Lipopolysaccharide로 유도된 Raw264.7 cell에서 Phellodendron amurense의 Jatrorrhizine에 의한 염증 억제효과 (Anti-inflammatory Effect of Jatrorrhizine from Phellodendron amurense in Lipopolysaccharide-stimulated Raw264.7 Cells)

  • 황백으로부터 n-BuOH 추출물에서 약 50%의 hyaluronidase 저해효과가 있음이 확인되었다. n-BuOH 분획 농축물을 Sephadex LH-20 및 MCI gel CHP-20 column chromatography로 정제한 결과 저해활성을 가지는 Jatrorrhizine을 분리, 동정하였다. 황백추출물과 Jatrorrhizine은 모두 농도 의존적으로 NO 발현을 억제하였고, 100 ${\mu}g/mL$의 농도에서 60% 이상의 억제 효과를 나타내었다. 황백추출물과 Jatrorrhizine의 iNOS와 COX-2의 억제력을 살펴본 결과, 100 ${\mu}g/mL$의 농도에서 45% 및 29%의 저해효과를 나타내었다. 따라서 황백추출물 및 Jatrorrhizine은 LPS로 유도되어진 대식세포주인 Raw264.7 세포에서의 염증반응의 억제효과를 기대할 수 있었다.

  • Protothalipine colorless needle crystal, mp $195-6^{\circ}$, $C_{21}H_{25}NO_{5}$) and jatrorrhizine chloride (Base A, orange needle crystal, mp $198^{\circ}$) are identified by chemical, spectral analysis, and comparison with authentic sample. Base B (reddish crystal, mp $192-4^{\circ}$) might be a artifact substance, which was changed from the jatrorrhizine chloride. Base C (yellowish needle crystal, mp $228~230^{\circ}$) and Base D (reddish needle crystal, mp $211~213^{\circ}$) were assumed to be thalifendine chloride, desoxythalidastine chloride by phytochemical and spectral analysis respectively.

A Study for the isolation of the Berberine-type Alkaloid from Coptidis Rhizoma and for their Antitumor Activities

  • The purpose of this study is the separation of biologically active ingredients from Coptidis Rhizoma which has been widely used as one of oriental herbal medicine for body fever. In this study, berberine-type alkaloids were tested on their biological activities in the aspect of antibacterial, antitumor, anti-herpetic and anti-HIV activity. Contents of five major alkaloids for the various origin of Coptidis Rhizoma were assayed by HPLC. As the results, the content of berberine from Coptis chinensis and Coptis japonica were 6.78% and 7.09%, respectively. The contents of coptisine, jatrorrhizine, berberastine from Coptis chinensis were higher than those of Coptis japonica. The amount of palmatine from both species were almost the same. Surprisingly for antitumor experiment, all compounds have been shown remarkable activity, especially against SNU-l(human stomach cancer) cell line. Among the compounds purified through column chromatography, palmatine, coptisine, and jatrorrhizine inhibited the growth of K-562(human chronic myelogenous leukemia) cell line whereas jatrorrhizine has been shown the effective inhibition of A-549 (human lung) cell line at the same time.

  • Examination of the alkaloids in the stem of Berberis amurensis $R_{UPRECHT}$ var. latifolia $N_{AKAI}$ was carried out. As tertiary bases, the biscoclaurin type base berbamine and oxyberberine as the berberine type were isolated, and jatrorrhizine, berberine, shobakunine as the berberine type and the aphorphine type magnoflorine were obtained as the quarternary bases.

매발톱나무의 성분에 관한 연구 (Studies on the Constituents of Berberis amurensis Ruprecht)

  • Berberis amurensis Ruprecht(Berberidaceae) is a medicinal plant indigenous to the middle and northern part of Korean peninsula. The woody parts of this plant have been used for the ocular, peptic and intestinal disorders. The stems of this plants were extracted with MeOH and the MeOH extract was partitioned between organic phases and water layer, successively to fractionated quarternary alkaloids. The acetone-soluble part of guarternary alkaloidal fraction had antibacterial activities and it contained four protoberberine alkaloids such as palmatine(I), Berberine(II), Jatrorrhizine(III) and coptisine(IV), and one aporphine alkaloid, magnoflorine(V). Although the isolations of the compounds I, II, IIII, IV and V from different sources were reported, this is the first report that Berberis amurensis contained the compounds. When the contents of compound I(palmatine) and II(berberine) were quantified and compared with those of other plant parts, cortex contained higher palmatine and berberine than any other part of the plant.

