Characteristics of Fibrinolytic Enzymes of Bacillus licheniformis CY-24 Isolated from Button Mushroom Compost (양송이 배지로부터 분리한 Bacillus licheniformis CY-24의 섬유소분해 효소의 특성)

  • The present study was performed to improve the technique used for fermenting the mushroom growth medium. Taxonomic analysis of 16S rDNA sequence from the predominant Bacillus strain CY-24 isolated during the fermentation phase of the rice straw medium identified it as Bacillus licheniformis. In addition, the growth environment of B. licheniformis was also examined in this study, which revealed the optimal growth temperature and pH to be 30 ℃ and 6.0, respectively. This study also revealed that carboxymethyl cellulase (CMCase) and polygalacturonase (PGase) enzymes isolated from B. licheniformis achieved their maximal activities at 50 ℃ and 60 ℃ respectively. Furthermore, the study confirmed that the two enzymes, i.e., CMCase and PGase in B. licheniformis are stable at temperatures above 60 ℃. The present study thus demonstrates that B. licheniformis CY-24 possesses excellent enzymatic properties. It also reveals that the action of enzymes during the production of growth mediums used for the cultivation of mushrooms is closely associated with the promotion of fermentation and softening of the rice straw. Overall, this study provides elementary information regarding the role of B. licheniformis enzymes during growth medium fermentation for Agaricus bisporus cultivation.

Isolation and identification of a tricin 4"-O-(threo-β-guaiacylglyceryl) ether producing microorganism from germinated rice (발아 벼로부터 tricin 4"-O-(threo-β-guaiacylglyceryl) ether 생성균주의 분리 및 동정)

  • This study was conducted to isolate and identify a microorganism that increases tricin-O-(threo-${\beta}$-guaiacylglyceryl) ether (TTGE) content in the hulls of rice (Oryza sativa L.). Bacteria from germinated rice were isolated by enrichment cultivation using yeast mold, luria bertani, potato dextrose and mannitol egg york polymyxin broths. The highest increase in TTGE content ($339.30{\mu}g/g$) was achieved by a microorganism isolated by PDA enrichment cultivation. On the basis of 16S RNA sequence homology and phylogenetic analysis, the isolated bacterium was identified to have 100% similarity with Burkholderia vietnamiensis. The isolated bacteria were short rods, negative for the Gram stain, and positive for the catalase test. The highest TTGE level was $435.86{\mu}g/g$ in 72-h fermented samples, representing a 2.5x increase compared with the control ($175.65{\mu}g/g$). In conclusion, the bacterium isolated from germinated rice extract was Burkholderia vietnamiensis, and the optimum fermentation period to maximize TTGE levels was 72 h. These findings might help in developing functional materials using rice hulls, a waste product of rice milling.

  • This experiment investigates the characteristics of microorganisms isolated from a medium during cultivation process and reveals the relationship between these microorganisms and the growth of Agaricus bisporus. The domestically grown strains of Agaricus bisporus displayed a higher inhibition growth rate against microorganisms isolated from straw, chicken manure, and medium than imported strains. As for inhibition of mycelial growth among mushroom cultivars of the microorganisms separated by each fermentation step from the mushroom medium, the domestic cultivar, 'Saedo,' grew more vigorously among other cultivars. As the fermentation progressed, it was confirmed that inhibitation of microorganisms against Agaricus bisporus was weakened. A total of 21 strains of microorganisms that promote mushroom growth were isolated in the 4th turning process, and the microorganisms isolated from the mushroom medium affect the growth and as yield of the mushroom through secretory substances.

Impact of Virus-resistant Trigonal Cactus Cultivation on Soil Microbial Community (바이러스저항성 삼각주 재배가 토양 미생물상에 미치는 영향)

  • BACKGROUND: Genetically modified(GM) trigonal cactus(Hylocereus trigonus Saff.) contained a coat protein gene of cactus virus X (CVX), which conferred resistance to the virus, phosphinothricin acetyltransferase (bar) gene, which conferred herbicide resistance, and a cauliflower mosaic virus 35S promoter (CaMV 35S). This study was conducted to evaluate the possible impact of GM trigonal cactus cultivation on the soil microbial community. METHODS AND RESULTS: Microorganisms were isolated from the rhizosphere of GM and non-GM trigonal cactus cultivation soils. The total numbers of bacteria, and actinomycete in the rhizosphere soils cultivated GM and non-GM trigonal cactus were similar to each other, and there was no significant difference. Dominant bacterial phyla in the rhizosphere soils cultivated with GM and non-GM trigonal cactus were Proteobacteria, Uncultured archaeon, and Uncultured bacterium. The denaturing gradient gel electrophoresis (DGGE) profiles show a similar patterns, significant difference was not observed in each other. DNA was isolated from soil cultivated GM and non-GM trigonal cactus, we analyzed the persistence of the inserted gene by PCR. Amplification of the inserted genes was not observed in the soil DNA, which was collected after harvest. CONCLUSION(S): This result suggests that the GM trigonal cactus cultivation does not change significantly the microbial community.

