Inhibition of Monamine Oxidase by a Flavone and Its Glycoside from Ixeris dentata Nakai

  • Ixeris dentata Nakai (Compositae) is a perennial herb which has been used as a folk medicine for treating diabetes and gastroenteric troubles in Korea. Active compounds were isolated from the aerial parts of Ixeris dentata through the bioassay-guided fractionation and isolation method evaluated for inhibition of monoamine oxidase (MAO) in vitro. The compounds were identified as 5,7,3',4'-tetrahydroxyflavone (1) and 5,7,3',4'- tetrahydroxyflavone 7-glucoside (2), based on physical and spectroscopic characteristics. Compounds 1 and 2 showed a selective inhibition of type B MAO (MAO-B) activity, with IC/sub 50/ values of 15.3 μM and 36.4 μM, respectively, but did not inhibit type A MAO (MAO-A) activity.

Effects of Ixeris dentata extract on the production of pro-inflammatory cytokines in the LPS stimulated rat and Raw 264.7 cells (씀바귀 추출물이 LPS투여 흰쥐 및 Raw 264.7세포에서 전염증성 cytokines 생성에 미치는 영향)

  • The purpose of this study was to investigate anti-inflammatory effect of Ixeris dentata ethanol extract in lipopolysaccharide-exposed rats and Raw 264.7 cells. Plasma concentrations of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ were lower in Ixeris dentata-treated groups than in control group. Concentration of plasma IL-10 was higher in Ixeris dentata-treated groups than in control group. Concentrations of liver IL-$1{\beta}$ and IL-6 were lower in the Ixeris dentata-treated groups than in control group. However, concentrations of liver TNF-${\alpha}$ and IL-10 were not significantly different among all treatment groups. In the study using lipopolysaccharide-exposed Raw 264.7 cells, concentrations of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ tended to be decreased, but concentration of IL-10 tended to be increased in Ixeris dentata-treated groups. Plasm concentrations of total protein and albumin appeared to be increased in Ixeris dentata-treated groups.

Effects of Ixeris dentata ext. on Lowering Lipid and Anti-oxidation (씀바귀의 지질강하 및 항산화효과)

  • To investigate effects of Ixeris dentata EtOH ext. on lowering lipid levels and anti-oxidation activity, hyperlipidemic rats were treated with Ixeris dentata EtOH ext. and lipid levels and anti-oxdation activity were measured. The Ixeris dentata EtOH ext. groups showed low concentration of plasma FFA, plasma triglyceride, plasma total cholesterol, and plasma LDL-cholesterol compared to control group. However, concentration of plasma HDLcholesterol was not significantly different among all the treatment groups. The Ixeris dentata EtOH ext. groups showed lower level of liver total cholesterol, liver triglyceride, plasma TBARS, and liver TBARS than those of control group. The Ixeris dentata EtOH ext. groups also showed higher level of GSH-Px activity, SOD activity, and CAT activity than those of control group. Moreover, the Ext. showed lower level of TNF-${\alpha}$, Apo-B, Apo-E, and leptin expression than those of control group. The results in this study shows that the Ixeris dentata EtOH ext. have positive effect in lowering lipid level, and anti-oxidative activity.

Antioxidant and Anti-inflammatory Activities of Different Parts of Ixeris dentata According to Extract Methods (씀바귀 부위별 용매추출 방법에 따른 항산화 활성 및 항염증 효과 비교)

  • This study was to investigate the antioxidant and anti-inflammatory activity of leaf and stem of Ixeris dentate extract by hot water and 70% ethanol. The total flavonoid and polyphenol in 70% ethanol extract of leaf and stem of Ixeris dentate were significantly higher than those of hot water extracts. DPPH and ABTS free radical scavenging activity of 70% ethanol extract of leaf was higher than that of hot water extract of leaf. 70% ethanol extract of leaf was the highest on the DPPH and ABTS activity. Nitric oxide(NO) production of 70% ethanol extract was higher than that of hot water extract. According to this study, it was found that there was difference in physiological activity due to extraction solvents of each part of Ixeris dentate and it is believed that it can be used as an ingredient of functional foods using Ixeris dentate.

