Development of Analytical Method for Fenoxycarb, Pyriproxyfen and Methoprene Residues in Agricultural Commodities Using HPLC-UVD/MS (HPLC-UVD/MS를 이용한 농산물 중 fenoxycarb, pyriproxyfen 및 methoprene의 분석법 확립)

  • Fenoxycarb, pyriproxyfen and methoprene are juvenile hormone mimic insecticide. These insecticides have been widely used for mosquito, fly, scale insects, and Lepidoptera. The purpose of this study was to develop a simultaneous determination procedure of fenoxycarb, pyriproxyfen and methoprene residues in crops using HPLC-UVD/MS. These insecticide residues were extracted with acetone from representative samples of four raw products which comprised brown rice, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and then n-hexane/dichloromethane partition was followed to recover these insecticides from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The analytes were quantitated by HPLC-UVD/MS, using a $C_{18}$ column. The crops were fortified with each insecticide at 3 levels per crop. Mean recovery ratios were ranged from 80.0 to 104.3% in four representative agricultural commodities. The coefficients of variation were less than 4.8%. Quantitative limit of fenoxycarb, pyriproxyfen, and methoprene was 0.04 mg/kg in crop samples. A HPLC-UVD/MS with selected-ion monitoring was also provided to confirm the suspected residues. The proposed simultaneous analysis method was reproducible and sensitive enough to determine the residues of fenoxycarb, pyriproxyfen and methoprene in the agricultural commodities.

Egg Development and Morphological Change of Larvae and Juveniles, Characteristics of Spawning Sites of the Korean Endemic Species, Coreoleuciscus splendidus (Cypriniformes: Gobioninae) in the Jo-jong Stream, Korea (조종천에 서식하는 한국고유종 쉬리 Coreoleuciscus splendidus (Cypriniformes: Gobioninae)의 난발생, 자치어 형태 발달 및 자연산란장 연구)

  • The egg development, early life history and spawning site characters of Korean endemic fish, Coreoleuciscus splendidus (Gobioninae), were investigated at the part of Jo-jong stream in Korea from May 2020. The fertilized eggs were 2.05~2.23 mm (mean, 2.13 mm) in diameter and had no oil globules. The embryo began to hatching about 98 hrs after fertilization under water temperature of 20±1℃. The newly-hatched larvae were 5.03~5.68 mm (mean, 5.31 mm) in total length (TL), and their mouth and anus were not opened. 4 days after hatching, Several rod-like cupulae were observed on the head and lateral side of the body at 6.95~7.89 mm (mean, 7.51 mm). 7 days after hatching, the post-larva stage were 8.39~9.29 mm (mean, 8.78 mm) in total length, and their york were completely absorbed. The cupulae were completely distinguished. They entered the juvenile stage when all fin-rays were formed at 29 days after hatching, and their TL were 14.16~17.04 mm (mean, 14.99 mm). Squamation was initiated on the caudal body at approximately 18.21~23.74 mm (mean, 30.28 mm), 38 days after hatching, and completed at 26.82~33.33 mm (mean, 19.42 mm), 73 days after hatching, the external characteristics from of juveniles were same to adults. The spawning site was characterized by bottom structure of pebble (64~16 mm) and gravel (16~2 mm), environmental conditions of the spawning sites were 6~18 cm in water depth, 0.43~0.73 m/sec in bottom water velocity. In spawning sites, an egg mass or separated eggs were located a small gap of under the pebble and gravel.

Development of Eggs, Larvae and Juveniles of the Ladislavia taczanowskii from Hongcheongang-River (홍천강에 서식하는 멸종위기종 새미 Ladislavia taczanowskii의 난발생 및 자치어 형태발달)

