The Anti-Cancer Effect of Apamin in Bee-Venom on Melanoma cell line SK-MEL-2 and Inhibitory Effect on the MAP-Kinase Signal Pathway (약침용(藥鍼用) 봉독성분(蜂毒成分) 중(中) Apamin의 항암효과(抗癌效果)와 MAP-Kinase 신호전달체계에 관한 연구(硏究))

  • Objective : To characterize the antitumorigenic potential of Apamin, one of the major components of bee venom, its effects on cell proliferation and the mitogen-activated protein kinase (MAPK) signal transduction pathway were characterized using the human melanoma cell line SK-MEL-2. Methods & Results : Cell counting analysis for cell death demonstrated that consistent with a previous results, SK-MEL-2 cells treated with $0.5-2.0{\mu}g/ml$ of Apamin showed no recognizable cytotoxic effect whereas detectable induction of cell death was identified at concentrations over $5.0{\mu}g/ml$. [3H]thymidine incorporation assay for cell proliferation demonstrated that DNA replication of SK-MEL-2 cells is inhibited by Apamin in a dose- and time-dependent manner. To explore whether Apamin-induced growth suppression is associated with the MAPK signaling pathway, phosphorylation of Erk, a function mediator of MAPK growth-stimulating signal, was examined Western blot assay using a phospho-specific Erkl/2 antibody. A significant increase of Erkl/2 phosphorylation level was observed in Apamin-treated cells compared with untreated control cells. Qantitative RT-PCR analysis revealed that Apamin inhibit expression of MAPK downstream genes such as c-Jun, c-Fos, and cyclin D1 but not expression of MAPK pathway component genes including Ha-Ras, c-Raf-1, MEK1, and Erk. Conclusion : It is strongly suggested that the antitumorigenic activity of Apamin might result in part from its inhibitory effect on the MAPK signaling pathway in human melanoma cells SK-MEL-2.

Development of the Hydraulic Performance Graph Model and its Application (수리거동곡선 모형의 개발 및 적용)

  • This paper presents a hydraulic performance graph model in which the flow carrying capacity of a channel system was determined by accounting the interacting backwater effect among channel reaches and incoming lateral flow. The method utilizes hydraulic performance graphs (HPGs), and the method is applied to a natural channel Nakdong River to examine its applicability. This research shows that estimation results using HPG are close to records from the stage station and the results from a widely-accepted model, HEC-RAS. Assuming that a water level gage site is ungaged, water level estimations by HPGs compared with observation show that with a flood event, the HPGs underestimate in the water level ascension phase, but in the recession phase they overestimate results. The accuracy of estimation with HPGs was greatly improved by considering the time difference of flooding between the observation and estimation locations.

  • FGF9/16/20 signaling pathway specify the developmental fates of notochord, mesenchyme, and neural cells in ascidian embryos. Although a conserved Ras/MEK/Erk/Ets pathway is known to be involved in this signaling, the detailed mechanisms of regulation of FGF signaling pathway have remained largely elusive. In this study, we have isolated Hr-Erf, an ascidian orthologue of vertebrate Erf, to elucidate interactions of transcription factors involved in FGF signaling of the ascidian embryo. The Hr-Erf cDNA encompassed 3110 nucleotides including sequence encoded a predicted polypeptide of 760 amino acids. The polypeptide had the Ets DNA-binding domain in its N-terminal region. In adult animals, Hr-Erf mRNA was predominantly detected in muscle, and at lower levels in ganglion, gills, gonad, hepatopancreas, and stomach by quantitative real-time PCR (QPCR) method. During embryogenesis, Hr-Erf mRNA was detected from eggs to early developmental stage embryos, whereas the transcript levels were decreased after neurula stage. Similar to the QPCR results, maternal transcripts of Hr-Erf was detected in the fertilized eggs by whole-mount in situ hybridization. Maternal mRNA of Hr-Erf was gradually lost from the neurula stage. Zygotic expression of Hr-Erf started in most blastomeres at the 8-cell stage. At gastrula stage, Hr-Erf was specifically expressed in the precursor cells of brain and mesenchyme. When MEK inhibitor was treated, embryos resulted in loss of Hr-Erf expression in mesenchyme cells, and in excess of Hr-Erf in a-line neural cells. These results suggest that zygotic Hr-Erf products are involved in specification of mesenchyme and neural cells.

