Objective: The objective of this experiment was to investigate the effect of dietary glucose oxidase (GOD), catalase (CAT), or both supplementation on reproductive performance, oxidative stress, and apoptosis in sows. Methods: A total of 104 multiparous sows were randomly assigned to four groups (n = 26) with each group given a basal diet, basal diet plus GOD at 60 U/kg, basal diet plus CAT at 75 U/kg, and basal diet plus GOD at 60 U/kg and CAT at 75 U/kg. Sows were fed the experimental diets throughout gestation and lactation. Results: Dietary GOD supplementation increased average daily feed intake of sows and litter weight at weaning (p<0.05). Dietary CAT supplementation reduced the duration of parturition, stillbirth, and piglet mortality and increased growth performance of weaned piglets (p<0.05). Dietary GOD and CAT supplementation enhanced antioxidant enzyme activities and lessened oxidative stress product levels in plasma of sows and elevated antioxidant capacity of 14-day milk and plasma in weaned piglets (p<0.05). Dietary GOD supplementation increased fecal Lactobacillus counts and reduced Escherichia coli counts of sows (p<0.05). Compared with the basal diet, the GOD diet reduced fecal Escherichia coli counts of sows, but the addition of CAT did not reduce Escherichia coli counts in the GOD diet. Dietary GOD and CAT supplementation reduced the apoptosis rate of the liver, endometrium, and ovarian granulosa cells in sows (p<0.05). In the liver, uterus, and ovary of sows, the mRNA expression of caspase-3 and caspase-9 was downregulated by dietary GOD and CAT supplementation (p<0.05). Conclusion: Dietary GOD and CAT supplementation could improve the antioxidant capacity of sows and weaned piglets, and alleviate hepatic, ovarian and uterine apoptosis by weakening apoptosis-related gene expression. Glucose oxidase regulated fecal microflora of sows, but supplementation of CAT to GOD could weaken the inhibitory effect of GOD on fecal Escherichia coli.
Objective: The objective of the study was to determine the influence of varying fibrous diets on fecal characteristics of growing pigs. Methods: A total of 104 pigs (initial weight $18{\pm}2.0kg$) were used in the study. They were housed in individual pens and fed on diets containing maize cob, grass hay, lucerne hay, maize stover, and sunflower husk. These fibers were included at 0, 80, 160, 240, 320 and 400 g/kg. Fecal and urine samples were collected. Results: Fecal output was largest amongst pigs fed on diets containing grass hay and maize stover (p<0.05). Nitrogen content was highest in feces from pigs fed on sunflower husk (p<0.05). Pigs fed on diets containing maize stover and maize cobs produced the largest concentrations of short chain fatty acids. Acetate concentration was high in feces of pigs fed maize stover than those fed grass hay and lucerne hay (p<0.05). As the level of fiber inclusion increased, fecal consistency and nitrogen content increased linearly (p<0.05). Urea nitrogen decreased as the inclusion level increased across all the fibers (p<0.05), with maize cobs containing the largest content of urea nitrogen. As dietary fiber content increased, fecal nitrogen content also increased (p<0.05). Conclusion: It was concluded that different fiber sources influence fecal characteristics, thereby having different implications on pig waste management. It is vital to monitor fiber inclusion thresholds so as to easily manage environmental pollutants such as butyrate that contribute to odors.
Purpose: The purpose of this study was to examine body composition and dietary self-efficacy in nursing students. Methods: The study was a descriptive, correlational study with a convenience sample of 197 female nursing students. This study was conducted from April 1st to May 31th in 2015. SPSS 21.0 program was used for data analysis. Results: The average of body protein was $7.90{\pm}1.17kg$ and normal range was 33%. A BMI's average was $21.02{\pm}2.94kg/m^2$ and 62.9 fell into standard range. The average of dietary self-efficacy of subjects was $3.42{\pm}0.53$. The subjects performed intense physical activities during three or more days had higher body protein (F=5.89, p=.003), body minerals (F=6.15, p=.003) and body-efficacy (F=4.13, p=.017) but remarkably lower body fat (F=4.04, p=.019). There is no noticeable correlation of body composition with dietary self-efficacy. However, it is obvious that each category of body composition had a significant positive correlation. Conclusion: This study demonstrated that students who were inclined to overeat had higher body protein, body minerals, body fat, and BMI. Students performed intense physical activities had high body protein, body minerals, and dietary self-efficacy, whereas they had low body fat. Each entry between the body composition had a significant positive correlation.
