Determination of Free Amino Acids in Isatidis Radix By HILIC-UPLC-MS/MS

  • A rapid, accurate and precise method for the determination of 22 amino acids in Isatidis Radix by Hydrophilic Interaction Ultra-High-Performance Liquid Chromatography Coupled with Triple-Quadrupole Mass Spectrometry (HILIC-UPLC-MS/MS) was established. Chromatographic separation was carried out on a Acquity UPLC BEH Amide column ($2.1mm{\times}100mm$, $1.7{\mu}m$) with gradient elution of acetonitrile (containing 0.05% formic acid and 2 mM ammonium formate) and water (containing 0.15% formic acid and 10 mM ammonium formate) at a flow rate of 0.4 mL/min; Waters Xevo$^{TM}$ TQ worked in multiple reaction monitoring mode. All components were separated in 17 min. All calibration curves were linear ($R^2$ > 0.991) over the tested ranges. The limits of detection (LOD) and limits of quantitation (LOQ) for these compounds were 0.21-79.55 and 0.72-294.23 ng/mL, respectively. The average recoveries were in the range of 93.75-104.16% with RSD value less than 6.56%. Therefore, this method could be an alternative assay for the determination of 22 amino acids in Isatidis Radix due to its rapidness, sensitivity, less sample and solvent consumption.

Review of Clinical Studies of Herbal Medicine Treatment for Molluscum Contagiosum (전염성 연속종의 한약 치료에 대한 임상 연구 동향 - 2000년대 이후 발표된 임상 연구 논문을 중심으로 -)

  • Objectives The purpose of this study is to investigate recent clinical studies on the effect of herbal medicine in molluscum contagiosum in China, and to seek better methods to treat and study for molluscum contagiosum in Korea. Methods We searched the clinical studies from the China Academic Journal (CAJ) in China National Knowledge Infrastructure (CNKI) by key words '傳染性軟?', '治療', '中?治療', '中藥' from January 2001 to August 2018. We analyzed the literature in regards to the treatment methods and results. Results Among the 103 searched studies, 9 randomized controlled trials, 2 controlled clinical trials, and 16 case studies were selected and analyzed. In most of the studies, the effectiveness of herbal medicine on molluscum contagiosum was significantly high. The most commonly used herbs for oral administration were Isatidis Radix (板藍根), Coicis Semen (薏苡仁), Cyperi Rhizoma (香附子), Equiseti Herba (木賊), Poria (茯?), Lithospermi Radix (紫草), Isatidis Folium (大靑葉) and Lonicerae Flos (金銀花). The most commonly used herbs for external applications were Isatidis Radix (板藍根), Lonicerae Flos (金銀花), Cyperi Rhizoma (香附子), Equiseti Herba (木賊), Dictamni Radicis Cortex (白鮮皮), Isatidis Folium (大靑葉) and Brucea javanica (鴉膽子). Conclusions Based on the results of the clinical studies from China, the use of Korean medicine for treatment of molluscum contagiosum has been shown to be effective in relieving symptoms. Based on the result of this study, it will be possible to widen the scope of Korean medicine on infectious disease, such as molluscum contagiosum. Additional clinical studies and experimental studies need to be performed to solidify these findings.

Experimental Study on the Antiinflammatory Activities of Bojeasodok-um subtracted Scrophulariae Radix, Lasiosphaera seu Calvatia, Isatidis Radix added indigo Naturalis, Lithospermi Radix (보제소독음가감방(普濟消毒飮加減方)의 소염작용(消炎作用)에 관한 실험적(實驗的) 연구(硏究))

  • Objective : Erysipelas is an acute inflammation caused by pyogenic bacteria. This mainly involves the upper part of dermis. It begins as erythematous patches with tenderness, followed by fever, headache, chills and fatigue etc. It may results in edema, obstruction of lymphatics and sepsis. So this experiment is carried out for test whether the Bojeasodok-um subtracted Scrophulariae Radix, Lasiosphaera seu Calvatia, Isatidis Radix added indigo Naturalis, Lithospermi Radix have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by Erysipelas. Method : Experimental animals made use of 4-5 week-age(weight 20-25g) ICR(male) mouse. In the breeding farm, the lighting time was controlled from 7:00 am till 7:00 pm, the temperature was controlled So we concluded that BS is prospected as an anti-inflammatory agent to cure inflammation induced bywithin 18-23$23^{\circ}C$ and water and food were not limited.The freezing lyophilization powder which were extracted from Bojesodok-Um divided low dose group(200mg/kg-BSL) and high dose group(500mg/kg-BSH) and after melting in water, it was orally administered to the mouse. Compared with inflammation induced group which were induced by triggering-inflammation reagent Carageenan and Zymosan and normal contrast group, we measured the edema decrement effect,macrophage and spleen cell activation. Result : 1. BS has suppress inflammatory reaction induced by Carageenan. 2. BS has suppress increasing activation of abdominal cavity macrophage in the Carageenan and Zymosan induced inflammation. 3. BS has suppress increasing activation of spleen cell in the Carageenan and Zymosan induced inflammation. Based on the above result, BS was improved its suppression effect to the inflammatory reaction through the suppression of spleen cell and macrophage activation. So we concluded that BS is prospected as an anti-inflammatory agent to cure inflammation induced by Erysipelas.

