The prevalence of potentially xenozoonotic viruses in the reproductive tract of female pigs in Korea was investigated by polymerase chain reaction (PCR). These viruses include porcine endogenous retrovirus (PERV), porcine reproductive and respiratory syndrome virus (PRRSV), swine hepatitis E virus (SHEV), porcine lymphotropic herpesvirus (PLHV), and porcine circovirus type 2 (PCV-2). Histopathological examination and PCR analysis were conducted using the ovaries of 70 slaughtered pigs that were collected from 14 farms in Jeju. Histopathologically, infiltrations of mononuclear inflammatory cells around the thick-walled coiled vessels in the ovarian medulla were observed in 15 cases. Based on the PCR method, PERV, PLHV, PRRSV, SHEV, and PCV-2 were detected in 69 (98.6%), 35 (50%), 5 (7.1%), 4 (5.7%), and 1 sample (1.4%), respectively. These results suggest that PERV and PLHV are the major xenozoonotic viruses in the porcine ovary. This study should aid in the development of a monitoring protocol for potential xenozoonotic agents and in the production of germ-free pigs for xenotransplantation.

Objective: The Jeju native pig (JNP) found on the Jeju Island of Korea is a unique black pig known for high-quality meat. To investigate the genetic uniqueness of JNP, we analyzed the selection signature of the JNP in comparison to commercial pigs such as Berkshire and Yorkshire pigs. Methods: We surveyed the genetic diversity to identify the genetic stability of the JNP, using the linkage disequilibrium method. A selective sweep of the JNP was performed to identify the selection signatures. To do so, the population differentiation measure, Weir-Cockerham's Fst was utilized. This statistic directly measures the population differentiation at the variant level. Additionally, we investigated the gene ontologies (GOs) and genetic features. Results: Compared to the Berkshire and Yorkshire pigs, the JNP had lower genetic diversity in terms of linkage disequilibrium decays. We summarized the selection signatures of the JNP as GO. In the JNP and Berkshire pigs, the most enriched GO terms were epithelium development and neuron-related. Considering the JNP and Yorkshire pigs, cellular response to oxygen-containing compound and generation of neurons were the most enriched GO. Conclusion: The selection signatures of the JNP were identified through the population differentiation statistic. The genes with possible selection signatures are expected to play a role in JNP's unique pork quality.
This study investigated the incidence rate of umbilical hernia (UH) in pig farms and examined the effect of using them as breeding pigs on reproductive traits. The occurrence of UH ranged from 0.1% to 3.0% in pig farms investigated. UH pigs were found in almost all farms except for a single farm in Jeju Island. Spontaneously occurring UH pigs were selected and used for cross breeding tests. UH-related crosses and their progeny showed significant (P < 0.05) differences in gestation period, the numbers of piglets born and alive, and body weights at birth and $21^{st}$ day comparing to those of the control population. UH-related crosses showed longer gestation period, reduced numbers of piglets, and lighter body weights than those from the control population. Interestingly, reduced number of piglets was about one fourth, suggesting that UH inheritance might play a critical role as a lethal gene during embryogenesis. In addition, UH incidence rate in UH-related crosses was significantly (P < 0.05) higher than that in the control except for UH-cross3. However, in the progeny of control cross, a pig also had UH appearance, indicating that porcine UH might be inherited in recessive inheritance mode. Taken together, the results of this study indicate that UH is one of recessively inherited genetic defect that occurs at ordinary times in pig farms, suggesting that the use of UH animals as sire and/or dam may lead to economic losses due to increased gestation period, reduced numbers of piglets born and alive, and lower growth rates after birth of pigs.
We conducted two studies to determine the effect of gender, gonadectomy (GDX) on growth and plasma cholesterol levels in pigs. In experiment 1, five sham-operated and five GDX female Landrace pigs (26kg) were allowed to have free access to water and feed up to market weight (approximately 100kg). Body weight and feed consumption were recorded biweekly, and daily body weight gain, daily feed intake and feed efficiency (gain/feed) were calculated during the feeding period. In experiment 2, 10 male (26kg) and 10 female (26kg) Landrace pigs were used; five male and five female pigs were assigned to sham-operated or GDX. Pigs were allowed to have free access to water and a diet without added cholesterol (Table 1) until they were 6 months old (male 104 and female 98kg) and thereafter they were fed a hypercholesterolemic diet (Table 1) containing 0.5% cholesterol and 0.1% cholate for 10 days. GDX of female pigs increased average daily gain (P<0.05), compared with their sham-operated counterparts during the growing-finishing period, but had no effect (P>0.05) on feed efficiency. Plasma cholesterol levels in pigs fed a hypercholesterolemic diet for 10 days were much higher (P<0.05) in females than in males (161 vs 104mg/100mL plasma), and were increased by GDX only in male pigs. HDL-cholesterol/LDL+VLDL-cholesterol ratio appeared to be higher in males than in females, and was not influenced by GDX in either sex. Results suggested that the lower growth rate of female pigs than their male counterparts is attributable to the ovarian activity, and the lower plasma cholesterol level in male than in female pigs fed a hypercholesterolemic diet is due to the testicular activity.

