홍색 비유황 광합성 세균 Rhodobacter sphaeroldes KD131의 수소생산에 미치는 빛 세기 및 질소원의 영향 (Effect of Light Intensity and Nitrogen Source on Hydrogen Production Using Rhodobacter sphaeroldes KD131)

  • Photobiological hydrogen production using Rhodobacter sphaeroides KD131 was studied on the effect of light intensities and nitrogen sources. Media containing malate and glutamate were shown higher hydrogen production rate than that containing succinate and $(NH_4)_2SO_4$ at the $110\;W/m^2$ illumination by halogen lamp at $30^{\circ}C$. Media lacking glutamate as the nitrogen source exhibited higher hydrogen production than that containing glutamate. Initial cell concentration was optimized to 1.0 at the absorbance of 660 nm. Hydrogen production was increased by increasing the light intensity from 0 to $216\;W/m^2$ but the increasing rate declined over $108\;W/m^2$.

  • KD₂PO₄ single crystals were grown from D₂O with reagent KH₂PO₄ and the crystal structure was determined by X-ray and neutron diffraction methods. The crystals are tetragonal at room temperature, I42d, with lattice parameters of a=7.4633(7), c=6.9785(5) Å and Z=4. Intensity data were collected on an Enraf-nonius CAD4 diffractometer with a graphite monochromated MoK/sub α/ radiation (λ=0.7107Å) and on the neutron four circle single crystal diffractometer with Ge(331) monochromated neutron beam (λ=0.997Å). The structure was refined by full-matrix least-square to final R and wR values of 0.030 and 0.072, respectively, for 204 observed reflections with I>2σ(I) by X-ray diffraction and to final R=0.041 and wR=0.096 for 144 observed relfecdtions by neutron diffraction. The O…O distance of 2.516(4)Å obtained by X-ray diffraction is the same as that of 2.515(4)Å by neutron diffraction. On the other hand, the O-D/H distance of 0.84(4)Å by X-ray diffraction is considerably shorter than 1.029(7) Åby neutron diffraction. Hydrogen and deuterium can be readily distinguished by neutrons. In this crystal 66% of H-positions were substituted by D and the rest 34% occupied by H. The phase transition temperature of DKDP obtained with deuteration levels is f193K. This value agrees fairly well with the result of DSC measurement. The nuclear density distribution by neutron diffraction provides an observation of the disordered state of D/H in KD₂PO₄ at room temperature.

  • 토양에 따른 제초제의 흡착을 구명하기 위하여 alachlor와 paraquat를 사용하여 공시토양 22에 대한 약제별 흡착정도 및 토양특성과 흡착정도의 상관을 조사한 결과는 다음과 같다. 1. 진탕시간에 따른 토양과 약제의 평형이 이루어지는 시간은 paraquat가 약 30분, alachlor는 약 4시간이었다. 2. 분배법칙에 의한 alachlor의 흡착계수는 5ppm 농도에서 0.81-33.83이었으며, 50ppm에서는 0.09-15.52의 범위를 나타내었다. 3. 토양특성중 alachlor의 흡착력은 O.M.함량, paraquat의 흡착력은 점토함량과 고도의 정의 상관을 보였다. 4. 우리나라 토양중 춘포통은 유기물과 점토함량이 낮음에도 특이한 특성 때문에 paraquat와 alachlor 모두 높은 흡착량을 나타내었다. 5. Freundlich의 흡착상수(K)는 분배법칙의 계수(Kd)보다 대체적으로 높으며, 평형농도가 증가함에 따라 K와 Kd값의 차이는 증가하는 경향이었다.

비정형 가와사키병의 임상 양상 (Clinical Features of Atypical Kawasaki Disease)

