Effects of Ixeris Sonchifolia H. Fiet on Lipid Metabolism and Liver Function of Rats Administered with Ethanol (고들빼기 첨가 식이가 알콜투여 흰쥐의 지방대사와 간기능에 미치는 영향)

  • To investigate the effects of Ixeris sonchifolia Hance diets on serum and hepatic lipid levles and enzyme activities in rats administered with ethanol chronically, Sprague-Dawley male rats were AIN-76 diet(control), control diet plus ethanol, control plus Ixeris sonchifolia Hance diet, or control plus Ixeris sonchifolia Hance diet plus ethanol for 30 days. Ixeris sonchifolia Hance diets significantly decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and GOT levels that were increased due to the chronic ethanol administration. In addition, Ixeris sonchifolia Hance diets significantly decreased the liver triglyceride and total lipid levels that were increased due to the ethanol administration. The present findings, combined with previous data showing differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the serum Y-GPT activity of rats(Cha and OH[2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505) raise the possibility that GABA in plants could have a nytraceutical role in the recovery of chronic alcohol-related diseases.   • Polyphenol oxidase was purified from acetone powder extract of the root of Ixeris sonchifolia. The enzyme obtained by ammonium sulfate fractionation and sephadex G-200 gelfiltration gave 51-fold purification over the crude extract. The purified enzyme showed activity toward chlorogenic acid, caffeic acid and pyrocatechol. The kinetics of thermal inactivation of the enzyme followed first-order reaction. Potassium cyanide and cysteine were potent inhibitors.

The Effects of Godulbaegi Extracts on the Fluidity of Phospholipid Liposomes by DSC (DPPC Liposome에 미치는 고들빼기 추출물의 DSC 연구)

  • Liposomes have been widely employed as biomembrane-mimetic system and drug-delivery system. In these applications, the low stability of liposomes has been the most serious problem. They have relatively short half-lives and easily lysed through interactions with biological components. This study was performed to investigate the effects of godulbaegi extracts on the fludity of phospholipid liposomes. We used dipalmitoyl phosphatidylcholine(DPPC) liposomes which make most stable liposomes among the other phosphatidylcholines. The thermograms of the DPPC liposomal bilayers incorporated with the hexane extract of godulbaegi(Ixeris sonchifolia H.) were obtained, and the enthalpy changes and the sizes of cooperative unit of the transition were calculated. The incorporation of the Ixeris sonchifolia H. into the liposomal bilayers effectively reduced the transition temperature at which the transition from gel state to liquid-crystalline state occurs, broadened the thermogram peaks, and reduced the ratio of van't Hoff to calorimetric enthalpies. These results indicate indicate that the godulbaegi extracts (Ixeris sonchifolia H.) have significant effects on the fluidity of biological membrance.

  • Three known sesquiterpene lactones were isolated from the n-BuOH traction of the roots of Ixeris sonchifolia. Their structures were identified as macrocliniside A (1), glucozaluzanin C (2), and ixerin H (3), by spectral analyses. Compounds 1 and 2 are guaiane-type, while compound 3 is a germacrane-type sesquiterpene lactone. Compounds 1-3 are first isolated from I. sonchifolia.

  • In our previous study, the leaves of Ixeris sonchifolia afforded two new and two known guaiane type sesquiterpene lactones by activity-guided fractionation. Now we report additional isolation of sesquiterpene lactones from the roots of Ixeris sanchi!a!ia. They are glucozaluzanin C and ixerin H. Ixerin H is germacranolide type sesquiterpene glucuside. Theil structures were determined by 1 D and 2D NMR spectroscopy. (omitted)

  • Effect of various fractions prepared from the methanol extract of Ixeris sonchifolia and flavonoids isolated from the extract on hypercholesterolemic mice were examined. When various fractions $(CHCl_3,\;Interphase,\;EtOAc,\;n-BuOH\;and\;H_2O)$were administered to mice, the Interphase material produced a significant activity. Total cholesterol level in the Interphase material-treated group given doses of 25 and 50mg was decreased significantly by as much as 16% and 30%, respectively. Among the isolated flavonoids, cynaroside caused a significant hypocholesterolemic activity.

