Effects of Saenghyetang on Learning and Memory Performances in Mice (생혜탕(生慧湯)이 흰쥐의 학습(學習)과 기억(記憶)에 미치는 영향(影響))

  • The effects of the oriental herbal medicine Saenghyetang(SHT, 生慧湯), which consists of Rehmanniae Radix (熟地黃 九蒸: was made by 9th steam) 40g, Corni Fructus(山茱黃) 16g, Polygalae Radix(遠志) 8g, Zizyphi Spinosae Semen(酸棗仁) 2g, Biotae Semen(柏子仁 去油: oil ingredient was removed) 20g, Poria Cocos(茯笭) 12g, Ginseng Radix(人蔘) 12g, Acori Graminei Rhizoma(石菖蒲) 2g, Sinapis Semen(白芥子) 8g, on learning ability and memory were investigated. Hot water extract(HWE) and ethanol extract(EE) from SHT were used for the studies. Learning ability and memory are related to modifications of synaptic strength among neurons that interactive. Enhanced synaptic coincidence detection leads to improved learning ability and memory. If the NMDA receptor, a synaptic coincidence detector, acts as a graded switch for memory formations, enhanced signal detection by NMDA receptors should enhance learning ability and memory. It was shown that NR2B was increased in the forebrains of oriental medicine-administrated mice, leading to enhanced activation of NMDA receptors and facilitating synaptic potentiation in response to stimulation at 10-100 Hz. These HWE-SHT treated mice exhibited that superior ability in learning and memory when performing various behavioral tasks, showing that NR2B is enhanced by HWE-SHT treatment and also is critical in gating the age-dependent threshold for plasticity and memory formation. NMDA receptor-dependent modifications, which were mediated in part by HWE administration, of synaptic efficacy, therefore, represent a mechanism for associative learning ability and memory. Results suggest that oriental medical enhancement of NR2B contributes to increase intelligence and memory in mammals On the other hand, to examine the effects of EE-SHT on the learning ability and memory in experimental mice, EE-SHT was tested on passive and active avoidance responses. The EE-SHT ameliorated the memory retrieval deficit induced by ethanol in mice, but not other memory impairments. EE-SHT(10, 20mg/100 g, p.o.) did not affect the passive avoidance responses of normal mice in the step through and step down tests, the conditioned and unconditioned avoidance responses of normal mice in the shuttle box, lever press performance tests and the ambulatory activity of normal mice in a normal condition. However, EE-SHT at 20 mg/kg significantly decrease the spontaneous motor activity during the shuttle box test, and also to extend the sleeping time induced by pentobarbital in mice. These results suggest that SHT has an ameliorating effect on memory retrieval impairments and a weak tranquilizing action.

Effect of Daebangpungtang herbal acupuncture on the carrageenan-induced arthritis in rats (대방풍탕(大防風湯) 약침(藥鍼)이 Carrageenan으로 유도한 관절염에 미치는 영향(影響))

  • Objectives : DaeBangPungTang(DBPT) is one of the prescriptions used for the treatment of knee arthritis in oriental medicine. The present study aimed to examine the analgesic effect of DBPT on a rat model of carrageenan-induced arthritis, and the relations between DBPT-induced analgesia and endogenous nitric oxide(NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. Methods : Carrageenan-induced arthritis rat model was used to test the effect of DBPT as a chronic pain model. After the induction of arthritis, rats subsequently showed a reduced stepping force of the affected limb for at least tile next 4 days. The reduced stepping force of the limb was presumably due to a painful knee, since oral infection of indomethacin produced temporary improvement of weight bearing. DBPT dissolved in normal saline was minted several acupoints. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 12 hours. Results : DBPT produced significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 9 hours. The magnitude of this improvement was equivalent to that obtained after an oral injection of 3mg/kg of indomethacin and this improvement of stepping force was interpreted as an analgesic effect. DBPT produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. Both NO production and iNOS, COX-2 protein expression increased by arthritis were suppressed by DBPT. DBPT on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the arthritis than either DBPT or EA did. Conclusion : The present study suggest that DBPT produces a potent analgesic effect on the chronic hee arthritis pain model in the rat and that DBPT-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.

