담액수경재배 시 재배후기 양액농도가 상추의 생장 및 수확후 품질에 미치는 영향 (Effect of Nutrient Solution Concentration in the Second Half of Growing Period on the Growth and Postharvest Quality of Leaf Lettuce (Lactuca sativa L.) in a Deep Flow Technique System)

  • 담액 수경재배에서 재배후반기 양액농도가 상추의 생장과 저장성에 미치는 영향을 구명하고자 4가지 양액농도(1/2S, 1S, 2S와 4S)를 '적치마'와 '청치마' 식물체에 처리한 후 생장과 수확후 특성을 조사하였다. 국화과 전용배양액($N-P-K-Ca-Mg=14.2-3.5-9-4-2me{\cdot}L^{-1}$)의 1배액으로 조성하여 3주간 재배한 후 2주간 양액 농도를 조절하여 재배한 결과 두 품종 모두 생체중 등의 생육량이 1배액이 좋았으며, 2배액과 1/2배액은 1배액보다 다소 떨어졌으며, 4배액에서는 생장 특성이 저하되는 것으로 나타났다. 색소 및 무기이온 함량에서는 4배액으로 처리한 상추가 높았다. 수확후 특성에서는 품종별로 2배액으로 처리한 상추가 저장기간 중 생체중 감소 및 SPAD 변화의 폭이 적었고, 외관과 같은 선도유지에 효과적인 것으로 나타났으며, 대체로 1배액, 1/2배액, 4배액 순으로 나타났다. 상추 수경재배 후기에 양액 농도교환에 따른 생육과 수확후 특성을 보면 재배 중에 최적의 생육량을 보인 양액 조성이 수확후 저장 및 품질유지 효과와 일관성을 보이지 않는 것으로 나타났다. 따라서 변화하는 농업 여건에 맞추어 수경재배에서 생산성 향상뿐만 아니라 향후에는 수확후 특성을 고려한 양액 조성과 재배 방법에 대한 개선이 필요한 것으로 보인다.

'Kowon', a New Korean Ginseng Cultivars with High Yield and Alternaria Blight Resistance

  • Recently, there has been increased attention to the development of new plant cultivars with enhanced resistance to biotic and abiotic stress. To develop ginseng cultivars with such traits, systematic breeding programs and comprehensive field studies are prerequisites. In this study, we applied a pure-line selection method to identify a ginseng cultivar with enhanced stress resistance. Phenotypic and agronomic characteristics, seed yield, and physiological responses to biotic and abiotic stresses were investigated according to the guidelines of the International Union for the Protection of New Varieties of Plants (UPOV). In the newly developed 'Kowon' cultivar, the time of emergence, flowering, and berry maturity were intermediate between those of the controls, 'Yunpoong' and 'Chunpoong'. The stem length of 'Kowon' was intermediate, whereas the root length was shorter and the main root diameter was greater than those of 'Chunpoong'. In local adaptability tests conducted in three regions, the yield of 'Kowon' was $666kg{\cdot}10a^{-1}$; 27% and 4% higher than that of 'Chunpoong' and 'Yunpoong'. Diseases such as Alternaria blight, Phytophthora blight, mulberry mealybug, and nematode infestation did not occur in 'Kowon'; and it also exhibited moderate resistance to damping-off and anthracnose. In these cases, yellow spots occurred on aerial parts and the rusty skin of the root, and it exhibited moderate resistance at high temperatures. Our study demonstrates that 'Kowon', which has a high root weight and enhanced biotic/abiotic stress resistance, is a superior cultivar that could increase farmers' income.

참지네고사리의 포자발아, 전엽체 및 포자체 번식에 영향을 미치는 배양 조건 (Culture Conditions Affecting Spore Germination, Prothallus Propagation and Sporophyte Formation of Dryopteris nipponensis Koidz.)

  • 본 연구는 참지네고사리의 기내 포자발아, 전엽체 증식 및 포자체 형성에 적합한 배지 및 배양토를 구명하고 균일묘를 생산하고자 수행되었다. 포자는 배지의 종류와 농도에 관계없이 모든 처리구에서 발아율이 우수하였다. 그 중 Knop배지에서는 MS 계열 배지에 비해 전엽체로의 발달이 빠르게 진행되었다. 포자를 발아시켜 획득한 전엽체를 재료로 배지의 구성물질을 달리하여 배양하였다. 전엽체의 증식율은 Knop배지에 비해 낮았으나, 장정기 등 생식기관의 형성이 활발한 1MS배지가 전엽체 증식용 배지로 적합한 것으로 생각되었다. 또한 MS배지의 sucrose 농도는 2%, 총 질소급원의 농도는 60mM, $NH_4{^+}$$NO_3{^-}$의 비율을 20:40mM로 조절하는 것이 전엽체의 생육 및 형태형성에 효과적이었다. 포자체 형성에 적합한 배양토를 선발하기 위하여 믹서기로 분쇄한 전엽체를 인공토양(원예상토, 피트모스, 펄라이트, 마사토)을 단용 또는 혼용한 토양에 분주하여 14주간 재배하였다. 그 결과, 원예상토를 사용한 처리구에서 포자체의 형성 및 생육이 우수하였으며, 특히 원예상토와 마사토를 2:1(v:v)로 혼용하였을 때 포자체의 형성(0.83개/$cm^2$)이 가장 왕성하였다.