  • From the leaves of Ixeris sonchifolia (Compositae), luteolin and its glucoside and apigenin glucuronide were isolated and their structures were characterized on the basis of spectral data. Besides these flavonoids, the composition and relative content of free amino, acids and related compounds, compared to standards determined under identical conditions was also investigated using automatic amino acid analyzer. Major free amino acids were glutamic acid, aspartic acid, serine, proline, valine and arginine.

Effect of Godulbaegi(Ixeris sonchifolia H.) Powder on Growth, Protein and Lipid Concentration in Rats (고들빼기가 흰쥐의 성장률, 단백질 및 지질농도에 미치는 영향)

  • The effects of godulbaegi(Ixeris sonchifolia H.) powder on body weight gain, lipid and protein concentrations, and enzyme activities were studied in growing male rats for 4 and 8 week. The fats (Sprague-Dawley) were fed diets containing 5% godulbaegi or without. The body weght gain, food efficiency ratio, protein efficiency ratio, hemoglobin concentration and hematocrit value of rats fed godulbaegi diet were similar to the control diet. The ratio of spleen weight to body weight in godulbaegi group for 8 weeks was greater than in that of control group, but they were similar both feeding groups in liver and kidney. The concentrations of the total lipid and triglyceride in serum were increased in rats fed godulbaegi diet for 4 week than in those fed control diet, but those were decreased in rats fed godulbaegi diet for 8 week than in those fed the control diet. The concentrations of the phospholipid, total cholestrol and HDL-cholestrol in serum were decreased in rats fed godulbaegi diet for 8 week than in those fed control diet. The HDL-cholestrol/total cholestrol ratio and athergenic index were similar to both diet groups. There were no differences in growth, serum total protein, albumin concentrations, aspartate aminotransferase and alanine aminotransferase activities between rats fed godulbaegi diet and those fed the control diet.

  • The ethyl acetate extract of the leaves of Ixeris sonchifolia afforded two new and two known sesquiterpene lactone glucosides of the guaiane-type, together with a known alkenol glucoside. The known compounds were identified as ixerin Z (1), ixerin Z-6'-p-hydroxyphenylace-tate (2), and (Z)-3-hexen-1-ol-$\beta$-D-glucopyranoside (3), respectively. The structures of the new compounds were elucidated as 11, 13a-dihydroixerin Z [4, 3-hydroxy-2-oxo-guaia-1 (10), $3-dien-5{\alpha},6{\beta},7{\alpha},11{\beta}H-12,6-olide-3-O-{\beta}-D-glucopyranoside],{\;}and{\;}3,10{\$beta}-dihydroxy-2-oxo-guaia-3,11(13)-dien-1{\alpha},5{\alpha},6{\alpha},7aH-12,6-olide-10-O-{\beta}-D-glucopyranoside$ (5), respectively. The cytotoxicity of these compounds against human hepatocellular carcinoma cell (HepG2) and human melanoma cell (SK-MEL-2) was evaluated.

Inhibitory Effects of Luteolin Isolated from Ixeris sonchifolia Hance on the Proliferation of HepG2 Human Hepatocellular Carcinoma Cells

  • We investigated the anti-proliferative effects of luteolin and apigenin, isolated from Ixeris sonchifolia Hance, on HepG2 human hepatocellular carcinoma cells. In MTT assay luteolin showed more efficient anti-proliferative effects on cells than apigenin did. According to propidium iodide staining and flow cytometry studies, we postulated that these effects might be a result of cell cycle arrest. Hence we examined the changes of protein expressions related to cell cycle arrest. Western blotting data demonstrated that the down-regulated expression of CDK4 was correlated to the increase of p53 and CDK inhibitor $p21^{WAF1/CIP1}$ protein. These data suggest that luteolin may have potential as an anti-cancer agent.