  • 목 적 : 최근에 고전적인 진단 기준을 만족하지 않는 비정형 가와사키병에 대한 보고가 증가하고 있어 이의 조기 진단 및 치료를 위한 진단 기준을 마련하고자 하였다. 방 법: 1998년 1월부터 2000년 6월까지 가와사키 병으로 진단 받은 환아 총 167명을 대상으로 비정형 가와사키병의 임상적 특징을 전형적 가와사키병과 비교 분석하였다. 결 과 : 1) 총 환아 167명 중 비정형 가와사키병은 28명(16.8%)이었고, 1세 미만 환아의 비율은 25.0%로 전형적 가와사키병에서의 16.5% 보다 높은 경향을 보였다(P=NS). 2) 전형군과 비정형군 간에 임상 증상의 발생 빈도를 비교하면 구강 점막 변화를 제외하고 안구 결막의 충혈, 부정형 발진, 사지 말단의 변화, 경부 림프절 종대는 비정형군에서 유의하게 낮았다. 비정형군의 임상 증상 중에서 구강 점막 변화가 제일 흔하며(96.4%), 다음으로 안구 결막 충혈(57.1%), 부정형 발진(46.4%)의 순이었다. 급성기의 검사 소견은 대부분 두 군간에 차이가 없었으며, 적혈구 용적 <35%, ESR >30mm/hr, CRP >3.1 mg/dL의 빈도는 두 군에서 모두 높았다. 3) 관상동맥 병변은 비정형군에서는 28명 중 7명(25.0%), 전형군에서는 139명 중 20명(14.4%)에서 발생하여 비정형군에서 빈도가 더 높은 경향을 보였다(P=NS). 4) 관상동맥 병변이 있는 환아 총 27명 중에서 7명(25.9%)이 비정형 가와사키병 환아였다. 5) 구강 점막 변화를 주기준, 안구 결막 충혈, 부정형 발진 및 적혈구 용적 <35%, ESR >30 mm/hr 또는 CRP >3.1 mg/dL을 부기준으로 할 때 발열과 주 기준 그리고 2개 이상의 부기준이 있을 때, 또는 발열과 4개의 부기준이 있을 때를 비정형 가와사키병의 진단 기준으로 정할 때 민감도는 89.3%였다. 결 론 : 비정형 가와사키병의 조기 진단을 위해 본 연구에서 제안한 진단 기준이 도움이 되리라고 생각된다.

Cardiovascular risk factors of early atherosclerosis in school-aged children after Kawasaki disease

  • Purpose: The aim of this study was to determine whether school-aged children with Kawasaki disease (KD) have an increased risk for early atherosclerosis. Methods: The study included 98 children. The children were divided into the following groups: group A (n=19), KD with coronary arterial lesions that persisted or regressed; group B (n=49), KD without coronary arterial lesions; and group C (n=30), healthy children. Anthropometric variables and the levels of biochemical markers, including total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), apolipoprotein A, apolipoprotein B, homocysteine, highsensitivity C-reactive protein (hs-CRP), and brachial artery stiffness using pulse wave velocity were compared among the three groups. Results: There were no significant differences in blood pressure and body index among the three groups. Additionally, there was no sex-specific difference. Moreover, the levels of triglyceride, HDL-C, apolipoprotein A, and hs-CRP did not differ among the three groups. However, the levels of total cholesterol (P =0.018), LDL-C (P =0.0003), and apolipoprotein B (P =0.029) were significantly higher in group A than in group C. Further, the level of homocysteine and the aortic pulse wave velocity were significantly higher in groups A and B than in group C (P=0.0001). Conclusion: School-aged children after KD have high lipid profiles and arterial stiffness indicating an increased risk for early atherosclerosis.

김치에서 분리한 Lactococcus lactis가 생산하는 박테리오신의 정제 및 특성 (Purification and Characterization of the Bacteriocin Produced by Lactococcus sp. KD 28 Isolated from Kimchi)

  • 김치로부터 분리한 L. sp. KD 28은 펩타이드성 항균물질인 박테리오신을 생산하였다. 이 박테리오신은 ${\alpha}$-chymotrypsin, proteinase K, lipase, ${\alpha}$-amylase, carboxypeptidase A 효소에 대해서 매우 민감한 반응을 보였다. 낮은 pH 2.0에서 약 염기성의 pH 8.0까지는 항균활성을 온전히 유지하였으나 pH 8.0 이상에서는 항균활성이 감소하기 시작하여 pH 12.0에서는 25%로 감소하였다. L. sp. KD 28는 16S rRNA 분자계통학적 분석을 한 결과 L. lactis와 99% 상동성을 보였다. 또한 김치에서 분리한 L. sp. KD 28이 생산하는 박테리오신은 acetonitrile, isopropanol, methanol, chloroform 및 acetone 등의 유기용매 최종 농도 50%에서 항균활성은 영향을 받지 않았다. 열 안정성의 경우 $80^{\circ}C$까지 한 시간의 열처리에 안정하였으나 $100^{\circ}C$에서는 20분 이상의 열처리에 의해 항균활성이 감소하여 50분간 열처리할 경우 항균활성이 나타나지 않았다. 또한 이 박테리오신은 그람 음성보다는 양성 세균인 M. luteus IAM 1056, L. delbrueckii subsp. lactis KCTC 1058, E. faecium KCTC 3095, B. cereus KCTC 1013, B. subtilis KCTC 1023, S. aureus subsp. aureus KCTC 1916, B. megaterium KCTC 1098, B. sphaericus KCTC 1184 및 L. ivanovii subsp, ivanovii KCTC 3444 등에 대해서 항균활성을 보였다. 박테리오신을 SP-Sepharose 이온교환크로마토그래피로 부분 정제한 후 RP-HPLC 를 통해서 최종적으로 정제하여 tricine-SDS-PAGE를 통해 분자량을 확인한 결과 약 3.4 kDa로 나타났다.