  • Objectives : This study was performed to investigate the effect of herbal acupuncture(Angelica gigas, Astragalus membranaceus) at Chun-wan(CV-12) and Chok-Samni(ST36) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL-cholesterol to total cholesterol ratio of rats fed high fat diet for diet for 5 weeks. Methods : Experimental groups were divided into normal group, high fat diet group(Control), high fat diet and Angelica gigas herbal acupuncture at CV12 and ST36 treated group(HA-1), high fat diet and Astragalus membranaceus herbal acupuncture at CV12 and ST36 treated Group(HA-2), high fat diet and Astragalus membranaceus herbal acupuncture at CV12 and ST36 treated Group(HA-3). Herbal acupuncture was bilaterally treated at the level of 16.7 ul/kg body weight per day. Results : Body weight were significantly decreased on 15th and 20th day in HA-1, HA-2, and decresed on 25th and 30th day in all the groups. Food intake were increased on 20th day and after were decreased on 30th day in HA-1 and HA-2. And food intake of HA-3 were decreased on 10th day and after were increased on 20th and 25th day. Total cholesterol, LDL-cholesterol, triglyceride and ALT were significantly decreased in HA-1, HA-2 and HA-3 groups compared with control group. HDL-cholesterol to total cholesterol ratio were significantly increased in HA-1, HA-2 and HA-3 groups compared with control group. And food efficiency were significantly decreased in HA-1 and HA-3 groups compared with control group. Especially HDL-cholesterol were increased in HA-1 Group. Conclusions : Herbal acupuncture using Angelica gigas and Astragalus membranaceus at CV12 and ST36 can manage obesity by controlling body weight, food intake and food efficiency ratio.

Estimation of the Optimum Installation Depth of Soil Moisture Sensor in an Automatic Subsurface Drip Irrigation System for Greenhouse Cucumber (시설오이 지중관비시 자동관수센서의 적정 매설깊이)

  • Vegetables production in greenhouse are typically intensely managed with high inputs of fertilizers and irrigation water, which increases the risk of ground-water nitrate contamination. In 2010 and 2011, a study was conducted to determine the appropriate depth of soil moisture sensor for automatic irrigation control to use water and nitrogen efficiently under subsurface drip irrigation (SDI) systems. The irrigation line for SDI placed 30 cm below soil surface and tensiometer was used as soil moisture sensor. Three tensiometer treatments placed at 10 (SDI-T10), 20 (SDI-T20) and 30 cm (SDI-T30) depths below soil surface under SDI. These are also compared to SUR-T20 treatment where tensiometer placed at 20 cm below soil surface under surface drip irrigation (SUR) systems. The growth of cucumber was not statistically different between SUR and SDI without SDI-T30 treatment. Fruit yields (Mg/ha) were 57.0 and 56.9 (SDI-T10), 56.0 and 60.5 (SDI-T20), 40.9 and 41.2 (SDI-T30) and 56.6 and 54.3 (SUR-T20) for 2010 and 2011, respectively. Slightly higher total yield was observed in tensiometer placed 20 cm below the soil surface, although no significant differences were found between SDI-T10 and SDI-T20 under SDI treatments. In addition, nitrogen application rates and daily irrigation rates were lowest in SDI-T20 compared with other SDIs and SUR treatments. Nitrogen and daily irrigation application under SDI-T20 was lower than that under SUR-T20 by 6.0%. These findings suggested tensiometer 20 cm depth under SDI systems was best for cucumber production in greenhouse.

Effects of Washing-water Temperature and Packaging Type on the Quality of Fresh-cut Crown Daisies (세척 쑥갓의 선도 유지에 미치는 세척수 온도와 포장 형태의 영향)

  • The effects of hydrocooling and packaging type on the quality attributes of fresh-cut crown daisies (Chrysanthemum coronarium var. spatiosum) were investigated by examining weight loss, respiration, vitamin C content, total chlorophyll content, microbial load, and sensory properties during storage at 4 and 10$^{\circ}C$. Fresh crown daisies were trimmed and washed with cold water (1 and 5$^{\circ}C$) as well as tap water (10$^{\circ}C$) 3 times each for 30 sec. They were then packaged in PP (polypropylene) film bags or PETE (polyethylene terephthalate) trays, and stored for 9 days at 4 and 10$^{\circ}C$, respectively. In general, weight loss was reduced as a result of the washing and packaging. The respiration rate increased slowly during storage at 4$^{\circ}C$, and the vitamin C and total chlorophyll contents of the crown daisies packaged in PETE trays decreased gradually during storage. Finally, the treatments consisting of hydrocooling and then packaging in PETE trays resulted in approximately 1-2 log CFU/g reductions in microbial load.

