Eighty one products from 36 kinds of vitamin and mineral feed supplement collected during August, 1984 to February, 1985 were examined for microbiological contamination. In addition, 83 strains of coliform isolated from the samples were tested for the resistance to 8 kinds of antimicrobial drugs and distribution of R plasmid. General bacteria were detected in all of samples tested. Bacterial population was varied from less than 10 per gram of the sample to 1,400,000 per gram and 34 (42%) of 81 samples were contaminated with 100 to 1,000 cells per gram. Coliform isolation, which was more frequent in samples with larger number of general bacteria, was possible in 14 (17.3%) out of 81 samples tested and 6 (33.3%) out of 18 companies were coliform positive in their products. Forty one (49.4%) out of 83 coliform isolates were fecal coliform. The frequency of resistant strains was the highest to sulfadimethoxine (Sa) with 92.8% and followed by streptomycin (Sm, 67.5%), tetracycline (Tc, 50.6%), kanamycin (Km, 26.5%), chloramphenicol (Cm, 18.1%) and ampicillin (Am, 15.7%). No strain was resistant to nalidixic acid (Na) and gentamicin (Gm). The resistance frequency of fecal coliform strains were higher compare to non-fecal coliform strains. There were minimum inhibitory concentration (MIC) of $3,200{\mu}g/m{\ell}$ or higher in 7 strains to Am, 3 to Sm and 3 to Km, and 70 strains had MIC of $1,600{\mu}g/m{\ell}$ of higher to Sa while Tc had MICs from $1.6{\mu}g/m{\ell}$ to $400{\mu}g/m{\ell}$. All strains had MICs of $6.3{\mu}g/m{\ell}$ of lower to Na and $3.1{\mu}g/m{\ell}$ of lower to Gm. Seventy nine (95.2%) of 83 strains were resistant to one or more drugs tested. The most frequent resistance patterns were SaSm (14.5%) and followed by SaSmTc(12%), SaSmTcKm(8.4%) SaTc (8.4%) and SaSmKm (7.2%) ; total 19 different patterns were noted. Thirty two (40.5%) of 79 resistant strains were transferred all of a part of their resistance to Escherichia coli ML 1410. The frequency of transferable resistance was high in Am (100%) and Cm (80%) while low in Tc (38.1%), Sa (18.2%), Sm (17.9%) and Km (4.5%).

In this study, changes in the microbial ecosystem of the Yeongheungdo island coastal waters were investigated for five years to collect basic data. To evaluate the influence of distance from the coast on the microbial ecosystem, four sites, coastal Site (S1) and 0.75, 1.5, and 3 km away from the coast, were set up and the changes in physicochemical and biological factors were monitored. The results showed seasonal changes in water temperature, dissolved oxygen, salinity, and pH but with no significant differences between sites. For nutrients, the concentration of dissolved inorganic nitrogen increased from 6.4 μM in April-June to 16.4 μM in July-November, while that of phosphorus and silicon phosphate increased from 0.4 μM and 2.5 μM in April-June to 1.1 μM and 12.0 μM in July-November, respectively. Notably, phosphorus phosphate concentrations were lower in 2014-2015 (up to 0.2 μM) compared to 2016-2018 (up to 2.2 μM), indicating phosphorus limitation during this period. However, there were no differences in nutrients with distance from the coast, indicating that there was no effect of distance on nutrients. Phytoplankton (average 511 cells mL-1) showed relatively high biomass (up to 3,370 cells mL-1) in 2014-2015 when phosphorus phosphate was limited. Notably, at that time, the concentration of dissolved organic carbon was not high, with concentrations ranging from 1.1-2.3 mg L-1. However, no significant differences in biological factors were observed between the sites. Although this study revealed that there was no disturbance of the ecosystem, further research and more basic data on the microecosystem are necessary to understand the ecosystem of the Incheon.