Ginseng (Panax ginseng Meyer) is a well-known health functional food used as a traditional herbal drug in Asian countries owing to its diverse pharmacological effects. Herb-drug interactions may cause unexpected side effects of co-administered drugs by the alteration of pharmacokinetics through effects on cytochrome P450 activity. In this study, we investigated the herb-drug interactions between Korean red ginseng extract (KRG) and five CYP-specific probes in mice. The pharmacokinetics of KRG extract induced-drug interactions were studied by cassette dosing of five CYP substrates for CYP1A, 2B, 2C, 2D, and 3A and the LC-MS/MS analysis of the blood concentration of metabolites of each of the five probes. The linearity, precision, and accuracy of the quantification method of the five metabolites were successfully confirmed. The plasma concentrations of five metabolites after co-administration of different doses of the KRG extract (0, 0.5, 1, and 2 g/kg) were quantified by LC-MS/MS and dose-dependent pharmacokinetic parameters were determined. The pharmacokinetic parameters of the five metabolites were not significantly altered by the dose of the KRG extract. In conclusion, the single co-administration of KRG extract up to 2 g/kg in vivo did not cause any significant herb-drug interactions linked to the modulation of CYP activity.
In this study, nutrient intake status and energy expenditure were examined to investigate the nutritional status of the elderly in a rural community. The results obtained by questionaries, the 24 hour recall method, and time-diary were as follows: The elderly men surveyed were 73.8 years old, on the average. The elderly women surveyed were 73.5 years old, on the average. The proportion of the elderly with diseases was 51.9%. Most of the subjects (86.1%) had a regular meal pattern of consuming three meals a day. The average daily energy intake of the rural elderly was much lower than the Korean RDA. The dietary assessment data showed that each energy intake of the males and the females was 79.5% and 84.3% of the RDA, respectively. The dietary intake of Ca, Fe, niacin, thiamin, and riboflavin was lower than the Korean RDA, and that of P and Vitamin C was adequate. The Fe intake was significantly different with respect to age and sex (p < 0.05). Although, in both elderly men and elderly women it decreased with age, the elderly men's intake was lower than the elderly women's. The heights of the elderly men and the elderly women was 159.7 cm and 147.5 cm, respectively, and the weights were 60.0 kg and 52.2 kg, respectively, and the BMI was in the moderate range. Heights significantly decreased with age (p < 0.05). According to daily living schedules, leisure time (11.0 hour) was the longest, physiological time (9.6 hours) was next, and work time (3.4 hours) was the shortest. Energy expenditure significantly decreased with age (p < 0.01). Energy intake also decreased with aging. Energy balance (energy expenditure/energy intake) was 93.4% in elderly men and 104.0% in elderly women. Especially, in elderly men in the 65 to 74 age range, the energy balance was the lowest, and the nutrient intake was also much lower than that of elderly women.
The variation of rumination response in steers fed a same amount of orcharograss hay was investigated. With three steers (6, 12, 24 months) of the Japanese Black Breed, rumination behaviour was measured continuously during a 5 days period by masseter EMG telemetry system. The results are summarized as follows: 1. The average daily ruminatio time of respective animals were 438,447 and 433 min. with small daily variation both within and among animal. But the rumination time per DM kg of hay consumed varied from 66 to 138 min., and calf spent longer than older calf. 2. Actural chewing time showed small variation, and percentage of actural chewing time spent in rumination time was from 88 to 93% on individual average. 3. The daily number of chewing in rumination was approximately 25,000 to 30,000 with large individual differences. The chews per DM kg intake varied from 3,800 to 9,600, and calf chewed more than older calf. 4. The average rate of chewing per 100 sec. (chewing speed), there was a large difference between animals I.e. 104, 114, 131 respectively, but very little variance between days in individual. 5. No relation between day to day variations in eating time and rumination behavior, but significantly positive correlation was observed in the relation among rumination time, chewing time, No. of boluses and No. of chews. 6. Active time of day in rumination altered considerably day to day, but variation in the amount of rumination per day as expressed by the rumination time was relatively small.
