Inhibitory Phlorotannins from the Edible Brown Alga Ecklonia stolonifera on Total Reactive Oxygen Species (ROS) Generation

  • Reactive oxygen species (ROS) play an important role in the pathogenesis of many human degenerative diseases such as cancer, aging, arteriosclerosis, and rheumatism. Much attention has been focused on the development of safe and effective antioxidants. To discover sources of antioxidative activity in marine algae, extracts from 17 kinds of seaweed were screened for their inhibitory effect on total ROS generation in kidney homogenate using 2',7'-dichlorofluorescein diacetate (DCFH-DA). ROS inhibition was seen in three species: UIva pertusa, Symphyocladia latiuscula, and Ecklonia stolonifera. At a final concentration of 25 $\mu\textrm{g}$/mL, U. pertusa inhibited 85.65$\pm$20.28% of total ROS generation, S. latiscula caused 50.63$\pm$0.09% inhibitory, and the Ecklonia species was 44.30$\pm$7.33% inhibition. E. stolonifera OKAMURA (Lam-inariaceae), which belongs to the brown algae, has been further investigated because it is commonly used as a foodstuff in Korea. Five compounds, phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5), isolated from the ethyl acetate soluble fraction of the methanolic extrclct of E. stolonifera inhibited total ROS generation.

산화물 반도체를 이용한 최신 호기센서 기술 동향 (Recent Developments in Metal Oxide Gas Sensors for Breath Analysis)

  • Breath analysis is rapidly evolving as a non-invasive disease recognition and diagnosis method. Metal oxide gas sensors are one of the most ideal platforms for realizing portable, hand-held breath analysis devices in the near future. This paper reviewed the recent developments in metal oxide gas sensors detecting exhaled biomarker gases such as nitric oxides, acetone, ammonia, hydrogen sulfide, and hydrocarbons. Emphasis was placed on strategies to tailor sensing materials/films capable of highly selective and sensitive detection of biomarker gases with negligible cross-response to ethanol, the major interfering breath gas. Specific examples were given to highlight the validity of the strategies, which include optimization of sensing temperature, doping additives, utilizing acid-base interaction, loading catalysts, and controlling gas reforming reaction. In addition, we briefly discussed the design and optimization method of gas sensor arrays for implementing the simultaneous assessment of multiple diseases. Breath analysis using high-performance metal oxide gas sensors/arrays will open new roads for point-of-care diagnosis of diseases such as asthma, diabetes, kidney dysfunction, halitosis, and lung cancer.

  • Phaleria macrocarpa (Scheff.) Boerl. is a dense evergreen tree of the family Thymelaeceae. This plant is popular with the name of Mahkota dewa, which is literally translated as God's Crown. All parts of this plant including fruits, seeds, stem, and leaves have well known therapeutic properties and have been extensively used in traditional medicine for the treatment of various diseases such as cancer, diabetes mellitus, allergies, kidney disorders, blood diseases, stroke, and acne with satisfactory results. Scientific findings on bioactivities of P. macrocarpa also demonstrated different pharmacological properties of various parts of this plant including cytotoxic, antidiabetic, antioxidant, anti-inflammatory, antibacterial, and antihypertensive activities. Phytochemicals studies of P. macrocarpa revealed the presence of several classes of compounds such as benzophenones, terpenoids, xanthones, lignans, acids, and sugars. This review aims to provide a critical overview on botanical description, traditional usage, phytochemicals, and pharmacological activities of P. macrocarpa.

1,3-비스페닐치오 프로판을 배위자로 한 백금 (II)착체의 선택적 세포독성 (Selective Cytotoxicity of New Platinum (II) Complex Containing 1,3-Bis-phenylthiopropane)

  • A new series of highly water soluble platinum(II) complexes {Pt(II)[1,3-bis(phenylthio) propane](trans- -1,2-diaminocyclohexane) (PC-1) and Pt(II)[1,3-bis-(phenythio)propane] cis-1,2-diaminocyclohexane(PC-2)} were synthesized, and characterized by their elemental analysis and by various spectroscopic techniques[infrared(IR), 13C-nuclear magnetic resonance (NMR)]. In vitro antitumor activity of new Pt(II) complexes was tested against P-388 and L-1210 mouse lymphocytic leukemia cell lines, PC-14 / P, PC-14/ADM and PC-14 / CDDP human pulmonary adenocarcinima, DU-145 human prostate carcinoma, HT-1376 human bladder carcinoma, ZR-75-1 human breast carcinoma, MKN-45/P and MKN-45/CDDP human gastric adenocarcinoma cell lines using colorimetric MTT[3-(4,5-dimethyl thiazol-2-yl)-2.5-diphenyltetrazoliumbromide] assay for cell survival and proliferation. PC-1 showed active against L-1210, P-388 leukemia, human lung, stomach, prostate, bladder and breast cancer cell lines, and the antitumor activity of these compounds were comparable or superior to those of PC-2 and displatin. The nephrotoxicities of PC-1 and PC-2 were found quite less than that of cisplatin using MTT and [3H] thymidine uptake in rabbit proximal tubule cells and human kidney cortical cells. Based on these results, this novel platinum (II) complex compound (PC-1) represents a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low nephrotoxicity.

