Alveolar Soft Part Sarcoma of The Lung Diagnosed by Fine Needle Aspiration Cytology - A Case Report - (세침흡인 세포검사로 진단한 폐의 포상 연부육종 - 1예 보고 -)

  • Alveolar soft part sarcoma(ASPS) is a rare malignant neoplasm with a distinct clinicopathologic entity of which fine needle aspiration(FNA) cytologic findings have been described in only a few reports. Although patients usually present with an isolated soft-tissue mass in the extremity, metastasis can occur in about 13 % of total cases and the most frequent metastatic site is the lung. We have recently experienced a FNA cytologic case of ASPS in the lung. A 23-year-old female patient was admitted to this hospital due to 2-month-history of cough She had been good in health before the visit. Chest computed tomography revealed multiple, variable sized, bilateral pulmonary nodules. Physical examination and other staging work up revealed no other lesions except for pulmonary nodules. A percutaneous transthoracic FNA was performed from the pulmonary nodules. The smear was cellular and most cells were arranged singly. In addition, a few clusters lined by thin-walled vasculature with a pseudoalveolar pattern were present. Some of the tumor cells were large and polygonal lo oval with abundant granular or vacuolated cytoplasm. Most cells were naked nuclei showing finely granular chromatin pattern with prominent central nucleoli.

Fine Needle Aspiration Cytology of Matrix Producing Carcinoma of the Breast - A Case Report - (유방의 기질형성 암종의 세침흡인 세포학적 소견 - 1예 보고 -)

  • Matrix producing carcinoma of the breast is a variant of heterologous metaplastic carcinoma which is defined as "overt carcinoma with direct transition to a cartilaoenous and/or osseous stromal matrix without an intervening spindle cell zone or osteoclastic cells". This tumor is very rare, occuring in less than 0.2% of total breast carcinoma, but the prognosis is better than other metaplastic carcinoma. We experienced a case of fine needle aspiration(FNA) cytologic finding of matrix producing carcinoma of the breast. A 75-year old woman, who presented a right huge breast mass$(9{\times}8cm)$ during 10months, was examined. Mammography reveals right lateral mass with even density without calcification. Breast ultrasonography shows multifocal hypoechogenic cystic change in the huge mass, suggesting resolving hematoma or carcinoma or sarcoma with necrosis. On cytologic finding of FNA, myxoid matrix was the dominant feature and the rest of the material was composed of scanty isolated atypical cells with large irregular nuclei. The histologic finding was moderately differentiated adenocarcinoma with abundant cartilagenous matrix and focal squamous metaplasia.

The Effect of Cadmium Administration in Seminiferous Epithelium of Mouse Testes : Electron Microscopic Study (카드뮴 투여가 생쥐정소의 정소관상피에 미치는 영향 : 전자현미경적 연구)

  • This study was carried out to investigate the effects of cadmium chloride on the spermatogenesis of male mouse. Cadmium chloride was administered as a single dose of 5mg/kg body weight by intraperitoneal injection. The testes were isolated from the experimental animals at 3 hours, 8 hours, 12 hours, and 24 hours respectively after administration of cadmium chloride. The major changes in ultrastructures of the seminiferous tubules observed after cadmium chloride administration include dilation of smooth endoplasmic reticulum, swelling of mitochondria and vacuolation in cytoplasm of the germ cells. Especially, cadmium chloride caused direct damages to spermatogonia such as degeneration of nuclei, nuclear membrane and plasma membrane. In addition, necrotic changes were observed in most germ cells at 24 hours after cadmium chloride administration. Therefore, it seems clear from these results that cadmium chloride induces acute irreversible degenerative changes in the seminiferous tubules of the mouse testis, so that the cadmium chloride ultimately causes necrosis in germ cells at all stages of the spermatogenesis.