Inhibitory Effects of Ixeris Dentata on the Mutagenicity of Aflatoxin $B_1$, N-methyl-N연-nitro-N-nitrosoguanidine and the Growth of MG-63 Human Osteosarcoma Cells (씀바귀 추출물들의 돌연변이 유발 억제 및 MG-63 암세포 성장 저해 효과)

  • Ixeris dentata was extracted with methanol and then the methanol extract was further fractionated to hexane, chloroform, ethyl acetate, butanol and aqueous fraction. The methanol extract of lxeris dentata had the strong antimutagenic effect on the aflatoxin B1(AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Ames mutagenicity test and SOS chromotest. Among the solvent extracted fractions from the methanol extract, the chloroform fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity of AFB1 with inhibition rate of 74 percent. The methanol extract of Ixeris dentata also revealed the inhibitory effect on the growth of MG-63 human osteosarcoma cells after 6 days of breeding at 37℃. The chloroform fraction and the ethyl acetate fraction from the methanol extract of lxeris dentata were most effective and inhibited the growth of MG-63 cells by 97 and 93 percent, respectively. It is suggested that the inhibitory effects of lxeris dentata on the mutagenicity and the growth of MG-63 human osteosarcoma cells are strong in the lipid soluble fractions.

A Preliminary Study on Hypocholesterolemic and Hypoglycemic Activities of Some Medicinal Plants

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

Volatile flavor components of Ixeris dentata and Amaranthus mangostanus (씀바귀와 비름의 휘발성 풍미성분)

  • Volatile components of Ixeris dentata and Amaranthus mangostanus, Korean wild vegetables, were collected by Steam Distillation-Extraction(SDE) method. Essential oils were analyzed by gas chromatography(GC)and combined gas chromatography-mass spectrometry(GC-MS). Sixty one components, including 13 alcohols, 9 esters, 12 aldehydes, 6 ketones, 9 hydrocarbons, 6 acids, 3 phenols and 3 miscellaneous ones were confirmed in Ixeris dentata. Fifty six components, including 15 alcohols, 5 esters, 13 aldehydes, 8 ketones, 3 hydrocarbons, 9 acids and 5 miscellaneous ones were confirmed in Amaranthus mangostanus.

Ixeris dentata (Thunb) Nakai Attenuates Cognitive Impairment in Scopolamine-induced Amnesic Mice

  • The purpose of the current study is to investigate the cognition-enhancing effects of Ixeris dentata (Thunb) Nakai in scopolamine-induced amnesic mice. Scopolamine (2 mg/kg, i.p.) was used to induce amnesia in mice. The cognitive-enhancing activity of the IDE (10, 20 and $40{\mu}g/mL$) was studied by passive avoidance response, elevated plus maze and Y-maze behavioral paradigm in normal and scopolamine-induced amnesic mice. Scopolamine-induced cognitive deficits were significantly reversed by IDE (P < 0.001 at 20 mg/kg) in a dose-dependent fashion in all the behavioral paradigms tested. IDE possesses cognitive-enhancing properties in amnesic mice due to its potent antioxidant action.

A Sensory Evaluation of the Bitter Compounds from Ixeris dentata Nakai (관능검사를 통한 씀바귀의 쓴맛 연구)

  • Ixeris dentata Nakai (Compositae) is a perennial herb and the young shoots when prepared properly are edible as a bitter appetizing vegetable in the early spring. The whole plant has been used as a folk medicine in treating diabetes in Korea. As a part of our studies on bitter substances, we have investigated the bitterness of the plant through a taste-guided fractionation method. Bitter substances were extracted by the solvent fractionation and the bitterness was evaluated by 10 trained panelists consisted with graduate students of Dept. of Foods and Nutrition. It has been revealed that the bitter compounds were distributed throughout the extracting fractions and the higher intensity of the bitterness was seen in the n-BuOH fractions. The bitter compounds were subfractionated by the silica gel column chromatography and the bitterness was evaluated by the trained panelists. The subfractions which were indicated as the bitter compounds will be further studied to isolate the compounds. The H$_2$O fractions were grouped according to the molecular weight through the ultrafiltration and the group with 1,000 < m.w. < 30,000 was evaluated as the most intense bitter subfraction.

Analysis of growth characteristics and carotenoids and phenolic compounds contents of Ixeris dentata

  • Ixeris dentata is a perennial plant belongs to Compositae family and it has been considered as a potential therapeutic agent in many biological activities like detoxification, elimination of blood stasis, anti-inflammatory, and analgesia. The I. dentata used in this experiment was collected in four areas of Yangpyeong, Dangjin, Chuncheon and Goesan, then transplanted to the farm of Department of Herbal Crop Research in National Institute of Horticultural and Herbal Science. The growth characteristics of I. dentata were investigated after harvesting. The aboveground biomass of plants collected from Chuncheon area was exhibited best, and the biomass of root in YangPyeong was showed best as 3.65 g/plant in dry weight. Based on the HPLC analysis, the leaf of I. dentata collected from Dangjin area was highest in carotenoids contents ($1,213{\mu}g/g$ dry weight), and the leaf of Chuncheon area was exhibited maximal phenolic compounds ($1,918{\mu}g/g$ dry weight). The results of this study could provide fundamental formation for selection of standard varieties of I. dentata and development of functional material by analysis of the growth characteristics and the contents of carotenoids and phenolic compounds.