  • This study was conducted to investigate the early life history by observing the egg development of Ladislavia taczanowskii in endangered fish and to use it as basic data for species conservation research. The broodstork used in the study was secured from the area of the Hongcheon River in Hongcheon-gun, Gangwon State. The broodstork, who was being raised in the laboratory, selected mature individuals in May 2021 and induced them to spawn by hormone injection. The size of the maturation egg was 1.50~1.79 (average 1.59±0.08, n=30) mm due to the circular invasive egg. The incubation time took 168 hours at 16.5℃ and 109 hours and 30 minutes at 25.5℃. Newly hatched larvae, the consonants had a total length of 5.55~6.31 mm (6.30±6.93, n=30) mm, and the mouth and anus did not open and had egg yolk. 5 days after hatching, the preflexion larvae had a total length of 9.91~10.8 (10.1±0.27, n=30) mm, and the mouth and anus opened, and feeding activities began. 8 days after hatching, the flexion larvae had a total length of 10.3~11.4 (10.8±0.38, n=30) mm, and the end of the vertebrae at the tail fin tip began to bend upward. 10 day after hatching, the postflexion larvae had a total length of 11.8~13.1 (12.3±0.43, n=30) mm, and the end of the vertebrae at the tail tip was completely bent at 45°. 18 days after hatching, the total length of the juveniles was 18.9~23.4 (20.4±1.69, n=30) mm, and the number of fins in each part was fin rays with 10 dorsal fins, 9 anal fins, 22 caudal fins, and 7 ventral fins. As a result of the study, the postflexion larvae showed differences in morphology from other Gobioninae fishes in the upper part of the tail's hypural, the shape of spots on the dorsal vertebrae, the vertical stripes developed on the head, and the irregularly deposited melanophore throughout the body.

  • Life cycle and seed production of the freshwater prawn, Macrobrachium nipponense, were studied and the results are as follows : 1. Larval development : Embryos hatched out as zoea larvae of 2.06 mm in mean body length. The larvae passed through 9 zoea stages in $15{\~}20$ days and then metamorphosed into postlarvae measuring 5.68 mm in mean body length. Each zoea stage can be identified based on the shapes of the first and second antennae, exo- and endopodites of the first and second pereiopods, telson and maxillae. 2. Environmental requirements of zoea larvae : Zoea larvae grew healthy when fed with Artemia nauplii. Metamorphosing rate was $65{\~}72{\%}$ at $26{\~}28\%$ and $7.85{\~}8.28\%_{\circ}Cl.$. The relationship between the zoeal period (Y in days) and water temperature (X in $^{\circ}C$) is expressed as Y=46.0900-0.9673X. Zoeas showed best survival in a water temperature range of $26{\~}32^{\circ}C$ (optimum temperature $28^{\circ}C$), at which the metamorphosing rate into postlarvae was $54{\~}72\%$ The zoeas survived more successfully in chlorinity range of $4.12{\~}14.08{\%_{\circ}}Cl.$, (optimum chlorinity $7.6{\~}11.6\;{\%_{\circ}}Cl.$.), at which the metamorphosing rate was $42{\~}76{\%}$. The whole zoeal stages tended to be longer in proportion as the chlorinity deviated from the optimum range and particularly toward high chlorinity. Zoeas at all stages could not tolerate in the freshwater. 3. Environmental requirements of postlarvae and juveniles : Postlarvae showed normal growth at water temperatures between $24{\~}32^{\circ}C$ (optimun temperature $26{\~}28^{\circ}$. The survival rate up to the juvenile stage was $41{\~}63{\%}$. Water temperatures below $24^{\circ}C$ and above $32^{\circ}$ resulted in lower growth, and postlarvae scarcely grew at below $17^{\circ}C$. Cannibalism tended to occur more frequently under optimum range of temperatures. The range of chlorinity for normal growth of postlarvae and juveniles was from 0.00 (freshwater) to $11.24{\%_{\circ}}Cl.$, at which the survival rate was $32{\~}35\%$. The postlarvae grew more successfully in low chlorinities, and the best growth was found at $0.00\~2.21{\%_{\circ}}Cl.$. The postlarvae and juveniles showed better growth in freshwater but did not survive in normal sea water. 4. Feeding effect of diet on zoea Ilarvae : Zoea larvae were successfully survived and metamorposed into postlarvae when fed commercial artificial plankton, rotifers, and Artemia nauplii in the aquaria. However, the zoea larvae that were fed Artemia nauplii and reared in Chlorella mixed green water showed better results. The rate of metamorphosis was $68\~{\%}75$. The larvae fed cow live powder, egg powder, and Chlorella alone did not survive. 5. Diets of postlarvae, juveniles and adults : Artemia nauplii and/or copepods were good food for postlarvae. Juveniles and adults were successfully fed fish or shellfish flesh, annelids, corn grain, pelleted feed along with viscera of domestic animals or fruits. 6. Growth of postlarvae, juveniles and adults : Under favorable conditions, postlarvae molted every five or six days and attained to the juvenile stage within two months and they reached 1.78 cm in body length and 0.17 g in body weight. The juveniles grew to 3.52 cm in body length and 1.07 g in body weight in about four months. Their sexes became determinable based on the appearance of male's rudimental processes (a secondary sex character) on the endopodites of second pereiopods of males. The males commonly reached sexual maturity in seven months after attaining the postlarvae stage and they grew to 5.65 cm in body length and 3.41 g in body weight. Whereas the females attained sexual maturity within six to seven months, when they measured 4.93 cm in body length and 2.43 g in body weight. Nine or ten months after hatching, the males grew $6.62{\~}7.14$ cm in body length and $6.68{\~}8.36$ g in body weight, while females became $5.58{\~}6.08$ cm and $4.04{\~}5.54$ g. 7. Stocking density : The maximum stocking density in aquaria for successful survival and growth was $60{\~}100$ individuals/$\ell$ for zoeas in 30-days rearing (survival rate to postlarvae, $73{\~}80{\%}$) ; $100{\~}300$ individuals/$m^2$ for postlarvae of 0.57 cm in body length (survival rate for 120 days, $78{\~}85{\%}$) ; $40{\~}60$ individuals/$m^2$ for juveniles of 2.72 cm in body length (survival rate for 120 days, $63{\~}90{\%}$) : $20{\~}40$ individuals/$m^2$ for young prawns of 5.2 cm in body length (survival rate for 120 days, $62\~90{\%}$) ; and $10\~30$ individuals/$m^2$ for adults of 6.1 cm in body length (survival rate for 60 days, $73\~100{\%}$). The stocking density of juveniles, youngs and adults could be increased up to twice by providing shelters.