LED가 고구마 바이러스 무병묘의 기내 생장에 미치는 영향 (Effects of Light-emitting Diodes on In Vitro Growth of Virus-free Sweet Potato Plantlets)

  • 고구마 바이러스 무병주의 기내 소식물체 생장에 미치는 LED의 영향을 조사하기 위하여, '맛나미', '신천미', '연황미'의 소식물체에서 10mm 크기의 마디를 잘라 $0.2mg{\cdot}L^{-1}$ BA를 첨가한 MS배지에 배양하였다. 광질은 형광등을 대조구로 하여, 적색(660nm), 청색(460nm), 백색, 적:청(8:2), 적:청(7:3) LED를 20cm 거리에서 $150{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 광합성유효광량자속(PPF)을 유지하였으며, 일장은 16/8(day/night)시간, 배양온도는 $25^{\circ}C$로 조절하였다. LED 광질에 대한 품종간 반응은 적색 LED에서 같은 방법으로 10품종의 마디배양을 하여 소식물체 생장을 조사하였다. 배양 3주 후에 줄기신장은 적색 LED에서 형광등보다 유의하게 증가하였으며, 청색광에서 가장 낮았다. 줄기두께는 적:청(7:3)과 백색 LED에서 증가하였다. 생체중 및 건물중도 적색광에서 가장 높았다. 또한 적색 LED에서 신초생육은 유의한 품종간 차이를 보였다. 줄기길이, 생체중 및 건물중 등은 '신천미', '맛나미', '신황미' 등에서 컸다. 따라서 LED 광원을 이용한 고구마 마디배양은 적색 LED에서 3주간 배양하여 5cm 이상 키운 소식물체를 토양이식하는 것이 대조구인 형광등보다 배양기간을 1주 이상 단축할 수 있었다.

Determination of the Optimal Nitrogen Concentration in Pre-planting Fertilizers for the Cultivation of Tomato Plug Seedlings

  • This study investigated the effect of pre-planting nitrogen (N) fertilization levels added to a soilless root medium on the growth of 'Dotaerang Dia' tomato seedlings. The N levels were varied for a total of 7 treatments: 0, 100, 250, 500, 750, 1,000, or $1,500mg{\cdot}L^{-1}$. The pH of the root media gradually rose in all treatments as the seedlings grew; however, the differences in the pH were not significant among the treatments. The electrical conductivity (EC) of the root media was significantly different among the treatments from sowing to week three, then drastically decreased after week four, which diminished the differences in the EC among the treatments. At week six, plant height, leaf length, leaf width, number of leaves, and fresh and dry weights of the shoot were highest for the treatment with $500mg{\cdot}L^{-1}N$. In contrast, the treatment with $1,500mg{\cdot}L^{-1}N$ had the lowest results for all growth measurements. The fresh weight was 67% heavier in the $500mg{\cdot}L^{-1}N$ treatment compared to the $1,500mg{\cdot}L^{-1}N$ treatment. The total N content in the tissues was lowest in the treatment with $0mg{\cdot}L^{-1}N$ and highest in the treatment with $1,000mg{\cdot}L^{-1}N$. The contents of calcium (Ca), magnesium (Mg), and metal micronutrients in the tissues were highest in the $250mg{\cdot}L^{-1}N$ treatment. A previous study demonstrated that adjusting the fertilization level to promote growth to over 90% of the maximum growth is a good strategy for lowering production costs and preventing damage due to excessive fertilizer absorption by crops. Our results indicated that the optimal pre-planting N fertilization level for tomato plug seedlings should be lower than $500mg{\cdot}L^{-1}$ and the optimum tissue N contents should be around 3.21% to 4.60%.

