Studies on the Transformation of Crop Plants. IV. Biochemical Characteristics of Embryogenic Callus in Rice (곡물류의 형질전환 유도에 관한 연구 (IV) 벼 배발생 세포의 생화학적 특징)

  • Rice (Oryza saliva L.) calli containing both embryogenic callus (EC) and non embryogenic callus (NEC) regions were initiated from the mature seed on MS medium supplemented with 2.0 mg/L 2,4-D, 0.5 mg/L kinetin. The calli were developed into two callus type which can be distinguished by visual examination depending on color and appearance. In order to illucidate the polypeptide composition between EC and NEC, the total protein extracted from two types of callus was analysed by electrophoresis. By one-dimesional anlaysis of SDS-PAGE and Isoelectric focusing, several protein bands showed quantitative and qualitative difference in each type of callus. The further analysis of the total protein with two-dimensional electrophoresis showed at least 20 EC specific protein and 10 NE specific protein. Also 3 specific protein spots showing micro heterogeneity of 90, 65, 50 kD were detected in EC, while a series of acidic heterologous protein spots were visualized in NEC.in NEC.

  • Free vibration analysis of arbitrary quadrilateral thick plates with internal columns and elastic edge supports is presented by using the powerful pb-2 Ritz method and Reddy's third order shear deformation plate theory. The computing domain of arbitrary quadrilateral planform is mapped onto a standard square form by coordinate transformation. The versatile pb-2 Ritz functions defined by the product of a two-dimensional polynomial and a basic function are taken to be the admissible functions. Substituting these displacement functions into the energy functional and minimizing the total energy by differentiation, leads to a typical eigenvalue problem, which is solved by a standard eigenvalue solver. Stiffness and mass matrices are numerically integrated over the plate by using Gaussian quadrature. The accuracy and efficiency of the proposed method are demonstrated through several numerical examples by comparison and convergency studies. A lot of numerical results for reasonable natural frequency parameters of quadrilateral plates with different combinations of elastic boundary conditions and column supports at any locations are presented, which can be used as a benchmark for future studies in this area.

  • In this study, it was investigated whether immortalized proximal tubule cells transformed with pRSVT could survive through the numerous passages. Results were as follows: 1. The cells transfected with pRSVT formed rapidly growing, multilayered colonies within 2 weeks in a hormone defined medium. Domes were also observed in some of the cultures. 2. r-glutamyl transpeptidase activity was equivalent to that observed in primary renal proximal tubule cell cultures. 3. Transformed cells with pRSVT form tubules in matrigel following 20 passages. 4. Genomic DNA of transformants was digested with either the restriction enzyme Xba or BamH1. A band of approximately 7.5kb was detected with Xba. Three BamH1 bands were detected at approximately 15 kb, 6.5 kb, and 3 kb.

  • We have obtained fertile transgenic rice plants resistant to the broad spectrum herbicide Bast $a^{(R)}$ (active ingredient phosphinothricin, PPT) by PEG-mediated transformation procedure. The plasmid pCaMV35S::Bar was used to deliver the bar gene into embryogenic suspension culture-derived protoplasts of rice (Oryza sativa L.). Transformed plants were regenerated and selected on medium containing 15 mg/l of phosphinothricin. Stable integration and expression of the bar gene in transgenic rice plants was confirmed by Southern and Northern blot analysis. Transgenic $R_1$ plants were also confirmed by assays for phosphinothricin acetyltransferase (PAT) activity. The bar gene was expressed in the primary transgenic rice plants and in the next generation progeny, in which it showed a 3 : 1 Mendelian inheritance pattern. Transgenic $R_1$ and $R_2$ plants were resistant to the herbicide Bast $a^{(R)}$ when sprayed at rates used in field practice.ice.

  • Protoplasts isolated from embryogenic cell suspensions were electroporated in buffered solutions containing plasmid DNA of pBI121. Transient GUS (beta-glucuronidase) activity measurement and selection for kanamycin resistent showed that expression of foreign genes and stable loransformation were achieved. GUS transient gene expression was increased by increasing DNA concentration of pBI121 plasmid and affected by the level of the applied voltage. An optimal level of GUS activity was obtained after electroporation with a pulse of 200 voltage/1180 uF. Protoplast viability was up to the 60% at the optimal voltage. Cell colonies resistent to 200$\mu\textrm{g}$/ml kanamycin were selected in agar medium and identified by histochemical GUS assay.

  • 급냉온도에 따른 전기 저항 측정으로 Cu-17, 25Zn-15AI 및 Cu-17.25Zn-15AI-1Ag형상기억합금의 열처리에 의한 마르텐사이트 변태온도의 영향을 연구하였다. DSC 측정으로 고온 모상에서의 상전이 온도롸 종류를 구별하였고 XRD측정으로 구조 변화를 연구하였다. 그리고 열처리에 의한 온도 변화의 원인을 연구하였17.25Zn-15AI 합금에서 고온 모상의 규칙-불규칙 전이온도인 $T_{2}$, $T_{L21}$은 각각 809K와 610K였다. CuZnAI의 경우 $T_{2}$근방에서의 급냉은 마르텐사이트 변태온도를 높이지만 $T_{L21}$ 근방에서의 급냉은 마르텐사이트 변태온도를 낮춘다. 실험결과 열처리에 따른 상전이온도 변화의 원인은 석출물의 형성이라기 보다는 급냉전의 모상의 구조에 가장 큰 영향을 받는다.받는다.

  • The occurrence of multidrug resistance (MDR) is one of the main obstacles in the successful chemotherapeutic treatment of cancer. In this study The gene expressions of multidrug resistance-associated protein (MRP), c-myc and c-fos were investigated in L1210 cells. Adriamycin- or vincristine-resistant L1210 cells, L1210AdR or L1210VcR, respectively, has been identified to overexpression of mdr1 gene. The expression leve of MRP gene in L1210AdR and L1210Cis was more decreased than that in L1210 cells. The c-myc and c-fos genes were expressed both in L1210 and resistant sublines. In L1210AdR, the expressions level of c-myc and c-fos genes were decreased than in L1210. However, in L1210VcR and L1210Cis, c-myc and c-fosgene expressionwere rather increased than L1210. These results suggested that MRP does not contribute in resistance of drug-resistant L1210 cells and there is no relations between MRP and mdr1 gene expression. The expression of c-myc and c-fos gene may be changed during transformation of L1210 to drug-resistant sublines.

  • Salt-tolerant transgenic Panax ginseng plants were produced by introducing the SAL1 geue (3'(2'), 5'-bis-phosphate nucleotidase) that confers tolerance to the salts through Agrobacterium tumefaciens co-cultivation. Cotyledon explants of immature ginseng zygotic embryos cultured on Murashige and Skoog medium lacking growth regulators formed somatic embryos directly with below 10%, but the 74% tranformation rate were observed at the treatment of phytohormone with 1.0 mg/l 2,4-D and 0.5 mg/l kinetin. Somatic embryos were initially cultured on MS medium supplemented with 250 mg/l cefotaxime for 3 weeks and subsequently subcultured five times to a medium containing 100 mg/l kanamycin and 250 mg/l cefotaxime. Upon development into the cotyledonary stage, these somatic embryos were transferred to on the medium containing 50 mg/l kanamycin and 10 mg/l gibberellic acid to induce germination and strong selection. Integration of the transgene into the plants was confirmed by polymerase chain reaction with specific primers. The ginseng transformants with well-developed shoots and roots were successfully acclimatized in a greenhouse when they were planted in soil.