Effects of Low Salinity acclimation on Oxygen Consumption in Giant Grouper, Epinephelus lanceolatus (염분변화가 대왕바리, Epinephelus lanceolatus의 산소소비에 미치는 영향)

  • Changes of oxygen consumption in juvenile giant grouper, Epinephelus lanceolatus were investigated in order to find out the physiological responses associated with freshwater and low salinities acclimation. Salinity changes in this experiment were set as follows; 35 psu (Exp. I) as control, decrease in the manners of $35{\rightarrow}20{\rightarrow}10{\rightarrow}0$ psu (Exp. II), $35{\rightarrow}10{\rightarrow}0$ psu (Exp. III) and $35{\rightarrow}0$ psu (Exp. IV). The average oxygen consumption at $27^{\circ}C$ in Exp. I (control) was $106.9{\pm}0.7mg$ $O_2/kg/h$. In Exp. II, according to the changes of $35{\rightarrow}20{\rightarrow}10{\rightarrow}0$ psu in freshwater acclimation, fish showed the gradual decrease in oxygen consumption as $108.1{\pm}2.1$, $99.6{\pm}2.9$, $74.6{\pm}0.9$ and $62.7{\pm}1.0mg$ $O_2/kg/h$, respectively. The fish in Exp. III also revealed the same tendency as $106.5{\pm}1.3$, $77.0{\pm}1.2$ and $64.7{\pm}1.2mg$ $O_2/kg/h$, respectively. In Exp. IV, fish showed the decreased oxygen consumption from $109.0{\pm}0.9mg$ $O_2/kg/h$ of 35 psu to $71.6{\pm}2.0mg$ $O_2/kg/h$ of 0 psu. Giant grouper is thought to be a diurnal fish species, because the level of oxygen consumption during light period was higher than that during dark period.

Early Life History and Spawning Behavior of the Gobiid Fish, Mugilogobius abei (Jordan et Snyder) Reared in the Laboratory (모치망둑, Mugilogobius abei(Jordan et Snyder)의 산란행동(産卵行動)및 초기생활사(初期生活史))

  • Spawning behavior and development of eggs and larvae of Mugilogobius abei were observed in the laboratory at Pusan, Korea. The adult male of Mugilogobius abei was observed making nest-like spawning-bed to lay eggs and showing territorial and courtship behaviors. The eggs were transparent and spherical in shape, measuring 0.40~0.50 mm in diameter. They have a bundle of adhesive filaments at their basal end and a cluster of small oil globules. The eggs became ellipsoid shape after the insemination and measured about 0.93~0.96 mm on the long axis. Hatching began about 110 hours after fertilization at water temperature of $24.5{\sim}25.5^{\circ}C$. The newly hatched larvae were 2.04~2.10 mm in total length, with 24~25(8~9+16) myomeres. Many melanophore and guanophore are distributed on eye cups, gas bladder, optic vesicle and the caudal region. Four days after hatching the yolk and oil-globule were completely absorbed and the larvae attained a total length 2.20~2.35 mm. The larvae swam actively in the aquarium and start to practice feeding on the rotifer. Twelve days after hatching, the larvae averaged 3.20 mm in TL and the caudal notochord flex at $45^{\circ}$. Rudimental second dorsal, anal, caudal and ventral fins are also formed. The larvae attained 10.40~10.80 mm in TL, 35 days after hatching, are found to start the bottom-life after having completely formed first dorsal and ventral fins. The larvae reached the juvenile stage at 50~60 days after hatching and attained 15.37~20.25 mm in TL. At this period all scales appeared on the body.

