Estimation of Pork Quality Traits Using Exsanguination Blood and Postmortem Muscle Metabolites

  • The current study was designed to estimate the pork quality traits using metabolites from exsanguination blood and postmortem muscle simultaneously under the Korean standard pre- and post-slaughter conditions. A total of 111 Yorkshire (pure breed and castrated male) pigs were evaluated under the Korean standard conditions. Measurements were taken of the levels of blood glucose and lactate at exsanguination, and muscle glycogen and lactate content at 45 min and 24 h postmortem. Certain pork quality traits were also evaluated. Correlation analysis and multiple regression analysis including stepwise regression were performed. Exsanguination blood glucose and lactate levels were positively correlated with each other, negatively related to postmortem muscle glycogen content and positively associated with postmortem muscle lactate content. A rapid and extended postmortem glycolysis was associated with high levels of blood glucose and lactate, with high muscle lactate content, and with low muscle glycogen content during postmortem. In addition, these were also correlated with paler meat color and reduced water holding capacity. The results of multiple regression analyses also showed that metabolites in exsanguination blood and postmortem muscle explained variations in pork quality traits. Especially, levels of blood glucose and lactate and content of muscle glycogen at early postmortem were significantly associated with an elevated early glycolytic rate. Furthermore, muscle lactate content at 24 h postmortem alone accounted for a considerable portion of the variation in pork quality traits. Based on these results, the current study confirmed that the main factor influencing pork quality traits is the ultimate lactate content in muscle via postmortem glycolysis, and that levels of blood glucose and lactate at exsanguination and contents of muscle glycogen and lactate at postmortem can explain a large portion of the variation in pork quality even under the standard slaughter conditions.

  • The most reasonable method of counting the number of the estuaries in Korea may be counting the number of streams flowing into the sea. The number counting of the streams should be carried out based on the river classes (National River, Regional 1st Class River and Regional 2nd Class River) given by the River Act. The National River scale estuaries are totaled to thirteen, i.e. Han River(including Imjin River), Anseongcheon, Sapgyocheon, Geum River, Mankyeong River, Dongjin River, Yeoungsan River, Tamjin River, Seomjin River, Gawhacheon, Nakdong River(including West Nakdong River), Taewha River, and Hyeongsan River estuaries. The Regional 1st Class River scale estuaries are counted as four, i.e. Yeoungdeok Osipcheon, Samcheok Osipcheon, Gangneung Namdaecheon, and Yangyang Namdaecheon. While, the Regional 2nd Class River estuaries are arranged as the Province and counted as total 444; Jeonnam, Gyeongnam, Chungnam, Jeju Province have the 108, 94, 52, and 35 estuaries, respectively. The counted estuary numbers, however, can be slightly increased/decreased on what references are applied, e.g. whether or not including the streams lost estuary functions by the sea-dike construction or integrated nearby streams, and so on.

Characterization of a New ${\beta}$-Lactamase Gene from Isolates of Vibrio spp. in Korea

  • PCR was performed to analyze the ${\beta}$-lactamase genes carried by ampicillin-resistant Vibrio spp. strains isolated from marine environments in Korea between 2006 and 2009. All 36 strains tested showed negative results in PCR with the primers designed from the nucleotide sequences of various known ${\beta}$-lactamase genes. This prompted us to screen new ${\beta}$-lactamase genes. A novel ${\beta}$-lactamase gene was cloned from Vibrio alginolyticus KV3 isolated from the aquaculture water of Geoje Island of Korea. The determined nucleotide sequence (VAK-3 ${\beta}$-lactamase) revealed an open reading frame (ORF) of 852 bp, encoding a protein of 283 amino acids (aa), which displayed low homology to any other ${\beta}$-lactamase genes reported in public databases. The deduced 283 aa sequence of VAK-3, consisting of a 19 aa signal peptide and a 264 aa mature protein, contained highly conserved peptide segments specific to class A ${\beta}$-lactamases including the specific amino acid residues STFK (62-65), SDN (122-124), E (158), and RTG (226-228). Results from PCR performed with primers specific to the VAK-3 ${\beta}$-lactamase gene identified 3 of the 36 isolated strains as V. alginolyticus, Vibrio cholerae, and Photobacterium damselae subsp. damselae, indicating the utilization of various ${\beta}$-lactamase genes including unidentified ones in ampicillin-resistant Vibrio spp. strains from the marine environment. In a mating experiment, none of the isolates transfered the VAK-3 ${\beta}$-lactamase gene to the Escherichia coli recipient. This lack of mobility, and the presence of a chromosomal acyl-CoA flanking sequence upstream of the VAK-3 ${\beta}$-lactamase gene, led to the assumption that the location of this new ${\beta}$-lactamase gene was in the chromosome, rather than the mobile plasmid. Antibiotic susceptibility of VAK-3 ${\beta}$-lactamase was indicated by elevated levels of resistance to penicillins, but not to cephalosporins in the wild type and E. coli harboring recombinant plasmid pKV-3, compared with those of the host strain alone. Phylogenetic analysis showed that VAK-3 ${\beta}$-lactamase is a new and separate member of class A ${\beta}$-lactamases.

