Studies on the Active Principles of Wild Vegetables on Biotransformation of Drug (야생 식용식물의 약물대사 활성성분에 관한 연구)

  • The effect of wild vegetables such as Allium tuberosum, Allium monanthum, Sedum sarmentosum, Ixeris dentata and Capsella-bursa pastoris on hexobarbital induced hypnosis was tested in mice. Among them, the methanol extract of Allium tuberosum exhibited significant lengthening of the barbiturate hypnosis. When various fractions prepared from the methanol extract of the Allium tuberosum were administered, the chloroform, ethylacetate and butanol extracts caused a significant activity. Through systematic fractionation by $SiO_2$ column monitoring by bioassays, $1,2,3,4-tetrahydro-{\beta}-carboline$ 3-carboxylic acid from the butanol extract was isolated as one of the active principles of this plant.

  • The effects of Aster scaber(Cham chyi) and Ixeris dentata(Sumbagui) of composite were studied on lipid metabolism in rats. Thirty rats were divided into five groups and fed diets containing 1% cholesterol, 0.25% sodium cholate, 10% coconut oil and 5% lard(control group) for 4 weeks. For each experimental diet added was 5% plant powder or extract of the plant which was equivalent to 5% Plant powder by dry weight. The lipid components of serum were assayed. The concentration of the total cholesterol was significantly lower in Cham chyi, and Sumbagui powder and the extract groups of those powder than the control group. The concentration of HDL-cholesterol was significantly higher in rats fed Cham chyi and Sumbagui powder than the control group. The concentration of LDL, LDL-cholestrol, VLDL and chylomicron were comparatively lower in Cham chyi and Sumbagui powder groups than those in the control group. The concentration of seum triglyceride was lower in Cham chyi powder fed group than the control group.

Cytotoxic and ACAT-inhibitory Sesquiterpene Lactones from the Root of Ixeris dentata forma albiflora

  • Ixeris dentata forma albiflora was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_{2}O$. Eight sesquiterpenes were isolated through repeated silica gel and octadecyl silica gel ($C_{18},\;ODS$) column chromatography of the EtOAc and n-BuOH fractions. Physicochemical analysis using NMR, MS and IR revealed the chemical structures of the sesquiterpenes, which were zaluzanin (1), 9a-hydroxyguaian-4(15), 10(14), 11 (13)-triene-6, 12-olide(2), $3{\beta}-O-{\beta}-D-glucopyranosyl-8{\beta}-hydroxyguaian$-4(15), 10(14)-diene-6, 12-olide (3), $3-O-{\beta}-D-glucopyranosyl-8{\beta}-hydroxyguauan$-10(14)-ene-6, 12-olide (4), ixerin M (5), glucozaluzanin C (6), crepiside I (7), and ixerin D (8). This is the first time that these sesquiterpene lactones have been isolated from this plant. Compounds 1, 2 and 7 revealed relatively high cytotoxicities on human colon carcinoma cell and lung adeno-carcinoma cell, while compounds 5 and 7 showed acyl-CoA: cholesterol acyltransferase (ACAT) inhibitory activity.

Effect of Antioxidant, Antimutagenicity and Anticancer of Root Extract from Ixeris dentata Nakai (씀바귀 뿌리 추출물의 항산화성, 항돌연변이원성 및 항암활성 효과)

  • Ixeris dentata root were extracted with methanol and then fractionated with n-hexane, EtOAc and BuOH to get active fractions. and their antioxidant and antimicrobial activities in each fraction were determined. Ethyl acetate fraction of Ixeris dentata root showed strong antioxidant activities, but aqueous fraction did not show any activities. But in the antimicrobial test, aqueous fraction showed strong antimicrobial activities except to Escherichia coli. especially, aqueous fraction showed the strongest activities against Hypocrea nigricans. and butanol fraction showed the strongest activities against Cladosporium herbarum. This study was performed to determine the antimutagenic and cytotoxic effect of Ixeris dentata root methanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using ames test and cytotoxicity assay, respectively. Cancer cell lines include human lung carcinoma(A549), human breast adenocarcinoma(MCF-7) and human hepatocellular carcinoma (Hep 3B). Futher fractionations with hexane, ethyl acetate, butanol and water from methanol extract of Ixeris dentata root were performed to obtain effective fraction, methanol extracts showed 79.94% inhibitory effect on the mutagenesis induced by N' -methyl- N' -nitro-N-nitrosoguanidine(MNNG) against TA100, while 89.99% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-1-oxide(4NQO) against TA98. In the meanwhile, butanol fraction showed 89.92% and 71.01% inhibitory effect on the mutagenesis induced by benzo(a)pyrene(B(a)P) against TA98 and TA100, respectively. Ethyl acetate fraction showed the strongest effect against A549, MCF-7 and Hep3B at the same concentration compared to those of other fration.

