Helicobacter pylori 항원을 이용한 면역우유의 열처리와 저장 중 항체의 안전성 (Stability of antibody during heat treatment and storage in immunized milk with Helicobacter pylori antigen)

  • Anti-H pylori 항체를 함유한 면역우유를 $60^{\circ}C$에서 30분 열처리한 것과 열처리하지 않은 Anti-H. pylori 항체를 함유한 원유의 면역활성 비교시 99.99(100)%의 면역 활성을 나타내었으며, $75^{\circ}C$ 이상의 열처리에 의해서는 면역 활성이 급격하게 감소하여 약 50%이하로 감소하였다. 열처리 온도가 높을수록, 열처리 시간이 증가할수록 총균수의 감소율이 증가하였다. Coliform bacteria의 경우는 원유에서 $10^2\;CFU/mL$이 관찰되었으나 모든 열처리 구에서는 관찰되지 않았다. 면역우유를 $2^{\circ}C,\;4^{\circ}C$ 그리고 $10^{\circ}C$에서 21일 동안 저장한 결과 면역우유의 저장 중pH의 변화는 저장 21일 동안 $2^{\circ}C$$4^{\circ}C$에 저장한 경우 저장온도에 따른 뚜렷한 변화는 관찰할 수 없었으며, $10^{\circ}C$에 저장한 경우 저장 7일 이후 급격히 감소하였다. 총균수의 변화는 저장 초기 $10^3\;CFU/mL$에서 $2^{\circ}C$$4^{\circ}C$에 저장한 열처리 면역우유의 총균수는 저장 21일 동안 뚜렷한 증가현상을 관찰할 수 없었으며, $10^{\circ}C$에 저장하였을 경우 7일 이후 급격히 증가하여 저장 14일째 $10^8\;CFU/mL$로 부패현상을 나타내었다. 면역우유의 저장 중 면역 활성의 변화는 저장 온도에 관계없이 저장 14일까지는 저장초기의 활성에 비해 뚜렷한 차이는 나타나지 않았으나 14일 이후 급격히 감소하는 경향을 나타내었다.

  • The effects of thyroxine (TX) or propylthiouracil (PPT) administration on the antibody forming activity agains t sheep red blood cell (SRBC) and Newcastle disease virus (NDV) were studied by using of hemagglutination and hemagglutination-inhibition techniques. Antibody titers to both SRBC and NDV increased significantly in the TX-treated group, whereas decreased in the PPT-treated group, compared with control. When TX was administered after antigen inoculatioon, antibody forming activity was significantly enhanced, compared with the TX administration before antigen inoculation.

신경모세종의 분화에 따른 인체 CD99의 표현의 증가 (The increased expression of CD99 in a differentiated neuroblastoma cell line)

  • Background: The human mic2 gene is a pseudoautosomal gene that encodes a cell surface antigen, CD99. High levels of CD99 constitute a tumor marker in Ewing s sarcoma (ES). We have recently demonstrated that CD99-induced apoptosis occurs only in undifferentiated ES cells, not in differentiated ES cells, raising the possibility of the involvement of CD99 in neural ontogeny. Methods: To elucidate the relations between the expression of CD99 and the differentiation of neural cells and the mechanism by which the expression of CD99 is regulated, we analyzed the differential patterns of CD99 expression in SH-SY5Y by treatment of 12-O-tetradecanoyl-13-phorbol acetate (TPA) and retinoic acid. In addition, to explore the transcriptional activity of CD 99 during neural cell differentiation, SH-SY5Y cells were transiently transfected with a CD99 promoter-driven luciferase construct, and treated with the inducers. Results: In immunoblotting and flow cytometry, the expression level of CD99 was increased on differentiated SH-SY5Y cells induced by TPA and retinoic acid. The luciferase activity was elevated by the treatment with TPA, known to mature SH-SY5Y cells toward a sympathetic neuronal lineage, whereas retinoic acid inducing a sympathetic chromaffin lineage displayed little effect. Conclusion: The result indicates that CD99 might be expressed only on cells maturing toward a neuronal lineage among differentiating primitive neuronal cells. In addition, the expression of CD99 seems to be regulated at the transcriptional level during the differentiation.