Multi-carbohydrase application into energy and amino acid deficient broiler diets: A strategy to enhance performance of broiler chickens

  • The effect of Multi-Carbohydrase (MC) supplementation on growth performance, visceral organ weights, blood metabolites, jejunum morphology, nutrient digestibility, and carcass parameters of broiler chickens fed nutrient-deficient corn soybean-meal based diets containing high levels of non-starch polysaccharides from wheat and wheat by-products was investigated. A total of 378 one-day-old Ross 308 broiler chickens were randomly assigned to one of seven dietary treatments to give six replicates per treatment (nine birds per pen). Dietary treatments were as follows: (1) positive control (PC; commercial standard diet); (2) negative control 1 (NC-1; PC-120 kcal/kg metabolizable energy); (3) NC-2 (PC-3% standardized ileal digestibility [SID] amino acids). The remaining four dietary treatments were formulated with the addition of MC (MC; Superzyme-CSTM) into two negative controls along with two supplementation levels of MC (i.e., 0.025% and 0.05%, respectively). Improved body weight, average daily gain, and feed conversion ratio (p < 0.05) were observed in broiler chickens fed a reduced energy diet supplemented with MC compared to birds fed NC-1 diet from days 1-35. Additionally, birds fed a reduced energy diet with 0.05% MC showed comparable (p > 0.05) growth performance with birds fed PC for 35-day post-hatch. Furthermore, the addition of MC into reduced amino acid diets improved (p < 0.05) growth performance. Broiler chickens fed MC supplemented nutrient-deficient diets showed a greater (p < 0.05) villus height to crypt depth ratio than birds fed diets without MC on days 21 and 35. Similarly, improved (p < 0.05) nutrient digestibility was observed in birds fed reduced energy diets supplemented with MC compared to birds fed NC-1 on days 21 and 35. Our results suggest that MC supplementation into reduced energy or reduced amino acid diets containing wheat and wheat by-products has the potential to improve growth performance and nutrient digestibility while maintaining healthier gut morphology in broiler chickens from 1 to 35 days of age.

Studies on the Foliar Application of Urea as Nitrogen Source of Rice Plant Nutrition (요소엽면살포(尿素葉面撒布)에 따른 수도(水稻)의 질소영양(窒素營養)에 관(關)한 연구(硏究))