Isolation and characterization of fowl adenovirus serotype 4 from chickens with hydropericardium syndrome in Korea

  • Four strains of fowl adenovirus (FAdV) were isolated from 4 flocks of broiler or layer chickens affected by hydropericardium syndrome in Korea. These FAdVs were classified as serotype 4 by restriction fragment length polymorphism patterns of hexon genes and whole genomes. The virus exhibited cytopathic effects consisting of rounding, ballooning and clustering in primary chicken embryo liver cell cultures. In transmission electron microscopy, virus particles in hexagonal shape aggregated exclusively in the nuclei of hepatocytes of the chickens as the typical appearances in adenovirus infections. Buoyant density of the virus in cesium chloride (CsCl) was 1.34 g/mL. The virus was stable to chloroform, ether, 50~70% ethanol, acidic condition at pH 3, 0.25% trypsin (1 : 250), heat at $50^{\circ}C$ for 30 min, but labile to 100% ethanol, heat at $52{\sim}60^{\circ}C$ for 30 min, 1 M $MgCl_2$ at $50^{\circ}C$ for 1 h, 1 : 2,000 formalin (37%). All of the physicochemical properties pertained to the characteristics of adenoviruses. Eight viral polypeptides were determined in CsCl-purified virus by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

  • Cell fusion between complementary mutants isolated from astaxanthin-producing yeast, Phaffia rhodozyma, was carried out to obtain astaxanthin-overproducing strains by protoplast fusion technique. The frequency of protoplast fusion was ranged from 2.3$\times$10-5 to 6.0$\times$10-5, and nuclear fusion in the cells of hybrids was demonstrated by several techniques such as isolation of recombinants after mitotic segregation of parental genetic markers, estimation of DNA content, direct observation of nuclei with nuclear staining, and comparison of survival rate to UV exposure. One of several hybrids, Fl, showed approximately 3-fold increase in astaxanthin content when compared with wild parent.

(-) Epigallocatechin gallate restores ethanol-induced alterations in hepatic detoxification system and prevents apoptosis

  • The present study was designed to estimate the protective effect of (-) epigallocatechin gallate (EGCG) on ethanol-induced liver injury in rats. Chronic ethanol administration (6 g/kg/day ${\times}$ 60 days) caused liver damage that was manifested by the elevation of markers of liver dysfunction - aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, bilirubin and ${\gamma}$-glutamyl transferase in plasma and reduction in liver glycogen. The activities of alcohol metabolizing enzymes such as alcohol dehydrogenase and aldehyde dehydrogenase were found to be altered in alcohol-treated group. Ethanol administration resulted in the induction of cytochrome p450 and cytochrome-$b_{5}$ activities and reduction of cytochrome-c reductase and glutathione-S-transferase, a phase II drug metabolizing enzyme. Further, ethanol reduced the viability of isolated hepatocytes (ex vivo) as assessed by trypan blue exclusion test and induced hepatocyte apoptosis as assessed by propidium iodide staining. Treatment of alcoholic rats with EGCG restored the levels of markers of liver injury and mitigated the alterations in alcohol metabolizing and drug metabolizing enzymes and cyt-c-reductase. Increased hepatocyte viability and reduced apoptotic nuclei were observed in alcohol + EGCG-treated rats. These findings suggest that EGCG acts as a hepatoprotective agent against alcoholic liver injury.

  • Hydrogen has potential for preventive and therapeutic applications as an antioxidant. However, micro- and macroparticles of hydrogen in water disappear easily over time. In order to eliminate reactive oxygen species (ROS) related with the aging process, we used functional water containing nanoparticle hydrogen. Nanoparticle hydrogen does not disappear easily and collapse under water after long periods of time. We used murine embryonic fibroblasts that were isolated from 12.5-day embryos of C57BL/6 mice. We investigated the ability of nanoparticle hydrogen in water to suppress hydroxyurea-induced ROS production, cytotoxicity, and the accumulation of ${\beta}-galactosidase$ (an indicator of aging), and promote cell proliferation. The accumulation of ${\beta}-galactosidase$ in the cytoplasm and the appearance of abnormal nuclei were inhibited by daily treatment of cells with hydrogen water. When the aging process was accelerated by hydroxyurea-induced oxidative stress, the effect of hydrogen water was even more remarkable. Thus, this study showed the antioxidant and anti-senescence effects of hydrogen water. Nanoparticle hydrogen water is potentially a potent anti-aging agent.