가정 폭력 경험이 남자 범죄 청소년의 남성성에 미치는 영향에 관한 연구

  • This exploratory qualitative study investigates the effects of experiencing domestic violence on male adolescent offenders' masculinities. Empirical and theoretical literature suggests that negative male role models in violent families result in male adolescents' experiencing conflict in constructing gender identities, especially masculinities. Moreover. criminologists argue that masculinities are often connected with crimes as a way to prove masculine competence. This study compares male adolescent offenders who have experienced domestic violence with those who have not experienced domestic violence and explores how domestic violence experiences influence the construction of gender identities among male adolescent offenders. The study used a secondary qualitative data analysis method. The data consisted of ethnographic in-depth interview transcripts, observational field notes, and formal facility records collected at a juvenile correctional facility in Minnesota. The process of data analysis was a "constant comparative method" that sought to understand differences and similarities in the expressed gender narratives and identity patterns between the two groups of offenders. This process also examined differences within each group. The qualitative data analysis revealed that domestic violence experiences in childhood may be related to the construction of gender identities during adolescence. The findings of this study showed that male adolescent offenders who had experienced domestic violence tended to attach themselves to oppressed mothers more readily than those who had not experienced domestic violence. Next, their attachment to mothers related to the construction of more relational gender identities although most participants, regardless of domestic violence experiences, had much in common regarding gender expression. Finally, despite these relational gender identities, male adolescent offenders who had experienced domestic violence tended to depend upon violence and crimes to show masculine competence, as did male adolescent offenders who had not experienced domestic violence. The study findings suggest a need for research to understand the construction of gender identities in the context of particular experiences and the importance of building theories that advance a comprehensive understanding of the construction of masculinities and youth crime. This study also discusses the development of social work programs that protect young men from adherence to exaggerated masculinity, which is often associated with crimes.

Knowledge, Attitude & Practice for Sexuality of Teachers -Based on over 50 year old teachers- (교사의 성 지식.태도.실태 연구 -50세 이상 교사를 중심으로-)

  • To correctly plant the value of sex to the juvenile, we cannot overemphasize the importance of the sexual education in schools. Therefore, the study for teachers' consciousness and view of sex is also necessary, because they are subjects of education and role-models of sex of their students. Especially, if teachers are over fifty in age, it is more important, because it is obvious, as they are in positions of managers and directors of education, that their view of sexual value and consciousness are influencing the aim of education and the course of education. Hereby this study was conducted to check their sexual consciousness, attitude, and condition and to prepare for the basic data needed for the development of a sexual education program suitable to them. The methods of the survey of this study are applied by modifying or supplementing those of precedented studies. They are used after analyzing reliance of items according to Cronbach's $\alpha$ figure calculating law, and modifying or supplementing items lower in reliance. The questionnaire survey was conducted among 1,000 randomly selected from teachers working in middle and high schools in Korea from June 14th in 2004 to June 30th. The survey analyzed 632 questionnaires suitable to it. The outcomes of the survey of this study are like the following: First, the objects of the survey consist of 64.6% men and 35.4% women. Among them, 41.1% are under fifty and the rest are over fifty. As for the periods of marriage, 15.5% are unmarried, 16.3% are under ten years, 18.0% are between ten and twenty, and 50.2% are over twenty. Second, with reagard to sex, 25.6% admit the necessity of a heterosexual friend. 32.0% say that they are conservative to sex. 54.1% are taught sexual education. 67.1% attribute sexual education to nursing teachers. Third, among those who answer that they enoughly know the concept of sex, seenig by age, 25.0% are under fifty and 58.1% are over fifty. Seening by the periods of marriage, 9.2% are unmarried, 28.2% are under ten, 49.1% are between ten and twenty, and 59.0% are over twenty. Among those who answer that they enoughly know the concepts of sexual harassment and sexual violence, seenig by age, in the turns by above-mentioned order, each 47.7% and 76.3%. Seening by the periods of marriage, each 9.2%, 28.2%, 49.1%, and 59.0%. Fourth, among those who answer that "teachers also need sexual education and sexual counsel," by age, each92.7% and 91.4%. By the periods of marriage, each 89.8%, 95.2%, 89.4%, and 92.4%. As a matter of course, we can infer the necessity of the sexual education and sexual counsel for teachers. Fifth, among those who answer that they are satisfied with their sexuality, by age, each 41.1% and 61.3%. By the period of marriage, each 4.1%, 63.1%, 64.1%, and 61.5%. As for the sexual desire like spiritual intercourse, physical intercourse containing sexuality, by age, each 71.9% and 93.5%. By the periods of marriage, each 54.0%, 81.5%, 90.3%, and 93.0%. The survey shows that those over fifty and having long marriage are feeling the stronger desire towards sexuality. Through the outcomes of this survey, a few suggestions are possible: First, it is necessary to check of the teachers' view of sexual value and to study deep about the sexual tendency of them in twenties, thirties, and forties. It is to make them teach their students on the firm basis, and also to develop suitable sexual education and counsel program.