  • This experiment was carried out as a part of the studies on reasonable application of nitrogen in rice plant to determine: (I) Nitrogen absorption. and rooting of rice seedlings as affected by urea foliar application at late seedling stage (II) Effect of leaf prunning and foliar application of urea at late heading stage on the maturation and yield of rice (III) Effect of foliar application of urea and its time during the stage of ear formation on yield of rice plant. Results obtained are summarized as follows. Exp.I: Nitrogen absorption and rooting of rice seedlings as affected be urea foliar application at late seedling stage. 1 : The foliar application of urea plots$(T_{1},T_2)$ snowed mare N-content than non-urea foliar application plot(T0) at lane seedling stage, being significant among treatments and foliar application of urea seemed more effective in increasing the N-content of seedlings. and promoted root settlement and early growth alter the transplanting. 2 : The carbon contents of the plants of $T_1$, and $T_2$ at late seedling stage increased than T0, and the carbon contents. of $T_1$ and $T_2$ plots became higher in amount in proportion to the nitrogen absorption as compared with those of $T_0$. 3 : C/N ratio appeared significant among soil application plots($N_1, \;N_2$) and foliar application of urea plots ($T_1$, $T_2$ and $T_0$). C/N ratio was lower in case of increased amount of nitrogen. The higher contents of nitrogen and carbon and lower C/N ratio resulted in the increment of root numbers and root lengths. Exp.II: Effect of leaf prunning and foliar application of urea at late heading stage on the maturation and yield of rice. 1 : There was a highly significant decrease in the maturing rate by severe leaf prunning. In the mean time, significant increase in maturing rate was observed with urea foliar application and it was found the more frequent application the more effective for higher maturing rate with a moderate significance. A correlationship between the level of prunning and maturing rate was enumerated to 0.961 of correlation coefficient, which indicated an increased maturing rate by the increased number of remaining leaves. 2 : The 1.000 grain weight, grain weight and hulled rice yield increased by leaf prunning in order (plot a$A_1$, $A_3$, $A_2$ and $A_0$ were 89.8%, 89.4%, 87.8% and 87.5% respectively, showing the highest of rate in $A_1$ and $A_3$ in methods of ear fertilization and being highly significant between its treatment. 3 : 1000 grain weights were highly significant between time of application, showing a tendency of increase of weights with the time lagging until days before earings as that of maturing rates. High significance was recognized between methods of ear fertilization, showing the highest in $A_2$ 23.18 gr. 4 : Yields per $3.3m^2$ were not significant between time of ear fertilization, whereas were highly significant between methods of ear fertilization. Those of $A_1$, $A_3$, $A_2$ and $A_0$ were 1.486 kg, 1.491 kg, 1.381 kg and 1.328 kg, respectively, showing the highest in $A_1$ and $A_3$. 5 : Hulling ratios showed significant different between time of ear fertilization, showing the highest in $T_2$, whereas those of methods of ear fertilization were highly significant between its treatment, Those of $A_1$, $A_3$, $A_2$ and $A_0$ were 84.72%, 84.06%, 83.29%, and 82.56% respectively, showing the highest m $A_2$ and $A_3$ among others. 6 : Yields of hulled rice per $3.3m^2$ showed significant different between time of ear fertilization, showing the highest in $T_1$ 1.192 kg. Whereas, those were highly significant between methods of ear fertilization. Those of $A_1$, $A_3$, $A_2$ and $A_0$ were 1.259 kg, 1.254 kg, 1.149 kg and 1.095 kg, respectively, showing the highest in $A_1$ and $A_2$. 7 : Contents of nitrogen on rice plant increased in case of nitrogen application as ear fertilizer and showed that the case of urea foliar application was more effective than that of soil application, showing the increased nitrogen content of rice plant was accompanied by carbon content.

Simultaneous Pesticide Analysis Method for Bifenox, Ethalfluralin, Metolachlor, Oxyfluorfen, Pretilachlor, Thenylchlor and Trifluralin Residues in Agricultural Commodities Using GC-ECD/MS (GC-ECD/MS를 이용한 농산물 중 Bifenox, Ethalfluralin, Metolachlor, Oxyfluorfen, Pretilachlor, Thenylchlor 및 Trifluralin의 동시 분석)

  • BACKGROUND: This experiment was conducted to establish a simultaneous analysis method for 7 kinds of herbicides in 3 different classes having similar physicochemical property as diphenyl ether(bifenox and oxyfluorfen), dinitroaniline (ethalfluralin and trifluralin), and chloroacetamide (metolachlor, pretilachlor, and thenylchlor) in crops using GC-ECD/MS. METHODS AND RESULTS: All the 7 pesticide residues were extracted with acetone from representative samples of five raw products which comprised apple, green pepper, Kimchi cabbage, hulled rice and soybean. The extract was diluted with saline water and directly partitioned into n-hexane/dichloromethane(80/20, v/v) to remove polar co-extractives in the aqueous phase. For the hulled rice and soybean samples, n-hexane/acetonitrile partition was additionally employed to remove non-polar lipids. The extract was finally purified by optimized Florisil column chromatography. The analytes were separated and quantitated by GLC with ECD using a DB-1 capillary column. Accuracy and precision of the proposed method was validated by the recovery experiment on every crop samples fortified with bifenox, ethalfluralin, metolachlor, oxyfluorfen, pretilachlor, thenylchlor, and trifluralin at 3 concentration levels per crop in each triplication. CONCLUSION: Mean recoveries of the 7 pesticide residues ranged from 75.7 to 114.8% in five representative agricultural commodities. The coefficients of variation were all less than 10%, irrespective of sample types and fortification levels. Limit of quantitation (LOQ) of the analytes were 0.004 (etahlfluralin and trifluralin), 0.008 (metolachlor and pretilachlor), 0.006 (thenylchlor), 0.002 (oxyfluorfen), and 0.02 (bifenox) mg/kg as verified by the recovery experiment. A confirmatory technique using GC/MS with selected-ion monitoring was also provided to clearly identify the suspected residues. Therefore, this analytical method was reproducible and sensitive enough to determine the residues of bifenox, ethalfluralin, metolachlor, oxyfluorfen, pretilachlor, thenylchlor, and trifluralin in agricultural commodities.