MGMT-B Gene Promoter Hypermethylation in Patients with Inflammatory Bowel Disease - A Novel Finding

  • Inflammatory bowel disease (IBD) is a disease strongly associated with colorectal cancer (CRC) as a well-known precancerous condition. Alterations in DNA methylation and mutation in K-ras are believed to play an early etiopathogenic role in CRC and may also an initiating event through deregulation of molecular signaling. Epigenetic silencing of APC and SFRP2 in the WNT signaling pathway may also be involved in IBD-CRC. The role of aberrant DNA methylation in precancerous state of colorectal cancer (CRC) is under intensive investigation worldwide. The aim of this study was to investigate the status of promoter methylation of MGMT-B, APC1A and SFRP2 genes, in inflamed and normal colon tissues of patients with IBD compared with control normal tissues. A total of 52 IBD tissues as well as corresponding normal tissues and 30 samples from healthy participants were obtained. We determined promoter methylation status of MGMT-B, SFRP2 and APC1A genes by chemical treatment with sodium bisulfite and subsequent MSP. The most frequently methylated locus was MGMT-B (71%; 34 of 48), followed by SFRP2 (66.6 %; 32 of 48), and APC1A (43.7%; 21 of 48). Our study demonstrated for the first time that hypermethylation of the MGMT-B and the SFRP2 gene promoter regions might be involved in IBD development. Methylation of MGMT-B and SFRP2 in IBD patients may provide a method for early detection of IBD-associated neoplasia.

  • The modern times is as special as from the river, the river is very important of our life. The importance to preserve the river environment has been issued and the river developing method is being changed to use potential function of nature as well as flood control. Essential element of the river restoration is a vegetation. The flow resistance by vegetation along the river banks is greatly increase the flood stage. Therefore, the flow resistance due to vegetation in the river and roughness coefficient changes to understand the hydraulic characteristics is an important elements in the river restoration. The purpose of this study is to analyze the flood stage and the aspects of riverbed changes due to the corridor restoration with river vegetation. In order to simulate the flood stage and riverbed changes, HEC-RAS, RMA-2, and SED-2D model were applied for the upstream and downstream in study reaches, respectively.

  • In recent years, the damage caused by flood has been gradually increased. The cause of the flood is unusual weather condition as well as topographical and geographical condition of our country. Flood area analyzing vulnerable flood areas on large regions without previous flood records were developed using GIS and hydraulic model, flood depth by return periods and topographical data such as DEM. The study area is the Munsan river basin. The results using GIS comparing the results using hydraulic model. The results in this study can be used to delineate the potential flooding areas in large regions and the damage from disasters can be reduced by making provisions with the obtained results.

Analysis of Flow Characteristics in Upstream Channel depending on Water Gate Operation of Nakdan Multi Functional Weir (수문운영에 따른 낙단보 상류하도 흐름특성 해석)

  • This study, examines the flow characteristics of upstream channel depending on water gate operation of Nakdan Multi-fuctional weir. The specific purpose of this study are to simulate the variation of flow velocity depending on the operation of the weir using 1-dimensional hydraulic model, HEC-RAS, and compare it with observed velocity. For discharge conditions from $50m^3/s$ to $3,500m^3/s$, it is observed that the velocity of upstream channel is almost constant, whereas for probability flood discharge, the velocity and froude number are increased as the discharge values are increased. The velocity values for downstream boundary condition EL, 40.0 m are more decreased than those for EL. 40.5m. From comparison on the variation of water stage depending on water gate operation, it is observed that the stage values are almost constant for discharges below $300m^3/s$, whereas 5 cm to 20 cm for discharges over $700m^3/s$. Flow velocity at streamflow gauging station. Nakdong, is decreased by more than 875% after installing the weir. The results obtained from this study indicate that the velocity of upstream channel is decreased and the discharge and velocity of downstream channel are significantly varied after installing the weir.