SH2D4A regulates cell proliferation via the ERα/PLC-γ/PKC pathway

  • SH2D4A, comprising a single SH2 domain, is a novel protein of the SH2 signaling protein family. We have previously demonstrated SH2D4A is expressed ubiquitously in various tissues and is located in the cytoplasm. In this study we investigated the function of SH2D4A in human embryonic kidney (HEK) 293 cells using interaction analysis, cell proliferation assays, and kinase activity detection. SH2D4A was found to directly bind to estrogen receptor $\alpha$ (ER$\alpha$), and prevent the recruitment of phospholipase C-$\gamma$ (PLC-$\gamma$) to ER$\alpha$. Moreover, we observed its inhibitory effects on estrogen-induced cell proliferation, involving the protein kinase C (PKC) signaling pathway. Together, these findings suggested that SH2D4A inhibited cell proliferation by suppression of the ER$\alpha$/PLC-$\gamma$/PKC signaling pathway. SH2D4A may be useful for the development of a new anti-cancer drug acting as an ER signaling modulator.

가공식품 나트륨 저감화 가이드라인 (A Guideline for Sodium Reduction of Processed Foods)

  • Sodium is a component of salt and naturally taken in the process of taking in table salt. For food processing, salt is very important. In general, salt adds flavor and taste including a salty taste and rheologically, it plays an important role in forming tissues. Also, it helps in improving preservability of food by controling growth of microorganisms. But excessive intake of salt has been blamed for outbreak of high blood pressure, heart disease, stroke, osteoporosis, kidney stone, stomach cancer and others. For this reason, there are active efforts to reduce sodium of processed foods all around the world. In Korea, a guideline for sodium reduction in 27 items and 44 kinds of foods including confectionery was suggested as part of the 'processed food sodium reduction guideline development project', which has been conducted since 2012.

Gastroprotective effect of cirsilineol against hydrochloric acid/ethanol-induced gastric ulcer in rats

  • This study was designed to evaluate the gastroprotective activity of cirsilineol in hydrochloric acid (HCl)/ethanol-induced gastric ulcer model. Cirsilineol was administered at the doses of 20 and 40 mg/kg in HCl/ethanol-induced rats. The gastroprotective ability was verified by determining the ulcer score, total acidity, hemoglobin, inflammatory cytokines, lipid peroxides, and enzymatic antioxidants superoxide dismutase (SOD) and catalase (CAT) in gastric tissue and serum biochemical analysis. The results showed a favorable increase in the hemoglobin level, antioxidant enzymes (SOD and CAT), restored electrochemical balance (carbon dioxide & anion gap) while a noticeable decrease in ulcer index, total acidity, lipid peroxides, inflammatory cytokines (interleukin-1 beta [IL-1β], IL-6, and tumor necrosis factor alpha) in rats treated with the cirsilineol. The serum biochemical analysis on liver markers (alkaline phosphatases, alanine aminotransferase, and aspartate aminotransferase), kidney markers (urea, creatinine, albumin, globulin, total protein), and lipid profile (triglyceride, high-density lipoprotein, total cholesterol) were attenuated by cirsilineol treatment in rats. Histopathology showed enhanced gastric protection and preserved the integrity of gastric mucosa upon cirsilineol administration. These results ultimately suggest that cirsilineol has gastroprotective effects that prevent the development of gastric ulcer.

위부분절제술 후 방사선치료에서 음식물 배출지연에 따른 영향 (Influence of Delayed Gastric Emptying in Radiotherapy after a Subtotal Gastrectomy)

  • 목 적: 위부분절제술 후 방사선치료를 받는 환자들 중에서 위 내 음식물의 배출지연으로 인한 위 용적 및 주변 장기의 위치 변화와 이것의 임상적 의의를 알아보고자 하였다. 대상 및 방법: 2005년 3월 1일부터 2008년 12월 31일까지 위암으로 위부분절제술을 받은 후 보조적 동시항암화학 방사선치료를 받았던 총 32명의 환자들 중, 전산화단층촬영모의치료 영상에서 12시간 이상의 금식에도 불구하고 위 내 음식물이 50 ml 이상 남아 잔존 위의 팽대를 보였던 경우들이 이번 연구 대상이었다. 이 환자들에 대해 방사선치료 기간 동안 주기적으로 시행된 컴퓨터단층촬영 자료를 바탕으로 잔존 위 용적 및 주변 장기들의 위치 변화를 평가하였다. 또한 방사선치료 설계 자료를 바탕으로, 잔존 위 및 주변 장기들의 변화를 고려하지 않고 치료를 지속했을 경우의 치료 표적 및 정상조직의 방사선분포 변화를 알아보았다. 결 과: 총 5명의 전산화단층촬영모의치료 영상에서 배출장애에 의한 50 ml 이상의 위 내 음식물의 저류가 확인되었다. 대상 환자들에서 위 용적은 치료과정 중에 다양한 변화양상을 보였고, 각 환자에서 최대 64.2~340.8 ml (평균, 188.2 ml)의 변화를 보였다. 또한 위 용적에 따라 좌측 신장이 각 환자별 최대 0.7~2.2 cm (평균, 1.2 cm)까지 상하 방향으로 이동하는 양상을 보였다. 모의치료 시의 설계를 위 용적의 변화에 따라 변경하지 않고 치료를 진행한 경우 유의하게 계획용표적체적(V43, 79.5${\pm}$10.4%)에 조사선량이 부족하고 좌측 신장(V20, 34.1${\pm}$12.1%; Mean dose, 23.5${\pm}$8.3 Gy)에 과도한 선량이 조사되는 결과를 얻을 수 있었다. 결 론: 위부분절제술 후 방사선치료를 받는 일부 환자에서 배출지연에 따른 위 용적의 변화와 좌측 신장의 위치변화를 확인할 수 있었다. 배출지연이 발생하는 환자를 선별하여 위 용적의 변화를 파악하고 이를 방사선치료 설계에 반영하는 것이 효과적일 것으로 생각된다.

