs such as liver, kidney, lung, and vascular tissues. HGF is also a novel member of neurotrophic factor in nervous systems. Together with the preferential expression of HGF in mesenchymal or stromal cells, and c-Met/HGF receptor In epithelial or endothelial cells, the HGF-Met coupling seems to orchestrate dynamic morphogenic processes through epithelial-mesenchymal (or-stromal) interactions for organogenesis and organ regeneration. HGF or HGF gene may well become unique therapeutic tools for treatment of patients with various organ failure, through its actions to reconstruct organized tissue architectures. This review focuses on recently characterized biological and physiological functions integrated by HGF-Met coupling during organogenesis and organ regeneration.

The Effects of Jehasuogamibang on Antioxidation Activity and Dietary Hyperlipidemia-induced Mice (제하수오가미방(製何首烏加味方)이 고지혈증 및 산화적 손상에 미치는 영향)

  • Objectives: This experiments were performed to determine the effects of the Jehasuogamibang (製何首烏加味方: JHGB) on antioxidationactivity and hyperlipidemia induced by a hypercholesterolemic diet in mice. Methods: After treatment with JHGB expert safety in cytotoxicity and toxicity of Human fibroblast cells and liver and kidney, effect on Reactive Oxygen Species, serum total cholesterol, LDL-cholesterol, triglyceride, glucose, HDL-cholesterol, lipid peroxid of liver tissue, significantly increased SOD and catalase. Results: 1. JHGB showed safety in cytotoxicity and toxicity of human fibroblast cells and liver and kidney. 2. JHGB showed significant inhibitory effect on reactive oxygen species. 3. JHGB significantly decreased serum total cholesterol, LDL-cholesterol, triglyceride, and glucose, and significantly increased serum HDL-cholesterol. 4. JHGB significantly decreased lipid peroxide of liver tissue and significantly increased SOD and catalase. Conclusions: These results suggest that Jehasuogamibang is effective in antioxidationactivity and dietary hyperlipidemia-induced mice.

Changes in Blood and Tissue Free Amino Acid Concentrations in Cats Adapted to Low-and High-protein Diets (단백질 섭취 수준에 따른 고양이의 혈액 및 조직의 유리 아미노산 농도의 변화)

  • Changes in free amino acid concentrations is blood and various tissues were evaluated in cats adapted to the low-protein diet(20% protein, LPD) or the high-protein diet(60% protein, HPD) for 5 weeks. Cumulative body weigth gain for the 5 week period was 463$\pm$43g, and -128$\pm$40g for cats fed HPD and LPD, respectively. Feeding HPD significantly increased the size of liver and kidney. Cats adapted to HPD for 5 weeks have significantly elevated plasma concrntrations of essential amino acids (branched-chain amino acides, threonine, trytophan, phenylalanine and methoionine), whereas plasma levels of non-essential amino acids(alanine, asparagine, glycine, glutamine and serine) were significantly reduced in animals adapted to HPD(p<0.01, or p<0.001) compared to the values for the cats fed LPD. Changes in free amino acid concentratioks in whole blood induced by the variations in dietary level of protein closely reflect the pattern seen in plasma. Amino acids such as branched-chain amino acids, proline and threonine were most difficult to maintain homeostasis and consistantly elevated in lever, kidney, skeletal muscle and brain, as well as in blood of cats adapted to HPD(p<0.01 or p<0.001). All of the free amino acids in jejunum, excluding taurine and ornithine, were significantly elevated in animals adapted to HPD, most probably due to the rapid absorption of large amount of amino acids across the epithelium of small intestine.

  • This study was attempted to observe the effect of Job's tear diet on change of body weight and tissue in rat. The changes of body weight were higher in Job's tear non-diet group(A) and seemed to lower in Job's tear diet group(c) as expected. The weight of each organ appeared to heavy more C group than A,Bgroup in Brain, Spleen. Lungs and Kidney. Liver was heavier in Job's tear to diet one day among six day group(B) and ovary was lighter in C group, The levels of each organ TBA-value were higher in Brain, Liver of A group and showed to higher Spleen, Lungs, Ovary of C group TBA-value of Kidney was revealed to lower more C group than A,B group.