The Construction of a Chinese Cabbage Marker-assisted Backcrossing System Using High-throughput Genotyping Technology

  • The goal of marker-assisted backcrossing (MAB) is to significantly reduce the number of breeding generations required by using genome-based molecular markers to select for a particular trait; however, MAB systems have only been developed for a few vegetable crops to date. Among the types of molecular markers, SNPs (single-nucleotide polymorphisms) are primarily used in the analysis of genetic diversity due to their abundance throughout most genomes. To develop a MAB system in Chinese cabbage, a high-throughput (HT) marker system was used, based on a previously developed set of 468 SNP probes (BraMAB1, Brassica Marker Assisted Backcrossing SNP 1). We selected a broad-spectrum TuMV (Turnip mosaic virus) resistance (trs) Chinese cabbage line (SB22) as a donor plant, constructing a $BC_1F_1$ population by crossing it with the TuMV-susceptible 12mo-682-1 elite line. Foreground selection was performed using the previously developed trsSCAR marker. Background selection was performed using 119 SNP markers that showed clear polymorphism between donor and recipient plants. The background genome recovery rate (% recurrent parent genome recovery; RPG) was good, with three of 75 $BC_1F_1$ plants showing a high RPG rate of over 80%. The background genotyping result and the phenotypic similarity between the recurrent parent and $BC_1F_1$ showed a correlation. The plant with the highest RPG recovery rate was backcrossed to construct the $BC_2F_1$ population. Foreground selection and background selection were performed using 169 $BC_2F_1$ plants. This study shows that, using MAB, we can recover over 90% of the background genome in only two generations, highlighting the MAB system using HT markers as a highly efficient Brassica rapa backcross breeding system. This is the first report of the application of a SNP marker set to the background selection of Chinese cabbage using HT SNP genotyping technology.

고당도 사계성 딸기 '장하' 육성 (Characteristics of New Ever-bearing Strawberry 'Jangha' Bred for High Soluble Solids Contents)

  • '장하'품종은 농촌진흥청 국립식량과학원 고령지농업연구소에서 2014년도 육성한 사계성 딸기 신품종이다. '장하'는 '고하'품종을 모본으로 하고 '엘시뇨' 품종을 부본으로 2008년 교배하여, 고온장일조건에서 당도가 높고 화방이 연속적으로 출현되는 우수한 사계성 개체를 선발한 것이다. 고랭지의 여름재배 작형에서 2011년 생산력 검정, 2012년 특성검정을 거쳐 '새봉 6호'로 계통명을 부여하고, 2013-2014년에는 2지역 적응성시험을 거쳐 '장하'로 명명하였다. '장하'의 초형은 반개장형이며, 엽형은 타원형이며, 초세가 중간이다. 과실모양은 원추형이며, 과색은 붉은 색이다. 엽수는 21.4매로 '플라멩고'의 55.8매 보다 34.4매 적다. '장하'의 당도는 8.9%로 '플라멩고'의 7.7%보다 1.2% 더 높다. '장하'의 평균과중은 11.7g으로 사계성 품종 중에서 중간크기에 속하는 편이고, 상품수량은 $19,013kg{\cdot}ha^{-1}$으로 '플라멩고' 품종보다 141% 더 많았다. '장하'는 식미가 좋아 생식용으로 적당한 사계성딸기 품종이다.

Growth, Flowering, and Nutrient Composition of Salvia Grown in Peat moss Media Containing Pellets Processed with Poultry Feather Fibers at Different Mixing Ratios

  • The objective of this study was to determine the effect of replacing perlite (PL) with pellets processed with poultry feather fiber as an inert material to prepare growing medium. The growth and flowering of Salvia splendens 'Vista Red' grown in individual growing medium $Biosangto^{(R)}$, peat moss (PM), PL, and two pellets (P45-1 and P45-2) were evaluated. Peat moss was mixed with PL, P45-1, or P45-2 at various ratios (1:0 to 1:3 or 3:1 by volume) to investigate the feasibility of replacing PL with pellets. Nutrient composition of the growing medium and leaf tissues was analyzed. The number of florets, inflorescence length, plant height, and fresh weight of plants grown in media containing P45-1 or P45-2 were reduced compared to those grown in individual growing medium PM or PL. As the mixing ratio of P45-1 or P45-2 to PM was higher, the growth of salvia, such as inflorescence length, plant height, number of leaves, and fresh weight was inhibited. Our results indicate that mixing three parts PM with one part of P45-1 (PM/P45-1/3:1) or P45-2 (PM/P45-2/3:1) accelerated flowering and increased the number of florets and leaves compared to other mixing ratios of PM and pellets media. The concentrations of phosphorus (P), calcium (Ca), boron (B), iron (Fe), and copper (Cu) in individual growing medium PL, P45-1, and P45-2 were significantly lower than those in PM. The concentration of N was the highest in leaves of plants grown in P45-1 or P45-2 amended media, and the concentrations of P, Ca, and zinc (Zn) in leaves were lower in individual growing medium P45-1 or P45-2 than in PM and PL. The pH of PM/P45-1/3:1 or PM/P45-2/3:1 media was maintained at optimal level (5.8-5.9) and the concentrations of macro- and micro-elements in the media and leaves were considered to be optimal levels. Therefore, mixing three parts PM with one part P45-1(PM:P45-1/3:1) or P45-2 (PM:P45-2/3:1) is recommended for improved growth and flowering in salvia. This suggests that P45-1 or P45-2 can replace PL as an inert material to prepare growing medium.