In recent years, South Korea has often witnessed damages by gusts caused by thunderstorms in summer. The Korea Meteorological Administration defines that a gust happens when the maximum instantaneous wind velocity is 10m/s or more and draws up hourly observation reports. When a cumulonimbus develops due to an ascending current and reaches the height of 12~16 km, the temperature of the cloud top drops and a lightening happens, which causes a gust accompanied by a thunderstorm and further regional meteorological damage. It's difficult to predict a regional gust with the mesoscale prediction model at the administration. Thus this study set out to analyze the damage cases by a gust accompanied by a thunderstorm and to make a contribution to the prediction and understanding of a gust by a thunderstorm. A gust by a thunderstorm happens where potential equivalent temperature converges or is higher than the surrounding areas. The convergence area of potential equivalent temperature matches the track of thunderstorm cells. The Kimje gust took place where high potential equivalent temperature converged, and the Jangsu gust did as the area of high potential equivalent temperature approached. There should be a good amount of vapor supply with the moisture flux converging at the bottom layer in order to bring instability. In addition, it should collide into a dry and cold atmosphere at 700 hPa. The moving track at the center of the low dew point spread corresponds to that of a gust.
Brain, heart, liver, lung, kidney and thymus etc. 12 organs were removed and homogenized from Dawley-Sprague rats after suffocation. After fractionation of the tissue cytosols, enzymatic activities of the key enzymes in metabolic inositol phosphates cycle, PLC, IPSK and Ins(1,4,5) P35-phosphatase, were measured respectively. Hybridoma monoclones producing anti-lP3K murine monoclonal antibodies were obtained by the fusion of SP2/Ag 0-14 and spleen cells of mouse immunized with purified 53KDa IPSK, screening and cloning procedures. 18 cloned hybridoma cells were obtained, background due to nonspecific binding was very low with 10 clones. These Abs were purified from ascitic fluids by using affi-gel 15, and determined subtype of Abs. When immunoreactivities for rat tissues IP3K were exercised by adding the mixed Abs of 19Gl and 19G2b, they showed an overall similarity with noncompetitive inhibition. Brain tissue has high sensitivity for anti-lP3K Ab, whereas heart tissue has very low activity. In kinetic parameters Km value was 1.58 mM and Vmx value was 5.41umol/min/ml, respectively Only one form of 40 KDa IPSK was detected in heart tissues, however rat brain contains at least three immunologically distinct IP3K (53, 51 and 40 KDa) in western blot analysis. Of them 53 KDa protein was major enzyme in enzymatic activity. Northern blot analysis with 32P-labeled CDNA probe which encodes 1.8 Kb IPSK gene was performed. These results suggest that IPSK are regulated at transcriptional level during rat tissue development.
The xylnX gene encoding a xylanase from Clostridium thermocellum ATCC27405 was cloned in the plasmid pJH27, an E. coli-Bacillus shuttle vector and the resultant recombinant plasmid, pJX18 was transformed into E. coli HB101. The overexpressed xylanase was found to be secreted into the periplasmic space of the recombinant E. coli cells. The crude enzyme was obtained by treating the E. coli cells with lysozyme, and purified by DEAE-Sepharose column chromatography. Molecular wieght of the xylanase was estimated to be 53 kDa by gel filtration. The pI value was determined to be pH 8.8. The N-terminal sequence of the enzyme protein was Asp-Asp-Asn-Asn-Ala-Asn-Leu-Val-Ser-Asn which was considered to be the sequence of that of the mature form protein. The Km value of the enzyme for oat spelt xylan was calculated to be 2.63 mg/ml and the Vmax value was $0.47 {\mu}mole/min$. The xylanase had a pH optimum for its activity at pH 5.4 and a temperature optimum at $60^{\circ}C$. The enzyme hydrolyzed xylan into xylooligosaccharides which were composed mainly of xylobiose (40%) and xyloltriose (12%) after 5 hour reaction. This result indicates that the xylanase from C. thermocellum ATCC27405 is an endo-acting enzyme.