  • Lipophilic fraction(LF) from Panax ginseng C.A. Meyer inhibited the aggregation of human platelets induced by th rombin(0.1u/$m{\ell}$). LF and Molsidomine(vasodilator) induced the stimulation of cGMP - elevation and 50KD - Phosphorylation. and then the inhibition of 20KD - Phosphorylation in human platelets activated by thrombin. LF also inhibited the $Ca^{2-}-influx$ into platelets. When rat(SD : male) was fed with LF, the level of cGMP was increased in rat platelets stimulated by collagen and thrombin. On the other hand. verapamil, $Ca^{2-}-antagonist$ increased cAMP level ;n platelet stimulated by thrombin. but LF does not affected. However LF potently inhibited the thromboxane $A_2(TXA_2)$ production. The results suggest that the inhibitory effects of LF are mediated by regulation the phosphorylatior. of 50KD via cGMP-elevation and depend upon the decrease of $TXA_2$ level.

  • In this paper, we designed and fabricated the ubiquitous thermal sensor unit for high voltage facilities. This sensor unit generates electric power from a current transformer which was mounted in the high voltage wires and transmitted thermal data by using a Zigbee module. We simulated the high voltage facilities that temperature was going up to $80^{\circ}C$ gradually. From the experimental results, it was confirmed that sensibility of the ubiquitous thermal sensor unit is in ranges from $-0.7^{\circ}C$ to $0.3^{\circ}C$.

Age-adjusted plasma N-terminal pro-brain natriuretic peptide level in Kawasaki disease

  • Purpose: Recent reports showed that plasma N-terminal pro-brain natriuretic peptide (NT-proBNP) could be a useful biomarker of intravenous immunoglobulin (IVIG) unresponsiveness and coronary artery lesion (CAL) development in Kawasaki disease (KD). The levels of these peptides are critically influenced by age; hence, the normal range and upper limits for infants and children are different. We performed an age-adjusted analysis of plasma NT-proBNP level to validate its clinical use in the diagnosis of KD. Methods: The data of 131 patients with KD were retrospectively analyzed. The patients were divided into 2 groups-group I (high NT-proBNP group) and group II (normal NT-proBNP group)-comprising patients with NT-proBNP concentrations higher and lower than the 95th percentile of the reference value, respectively. We compared the laboratory data, responsiveness to IVIG, and the risk of CAL in both groups. Results: Group I showed significantly higher white blood cell count, absolute neutrophil count, C-reactive protein level, aspartate aminotransferase level, and troponin-I level than group II (P<0.05). The risk of CAL was also significantly higher in group I (odds ratio, 5.78; P=0.012). IVIG unresponsiveness in group I was three times that in group II (odds ratio, 3.35; P= 0.005). Conclusion: Age-adjusted analysis of plasma NT-proBNP level could be helpful in predicting IVIG unresponsiveness and risk of CAL development in patients with KD.

Polymorphisms of methylenetetrahydrofolate reductase are not a risk factor for Kawasaki disease in the Korean population

  • Purpose: Hyperhomocysteinemia is known as a risk factor for atherosclerosis. Preclinical arteriosclerosis is noted and premature atherosclerosis is known to be accelerated in Kawasaki disease (KD) patients. Genetic polymorphisms in the 5,10-methylenetetrahydrofolate reductase (MTHFR) gene result in elevated plasma homocysteine concentrations and are known to be associated with the development of coronary artery disease. Our hypothesis is that single nucleotide polymorphisms (SNPs) of the MTHFR gene are related to the development of KD and coronary artery lesions (CALs). Methods: For this study, we selected 3 candidate single nucleotide polymorphisms (SNPs) (rs2274976, rs1801131, and rs1801133) of MTHFR. These SNPs are located on chromosome 1p36.3. We included 101 KD patients and 306 healthy adults as controls in this study. CALs were seen in 38 patients. Genotypes of the selected SNPs were determined by direct sequencing and analyzed with SNPAlyze. Results: The genetic distribution and allelic frequency of the 3 MTHFR SNPs (rs2274976, rs1801131, and rs1801133) were not significantly different in patients with KD compared to the control group (P=0.71, 0.17, and 0.96, respectively). There was no difference in the genetic distribution of the MTHFR SNPs between the normal control group and the CAL group (P=0.43, 0.39, 0.52 respectively). Conclusion: The genetic distribution of the MTHFR SNPs (rs2274976, rs1801131, and rs1801133) was not different in the KD group compared to the control group. In addition, the genetic distribution of these SNPs was not different in the CAL group compared to the control group in the Korean population.