  • The prevalence of diarrhea caused by enterotoxigenic Escherichia coli was surveyed on 445 calves in 6 farms which were located in the central part of Korea. The incidence of enterotoxigenic Escherichia coli in calves with diarrhea was investigated by detecting the K99 and F41 antigens from the isolated strains of Escherichia coli The incidence of colibacillosis in calves was 23.3%. Of 238 strains of Escherichia coli isolated from calves with diarrhea, 73 strains(30.6%) were proved possessing the K99 antigen by mannose-resistant hemagglutination(MRHA) using horse red blood cells and 79(33.1%) possessing F41 antigens by MRHA using guinea-pig red blood cells. The minca medium, nutrient broth, tryptic soy broth and brain heart infusion were tested for yield of K99 and F41 pili. The production of pili was greatest in minea medium. The best detachment method of the K99 and F41 pili from the cells was heat treatment for 20 minutes at $60^{\circ}C$ and concentration by precipitation with ammonium sulfate. The purified antigens of K99 and F41 were polypeptides with molecular weights of 18,500 and 29,500, respectively by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

Salmonella enteritidis의 편모항원에 대한 난황항체의 ,생산 x Production of Egg Yolk Antibodies against Flagella Antigen of Salmonella enteritidis

  • 가금 및 가축의 살모넬라중을 일으키는 에서 편모항원을 순수분리하고 이를 면역원으로 산란계에 면역하여 난황 중에 형성된 항체와 혈청간에 면역 반응을 조사하고 생산된 에 대한 특이난황 항체의 특이성 검사와 항체회수율 검사를 실시한 결과S. enteritidis는 다음과 같다. 1. S. enteritidis에서 순수 분리한 flagella protein 의 분자량은 54.6 kDa으로 나타났다. 2. 산란계의 혈청중 항체가 형성은 면역 후 2주경부터 급격히 증가하기 시작하여 6주경에 최고의 항체가 수준에 도달하였다. 반면에 난황중 항체가는 면역 후 4주경부터 급격히 증가하기 시작하여 6주경에 혈청항체가 수준과 동일하게 도달되며, 10주 이후에는 혈청항체가 수준보다 높게 형성되어 일정한 수준을 유지하였다. 3. S. enteritidis의 flagella protein을 면역원으로 산란계에 면역하여 생산된 난황항체는 주로 S. enteritidis 균주에서만 특이적으로 반응하며 E. coli (Kl2:K99, K88+, 987P) 균체에서는 반응이 전혀 없는 것으로 나타났다. 4. 한개의 난으로부터 분리한 IgY의 함량은 106 mg이었으며 회수율은 88.3%였다.

  • The term allergy was originally defined by Clemens Yon Pirquet as ‘an altered capacity of the body to react to foreign substance’, which was an extremely board definition that included all immunological reactions. Allergy is now defined in a much more restricted manner as ‘disease following an immune response to an otherwise innocuous antigen’. Allergy is a member of a class of immune responses that have been termed hypersensitivity reactions; these are harmful immune responses that produce tissue injury and may cause serious disease. 송아지 대장균 백신개발에 관한 연구 (Studies on Protective Efficacy of Escherichia coli Vaccines)

  • The oil emulsion and alhydrogel vaccines were prepared from a strain of enterotoxigenic Escherichia coli isolated from calves with diarrhea and their protective efficacy and immunogenicity were tested in Guinea-pigs. Enterotoxigenic Escherichia coli, isolated from calves with diarrhea, has K99 and F4l antigen as 46.2% and 50.9% with 48 and 53 strains respectively out of 104 strains. The protective efficacy of the gel and oil vaccines were 60% and 80% respectively. Agglutinin titers to sera of Guinea-pigs vaccinated with experimental gel and oil vaccines peaked at 5 and 6 weeks after vaccination.