  • The yeast strain was isolated from food industry waste water and its identification and biological characteristics were investigated. The optimum condition for cultivations and its activities for the reduction of B.O.D. on the food industry waste water were also confirmed. The results are as follows; 1) The isolated was identified as Candida curvata. 2) Candida curvata grew well in all of the experimented media, so and it can be regarded as a useful strain in the treatment of food industry waste water. 3) There was only a slight difference in the induction period between sterilized cultivation and unsterilized cultivation. But in the ice cream waste water, the period was considerably longer in unsterilized cultivation. 4) Specific rate of growth of Candida curvata in sugar waste water was 0.50/hr, ice cream waste water 0.50/hr, and beer waste water 1.0/hr. 5) Increasing of innoculum reduced the induction period in unsterilized cultivation. 6) The amount of dried yeast from sugar waste water were $175mg/{\ell}$, ice cream waste water $628mg/{\ell}$, and beer waste water $857mg/{\ell}$. Crude protein content in the dried yeast from sugar waste water were 52%, ice cream waste water 54%, and beer waste water 54%. 7) The rate of BOD reduction in sugar waste water were 49%, ice cream waste water 80%, and beer waste water 64%.

Characterization of Photosynthetic Bacteria in Swine Wastewater (양돈 계수에서 광합성 미생물의 특성)

  • Photosynthetic bacteria (PSB), strains IP-4 and IP-6-7 were isolated from nature, and were studied for swine wastewater treatment. It was confirmed that these PSB were very effective for the COD treatment. Growth of those PSB were increased to 4.5 fold in organic-acids (acetate, propionate and butyrate) added medium than cultivation in Lascelles basal medium and the amount of bacteriochlorophyll a were increased to 5 folds. The COD removal rate in swine wastewater using PSB, strains IP-4 and IP6-7 were obtained 91% and 85%, respectively.

The saponin contents of cultivated Anemarrhena asphodeloides B$_{UNGE}$ in Korea (재배지모의 saponin 함량에 관한 연구)

  • Optimum condition for cultivation of Anemarrhena asphodeloides B$_{UNGE}$ and seasonal variation of saponin and sapogenin contents in the rhizome were investigated. It is confirmed that the best harvest time is 3rd year. Average contents of saponin in rhizome was 8.5% and 5.0%, in fibrous roots, being highest in winter. The facotrs of saponin content variation were seemed to be temperature and humidity. It was also proved that asphonin was the mixture of timosaponin A-III, timosaponin A-I and mangiferin. Mangiferin and isomangiferin were isolated from the flower.

  • Seven bacterial strains capable of conversing Gardenia irridoidglucoside into blue color was isolated on nutrient agar plates with 0.1% water extracted solution of Gardenia's dryad seed. In the seven, strain No. C2 was most effective in the production of blue color. The optimal conditions in production of blue color were when initial pH of medium was 7.0 and cultivation temperature was 35$^{\circ}C$. In 5 $\ell$-Jar fermantor, the powder of blue color was produced about 15% (W/W). And the color was relatively stable in our test.

Characteristics and breeding of a new brown variety 'Geumhyang' with short cultivation period in Flammulina velutipes (재배기간이 짧은 갈색 팽이버섯 신품종 '금향'의 육성 및 특성)

  • 'Geumhyang', a new variety with short cultivation period and yellowish ivory color in Flammulina velutipes, was bred by crossing between monokaryons isolated from 'Garlmoe' and wild strain 'CBMFV-33'. In bottle cultivation, the period of mycelial growth, primordia1 formation, and growth of fruit body was required 23 days, 7 days, and 17 days, respectively. Total culvivation period was 47 days, showing 7 days shorter than that of 'Garlmoe'. Pileus diameter was greater than that of 'Garlmoe', and pileus color showed yellowish ivory. Fruit body production per 850 ml bottle was 160 g, which was 8% higher than that of 'Garlmoe'. The taste, derived from 'Garlmoe' and wild strain, was excellent because of better chewing feeling than white ones. 'Geumhyang' with differentiated color and taste, has an advantage to target a niche market of white ones.