Effects of Isatidis Radix and it's Active Component, Tryptanthrin on the Production of Inflammatory Mediators in Lipopolysaccharide-activated Raw264.7 Cells (LPS로 활성화된 Raw264.7 cell에서 판람근 및 Tryptanthrin의 염증매개물질억제효과)

  • Objectives : 판람근(板藍根)은 십자화과에 속하는 대청(大靑) 또는 숭남의 근(根)을 건조한 것이다. 본 연구는 판람근(板藍根)이 청열해독(淸熱解毒)함에 근거하여, LPS로 활성화된 Raw264.7 cell에서 판람근(板藍根)과 그 성분중의 하나인 tryptanthrin이 염증매개물질에 미치는 효과를 살펴보고자 하였다. Methods : 세포생존율은 MTT, nitric oxide (NO)는 Griess reagent를 사용하여 측정하였으며, 각 단백질의 발현량은 Western blot 방법을 사용하였으며, cytokine 및 cyclooxygenase-2 (COX-2)는 ELISA방법을 사용하여 측정하였다. Results : LPS는 NO 및 prostaglandin E2 (PGE2)를 유의하게 상승시켰으며, 판람근(板藍根)추출물 (IRE) 및 tryptanthrin 은 이들을 유의하게 억제하였다. 그러나 판람근(板藍根)의 또 다른 성분인 indigo는 유의한 결과를 나타내지 못하였다. IRE와 tryptanthrin은 inhibitory kappa B alpha의 인산화를 억제하여, nuclear factor-${\kappa}$B (NF-${\kappa}$B)의 핵으로의 전위(轉位)를 억제하여, iNOS 및 cytokine을 억제하였다. IRE와 tryptanthrin의 PGE2 억제는, COX-2의 발현억제에서가 아니라, COX-2의 활성을 억제함에서 기인하였다. Conclusion : 이러한 결과는 판람근(板藍根)이 NF-${\kappa}$B pathway를 경유하여 iNOS의 발현 및 COX-2의 활성을 억제함을 나타내며, 이러한 판람근(板藍根)의 항염증효능은 일부 tryptanthrin의 작용에서 기인함을 시사한다.

Review of Clinical Studies for Herbal Medicine Treatment on Flat Warts (편평 사마귀의 한약치료에 대한 임상 연구 동향 - 중의학논문을 중심으로)

  • Objective The purpose of this study is to investigate recent clinical studies on the effect of herbal medicine in flat warts in China, and to seek better methods to treat and study for flat warts in Korea. Methods The clinical studies from the CAJ (China Academic Journal) of CNKI (China National Knowledge Infrastructure) by key words '扁平疣' in recent 5 years (From January 2011 to December 2015) were searched. Results 16 articles were selected from 122 clinical studies, and analyzed. The most commonly used herbs were Isatidis Radix (板藍根), Equiseti Hiemalis Herba (木賊), Cyperi Rhizoma (香附子), Carthami Flos (紅花), Glycyrrhizae Radix (甘草) etc., and the articles all showed effectiveness over the control group. Conclusion Based on analyzing the studies, herbal medicine has shown effective in flat wart. This study may be useful not only as clinical data, but also to determine better treatment going forward.