This study was conducted to investigate the hygienic quality of commercial Kwamegi, a Korean traditional food, made from semi-dried Pacific saury. The hygienic qualities of 10 Kwamegi samples were examined from the 15 commercially available types. Samples with code 3, 4, 6, 11 and 12 were acceptable based on criteria suggested by Ministry for Food, Agriculture, Forestry and Fisheries (i.e. moisture below 50%, total mercury below 0.5 ppm, lead below 2.0 ppm, viable cells below $1.0{\times}10^5$ CFU/g and coliform count below $1.0{\times}10^3$ MPN/100g. Peroxide value of commercial Kwamegi ranged from 15.3-104.1 meq/kg. Agmatine sulfate, cadaverine, histamine, tyramine, spermidine, dopamine and spermine were found in all of the 15 commercially available Kwamegi, while tryptamine and 2-phenylethylamine were not detected. Putrescine dihydrochloride was only detected in 7 samples. The histamine content of commercial Kwamegi ranged from 33-124mg/kg, which is believed to be an acceptable limit according to guidelines from other countries. Cadaverine can be used as a potential index for freshness of commercial Kwamegi as there is a good correlation between volatile basic nitrogen and biogenic amine contents.
The mouse bioassay has been used widely for the monitoring of paralytic shellfish toxins (PSTs) in many countries. However, this method shows low sensitivity and high limit of detection (LOD), as well as it cannot confirm toxic profiles. Recently, LC-MS/MS method was studied for the quantitative of PSTs, however, the method has any problems with unstable retention times by ionization suppression caused by high salt concentration in shellfish extracts. To establish an alternative method for PSTs analysis, we tried to original LC-MS/MS methods adding desalting operation using amorphous graphitized polymer carbon solid-phase extraction cartridges. The method validation was conducted to determine linearity, limit of detection, limit of quantification (LOQ), accuracy, and precision in quantifying PSTs. The correlation coefficients for all tested PSTs maintained over 0.999. The LODs and LOQs for all PSTs were about 0.19-1.05 ㎍/kg and 0.58-3.18 ㎍/kg, respectively. The accuracies for PSTs were 95.4-107.7% for saxitoxin group, 97.1-100.9% for gonyautoxin group, 99.0-100.8% for N-sulfocarbamoyl toxin group, and 96.8-104.6% for decarbamoyl toxin group. These results indicate that the modified LC-MS/MS method was appropriate for analyzing the PSTs in shellfish and tunicates.
본 연구는 혹명나방 저항성 유전자 변형 낙동벼(GMO)의 식품으로서의 안전성을 평가하고자 13주 경구 반복투여 독성시험을 수행하였다. SD 랫드를 0, 5, 20%의 유전자 변형 낙동벼와 20% 낙동벼(Non-GMO)를 투여군으로 분리하여 13주간 투여한 후 체중, 사료섭취량, 요, 혈액 및 혈액생화학, 부검 및 조직병리학적 검사를 수행하였다. 시험종료시까지 모든 동물은 생존하였으며, GMO 벼의 투여와 관련된 독성은 관찰되지 않았다. 따라서 혹명나방 저항성 유전자 변형 낙동벼의 무독성량은 10,000 mg/kg/day 이상일 것으로 판단되며, 13주 경구 반복투여 독성은 없는 것으로 평가되었다.
Sunjeonhwadok-Tang was a drug that treated carbuncle and cellulitis. So, the purpose of this study was to investigate effects of Sunjeonhwadok-Tang on the proliferation of cancer calls and immunocytes focusing around combined effects of anticarcinogen. We used Sunjeonhwadok-Tang extract(SHT) with freeze-dried, 8wks-old male balb/c mice and cancer cell lines(L1210, Sarcoma-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results : 1. SHT was significantly showed cytotoxicity on the L1210 cell lines. 2. SHT was significantly increased proliferation of thymocytes and splenocytes in vitro. 3. In combined effects of SHT and vincristine(0.005 mg/kg), SHT was significantly inhibited proliferation of L1210 cell lines, but was not inhibited proliferation of Sarcoma-180 cell lines compared with positive control group. 4. In combined effects of SHT and vincristine(0.005 mg/kg), SHT was significantly increased proliferation of thymocytes and splenocytes compared with positive control group. 5. In combined effects of SHT and vincristine, SHT was significantly increased proliferation of thymocytes and splenocytes in normal mice. 6. In combined effects of SHT and vincristine, SHT was significantly inhibited proliferation of L1210 cells in L1210 cells transplanted mice 7. In combined effects of SHT and vincristine, SHT was significantly increased proliferation of L1210 cell in L1210 cells transplanted mice. The present author thought that SHT had action of anti-cancer and immuno-activity, and in combined effects of vincristine, SHT had recoverable effects on damage by anticarcinogen.