Increasing human activity due to rapid economic growth and land use change alters the patterns of the Asian monsoon, which is key to crop yields in Asia. In this study, we tested the performance of regional climate model (RegCM3) by simulating important components of Indian summer monsoon, including land-ocean contrast, low level jet (LLJ), Tibetan high and upper level Easterly Jet. Three contrasting rain years (1994: excess year, 2001: normal year, 2002: deficient year) were selected and RegCM3 was integrated at 60 km horizontal resolution from April 1 to October 1 each year. The simulated fields of circulations and precipitation were validated against the observation from the NCEP/NCAR reanalysis products and Global Precipitation Climatology Centre (GPCC), respectively. The important results of RegCM3 simulations are (a) LLJ was slightly stronger and split into two branches during excess rain year over the Arabian Sea while there was no splitting during normal and deficient rain years, (b) huge anticyclone with single cell was noted during excess rain year while weak and broken into two cells in deficient rain year, (c) the simulated spatial distribution of precipitation was comparable to the corresponding observed precipitation of GPCC over large parts of India, and (d) the sensitivity experiment using NIMBUS-7 SMMR snow data indicated that precipitation was reduced mainly over the northeast and south Peninsular India with the introduction of 0.1 m of snow over the Tibetan region in April.
This glucose isomerizing enzyme, which Actinomyces sp. (strain, K-17) produced, was inhibited by citrate, oxalate, ethylene diamine tetraacetic acid and cysteine on the enzyme reaction. This enzyme isomerized xylose to ketose as well as glucose. The Michaelis constant of this enzyme was $7.2X\;10^{-1}M.$. on D-glucose. The enzyme activity of intact cells which were harvested in the none xylose containing medium was very weak. If these intact cells of low activity were treated in the buffered xylose solution, its activity was considerably increased. After fifteen hours at $32^{\circ}C$. on xylose treatment, the enzyme activity was increased to equilibrium and the treating condition was proper at neutral pH and in aerobic state. The adequate concentration of xylose on the treatment was about 0.5 percent.
British Hydraulics research Station's Sparse Scheme was suggested for the solution of two dimensional finite difference tidal equations. 2D-numerical tidal model formed with 4.8km rectangular cells was set up to find out the barrage effects on tidal ranges of some possible areas for tidal power generation in Korea. The comparisons were made hetween spring tidal ranges with and without barrages at the entrance of Asan Bay, Garorim bay, Cheonsu bay and near Incheon Bay. Because of the coarseness of the grid, the model results could not represent the values of changes precisely. However the results show that the tidal amplitude near Incheon Bay increased by about 40cm by setting a barrage and about 50cm increase near the entrance of Asan Bay, respectively. But there were only a few centimeter changes in other areas.
Acetyl xylan esterase was produced by E. coli HB101 harboring a recombinant plasmid pKMG6 which contained the estI gene of Bacillus stearothermophilus. The maximum production was observed when the E. coli strain was grown at 37$\circC for 12 hours in the medium containing 0.5% acetyl xylan, 1.0% tryptons, 1.0% sodium chloride, and 0.5% yeast extract. The esterase produced was purified to homogeneity using a combination of ammonium sulfate fractionation, DEAE Sepharose CL-6B ion exchange chromatography and Sephacryl S-200 gel filtration. The native enzyme had an apparent molecular mass of 60 kd and was composed of two identical subunits of 29 kd. The N-terminal amino acid sequence of the polypeptide was Ala-X-Leu-Gln- Ile-Gln-Phe-X-X-Gln. The acetyl esterase displayed a pH optimum of 6.5 and a temperature opti- mum of 45$\circC. The heavy metal ions such as Ag$^{++}$, Hg$^{++}$ and Cu$^{++}$ inhibited nearly completely the activity of the esterase, and no specific metal ion was found to be required for the enzyme activity. The enzyme readily cleaved MAS, $\beta$-D-glucose pentaacetate, $\alpha$-naphthyl acetate, $\rho$-nitrophenyl acetate as well as acetyl xylan, but had no activity on $\rho$-nitrophenyl propionate, $\beta$-nitrophenyl butyrate or $\beta$-nitrophenyl valerate. The Km and Vmax values for MAS were 2.87 mM and 11.55 $\mu$mole/min, respectively. Synergistic behavior was demonstrated with a combination of xylanase and esterase from B. stearothermophilus in hydrolyzing acetyl xylan.

