보건소 간호조직문화가 직무만족과 조직몰입에 미치는 영향 (The Influence of Nurses' Organizational Culture on Their Job Satisfaction and Organization Commitment at the Public Health Center)

  • Purpose: This study examines how the types of organizational culture at a public health center affect job satisfaction and organization commitment of nurses. Method: The study selected 139 nurses from six public health centers located in G city, J province as subjects. The data was collected from April 1 to May 31 in 2008. Result: In regard to type of organizational culture had significant correlation with both job satisfaction and organization commitment, job satisfaction and organization commitment also showed high correlation. Concerning type of organizational culture, two variables of affiliated culture and innovative culture explained 26.3% of job satisfaction and 29.3% of organization commitment. Conclusion: The job satisfaction and organization commitment of nurses varied according to types of organizational culture of a public health center, and showed high correlation. The more affiliated and innovative the organizational culture was, the higher job satisfaction and organization commitment turned out. Therefore, it is advisable to develop a strategy that systematically creates a affiliated and innovative organizational culture that pays attention to goal achievement of the nurse.

소방공무원의 조직문화가 직무만족도에 미치는 영향 (The Influence of Organizational Culture on Job Satisfaction of Fire Officials)

  • Purpose : The purpose of this study was to investigate the influence of organizational culture on job satisfaction of fire officials. Method : The subjects of this study were 251 fire officials in Gongju and Choengju province and Daejeon City. Data were collected using self-administering questionnaire from February 20 to March 20 in 2006. Collected data were analyzed using SPSS 14.0 program. Real number, percentage, mean and standard deviation were calculated, and t-test, ANOVA, Pearson correlation coefficient, multiple regression were analyzed. Result : The results of this study were as follows : 1. The job satisfaction was found to be significant positive correlations with full organizational culture(r = .595, p = .000). 2. The job satisfaction was found to be in significant positive with rational goal culture, open system culture and human relation culture. 3. How organizational culture affects the job satisfactions was as follows; 25.2% of core values of manpower, 6.3% of rational goal culture, and 4.9% of job initiative. Conclusion : Enhancing the job satisfaction of fire officials would improve personalized factors and changes rational goal culture.

Production of Tropane Alkaloids by Two-stage Culture of Scopolia parviflora Nakai Adventitious Root

  • Scopolia parviflora Nakai, a rare and endangered species, is the sole plant producing tropane alkaloids (TA) among the Korean native species. In order to enhance TA productivity the SP72 root line was selected by screening 100 of root line, and the optimal culture media for root growth and TA production were investigated with the SP72 roots. Based on the several media, SH and 2B5 medium were determined as growth medium and White and NN medium as production medium. Among the four combinations of two-stage culture, 2BN (2B5 as growth medium plus NN as production medium) showed more enhanced root growth and TA production as compared with production media of White and NN medium and growth media of SH and 2B5 medium, respectively. However, bubble column bioreactor (BCB) cultures applying two-stage culture did not reveal the effective results despite of the each successful operation of two-stage culture in conical flasks and BCB cultures.

Control of Glucose Concentration in a Fed-Batch Cultivation of Scutellaria baicalensis G. Plant Cells a Self-Organizing Fuzzy Logic Controller

  • A self-organizing fuzzy logic controller using a genetic algorithm is described, which controlled the glucose concentration for the enhancement of flavonoid production in a fed-batch cultivation of Scutellaria baicalensis G. plant cells. The substrate feeding strategy in a fed-batch culture was to increase the flavonoid production by using the proposed kinetic model. For the two-stage culture, the substrate feeding strategy consisted of a first period with 28 g/I of glucose to promote cell growth, followed by a second period with 5 g/I of glucose to promote flavonoid production. A simple fuzzy logic controller and the self-organizing fuzzy logic controller using a genetic algorithm was constructed to control the glucose concentration in a fed-batch culture. The designed fuzzy logic controllers were applied to maintain the glucose concentration at given set-points of the two-stage culture in fed-batch cultivation. The experimental results showed that the self-organizing fuzzy logic controller improved the controller\`s performance, compared with that of the simple fuzzy logic controller. The specific production yield and productivity of flavonoids in the two-stage culture were higher than those in the batch culture.

한국전통누룩에서 분리한 유용곰팡이의 특성 (Characteristics of Useful Fungi Isolated from Traditional Korea Nuruk)

  • For the standardization and quality improvement of traditional Korean Nuruk, 120 strains of fungi were isolated from Nuruks and 18 strains of them were selected as strains analysing the amylase and flavor. The genera of these fungi were identified as Aspergillus sp. (14 strains), Penicillium sp. (3 strains) and Rhizopus sp. (1 strains) by the conventional slide culture. Most of these fungi showed a better productivity of the saccharogenic and dextrinogenic enzymes in raw wheat bran culture than in cooked wheat bran culture. The ability of acid and flavor production was good in the raw wheat bran culture, and aflatoxins were not produced under the same culture. Penicillium sp. and Rhizopus sp. had high cell growth and amylast activity. Mixed culture of Aspergillus sp. No.3-6 and Penicillium sp. No.7-7 revealed a high liquefying and saccharifying activity as well as high flavors production. These results indicated that these fungi was proper strains for making Nuruk of good quality.

임상현장에서의 긍정적인 간호조직문화 평가지표 개발 (Development of the Evaluation Indicators of Positive Nursing Organizational Culture in a Clinical Setting)

  • Purpose: The purpose of this study was to develop the evaluation indicators of positive nursing organizational culture in a clinical setting. Methods: The evaluation indicators of positive nursing organizational culture were developed from a literature review and a focus group interview. The content validity testing was done using a clinical expert panel. The content utility testing was done using a survey questionnaire. Results: The evaluation indicators of positive nursing organizational culture consists of 88 indicators representing the eight domains with the 24 categories. The average scores in evaluation indicators of positive nursing culture included the importance (3.29 points in average), the potential for further utilization (3.14 points in average) and the current state of extension agency (2.80 points in average). Conclusion: The developed evaluation indicators can be applied to measure the nursing organizational culture, which would be the basic data to manage human resources effectively in a clinical setting.