갈색세포종 환자에서 Medical Internal Radiation Dose법을 이용한 I-131 Metaiodobenzylguanidine 치료 후 흡수선량 평가 (Radiation Absorbed Dose Measurement after I-131 Metaiodobenzylguanidine Treatment in a patient with Pheochromycytoma)

  • 목적: 근치적 절제술이 불가능하거나 전이를 동반한 악성 갈색세포종 환자의 I-131 MIBG 치료 시 흡수선량의 평가는 치료 효과 예측 및 치료용량 결정에 중요하다. 저자들은 갈색세포종 환자에서 I-131 MIBG 치료 후 감마카메라와 MIRD법을 이용하여 흡수선량을 평가하고자 본 연구를 시행하였다. 대상 및 방법: 종격동, 우측 신장 및 대동맥주위 림프절등에 전이가 확인된 악성 갈색세포종 환자에서 74 GBq I-131 MIBG 투여 후 양쪽 목옆에 37 MBq 와 74 MBq의 표준선원을 놓고 각각 0.5, 16, 24, 64, 145시간에 감마카메라로 전 후면영상 얻었다. 배후방사능 보정과 불응시간 보정 그리고 감쇠 보정후 MIRD법을 이용하여 표적조직의 흡수선량을 구하였다. 결과: 병소 각각에서 약 $32{\sim}63$ Gy/74 GBq의 흡수선량을 얻었으나 완전관해에 필요한 $150{\sim}200$ Gy에는 도달하지 못하였다. 치료 1달 후 X선 전산화 단층촬영상에서 병소의 감소를 확인하였고 치료 전과 비교한 I-123 MIBG 영상에서 MIBG 섭취감소를 확인하였다. 결론: 갈색세포종 환자에서 I-131 MIBG 치료 후 치료용량결정 및 치료계획을 수립하는 데 있어서 흡수선량평가가 필요하며 본 연구에서처럼 감마카메라와 MIRD법을 이용한 방법은 보다 간편하게 흡수선랑을 평가할 수 있어 임상적인 환경에서 유용할 것으로 여겨진다. 앞으로 본 연구에서 얻은 흡수선량의 정확성을 검증하고, 보다 많은 수의 환자에 적용하여 흡수선량과 치료 효과와의 관계를 규명하는 연구를 하여야 할 것이다.

산천어(Oncorhynchus masou) 에탄올 추출물의 in vitro 및 in vivo에서 항암활성 (Anticancer Activity on Ethanolic Extract of the Masou Salmon (Oncorhynchus masou) in vitro and in vivo)

  • 산천어 70% 에탄올 추출물(MSE)이 암세포 성장억제와 Balb/c 마우스에서의 종양 성장 억제에 미치는 영향을 알아보았다. 인간 자궁암(HeLa), 간암(HepG2), 유방암(MCF-7), 위암(AGS), 폐암(A549) 세포 그리고 인간신장 정상세포 (293)에 MSE을 처리했을 때 세포생존율이 감소하였고 이들 감소율을 MTT assay로 알아보았다. MSE는 정상세포 293에서보다 인간 암세포주인 MCF-7, A549, HepG2, AGS 그리고 HeLa세포에서 현저하게 더 높은 세포독성이 나타났다. 시료 최고농도인 1 mg/mL를 인간 폐암세포(A549), 유방암세포(MCF-7), 간암세포(HepG2) 및 위암세포(AGS)에 처리한 결과 각각 9.2%, 12.7%, 14.6% 및 16.9%의 세포 생존율을 나타내었다. 더하여 복수암 세포(sarcoma-180)를 이용한 실험동물(Balb/c mice)에서의 고형암 성장 억제 효과를 알아보았으며, 각각 25 mg/kg body weight와 250 mg/kg body weight의 MSE를 투여하였을 때 종양 성장 억제율은 각각 44.7%와 55.7%였다. 따라서 우리는 MSE가 암 예방을 위해 인간에게 유익한 기능성 소재일 것이라는 것을 제안하였다.