Effects of Red Ginseng on Methyl Mercury Toxicities to Lipoprotein and Tissue Protein in Mouse (생쥐의 지단백질과 조직단백질에 미치는 메틸수은 독성에 대한 홍삼의 영향)

  • In order to investigate the effects of red ginseng extract to methyl mercury toxicities in mice, the serum lipoproteins, tissure protein patternsm and growth rates were studied. Animals wee divided into 3 groups of the control, group I treated with methyl mercury chloride only, and group II treated together with methyl mercury chloride and red ginseng extract. In serum lipoprotein fractions of group I, beta lipoprotein fraction was increased and pre-beta lipoprotein fraction was decreased in comparision to those of the control. However, there was almost no difference in quantities of serum lipoprotein fractions between the control and group II. Total pretein contents of groups I and II were increased in liver and those of groups I and II in the kidney were decreased. However, in comparison to group I, total protein contents of group II in the liver and kidney were similar values with those of the control. Percentage of tissue protein fractions between control and group I in the liver and kindey showed considerable difference. On the other hand, the percentage of protein fractions of group II approximated to that of the control. Daily average growth rate of body weight in group II was similar to the control, but that of group I was decreased significantly in comparison to the other 2 groups.

An investigation on the infection of Encephalitozoon cuniculi and pathological changes in laboratory rabbits (실험용 토끼에서 Encephalitozoon cunuculi의 감염과 병리학적 변화)

  • Eighteen NZW rabbits used for local skin irritation study were examined grossly and microscopically for natural infection with Encephalitozoon cuniculi. For microscopic tissue evaluation histochemical techniques such as PAS Gram iron hematoxylin and HE stain were used. Although rabbits in the study had no abnormal clinical signs 7/18(38.9%) animals were microscopically infected with E. cuniculi. The affected rabbits had gray-whitish and depressed approximately 0.1∼0.6 cm diameter lesions in the kidneys. All other organs examined were grossly unremarkable. Histopathologically however in addition to segmental interstitial nephritis focal lymphocytic myocarditis and granulomatous inflammatory reaction in portal areas of the liver multifocal granulomatous foci with vasculitis were present in the brain kidney and lungs. Aggregates of minute oval organisms were observed in brain and kidney sections frequently within the granulomatous foci and sometimes without any inflammatory reaction particularly in the renal tubules. in histochemical stains the organisms were gram positive stained with iron hematoxylin and had PAS positive granule at one pole. They measured approximately 1.5×2.5μm consistent with E. cuniculi. Histochemical characterization is important to differentiate E. cuniculi from other common protozoal infection such as Toxoplasma gondii. This study demonstrate the importance of subclinical. E. cunuculi infection and the associated histological alterations may interfere with tissue evaluation in toxicologic studies.

  • This study was examined chromosome structure of kidney, fin, blood cell, sexual organ that were differently examined chromosome of a good cell activity from organ, medium, staining methods, instruments with rainbow trout 100. And so we obtained following conclusion. 1. The difference from each organ and medium is that a good cell activity of fin, kidney, sexual organ were obtained in TC-199 medium and a good cell activity in TC-199 medium+PHA(5%). 2. In the difference by staining methods, G-banding and C-banding were analyzed by electron microscope or cytoscan. Among them, heterochromatin of centromere analyzed by C-banding. 3. The zygotene and the leptotene were analyzed in this study. 4. Karyotype, heterochromatin and each stage of cell division were clearly analyzed by cytoscan.

The Distribution of Estrogen Receptor in Various Organs of Rabbit (토끼 각장기의 Estrogen 수용체의 분포)

  • For clinical application of radioreceptor assay, we studied preliminarily the distribution of estrogen receptor in various organs of rabbit by a dextran-charcoal method using $6,7-^3H-estradiol$. The results were expressed as binding index, which is the ratio of specific estradiol receptor binding radioactivity to total radioactivity. The materials consist of 5 female rabbits and 3 male rabbits. The results were as follows: 1) Female rabbits The binding index was highest in the uterine tissue. This binding index of the uterine tissue was 9.4 times that of the liver, 21.9 times that of the kidney, 24.6 times that of the brain, 28.1 times that of the lung and 65.7 times that of the muscle. 2) Male rabbits The binding index was highest in the liver and decreased in the order of the kidney, the testis, the lung, the brain and the muscle. It is suggested that the estrogen receptor is not confined to any specific target organ but is widely distributed in the various organs, to a different degree.

Quantitative analysis of a myxosporean parasite, Parvicapsula sp. detected from emaciated olive flounder, Paralichthys olivaceus in Korea (국내 여윔 넙치에서 검출된 점액포자충 Parvicapsula sp.의 정량적 분석)

  • Quantitative analysis of a myxosporean parasite, Parvicapsula sp. in internal organs (kidney, intestine, spleen, brain and liver) from non-emaciated (farm-A) or emaciated (farm-B and farm-C) olive flounder Paralichthys olivaceus were performed by real-time PCR. The highest DNA copy number ($1.7{\times}10^7copies/mg$ tissue) was detected in kidney of the emaciated olive flounder from farm-C, while the DNA copy number was below detection limit in all the organs of the olive flounder from farm-B. There was not positive result in all of organs from olive flounder in farm-A. PCR and histopathological analysis were also performed using the same specimen and showed same results as those by real-time PCR.