Quantitative and Qualitative Analysis of Alkaloids in Coptis chinensis (Coptidis Rhizoma) by LC-DAD and LC-ESI/MS

  • The quality control of natural products is principal key to guarantee the Good Manufacturing Practices (GMP) and Good Clinical Practices (GCP) for the functional food, pharmaceuticals and cosmeceuticals in the industry. In this study, we examined the quantitative analysis of berberine as marker substance of Coptidis Rhizoma by high performance liquid chromatography-photodiode array detector (HPLC-DAD). The HPLC method was validated and met all the requirements for the quality control analysis recommended by FDA and ICH. The berberine was separated on a Xterra $C_{18}$ column ($5{\mu}m$, $4.6{\times}250mm$) using mobile phase consisting of distilled water and acetonitrile with $KH_2PO_4$ (3.4 g) and $Na_2SO_4$ (1.7 g). Calibration curve of berberine has been estimated (y = 42293.47x-41589 with the correlation coefficient 0.9999). The amount of berberine was calculated as 4.25%. And berberastine, palmatine, columbamine, jatrorrhizine, epiberberine, berberine and coptisine in the Coptidis Rhizoma were identified by high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-ESI-MS) method.

In Vitro Effect of Methanolic Extract of Argemone mexicana against Trichomonas vaginalis

  • Infections caused by Trichomonas vaginalis in humans are one of the main public health problems caused by sexually transmitted diseases. Objective of this study was to evaluate potential biological activity of the medicinal plant Argemone mexicana (Mexican poppy) on T. vaginalis. Methanolic extracts of the stems and leaves of A. mexicana, and different fractions were prepared with solvents of different polarities. The extracts and functional groups were detected containing sterols, triterpenes, quinones, flavonoids and, alkaloids. Extracts from both the stems and leaves of A. mexicana inhibited the growth of T. vaginalis with half-maximal inhibitory concentration value of 70.6 and 67.2 ㎍/ml, respectively. In the active fractions, the most abundant compounds were berberine and jatrorrhizine, with presumed antiparasitic activity.

  • By screening inhibitory activities on the melanin polymer biosynthesis in B-16 mouse melanoma cell lines, MeOH extract of Phellodendri Cortex was found to have inhibitory effect on melanin polymer biosynthesis. Twelve compounds were isolated from the MeOH extract of P. Cortex. They were identified as obacunone (1), limonin (2), ${\beta}-sitosterol$ (3), bis(2-methylheptyl) phthalate (4), cycloeucalenol (5), berberine (6), palmatine (7), jatrorrhizine (8), syringin (9), umbelliferone (10), rutaecarpine (11) and scopoletin (12) by comparison of their physical and spectral data with those of authentic samples. Among the isolated compounds, berberine (6) and palmatine (7) showed potent inhibitory effect on the melanin polymer biosynthesis in cultured B-16 mouse melanoma cell lines, with Inhibition rate of 96% and 90%, respectively. As a positive control, arbutin exhibited an inhibition rate of 56%.

고지방식이를 통해 비만이 유발된 마우스에서 C-DM3 복합추출물의 항비만 및 항당뇨 효능 연구 (The Herbal Formula C-DM3 Improves the Changes of Diabetes-Related Biomarkers in High-Fat Diet-Induced Obese Mice through Regulation of the IRS1/PI3K/AKT and AMPK Signaling Pathways in the Liver and Pancreas)

  • Objectives: In the present study, we investigated the effects of clean-diabetes mellitus 3 (C-DM3), a herbal formula with Trichosanthis Radix, Coptidis Rhizoma, Crataegi Fructus, and Cinnamomi Cortex, on the pathological and serological symptoms of diabetes and its related molecular mechanisms in diet-induced obese mice. Methods: We prepared an obese mouse model using a high-fat diet for 8 weeks and then administered the C-DM3 extract for 4 weeks. The changes of pathological and serological biomarkers for diabetes assessment were measured in the mice and histological changes were observed in the liver and pancreas tissues. We also identified the main compounds in the C-DM3 extract using high pressure liquid chromatography (HPLC) and analyzed the molecular mechanism of the disease condition by network pharmacological analysis. Results: In the in vivo, the administration of C-DM extract to obese mice significantly reduced body weight gain, fatty liver symptoms, and muscle loss, and decreased the levels of fasting blood glucose, insulin, aspertate aminotransferase, triglycerides, and low-density lipoprotein-cholesterol. In addition, C-DM extract significantly increased the phosphorylation of insulin receptor substrate 1, protein kinase b (AKT), phosphoinositide 3-kinase (PI3K), adenosine monophosphate-activated protein kinase, and glucose transporter 4 in all pancreatic and liver tissues, with inhibition of histopathological changes in obese mice. HPLC analysis identified hyperoside, berberine, epiberberine, columbamin, coptisine, coumarin, jatrorrhizine, and citric acid as the main compounds. In the network pharmacological analysis, the molecular targets of C-DM3 extract on obesity and diabetes were shown as the insulin, AKT, PI3K, and mitogen-activated protein kinase pathways with the regulation of inflammatory molecules interleukin 6 (IL-6), jun proto-oncogene, and IL-1β, which matched our in vivo targets. Conclusions: Based on these results, C-DM3 extract is expected to be effective in improving obesity and preventing diabetic progression.