Post-weaning multisystemic wasting syndrome (PMWS) is a new emerging disease affecting nursery and growing pigs in worldwide. Porcine circovirus type 2 (PCV-2) is a most important pathogen associated with PMWS. This study was carried out to investigate the pathological changes in the pancreas of pigs diagnosed as PMWS. To detect the PCV-2 antigen and nucleic acid in the tissue, immunohistochemistry and polymerase chain reaction (PCR) was conducted, respectively. 24 pigs of 4-10 weeks old showed clinical signs of PMWS such as chronic wasting, respiratory distress and diarrhea were examined. Histopathologically, interstitial and periductular mononuclear cells infiltration were observed in pancreas. Multifocal to diffuse necrosis of acinar tissues or necrotizing to granulomatous pancreastitis with numerous syncytial cells infiltration were examined in severe cases. PCV-2 nucleic acid was detected from all tested pancreas using PCR. The PCV-2 antigen in 12 pancreas sections was detected by immunohistochemical staining. PCV-2 has a tropism for vascular endothelial cells and infiltrated macrophages. Although gross lesions are uncommon in the pancreas of pigs with PMWS, histopathological changes and the presence of PCV-2 in this tissue may be related to clinical signs associated with digestive disorders.
This study was conducted to investigate the genotypes and frequencies of Melanocortin 1 Receptor(MC1R) genes in pigs which plays a central role in regulation of eumelanin (black/brown) and phaeomelanin(red/yellow) pigment synthesis within the mammalian melanocytes. Four different breeds of pigs(20 Landrace, 20 Yorkshire, 20 Duroc, and 93 Jeju native black pigs) were used and PCR-RFLP analysis of MC1R gene was also carried out. Two regions of MC1R genes (428bp and 405bp) were amplified using two specific primers (MERL1-EPIG2, EPIG1-EPIG3), respectively and MC1R allele were determined using 2 restriction enzymes (BspHⅠ, AccⅡ). The results of this experiment indicated that MC1R allelic type in Landrace, Large Yorkshire and Duroc were MC1R *2 (Ep), MC1R *2 (Ep), MC1R *4 (e), respectively. However, various allelic types of MC1R genes were detected in Jeju native black pigs. MC1R allelic type of Jeju black pigs was MC1R*2 type as in Meishan and Large black breeds or MC1R*3 type as in Hampshire and Berkshire breeds and the gene frequencies of ED1 and ED2 were 0.554 and 0.446 in average.
Multiplex PCR and immunohistochemistry were used to detect and differentiate between porcine circovirus (PCV) type-I and the PCV associated with postweaning multisystemic wasting syndrome (PMWS). Unique DNA product to PCV type-II was confirmed the some organs including lymph nodes, tonsil and spleen from eight pigs in Jeju by multiplex PCR. In this study, the samples were tested by a multiplex PCR assay to determine the type of PCV in each case; all cases were PCV type-II positive. PCV type-II was identified not only in typical PMWS cases, but also in field cases submitted with various clinical histories, some of which were not suggestive of PMWS. Immunohistochemically PCV type-II antigen was detected in macrophage-like cells in the tonsil, liver, lymph nodes and spleen, while some hepatocytes and renal tubular epithelial cells were also positive to the virus. This study suggested that PCV type-II is one of the causative agents of PMWS as well as the major type of PCV in the affected pigs in Jeju.

Tylosin has been used as a livestock feed additive and antibiotic growth promoter for many years. However, the mode of action by which tylosin enhances animal growth is unclear. We used high-throughput sequencing of 16S rRNA genes to investigate the effects of tylosin as a feed additive on swine gut microbiota. No significant difference in the rate of weight increase was observed between control and tylosin-treated pigs during a 10-week feeding trial. However, tylosin-treated pigs showed rapid increases in the relative abundance of the phylum Firmicutes. Increases in Firmicutes species are associated with (so-called) obese-type gut microbiota. The abundance of species of four families of the phylum Firmicutes (Streptococcaceae, Peptococcaceae, Peptostreptococcaceae, and Clostridiaceae) correlated positively with host weight gain. The abundance of Streptococcaceae family bacteria was least affected by tylosin treatment. Distribution analysis of operational taxonomic units (OTUs) showed that both control and tylosin-treated pigs exhibited similar OTU alterations during growth. However, the tylosin-treated group showed distinctive alterations in gut microbiota when the host weighed approximately 60 kg, whereas similar alterations occurred at around 80 kg in the control group. Our results suggest that use of tylosin accelerates maturation of swine gut microbiota rather than altering its composition.

Forty guinea pigs were divided into four groups and fed 0.04% cholesterol based control diet, plus 0.05% simvastatin, and statin plus 0.1% CoQ10 or 10% Ardisia Japonica Blume (AJB) leave powder for 4 weeks. Plasma total cholesterol levels decreased significantly in all groups fed the statin-containing diet compared with that in guinea pigs fed the control diet (P < 0.01). Plasma and liver triglycerides decreased significantly in the statin plus CoQ10 group compared with those in the control (both P < 0.05). Maximum platelet aggregation was significantly higher in the statin plus CoQ10 group than that in the other groups (P < 0.05). Na-K ATPase activity increased in the statin group and decreased in the statin plus CoQ10 group (P < 0.01). Na-K co-transport and Na passive transport decreased significantly in the control group compared with those in the other groups (both P < 0.05). Intracellular Na was highest in the statin group and lowest in the statin plus CoQ10 group and was correlated with Na-K ATPase activity. Thiobarbituric acid reactive substance production in platelet-rich plasma and liver tended to decrease in the statin plus CoQ10 group compared with those in the other groups. Plasma glutamic-pyruvic transaminase and glutamic-oxaloacetic transaminase increased significantly in the statin group compared with those in the control (P < 0.05). These result suggest that antioxidant rich AJB did not have positive effects on cardiovascular disease parameters. The statin plus CoQ10 seemed to decrease cholesterol more efficiently than that of statin alone.