Decolorization of Blue-Stain by Dual Culture of Blue Staining and Basidial Fungi

  • This study was performed to understand the interaction between Ophiostomataceae and basidiomycetes fungi during cultures, and whether the basidiomycetes fungi inhibit the growth and decolorize dark pigments of blue staining fungi. The conjoint cultivation was studied on 2% malt extract agar. The ability of basidial cultures to decolorize dark pigments of ophiostomatoid fungi was the main characteristics estimated during this study. More than half of basidial cultures were characterized by deadlock interaction with blue staining fungi. In the dual cultures, where basidial partners were presented by Agaricus bisporus(64), Laetiporus sulphureus(L01/89), Trametes versicolor(09) and unknown fungus(02), antagonism was found at the phase of primary contact of colonies. Replacement interaction resulted usually in decreasing dark colour of substrate was observed for 11 basidial cultures that were belonging mainly to white-rot fungi. Among them Abortiporus biennis(123), Antrodiella hoehnelii(S28/91), Bjerkandera fumosa (137), and Gleophyllum odoratum(124) were characterized by the absence of deadlock-phase: they began to grow over dark colonies of their partners just after primary contact. Basidiomycetes did not affect strongly the pigments of Ceratocystis spp. and Leptographium sibirica isolates, but completely decolorized colonies of Ophiostoma ips and to a smaller degree Ophiostoma minus. Antrodiella hoehnelii(S28/91), Bjerkandera fumosa(137), Gleophyllum odoratum(124) and Trametes versicolor(B18/91) cultures were found to be the most active in decreasing dark color of blue staining fungi colonies. The cultures were recommended for further development as agents of biopulping of wood chips and bio-control of blue stain in woods.

Water Level Variation Analysis in the Cooling Water Discharge Channel of Power Plant due to Installation of Ocean Small Hydropower Plant (해양소수력 건설에 따른 방류수로의 수위 변화 특성 분석)

  • A small hydropower plant(SHP) using cooling water discharged from the power plant was constructed in Samcheonpo. This study presents predicted and measured hydrological data in the construction process of small hydropower plant in order to evaluate characteristics of water level variation of cooling water discharge channel which is a key factor in the design of SHP since the water level rise of channel is related to impact on circulating water system of the existing power plant. Various methods were applied for prediction of water level variation in the design stage from simple empirical formula to sophisticated 3-dimensional CFD method. Measured results reveal that mean value was similar between measured and predicted, but measured results were larger than predicted in deviation. Moreover, simple formula, i.e. standard weir equation and Honma equation, were more useful before installation of SHP, but sophisticated methods during operation of SHP.

Association Study Between the C3123A Polymorphism of the Angiotensin II Type 2 Receptor Gene in the Human X Chromosome and Essential Hypertension in Koreans (한국인에서 Angiotensin II Type 2 Receptor 유전자에 존재하는 C3123A 다형선과 본태성 고혈압과의 관련성에 관한 연구)

  • Renin-angiotensin system (RAS)은 혈압 조절에 중요한 역할을 수행하는 생리적 조절계로써, 이 system 을 구성하는 유전자들의 이상은 본태성 고혈압의 발병과 유의하게 관련된 것으로 알려졌다. RAS의 주요한 구성 성분인 angiotensin II는 2종류의 수용체인 angiotensin II type I receptor(AT₁R)와 angiotensin II type I receptor(AT₂R)에 의해 그 효과가 매개되기 때문에, 이 수용체를 암호하는 유전자는 본태성 고혈압의 유력한 후보 유전자라고 볼 수 있다. 현재가지의 연구에 의하면, AT₁R 유전자에 존재하는 유전적 변이와 본태성 고혈압과의 관련성에 관해서는 많은 보고들이 있었지만, AT₂R 유전자에 존재하는 유전적 변이 가 본태성 고혈압에 유의한 효과를 나타내는 지에 관해서는 이렇다할 연구 성과가 별로 없는 실정이다. 이에 본 연구에서는 한국인 집단을 대상으로 하여, AT₂R 유전자에 존재하는 C3123A 다형성이 한국인 집단에서 본태성 고혈압과 유의한 관련성이 있는 지를 분석하였다. 이 유전자는 인간의 X 염색체에 존재하기 때문에, 여성인 경우에는 CC, CA및 AA로 이루어진 3유전자형이 존재하지만, 남성인 경우에는 C와 A로 이루어진 2종류의 대립 유전자로 구성되어 있기 때문에, 본 연구에서는 남성과 여성을 개별적으로 나누어서 분석하였다. 연구 결과, AT₂R 유전자에 존재하는 C3123A 다형성은 남녀 모두에서 본태성 고혈압과 유의한 관련성을 나타내지 않았다(P>0.05). 그렇지만, 이 다형성에 대한 대립 유전자 빈도를 서양인 집단과 비교했을 경우에는, 한국인을 대상으로 한 본 연구에서 A 대립 유전자 빈도가 0.33인 반면에 서양인 집단은 그 빈도가 0.43~0.48로 한국인 집단보다 높은 값을 나타내었다. 따라서, AT₂R 유전자에 존재하는 C3123A 다형성과 본태성 고혈압과의 관련성에 대해서는 한국인과 유전적 배경이 다른 서양인 집단을 대상으로 한 추시가 필요할 것으로 사료된다.