A Study on the Growth Environment and Tissue Culture of Gyrophora esculanta MIYOSHI in Korea (한국산(韓國産) 석이(石耳)의 생육환경(生育環境)과 조직배양(組織培養)에 관(關)한 연구(硏究))

  • The objectives of this study were to investigate the growth of Gyrophora esculanta and to establish a method of tissue culture of the plant. The results obtained were as follows : 1. The Gyrophora esculanta was found growing mostly on the rock slopes of 722 m to 1915 min elevation on mountains in Korea. 2. Trees growing in the vicinity of the G. esculanta were mainly Quercus spp., Pinus thunbergii, Acer spp. and Lespedeza spp, Especially Quercus spp. was found growing in all of the study site. 3. The average Length of the rock slopes with G. esculanta growing on was 14 m and their aspects were mostly south. 4. The G. esculanta were found growing on rocks of Crystalline Schist, Quartz, Liparite, Granite, ete. Particularly they were mostly found on granites. The gradient of the rock slopes was in the range of 22-90 degrees. 5. The mean number of individuals of G. esculanta per one rock slope ranged from 14 at Mt. Bukhan to 70 at Mt. Jrri. Their mean diameter ranged from 1.8cm at Mt. Munsu to 4.6cm at Mt, Sokri. 6. The average percentage of G. esculanta with fruit body was 17.6%. The highest value was found at Mt. Cheonhwang (24.0%). 7. When the 100 segments of rhizoid of Gyrophora esculanta cultured in Detmer's medium supplemented with kinetine 5mg/l and 2, 4-D 3mg/l, n callus of microspore origins were induced from about 20% of the segments. As the induced n callus was transplanted on the six different types of rocks, it was observed that the juvenile G. esculanta grew best on granite and the development rate of G. esculanta on the granite was about 55%.

  • The bitterling (Cyprinidae, Acheilongnathinae) is a temperate freshwater fish with a unique spawning symbiosis with host mussels. Female bitterlings use their extended ovipositors to lay eggs on the gills of mussels through the mussel's exhalant siphon. In the present study, in April of 2020, we investigated spawning frequencies and patterns of three bitterling fish species in host mussel species in the Nakdong River basin (Hoecheon). During field surveys, a total of four bitterling and three mussel species were found. We observed bitterling's spawning eggs/larvae in the three mussel species: Anodonta arcaeformis(proportion spawned: 45.5%), Corbicula fluminea(12.1%), and Nodularia douglasiae (45.2%). The number of bitterlings' eggs/larvae per mussel ranged from 1 to 58. Using our developed genetic markers, we identified the eggs/larvae of each bitterling species in each mussel species (except for A. macropterus): A. arcaeformis (spawned by Acheilognathus yamatsutae), C. fluminea (A. yamatsutae and Tanakia latimarginata), and N. douglasiae (A. yamatsutae, Rhodeus uyekii, and T. latimarginata). Approximately 57.6% of N. douglasiae mussel individuals had eggs/larvae of more than one bitterling species, suggesting that interspecific competition for occupying spawning grounds is intense. This is the first report on bitterling's spawning events in the Asian clam C. fluminea from Korea; however, it should be ascertained whether bitterling's embryo undergoes successful development inside the small mussel and leaves as a free-swimming juvenile.