Studies on Ultrastructure and Virus Infection of Aspergillus ochraseus (Aspergillus ochraseus의 미세구조(微細構造) 및 바이러스 감염(感染)에 관(關)한 연구(硏究))

  • These studies were carried out to detect the presence of infected virus- like particles and also were observed the ultrastructures of Aspergillus ochraseus isolated from kokja and Korean ginseng. The results of ultrastructures of Aspergillus ochraseus are summarized as follows: 1. In fungal cells, nuclei were enclosed by a irregular double membrane and nucleoli in the nucleus. 2. In cytoplasm, mitochondria, rough endoplasmic reticulum with ribosomes and glycogen were scattering distributed and many lomasomes also observed. 3. The osmiophilic bodies of fungal cells existed in the vesicles. 4. The cell walls were composed of a low electron dense materials. 5, Conidia cell walls were extremely thick and possessed the high electron density of outer coat. The virus-like particles were observed in the hyphae of Penicillium chrysogenum Q-176. These virus-like particles measured $350{\AA}$ in diameter. But strains of Aspergillus ochraseus, showing some vesicle particles were also observed about $800{\AA}$ in diameter in the central region of young fungal hyphae. Based on the results of these experiments, it can not be determined virus particles or not. The further studies to determination of virus particles will be proceeded by the chemical, physical and biological assay methods.

Chromatin Structural Rearrangement during Dedifferentiation of Protoplasts of Cucumis sativus L.

  • This paper reports on the structural rearrangement of satellite DNA type I repeats and heterochromatin during the dedifferentiation and cell cycling of mesophyll protoplasts of cucumber (Cucumis sativus). These repeats were localized in the telomeric heterochromatin of cucumber chromosomes and in the chromocenters of interphase nuclei. The dramatic reduction of heterochromatin involves decondensation of subtelomeric repeats in freshly isolated protoplasts; however, there are not a great many remarkable changes in the expression profile. In spite of that, reformation of the chromocenters, occurring 48 h after protoplast isolation, is accompanied by recondensation of satellite DNA type I; however, only partial reassembly of these repeats was revealed. In this study, FISH and a flow cytometry assay show a correlation between the partial chromocenter and the repeats reassembly, and with the reentry of cultivated protoplasts into the cell cycle and first cell division. After that, divided cells displayed a higher variability in the expression profile than did leaves' mesophyll cells and protoplasts.

Liver Plasma Membrane and Nuclear $T_{3}$ Receptor Binding in the Obese (ob/ob) Mouse (비만 쥐(ob/ob mouse)의 간 세포막과 핵에 있는 $T_{3}$ 수용체의 결합능력에 관한 연구)

  • $L-Triiodothyronine(T_3)$ binding to purified plasma membrane and to isolated nuclei from the same liver in obese(ob/ob) mice and their lean littermates was examined. The maximal binding capacity(Bmax) for $T_3$ receptor of liver nuclei, as compared to lean control, was significantly lower in the obese mouse$(obese 527{\pm}80fmol/mg\;DNA ; lean 883{\pm}62fmol/mg\;DNA)$, without an apparent difference in dissociation constant(Kd). The finding that obese mice have fewer liver nuclear $T_3$ receptors confirms previous reports. The Bmax and Kd of liver plasma membrane $T_3$ receptor were not significantly different between obese and lean mouse, which suggests no defect to be occurring in the function of the plasma membrane $T_3$ receptor and reinforces the view that the peripherally impaired thyroid hormone action in obese mice is a post plasma membrane receptor event. These results further support the hypothesis that the major defect of the thyroid hormone metabolism in genetic obesity occurs at the level of the nuclear receptor.