돼지 유래 대장균의 생물학적 특성과 plasmid profile에 대하여 (Studies on biological characters and plasmid profiles of Escherichia coli isolated from pigs)

  • The purpose of this study was the examination for presence of pilus antigen, O serogroups, colicin production, antibiotic susceptibility and plasmid profiles among E coli isolated from diarrheal piglets and fattening pigs in Taegu province. Of 145 E coli isolated, 98 strains (67.4%) possesed pilus antigens which belonged to either K88 (47.6%), K99 (11.7%) or 987P (8.3%) types. Fifty-nine strains (40.7%) were classified into tenO serogroups and their types were O8 (22.0%), O20(16.9%), O141(15.3%), O9(10.2%), O45(10.2%), O139(8.5%), O064(6.8%), O149(5.0%), O157(3.4%), and O115(1.7%). Thirty-three strains (22.8%) were colicinogenic and 6 strains (4.1%) were hemolytic. One hundred and thirty-nine strains (95.9%) of 145 E coli isolates were resistant to ampicillin, chloramphenicol, gentamicin, kanamycin, streptomycin, tetracycline, rifampicin and nalidixic acid, alone or in combination thereof. Ninety strains (64.7%) of 139 drug resistant strains carried R factor (R) which were transferable to the recipient by conjugation. In gel electrophoresis for the isolation of plasmid DNA, the number of plasmid DNA band varied from 2 to 11 in 16 E coli with pilus antigen. It's molecular weight ranged from 1.0 to 60.0 megadalton.

혈액, 초음파 검사 결과를 이용한 전립선이행대용적의 상관관계 연구: 기초질환이 없는 30대를 대상으로 (A Study on the Correlations between Total Prostate Volume and Prostate Transition Zone Volume Assessed Using Blood and Ultrasound Tests: in the Healthy Korean Men in Thirties)

  • 본 연구는 혈액 검사 결과인 전립선특이항원(PSA) 및 전립선특이항원밀도 (PSAD), 전립선 경직장 초음파 (TRUS)를 이용한 전립선 전체용적 (TPV), 나이등 변수와 전립선이행대용적 (PTZV)과 상관관계를 연구하여 임상적 지표로 활용하고자 한다. 2007년 6월부터 2016년 4월 까지 전립선에 대한 치료력이 없는 건강한 30대 성인 남성으로 TRUS와 PSA, PSAD 혈액 검사를 실시한 총 68명을 후향적으로 분석하였다. 초음파 장비는 Siemens Acuson sequoia 512)와 탐촉자 Siemens EC~10C5 Endocavitary를 이용하였다. 통계 처리의 경우, SPSS 18.0 각 변수와 표준 편차의 평균을 계산하는 데 사용하고, 피어슨 상관관계 분석을 수행 하였다. 변수들의 기술 통계량은 TPV; $24.27{\pm}6.60$, PTZV; $6.99{\pm}6.60$, PSA; $2.12{\pm}2.76$와 PSAD; $0.281{\pm}0.1$이고. 전립선 이행용적과 변수의 상관 계수는 PSAD; 0.831, TPV; 0.707, 나이; 0.398, 그리고 PSA; 0.118이었다. PSA 및 연령과의 양의 크기의 상관관계지만, PSAD, TPV가 높은 상관관계를 보였다. 따라서, 기저 질환이 없는 30대 남성의 PTZV는 TPV와 PSAD를 통해 예측할 수 있다.

기관지 세척액 검사를 이용한 MAGE 유전자 검사의 임상적 의의 (The Clinical Implication of MAGE Gene Detection in Bronchial Washing Fluid in Routine Practice)

  • Background: Melanoma antigen genes (MAGE) are expressed in many human malignant cells and are silent in normal tissues other than in testis and in placenta. But MAGE expression in benign lung diseases, such as pulmonary tuberculosis or cases with severe inflammation, needs further evaluation to overcome false-positive findings. We evaluated detection rates of the melanoma antigen genes (MAGE) RT-nested PCR in bronchoscopic washing samples from patients with benign lung disease, as well as in patients with malignancies. Methods: Bronchial washing fluid from 122 patients was used for cytological examination and MAGE gene detection using RT-nested-PCR of common A1-6 mRNA. We compared the results from the RT-nested PCR and the pathologic or bacteriologic diagnosis. We also analyzed the expression rate and false positive rate of MAGE gene. Results: Among 122 subjects, lung cancer was diagnosed in 23 patients and benign lung disease was diagnosed in 99 patients. In patients with lung cancer, the positive rate of MAGE expression was 47.8% (11/23) and in benign lung disease group, the expression rate was 14.1% (14/99). Among benign lung disease group, the expression rate of MAGE gene (25.0%) in patients with pulmonary tuberculosis (11/44) was especially high. Conclusion: MAGE A1-6 RT-nested PCR of bronchial washing fluid can be used as a complementary method in lung cancer, but that test results in a high false positive rate in tuberculosis patients.