  • To isolate ergot strains which are capable of growing and producing alkaloids in submerged culture, strains were isolated from the sclerotia parasitizing the Graminae plants in Korea and the experiments of submerged culture of these strains yielded the following results: 1) The mycelia which were respectively isolated from the sclerotia parasitizing Agropyron semicostatum $N_{EES}$, Arundinella hirta $T_{ANAKA}$ var. ciliata $K_{OIDSUMI}$, Ischaemum anthephoroides $M_{IQ}$. var. eriostachyum $H_{ONDA}$, and Phleum pretense L. grew in Medium D by submerged culture. 2) When the strain of the ergot of Agropyron semicostatum was inoculated into six different nutrient solutions for submerged culture, its mycelium grew well in Media C, D and F, but produced alkaloids only in Medium C, indicating that Medium C is relatively suitable for the strain. 3) The extraction of the alkaloids from the culture broth by ether and T.L.C. analysis of the extract showed that it contained at least two types of alkaloids.

  • 본 실험은 여름딸기 무병묘 대량증식을 위해 bioreactor배양의 증식 및 경제성 효과를 비교하고자 실시하였다. 배양 방법은 반고체, 고체, 현탁배양 및 bioreactor 배양 등 4가지 방법을 이용하였다. 배양 6주 후, 식물체의 초장은 고체배양이 3.6cm로 가장 짧았으며, bioreactor 배양이 8.3cm로 가장 길었다. 생체중과 건물중은 bioreactor 배양이 2,261mg과 525mg으로 다른 배양방법에 비하여 가장 무거웠다. 액아는 반고체, 고체 및 현탁배양은 거의 발생하지 않았으나, bioreactor 배양은 주당 7개의 액아가 발생하였다. 경제성 분석 결과 기본식물 생산 시 bioreactor배양이 303원/주으로 고체배양의 845원/주보다 542원/주 적었다. 따라서 딸기 무병묘 생산 시 bioreactor배양이 대량증식 및 경제적인 면에서 효율적이었다.

간호사가 지각하는 간호조직문화와 조직몰입간의 관계 분석 (Analysis of Relationship between Nursing Organization Culture and Organizational Commitment)

  • Nursing organization culture is common patterns of believing, thinking and behaving resulted from a variety of experiences and interaction nurses in the same setting. This study was performed based on the assumption of existence of different subcultures within meta-culture, to identify the differences of subculture among 5 nursing units and to analyze the relationship between nursing organization culture and organizational commitment In this study, two kinds of instruments were used. One was the instrument of nursing organization culture developed by researcher through literature review and interview with nurses. The other one was Mowday's Organizational Commitment Questionnaire to measure organizational commitment of nurses. Both of them were structured with 5 scale. The answers were analyzed using SPSS program. The results were as follows : The meta-culture of the nursing organization culture was the people stable culture. There were significant differences in people development culture and people stable culture among 5 nursing units and all 5 units had different culture score. Especially, emergency room had strong development culture, and stable culture was dominant in operating room. Other units except emergency room and operating room had high scores in people stable culture than other culture types, but revealed different distribution. There were significant differences of the nursing organizational culture types -people development, task development, people stable, task stable-among 5 units. Four types of nursing organizational culture consisted of competing values in one organization. Nurses's organizational commitments were sign ificantly different among the units. The score per ceived by nurses who work in emergency room, operating room and leu was higher commitment score than that of medical/surgical nursing units Nurse's commitment to nursing organization was also related to total work period as nurse in any setting and work period in this hospital. Organizational commitment was significantly different among the nursing culture types, indicating that the scores of developmental culture were higher than stable culture. In conclusion, there were many different subcultures in nursing organization. In subculture, the organizational commitment was different. Therefore, the change of nursing organization culture or nursing unit culture needs to be considered to hire, give orientation, teach. and reallocate nurses efficiently. Research on nursing organization culture using both qualitative and quantitative method needs to be further considered. Furthermore, the strategy in nursing organization culture for nursing administrator to manage human resources efficiently and to change nursing unit effectively, needs to be developed.

Effect of Osmolarity of Culture Medium on the Preimplantation Development of Porcine NT and IVF Embryos

  • In vitro development of porcine embryo is affected by culture condition. One possible factor is osmolarity of culture medium. 1his study examined whether high osmolarity of culture medium at the early culture stage improves development of preimplantation porcine in vitro fertilization (IVF) and nuclear transfer (NT) embryos. NT and IVF embryos were divided into three groups and the basic medium was PZM-3 ($250{\sim}270$ mOsmol, control group). The control group of embryos was cultured in PZM-3 for whole culture period. Other two groups of embryos were cultured in a modified PZM-3 with 0.05 M sorbitol or 0.05 M sucrose ($300{\sim}320$ mOsmol, sorbitol or sucrose group) for the first 2 days, and then cultured in PZM-3 for further culture. NT embryos cultured in sucrose group showed a significantly higher developmental rate to the blastocyst stage with a decreased apoptosis rate compared to the sorbitol (p<0.05). For IVF, sucrose group showed a significantly increased the blastocyst formation rate with a decreased apoptosis rate compared to the control (p<0.05). This study represents that the high osmolarity in the early embryo culture stage can enhance the in vitro development of porcine NT and IVF embryos to the blastocyst stage with reduced apoptosis of cells.
