항암단의 안전성(安全性)에 관(關)한 실험적(實驗的) 연구(硏究) (Study on the Toxicity of Hangam-dan(HAD) in Mice)

  • This study was carried out to investigate the oral toxicity of HAD in mice. The results were summarized as follows: 1. HAD did not induced any toxicological effect in the body weight changes of mice. 2. HAD(2.5g/kg, 0.25g/kg) treated group were increased as compared with control group in the hematological values of mice. 3. ALT, AST, BUN, creatinine, CPK were not increased in HAD(2.5g/kg, 0.25g/kg) treated group as compared with control group in the serological values of mice. 4. Brain, Heart, Liver, Spleen, Kidney's weight were not increased in HAD(2.5g/kg, 0.25g/kg) treated group as compared with control group in the organ's weight of mice. 5. HAD(2.5g/kg, 0.25g/kg) treated group did not induced any toxicological effect in the gross findings in mice's any organs. 6. HAD(10g/kg , 5g/kg, 2.5g/kg, 1.25g/kg) treated group did not induced any toxicological effect in the histopathological findings in mice's any organs. 7. HAD(2.5g/ kg, 0.25g/ kg) treated group did not induced any toxicological effect in the histopathological findings in mice's any organs. From above results, HAD has not toxicological effects to mice.

항암단(抗癌丹)을 투여(投與)한 폐암(肺癌) 환자(患者) 62례(例)에 대한 임상보고(臨床報告) (The Clinical study in 62 cases for lung cancer patients on the effects by Hangamdan(抗癌丹))

  • Clinical studies were carried out 62 cases of patients with lung cancer treated by Hangamdan(抗癌丹) from January 1th 1998 to September 30th 2000. The results were summarized as follows; 1. Distribution of those attached by lung cancer, by sex, showed that Male is more then Female, by age, showed that the number of sixties is majority. 2. Distribution of diagnostic stage, in descending order; stage IV(43.6%,top), stage III(35.5%), stage II(17.7%), stage I(3.2%). 3. The effects of maintenance and improvement in the symptoms with traditional oriental therapy(83.3%) and combined treatment of western and oriental therapy(84.0%) were observed. The effects of the symptoms were as follows; cough(50.0%), anorexia(48.4%), chest discomfort(31.0%), sputum(24.2%), general body weakness(11.3%), hemoptysis(9.7%) and etc. in orders. 4. Analysis of hematology attached by lung cancer, maintenance and increasing of WBC(98.4%), RBC(74.2%), Hgb(71.0%), Platelet(96.7%) were observed. After taken Hangamdan, the safety of the liver and kidney were as follows; maintenance and decreasing of AST(91.5%), ALT(93.2%), ${\gamma}-GTP$(95.0%), BUN(82.7%), Creatinine(93.3%) were observed. 5. Analysis of IL-12 and $IFN-{\gamma}$ attached by lung cancer, increasing of IL-12(31.3%), $IFN-{\gamma}$(72.7%) were observed. 6. Analysis of QOL attached by lung cancer, maintenance and improvement of combined treatment of western and oriental therapy(94.0%), traditional oriental therapy(91.7%) were observed. 7. Analysis of survival in patients with IV stage of lung cancer, above 7 months(22.2%), 12 months(70.4%). 8. Analysis of antitumor effects, maintenance of traditional oriental therapy(50.0%) and maintenance and improvement of combined treatment of western and oriental therapy(80.0%) were observed. 9. Analysis of curative valuation, maintenance and improvement of traditional oriental therapy(50.0%), combined treatment of western and oriental therapy(60.0%) were observed. From the above results, it is suggested that Hangamdan has significant effects of antitumor and immune activity, also could be usefully applied for lung cancer patients by combination with western therapy or alone.

Antimutagenic and Antileukemic Activities Aloe vera L.

  • We investigated that the extract of Aloe vera L. and its fractions exert antimutagenic activity against Salmonella typhimurium TA98 and TA100, and antileukemic effect against K562 human leukemia cell line. The aqueous ethanolic extract of A. vera L. was revealed to have antimutagenic effect on the AF-2 (2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide) in Salmonella mutation assay. Among the three fractions (fractions A, B and C) separated by silica gel chromatography, fraction C $(50\;{\mu}g/plate)$ exhibited the greatest antimutagenic effect on the AF-2 with inhibition rate of 84 and 90% in Salmonella typhimurium TA98 and TA100, respectively. The fraction C $(500\;{\mu}g/ml)$ inhibited the growth of K562 human leukemia cell line by 93% in MTT assay. However, the components of A. vera L. did not exhibit cytotoxic effect against MDBK bovine normal kidney in MTT assay.