  • Ixeris dentata var. albiflora Nakai, a herbal plant, is often used to make a strong stomach as an antiphlogistic used when dyspepsia and to improve appetite in Korea and China. And also it is used for adult diseases such as diabetes and liver diseases as Korean traditional medicine. In this study, the composition and DPPH radical scavenging activities of the root of Ixeris dentata var. albiflora Nakai and its effects on cell viability on vero and chang cells were investigated. Moisture, crude ash, crude protein and crude lipid were 79.14, 2.49, 8.28 and 2.56 g/100 g respectively. The highest mineral content was K. The major free sugars were glucose, fructose and sucrose. Major fatty acid are linoleic acid, palmic acid and linolenic acid. Major amino acids were glutamic acid, arginine and aspartic acid and the total contents of amino acids were 28.12 mg/g. The methanol extracts were further fractionated with n-hexane, chloroform, ethylacetate, butanol and water to get an active fraction. In addition, cell viabilities in each fraction were determined. Methanol extract, butanol, and aqueous fraction showed strong survival rates in vero cell and chang cell viability test, and hexane, chloroform, and ethylacetate fraction were examined for toxin in a cell. The root of Ixeris dentata var. albiflora Nakai had scavenging activities against DPPH radicals in a dose-dependent assay. Ethylacetate fraction's SC50 was $6.8\{\mu}g/mL$, very strong DPPH radical scavenging activities, but water fraction did not show any activity.

  • The antioxidative effects of the extracts from Salicornia herbacea and Ixeris dentata were analyzed with 2,2-diphenyl-1-picrylhydrazyl (DPPH). The RC$_{80}$ of the ethanol-extract from I. dentata was 32.26$\mu\textrm{g}$/$m\ell$, while the RC$_{80}$ of ascorbic acid and glutathione, well-known antioxidants were 14.16 and 64.69$\mu\textrm{g}$/$m\ell$, respectively. However, in the case of the ethanol-extract from S. herbacea it was 108.29$\mu\textrm{g}$/$m\ell$. The cell viability in the group treated with the extracts from I. dentata leaves increased by 174% compared with the control 24 hr after irradiation. On the whole, the ethanol extracts resulted in a higher scavenging activity than the water extracts. Especially, the ethanol-extract from I. dentata leaves had the highest activity among all the extracts studied. It is suggested that S. herbacea and I. dentata are useful source materials for nutraceutical and pharmaceutical applications.ons.

Quantitative Determination of Five Phenolic Peroxynitrite-scavengers in Nine Korean Native Compositae herbs

  • Peroxynitrite (ONOO)-scavenging activities of nine Compositae herbs consisting of three Ixeris, two Youngia, two Cirsium and one of each Lactuca and Taraxacum species were evaluated. The contents of their ONOO scavengers in the extracts were also determined on a HPLC using seven standard compounds, chlorogenic acid (CGA), chicoric acid (CA), luteolin 7-glucoside (luteolin-7-glc), luteolin 7-glucuronide (luteolin-7-glcU), luteolin, linarin and pectolinarin. Five of those compounds exhibited potent ONOO-scavenging activities: IC50, CA (0.76 μM), CGA (1.34 μM), luteolin (0.81 μM), luteolin-7-glc (0.86 μM) and luteolin-7-glcU (3.13 μM). Both CA and luteolin-7-glc were highly contained in I. dentata (19.71 mg/g and 13.58 mg/g, respectively), I. dentata var. albiflora (17.58 mg/g and 23.83 mg/g, respectively) and I. sonchifolia (65.71 mg/g and 6.99 mg/g, respectively). Among the nine herbs, those three Ixeris species had very low IC50 values over the range of 0.48 - 1.74 μg/mL, suggesting that they could be potential therapeutic vegetables, particularly for preventing diabetic complications or obesity, which can be caused by an excess production of ONOO.

Antioxidant Activity of Ethyl acetate Fraction of Ixeris dentata (Thunb.) Nakai in Caenorhabditis elegans (씀바귀 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과)

  • Through the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of Ixeis dentata was investigated. The ethyl acetate soluble fraction of the I. dentata methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain (CF1553). As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

Studies on the Germination Characteristics in the Several Weeds of Compositae (국화과(菊花科) 잡초(雜草)의 발아특성(發芽特性)에 관(關)한 연구(硏究))

  • This experiment was carried to investigate the germination pattern in relation to temperatures and lights, and the emergence pattern in relation to seeding depths, lights and the alpha amylase activity of Youngia sonchifolia, Lactuca indica var. laciniata, Ixeris dentata var. albiflora and Ixeris polycephala. In Y. sonchifolia, the optimum germination temperature was $25^{\circ}C$, the optimum seeding depth to emerge was 0 mm and it could emerge in 0-5mm. In L. indica var. laciniata under cool storage, the optimum germination temperatures were $19^{\circ}C-28^{\circ}C$, the optimum seeding depth was 5mm and it could emerge in 0-20mm. In L. indica var. laciniata under room storage, the optimum germination temperature was $25^{\circ}C$ the optimum seeding depth was 5mm and it could emerge in 0-10mm. In I. dentata emerge was and 0mm and it could emerge in 0-5mm. In I. polycephla, the optimum temperatures were $16^{\circ}C-19^{\circ}C$, the optimum seeding depth to emerge was 0mm and it could emerge in 0-5mm. The alpha amylase activity was lower Y. sonchifolia, L. indica var. laciniata and I. dentata var. abiflora than barley cultivar Dongbor#1. And the increased pattern of alpha amylase activity was likely to it of germination rate.