  • Chinese herbs have been used for a long period of time and less side effects than synthesized chemical drugs. Therefore, using Chinese herbs as natural additives in cosmetics becomes popular in recent years. The methanol extracts of Scutellariae Radix, Lithospermi Radix, Lonicerae Flos, Andrographitis. Herba, Angelicae Dahuricae Radix, Ligustici Rhizoma et Radix, Hedyotis Diffusae Herba, Isatidis Folium, Magnoliae Liliflorae Flos, Forsythiae Fructus, Anmarrhenae Rhizoma, Spirodelae Herba, Gardeniae Fructus, Sophorae Flavescentis Radix, Coptidis Rhizoma, Prunellae Spica, Equiseti Hiemalis Herba, Gentianae Radix, Moutan Radicis Cortex, Fraxini Cortex, Lycii Radicis Cortex, Violae Herba, Lophatheri Herba, Matricariae chamomillae Flos, Taraxaci Herba and Scutellariae Barbatae Herba are used to test the efficiency of inhibiting acne pathogens. Twenty-six Chinese herbs are extracted by methanol, and then condensed to dried powder. These extracts are divided into water-soluble part and DMSO soluble part. These two type solutions are tested for the effect on acne pathogens by paper disc diffusion method. The results show that the substances of water soluble part which are Coptidis Rhizoma, Moutan Radicis Cortex, Scutellariae Barbatae Herba have medium to high activity of inhibiting acne pathogents, and the substances of DMSO soluble part which are Coptidis Rhizoma, Ligustici Rhizoma et Radix, Sophorae Flavescentis Radix, Moutan Radicis Cortex, Scutellariae Radix, Scutellariae Barbatae Herba also have medium to high activity of inhibiting acne pathogens. Using Chinese herbs as natural additives in cosmetics is convenience and valuable application in cosmetceutical research and development. Therefore, it is worth that re-investigation and find out the potential of Chinese herbs being use in cosmetics.

Review of Recent Clinical Studies of Herbal Medicine Treatment for Pediatric Tonsillitis - Focused on Chinese Randomized Controlled Trials - (소아 편도염의 한약 치료에 대한 최신 임상 연구 동향 - 중국 RCT 연구를 중심으로 -)

Methods We searched clinical studies from the China National Knowledge Infrastructure (CNKI) by search formula (SU='扁桃体炎'+'扁桃炎'+'扁桃腺炎'+'乳蛾'+'喉蛾'+'蚕蛾'+'石蛾'+'珠蛾'+'肉蛾'+'乳蛾風'+爛乳風'+'tonsillitis') and (SU='中樂'+'中医樂'+'本草'+'湯'+'丸'+'散'+'方'+'顆粒'+'膠囊'+'自擬'+'herb'+'herbal'+'decoction'+'remedy'+'Chinese medicine'+'Korean medicine'+'kampo'+'formula'+'herbal drug'+'Chinese drug'+'plant'+'Chinese prescription'+'traditional medicine'+'Medicine, East Asian Traditional'+'Herbal Medicine') in professional search from January 2016 to August 2020. We analyzed the literature focusing on the treatment methods and results. Results Among 1464 searched studies, 35 randomized controlled trials were selected and analyzed. In most studies, the effectiveness of oral administration of herbal medicine on pediatric tonsillitis was significant. Most commonly used herbs were Fructus Forsythiae (連翹), Radix Glycyrrhizae (甘草), Radix Scutellariae (黃芩), Herba Menthae (薄荷), Fructus Arctii (牛蒡子), Radix Scrophulariae (玄蔘), Radix Platycodi (桔梗), Flos Lonicerae (金銀花), Radix Isatidis (板藍根), Radix Bupleuri (柴胡), Fructus Gardeniae (梔子), Rhizoma Belamcandae (射干), Radix et Rhizoma Rhei (大黃), Gypsum Fibrosum (石膏). Conclusions By analyzing the improvement of indicators such as total effective rate, cured rate, symptom disappearance time and symptom score, we found that herbal medicine treatment can help improve pediatric tonsillitis. However, additional studies are needed to solidify these findings.

Medicinal herb extracts ameliorate impaired growth performance and intestinal lesion of newborn piglets challenged with the virulent porcine epidemic diarrhea virus

  • The objective of this study was to evaluate effects of a combined use of extracts of medicinal herbs Taraxaumi mongolicum, Viola yedoensis Makino, Rhizoma coptidis, and Radix isatidis (MYCI) on porcine epidemic diarrhea (PED). Twenty-two 3-day-old piglets received an oral challenge with $3{\times}10^{3.5}$ $TCID_{50}$ of the virulent PED virus (PEDV) in PBS or PBS only and daily oral administration of 60 mg of the MYCI mixture suspended in milk replacer or the vehicle for 7 days in a $2{\times}2$ factorial arrangement of treatments. Average daily gain (ADG) increased (p < 0.05) in response to the MYCI treatment in the PEDV-challenged piglets (-18 vs. 7 g for the vehicle- vs. MYCI-administered group), but not in unchallenged animals (27 vs. 28 g). Diarrhea score and fecal PEDV shedding, however, were not influenced by the MYCI treatment. The PEDV challenge caused severe intestinal villus atrophy and crypt hyperplasia, both of which were alleviated by administration of the MYCI mixture as indicated by an increase in the villus height and a decrease in the crypt depth due to the treatment. Overall, medicinal herb extracts used in this study ameliorated impaired growth performance and intestinal lesion of newborn piglets challenged with the virulent PEDV. Therefore, our results suggest that the MYCI mixture could be used as a prophylactic or therapeutic agent against PED.