Conditional Replication of a Recombinant Adenovirus Studied Using Neomycin as a Selective Marker

  • An E1B-defective adenovirus, named r2/Ad carrying the neo expression cassette, was constructed by homologous recombination. The construction, selection (using neomycin as a selective marker), and propagation of the recombinant virus was performed in human embryonic kidney 293 cells (HEK 293). An in vitro study demonstrated that this recombinant virus has the ability to replicate in and lyse some p53-deficient human tumor cells such as human glioma tumor cells (U251) and human bladder cells (EJ), but not in some cells with functional p53, such as human adenocarcinoma cells (A549) and human fibroblast cells (MRC-5). Also, based on the cytopathic effect (CPE), it was demonstrated, under identical conditions, that the U251 cells were more sensitive to r2/Ad replication than the EJ cells. In this paper, we report that r2/Ad could be very useful in studying the in vitro selective replication of E1B-defective adenovirus and has great potential in cancer gene therapy.

소음인 A형 인플루엔자 감염 치험례 (Treatment of Influenza A Virus Infection in a Soeumin Patient; a Single Case)

  • Objective This study aimed to report an improvement of influenza virus infection managed with Sasang Constitutional Medicine in a Soeumin patient with thrombocytopenia and histories of acute myeloid leukemia and interstitial pulmonary disease. Methods The medical records of patient including vital signs, laboratory test, simple x-ray, and symptoms were reviewed retrospectively. Patient was treated with Gunggwihyangso-san gamibang without any antiviral agents under the diagnosis of Congestive Hyperpsychotic symptomatology of Kidney Heat-based Exterior Heat disease. Results At admission, symptoms of shivering, fever, cough and myalgia were presented and her skin temperature was over $39^{\circ}C$. Influenza A virus was detected and inflammation biomarkers were elevated. After administration of herb medication, her vital signs and symptoms were improved and elevated inflammation biomarkers were decreased. There was no secondary complication during the treatment period. Conclusions This case treated with Gunggwihyangso-san gamibang showed a significant improvement from viral infection without any complication.

Application of Laparospic Ultrasonography in Surgery of Small Renal Cell Carcinoma

  • Purpose: : To assess the clinic value of application of laparospic ultrasonography (LU) in partial nephrectomy of small renal cell carcinoma. Materials and Methods: From 2007 to 2011, 28 small renal cell carcinoma patients in ou clinic underwent laparoscopic partial nephrectomy with LU. For comparison with preoperative conventional ultrasound and CT, we collected ultrasonic performance of the affected side kidney, renal tumor location, size, echo change, blood supply situation and the relationship with the surrounding tissue. Results: LU could more clearly show the tumor interior structure and blood supply, as well as the relationship with the surrounding tissue. It also can provided doctor assistance with real-time tumor resection, reducing operative complications. Conclusions: LU can clearly show tumor internal structure and blood supply, which is helpful for explicit diagnosis. Moreover, it supplies accurate information for surgeons and assists surgery. Therefore LU has an important guiding value in partial nephrectomy for small renal cell carcinoma.

Expressional Subpopulation of Cancers Determined by G64, a Co-regulated Module

  • Studies of cancer heterogeneity have received considerable attention recently, because the presence or absence of resistant sub-clones may determine whether or not certain therapeutic treatments are effective. Previously, we have reported G64, a co-regulated gene module composed of 64 different genes, can differentiate tumor intra- or inter-subpopulations in lung adenocarcinomas (LADCs). Here, we investigated whether the G64 module genes were also expressed distinctively in different subpopulations of other cancers. RNA sequencing-based transcriptome data derived from 22 cancers, except LADC, were downloaded from The Cancer Genome Atlas (TCGA). Interestingly, the 22 cancers also expressed the G64 genes in a correlated manner, as observed previously in an LADC study. Considering that gene expression levels were continuous among different tumor samples, tumor subpopulations were investigated using extreme expressional ranges of G64-i.e., tumor subpopulation with the lowest 15% of G64 expression, tumor subpopulation with the highest 15% of G64 expression, and tumor subpopulation with intermediate expression. In each of the 22 cancers, we examined whether patient survival was different among the three different subgroups and found that G64 could differentiate tumor subpopulations in six other cancers, including sarcoma, kidney, brain, liver, and esophageal cancers.