Patterns of Leaching and Distribution of Cations in Reclaimed Soil according to Gypsum Incorporation Rate (석고 혼합량에 따른 간척지토양의 양이온 용탈 및 분포)

  • Serious problems in reclaimed land for agriculture are high soil salinity and very poor vertical drainage. However, desalinization in these soils is very difficult. To identify the change of soil permeability by the gypsum incorporation in soils we observed elution patterns and salt distribution of the soil using soil columns packed with reclaimed saline soil with various rates of gypsum amendment. Saturated hydraulic conductivity ($K_{sat}$) of the top soil of reclaimed saline soils without gypsum incorporation was close 0 cm $hr^{-1}$ while $K_{sat}$ increased up to 0.3 cm $hr^{-1}$ with increasing amount of gypsum for 0.4% or more gypsum. Also $K_{sat}$ of the reclaimed saline soils for top soil was drastically increased to 1.0 cm $hr^{-1}$ or slightly greater after 141 hour's elution experiment. The cumulative amount of cation recovered in the effluent also increased in the order of gypsum incorporation rate 0.8% > 0.6% > 0.4 > 0.2%. Soil EC in soil columns decreased from initial 33.9 dS $m^{-1}$ to less than 0.4 dS $m^{-1}$ and exchangeable Ca 2+ increased by 32~140% according to gypsum incorporation rate.

A Study on the Physical Growth and Health Status of the Freshmen in a Private University (일부사립대학교(一部私立大學校)의 학생(學生)의 성장(成長).발육(發育) 및 건강상태(健康狀態)에 관(關)한 조사연구(調査硏究))

  • For the purpose of ascertaining the status of physical growth and health status of the University Students, an intensive survey was conducted by the authors towards a total of 1,250 (Male 792, Female 458) who passed the written entrance examination at the Kyung Hee University in 1972. The items included the measurements on physical growth, various physical and nutritional indices, status of visual distourbance, dental status and tuberculosis. The findings and results cad be summarized as follows; 1. Physical Growth and Develspment. i) The average of body height by anthropometric was $169.39{\pm}5.05cm$ in male and $157.45{\pm}4.43cm$ in female. ii) The averages of body weight by sex were $59.42{\pm}6.47Kg\;and\;51.81{\pm}5.21Kg$, respectively. iii) The averages of chest-girth were $87.18{\pm}5.30cm\;and\;80.51{\pm}4.53cm$. iv) The averages of sitting-height were $92.21{\pm}2.79cm\;and\;86.28{\pm}2.57cm$. In all cases the male measurements were higher than female. 2. Various ludices about Physical Growth and Development; i) Relative body weights by sex were 35.08 and 32.91, relative chestgirth 51.47 and 51.13, and relative sitting-height 54.43, 54.79, respectively. ii) Rohrer index was 1.222 in males and 1.327 in females, Kaup index 2.071 and 2.089, Verveck index 86.54 and 84.04, and Pelidisi index 91.15 and 93.08, respectively 3. Status of visual distourbance The vision of the students under 0.8 with Landolt's testing chart was 45.4% in left eye and 46.6% in right eye(male: 44.4% and 45.7%, female: 47.2% and 48.0%), while under 0.6 in both visions was 41.0% and 40.5% (male: 40.8% and 40.7%, female: 41.5% and 40.2%), respectively. 4. Dental Status Out of total 19.4% (male 19.2%, female 19.6%) had gingivitis, the female incidence rate of gingivitis was higher than male. Average number of teeth in each subject was 29.6 teeth: male had 29.9 teeth and female had 29.1 teeth. The caries rate was 78.9%(male 75.6%, female 84.5%), the female caries rate was higher than male. Total filing per subject was 35.6% (male 34.0%, female 37.1%), female incidence was higher shan male. The average number of i) Caries per tooth rate was 8.5% in male and 9.2% in female, ii) Missing per tooth rate was 0.3% and 0.2%, iii) Filling per tooth rate was 3.9% and 4.2%. in all cases, the female incidences were higher than the male. The average number of D.M.F. was 3.8 teeth in male and 4.0 teeth in female, female was higher than male. The average rate of D.M.F. was 12.7% in male and 13.7% in female, female was higher than male. 5.