Different Oxygen Transmission Rate Packing Films During Modified Atmosphere Storage: Effects on Asparagus Spear Quality

  • The aim of this study was to determine suitable oxygen transmission rate (OTR) films for modified atmosphere (MA) storage of asparagus spears (Asparagus officinalis L.), and to examine the related changes in asparagus spear quality. Asparagus spears were packed with 10,000, 20,000, 40,000, 70,000, and $100,000cc/m^2{\cdot}day{\cdot}atm$ OTR film treatments for MA storage, and perforated film (conventional storage) was used as the control. The fresh weight loss rate was less than 1% in all OTR film treatments but not in the control. In all OTR film treatments, ethylene content rapidly increased on the first day of storage, and gradually decreased thereafter to $2.0{\mu}L{\cdot}L^{-1}$ by the final storage day. In the 10,000 cc and 20,000 cc OTR film treatments, carbon dioxide content was maintained within the permissible range for asparagus spears under recommended controlled atmosphere (CA) and MA conditions (5-12%). The oxygen content was maintained between 12% and 20% in all OTR film treatments. Hue angles and total chlorophyll content were highest in the 10,000 cc OTR film treatment in both tips and stems. Visual quality was maintained at a saleable level only in the 10,000 cc OTR film treatment until the final storage day (25 days). Based on sensory evaluation, the least off-odor was detected in the 10,000 cc OTR film treatment, and the most in the control treatment. While the soluble solids content decreased with all film treatments, it was maintained at the highest level in the 10,000 cc OTR film treatment. Vitamin C content decreased with all film treatments during storage, but was maintained at higher levels in the 10,000, 20,000, and 40,000 cc OTR film treatments. The firmness of tips and stems increased with storage duration in all film treatments, stems were firmer under the perforated film, 10,000, and 20,000 cc OTR film treatments. Given these results, the 10,000 cc OTR film treatment is considered the most appropriate for MA asparagus spear storage since visual quality and soluble solids content were maintained, and fresh weight loss, vitamin C content, off-odor, and yellowing were suppressed during storage.

Characterization of the Lsi1 Homologs in Cucurbita moschata and C. ficifolia for Breeding of Stock Cultivars Used for Bloomless Cucumber Production

  • Bloomless cucumber fruits are commercially produced by grafting onto the pumpkin stocks (Cucurbita moschata) to restricted silicon ($SiO_2$) absorption. Inhibition of silicon absorption in bloomless stocks is conferred by a mutant allele of the CmLsi1 homologous to Lsi1 in rice. In this study, we characterized the Lsi1 homologs in pumpkin (C. moschata) and its cold-tolerant wild relative C. ficifolia ('Heukjong') in order to develop a DNA marker for selecting a bloomless trait and to establish the molecular basis for breeding bloomless stock cultivars of C. ficifolia. A Cleaved amplified polymorphic sequence (CAPS) marker (CM1-CAPS) was designed based on a non-sysnonymous single nucleotide polymorphism (SNP, C>T) of the CmLsi1 mutant-type allele, and its applicability for Marker-assisted selection (MAS) was confirmed by evaluating three bloom and five bloomless pumpkin stock cultivars. Quantitative RT-PCR of the CmLsi1 for these stock cultivers implied that expression level of the CmLsi1 gene does not appear to be associated with the bloom/bloomless trait and may differ depending on plant species and tissues. A full length cDNA of the Lsi1 homolog [named CfLsi1($B^+$)] of 'Heukjong' (C. ficifolia), was cloned and sequence comparison between CmLsi1($B^+$) and CfLsi1($B^+$) revealed that there exists total 24 SNPs, of which three were non-synonymous. Phylogenetic analysis of CfLsi1($B^+$) and Lsi1 homologs further revealed that CfLsi1($B^+$) is closesly related to Nodulin 26-like intrinsic proteins (NIPs) and most similar to CpNIP1 of C. pepo than C. moschata.