대장암 세포가 이식된 동물모델에서 사리장의 항암효과에 대한 연구 (Anti-cancer effect of Sarijang on colorectal cancer cells in a xenograft nude mouse model)

  • 본 연구는 사람유래 대장암세포가 이식된 동물모델을 사용하여 사리장의 항암효과를 확인하기 이하여 수행하였다. 사리장은 죽염과 서목태, 마늘과 유근피를 혼합 추출한 물질로써 아이소플라본을 포함하고 있으며 항염, 항산화 등의 효능이 보고된 물질이다. 이러한 사리장을 종양이 이식된 동물모델에 4주간 투여하면서 종양 크기의 변화, 종양 성장인자의 변화와 사리장의 안전성을 평가하였다. 그 결과 사리장은 종양의 크기를 감소시켰으며, 종양 성장과 관련된 ERK와 p16의 발현량을 조절함으로써 종양의 성장을 억제하는 효과를 나타냈다. 또한 사리장을 투여한 시험군에서는 체중, 장기무게, 혈청생화학적검사와 조직학적 검사에서 독성으로 인한 변화가 관찰되지 않았다. 이러한 결과는 사리장이 장기복용이 가능한 항암 보조식품의 소재로써 활용될 수 있다는 것을 의미한다고 할 수 있다.

전이성 폐암의 외과적 치료 (Surgical Treatment of Metastatic Lung Cancer)

  • In spite of recent progress in anticancer chemotherapy, the survival of patients with metastases to the lung treated nonsurgically has been extremely poor. So we adopted more aggressive surgical approaches for the treatment of patients with pulmonary metastases since 1985. We experienced 22 operations of metastatic lung cancer in 19 patients in the department of Thoracic & Cardiovascular Surgery in Kosin Medical College since 1985, so we reviewed the results of treatment retrospectively. The results were as follows: 1. The primary organs of metastatic lung cancer were 4 cases in each of the breast, uterus, and extremities, 3 cases in the rectum, 2 cases in the kidney, 1 case in each of the pelvis and liver, and the pathological findings were 13 cases in carcinoma and 6 cases in sarcoma. 2. The treatments for primary lesions were 15 cases of the operations with anticancer chemotherapy or radiation therapy, 2 cases of choriocarcinoma with anticancer chemotherapy only, 1 cases of uterine cervical carcinoma with chemo-radiation therapy, and 1 case of pelvic synovia sarcoma with intra-arterial anticancer chemotherapy. 3. Disease free intrerval were as follows: 7 cases were in 2 years to 4 years, 4 cases were in 1 year to 2 years, and 5 cases were beyond one year, of them one case was discovered primary lesion and metastatic lung tumor concomittently. 3 cases were above 4 years, of them one case of breast cancer were above 13 years especially. 4. The sites of metastatic lung cancer was 15 lesions in the right lung, and 9 lesions in the left lung, And the lobar sites were 10 lesions in the upper lobe, 2 lesions in the middle lobe, and 12 lesions in the lower lobe. 5. The operative methods of metastatic lung cancer were 7 case of partial resection of lung, 12 cases of pulmonary lobectomy, 1 case of pneumonectomy and 1 case of dissection of mediastinal lymph node. 6. The postoperative complications were 1 case of mild respiratory insufficency, 1 cases of pyothorax, and 1 case of urethral stricture. 7. Postoperative adjuvant therapy were as follows: No adjuvant therapy were 4 cases, anti-cancer chemotherapy were 8 cases, radiation therapy was 1 case, and combined with chemo k radiation therapy were 8 cases. 8. The results of long term follow-up were as follows: The 5 patients were died at 2 months, 22 months, 24 months, 32 months, and 49 months postoperatively, so mean survival period was 32 months postoperatively excluding one patient who was died at 2 months postoperatively. And 14 patients are aliving, of them 3 patients are living in recurred state, and the other 11 patients are living without any evidence of recurrence.

Somatic Mutations of the ENPP2 (Autotaxin/lysoPLD) Gene in Breast Cancer

  • ENPP2, a 125 kDa secreted lysophopholipase D which originally identified as a tumor-motogen, Autotaxin, enhances cellular locomotion, cell proliferation, angiogenesis and cell survival by generating the signal molecule lysophosphatic acid or sphingosine-1-phosphate. Previous studies have suggested that expression of Autotaxin is associated with invasive phenotype in advanced breast carcinomas. Thus, to determine whether genetic alterations of ENPP2 gene are involved in the development or progression of breast cancer, we analyzed its somatic mutation in 85 breast carcinomas by single-stranded conformational polymorphism and sequencing. Overall, six ENPP2 mutations were found (7.0%), comprising five missense and one nonsense mutation (s). To our knowledge, this is the first report on ENPP2 mutation in breast carcinoma, and the data indicate that ENPP2 is occasionally mutated in breast carcinomas, and suggest that ENPP2 mutation may contribute to the tumor development in some breast carcinomas.