본 연구에서는 수치모델을 이용하여 대하천서 발생되는 조류의 공간적 농도 분포를 예측하였고, 현장실험을 통해 모델을 검증하였다. 국내하천은 다수의 지류가 본류로 유입됨에 따라 오염물질의 생산과 공급이 지속적으로 발생하고, 하천의 유로연장과 하폭에 비해 수심이 낮은 지형학적 특성을 지닌다. 따라서 지류 유입 이후 발생되는 조류의 거동 특성을 분석하기 위해 수심 적분된 2차원 이송-확산 모델을 사용하였다. 광합성 성장을 이루는 조류의 성장속도 계산을 위해 영양염류, 수온, 일사량과 수심 등을 변수로 하는 성장속도 함수들을 위의 모델과 결합하였다. 본 연구의 대상구간은 낙동강과 금호강 합류부를 포함한 강정고령보 하류 약 9.2 km 구간으로 모델 검증을 위한 현장실험을 수행하였다. 2차원 이송-확산 모델의 입력 값인 유속 및 수심을 계산하는 수리동역학 모델 검증을 위해 미국 Sontek사의 M9을 이용하여 낙동강과 금호강 각각 32개, 12개 측선에 대하여 수리량을 측정하였다. 수리량 측정결과, 금호강과 낙동강의 평균 유량은 각각 $240m^3/s$, $60m^3/s$로 측정되었고 측정된 유량을 모델의 상류단 경계조건으로 사용하여 측정 유속 및 수심과 유사한 결과를 모델로부터 취득할 수 있었다. 조류 농도 측정을 위해 독일 bbe사의 AlgaeTorch 10을 사용하였으며, 수리량 측정과 동일한 측선서 총 조류 세포수(cells/ml)를 측정하였다. 농도 측정결과, 하류로 내려감에 따라 조류의 농도가 증가하는 경향이 나타났고 금호강 합류 후 최대농도는 측정구간 최하류 우안서 4,460 cells/ml로 나타났다. 주 흐름이 발생하는 하천 중앙부에 비해 유속이 느린 하안서 상대적으로 높은 농도가 측정되었으며, 이와 같은 경향은 하류로 내려감에 따라 강하게 나타났다. 측정된 조류 농도를 이용한 2차원 이송-확산 모델 검증결과, 합류부 최상류 측선서 MAPE = 10.5 %의 최대오차가 발생하였고 최하류 측선서 MAPE = 6.7 %의 최소오차가 발생하였다. 인과 질소와 같은 영양염류의 농도가 높고 횡 방향 수온 분포가 균일한 대상구간의 특성상 영양염류 함수와 수온 함수로부터 계산된 성장속도 가중치 범위는 각각 0.8~1.0, 0.91~1.09로 공간적 변동성이 크게 나타나지 않은 반면, 수심을 변수로 하는 일사량 함수의 성장속도 가중치 범위는 0.05~1.00으로 상대적으로 매우 높은 공간적 변동성이 나타났다. 수심이 4 m 이하인 하천 양안서 0.8 이상의 가중치가 나타났으며, 수심이 7 m 이상인 하천 중앙서 0.4 이하의 가중치가 나타났다. 본 연구의 수치모의 결과, 수리동역학 모델로부터 계산된 수심이 모델 결과 값에 큰 영향을 미치는 것으로 판단된다.