Inclusion effect of soybean meal, fermented soybean meal, and Saccharina japonica in extruded pellet for juvenile abalone (Haliotis discus, Reeve 1846)

  • Inclusion effect of soybean meal (SBM) and fermented SBM (FSM) in extruded pellet for juvenile abalone (Haliotis discus) was compared in abalone farm. Dietary inclusion effect of the combined macroalgae (MA) (Undaria pinnatifida and Hizikia fusiforme) and a single Saccharina japonica on abalone was also compared. Three thousand six hundred juvenile abalone were purchased from a private hatchery and acclimated to the experimental conditions for 2 weeks. Six 5-ton flow-through raceway tanks were used, and abalone were randomly distributed into tanks (n = 600 per tank). Three experimental diets were prepared in duplicate. Fish meal, FSM, corn gluten meal, and shrimp meal and wheat flour and dextrin were used as the protein and carbohydrate sources, respectively, in the FSM diet. MA was also included in the FSM diet. FSM and MA in the FSM diet were substituted with SBM at the expense of wheat flour and S. japonica, referred to as the SBM and SJ diets. The experimental diets were pelletized by an extruded pelleter. Water stability of nutrients in the experimental diets was monitored at 12, 24, and 48 h after seawater immersion. The experimental diets were fed to abalone once a day to satiation with a little leftover for 120 days. The retained crude protein and lipid and ash content of the extruded pellets were changed over all period of time. Weight gain and specific growth rate (SGR) of abalone fed the SBM diet were greater than those of abalone fed the FSM and SJ diets. Weight gain and SGR of abalone fed the SJ diet were also greater than those of abalone fed the FSM diet. The longest shell length, widest shell width, highest shell height, and greatest soft body weight were obtained in abalone fed the SBM diet, followed by the SJ and FSM diets. Proximates of the soft body of abalone were not different among the experimental diets. In conclusion, SBM was a superior protein source to FSM in extruded pellet for growth performance of abalone. Dietary inclusion of a single S. japonica was superior to the combined inclusion of U. pinnatifida and H. fusiforme in the production of abalone.

Study on the Properties of White Pan Bread Made by Adding Preferment Prepared by Lactic Acid Bacteria Isolated from Kimchi (김치 유산균으로 제조한 preferment 첨가 수준에 따른 white pan bread의 품질특성)

  • The effects of preferment levels prepared with lactic acid bacteria isolated from kimchi on the quality of white pan bread were studied. The aim was to investigate how preferment influenced the baking-technological, textural, and sensory properties of white pan bread. During the process of making white pan bread, the pH decreased as the preferment levels increased, whereas the titratable acidity increased. The loaf specific volume increased from 4.66 to $5.59\;cm^3/g$ as preferment content increased from $10%{\sim}25%$. The moisture content and water activity of the bread ranged from 40.77 to 41.49%, and 0.961 to 0.966, respectively, indicating that no appreciable differences were related with the preferment levels (p<0.05). The textural characteristics of white pan bread were highly correlated with the amount of preferment added. White pan bread containing preferment showed a decrease in hardness and gumminess, and an increase in springiness. The color of white pan bread was not significantly different from that of the control. In sensory evaluation, the values for volume, springiness, and umami were highest in the white pan bread with 20% preferment, and the sour and umami tastes increased with the amount of preferment.

Distribution of the Kentish Plover (Charadrius Alexandrinus) Based on the 3rd National Ecosystem Survey and Its Adequacy as a Bioindicator (제 3차 전국자연환경조사를 이용한 흰물떼새(Charadrius alexandrinus)의 분포현황과 생물지표종의 제안)

  • In this study we analyzed the spatial and temporal distribution and preferred habitat type of the Kentish plover (Charadrius alexandrinus) based on the 3rd National Ecosystem Survey. Kentish plovers were observed in 97 maps out of a total 842 maps (11.8%) between 2006 and 2012, mainly along the western and southern coasts of Korea. They were also observed in the eastern coast of Korea, inland rivers (Han, Geum, Nakdong, Seomjin, and Yongsan River), the western and eastern coast of Jeju island, and Daecheong Island in the Yellow Sea. The observations were mainly made during the spring breeding season and migration seasons in spring and autumn. The occurrence of kentish plovers was positively influenced by the area of water and wetland according to the middle classification level of land cover type analysis and the area of coastal wetlands in the detailed classification level of land cover types. Most (90%) of the kentish plovers recorded maps had coastal wetlands. Kentish plovers were known to be susceptible to change of habitat. As the occurrence of kentish plovers could be associated with the habitat-change of coastal wetlands and it is possible to estimate the number of individuals, it is recommended that kentish plovers be used as a bioindicator species for the ecological assessment of ecosystem in intertidal zones.

Effects of Long Term Tangerine Peel Consumption on the Physicochemical Properties and Palatability of Crossbred Pig Meats (감귤피 첨가 사료를 장기간 급여한 교잡종 돼지고기의 물리화학적 특성과 기호성)

  • Crossbred pigs (female, 198 days old, $102{\sim}118kg$, Tamra Marketing Farm) were fed a diet supplemented with tangerine peel fer an extended period to study its effect on the physicochemical properties and palatability of the pork. The samples for this study consisted of the pork from pigs fed a diet without tangerine peel (T0), and the pork from pigs fed a diet containing 8% tangerine peel during the entire breeding period, from early pregnancy through the finishing period (T1). The pH Hunter's $L^*$ value, water holding capacity, freezing loss, thawing loss, cooking loss, hardness, springiness, cohesiveness, sensory raw meat color and cooked meat palatability of loin and belly were not significantly different between T0 and T1 pork (p>0.05). However, the Hunter's $a^*\;and\;b^*$ value, gumminess, shear force value (SFV) and smell were significantly higher in meat from loin of T1 than T0 pigs (p<0.05). Thus T1 pork exhibited an improved SFV, DPPH reduction and aroma of cooked belly meat (p<0.05).

Dormancy and Germination Characteristics of Alpine Modest Primrose (Primula modesta var. hannasanensis T.Yamaz.) Seeds (설앵초의 종자 휴면과 발아특성)

  • This research was performed to develop mass propagation method of Alpine modest primrose (Primula modesta var. hannasanensis T.Yamaz.) for improving the conservation and utilization of the species. Seeds were collected on August 2011 in Jeju-city and well-selected seeds were dry-stored at $4{\pm}1.0^{\circ}C$. Seed size ranged $0.44{\pm}0.07{\times}0.61{\pm}0.04mm$, and weight of 1,000 seeds was $51.78{\pm}0.021mg$. Thus the seed was classified as 'dwarf seeds'. As the result of dormancy characteristics, moisture content of freshly matured seeds increased rapidly by water-soaking treatment and seeds did not germinate at $20^{\circ}C$ for 4 weeks under the light condition. Therefore, seeds were estimated to have physiological dormancy. Germination conditions of dry-stored seeds were found to be $20^{\circ}C$ and light condition, and seeds did not germinated under dark condition regardless of all temperature regimes. Therefore, Alpine modest primrose seed was considered as photoblastic type. Percent germination (PG) and germination energy (GE) were greatly improved by soaking the seeds in $GA_3$ and kinetin for 24 hours. Especially, 200~500 mg/L $GA_3$ treatment resulted in the highest PG (95.5%) and GE (98.3%). So, chemical treatment such as $GA_3$ was thought to be a useful method for raising seedling uniformly.

A Survey on Insect Diversity of Mulyeongari-Oreum Wetland, on Jeju Island, South Korea (제주도 물영아리오름 습지의 곤충다양성에 관한 연구)

  • The purpose of this study is to identify insects inhabiting Mulyeongari-Oreum wetland and to provide basic information for conservation of wetlands. As a result, a total 229 species, 73 families, 9 orders of insects were collected: 136 species, 39 families, 6 orders from Mulyeongari-Oreum Wetland and 121 species, 52 families, 9 orders from Mt. Sooryeong. Of the 229 species of insects investigated, Copris tripartitus Waterhouse, an endangered species of wildlife fauna in Korea, was found with many individuals. Also, Lychnuris rufa(Olivier), having a narrow habitat near unpolluted water and mountains, was also recorded with 3 individuals and then should be considered as a protected species in Wetlands. To identify the community structure of moth on Mulyeongari-Oreum wetland, a light trap was examined and a total 76 species of moths were collected. Total dominant species was Hydrillodes morosa(Butler) with index of 0.12 and the species diversity on the moth community from survey area was the highest in July with index of 3.42.

Estimation of Markov Chain and Gamma Distribution Parameters for Generation of Daily Precipitation Data from Monthly Data (월 자료로부터 일 강수자료 생성을 위한 Markov 연쇄 및 감마분포 모수 추정)

  • This research was to elucidate the generation method of daily precipitation data from monthly data. We applied a combined method of Markov chain and gamma distribution function using 4 specific parameters of ${\alpha}$, ${\beta}$, p(W/W) and p(W/D) for generation of daily rainfall data using daily precipitation data for the past 30 years which were collected from the country's 23 meteorological offices. Four parameters, applied to use for the combination method, were calculated by maximum likelihood method in location of 23 sites. There are high correlations of 0.99, 0.98 and 0.98 in rainfall days, rainfall probability and mean amount of daily rainfall between measured and simulated data in case of those parameters. In case of using parameters estimated from monthly precipitation, correlation coefficients in rainfall days, rainfall probability and mean amount of daily rainfall are 0.84, 0.83 and 0.96, respectively. We concluded that a combination method with parameter estimation from monthly precipitation data can be applied, in practical purpose such as assessment of climate change in agriculture and water resources, to get daily precipitation data in Korea.
