Effects of Low Incident Energy Levels of Infrared Laser Irradiation on the Proliferation of Candida Albicans Part III : A Study on the Interval of Irradiation

  • This study was performed to investigate the biostimulation effects of low level laser therapy (LLLT) on the fungus, Candida alvicans, according to the interval of irradiation during the cell cycle. Samples were divided into 5 groups which were P7-2Hr, P7-4Hr, P15-2Hr, P15-4Hr and Co. Sample was irradiated for 1 minute with 2 hours or 2 minutes with 4 hours of elapsed time during 28 hours of the cell cycle of Candida albicans, and the optical density was assessed by spectrophotometry every 2 hours. It was found that there was significant difference, in the cell growth, between 2 hour and 4 hour interval laser irradiation, although the total energy of each group using identical pulse during exam was equal.

Effect of 8 mW 525 nm LEDs Light Irradiation on the Defect Reduction in the Skin Wound of SD-rat

  • The purpose of this study is to develop the Photodynamic Therapy Equipment for medical treatment. We developed the equipment which was helpful in palpating wound healing by using 525 nm LEDs. The equipment was assembled with a micro-controller and green color LEDs, and designed to enable us to control light irradiation time, intensity and so on. In this study, the designed device was used to find out how 525 nm LEDs light affected the skin wound of SD-Rat(Sprague-Dawley Rat). We divided the participants into two groups; irradiation group which was irradiated one hour a day for 9 consecutive days, and none irradiation group. The results showed that the study group had lower incidence of inflammation and faster recovery, compared with the control group.

  • The liquid crystal (LC) alignment and pretilt angle generation using photo-alignment method were investigated in this study. We obtained that the monodomain alignment of nematic(N) LC increases with increasing irradiation time in a cell with linearly polarized ultraviolet (LPUV) light irradiation on polyimide (Pl) surfaces. We consider that the monodomain alignment is able to be anisotropic dispersion force due to photo-depolymerization of polymer with LPUV light irradiation on Pl surface. Also, we observed that the uniform alignment of NLC is obtained in a cell with normally LPUV light irradiated on Pl surface at 30 min. and then oblique LPUV is secondly irradiated on Pl surfaces rotated by 90$^{\circ}$. The pretilt angle of NLC is generated about 1.5$^{\circ}$ with oblique angle 60$^{\circ}$.

  • A varying degree of eccentricity always exists in the initial configuration of a nuclear fuel rod. Its impact on traditional LWR fuel is limited as the radial gap closes relatively early during irradiation. However, the effect of misalignment is expected to be more relevant in rods with highly conductive fuels, large initial gaps and low conductivity filling gases. In this paper, we study similar characteristics in the experimental setup of two fuel disc irradiation campaigns carried out in the OECD Halden Boiling Water Reactor. Using the multi-dimensional fuel performance code OFFBEAT, we combine 2-D axisymmetric and 3-D simulations to investigate the effect of eccentricity on the fuel temperature distribution. At the same time, we illustrate how the advent of modern tools with multi-dimensional capabilities might further improve the design and interpretation of in-pile separate-effect tests and we outline the potential of such an analysis for upcoming experiments.

Sequence and Time Interval in Combination of Irradiation and Cis-Diamminedichloroplatinum in C3H Mouse Fibrosarcoma (C3H 마우스 섬유육종에 있어서 방사선 조사와 Cis-diamminedichloroplatinum의 병용시 순서 및 시간간격의 영향)

  • Experiments have been carried out with C3H mouse fibrosarcoma (FSa II) to determine the effect of different sequence and time intervals between irradiation and administration of cis-diammihedichloroplatinum (cis-DDP) with gross tumors (6 mm in diameter), microscopic tumors (3 days after transplantation of $10^3$ cells) and cells in culture. The drug was administered either 24, 12, 8, 4, 2, 1, 0.5 hour before irradiation, immediately before irradiation, or 0.5, 1, 2, 4, 8, 12, 24 hours after irradiation. In case of in vivo studies, tumor growth delay was used as an end point. Clonogenic cell surviving fraction was used for in vitro studies. Tumor growth delay for gross tumor after 10 Gy radiation plus 10 mg/kg cis-DDP ranged from 6.3 to 10.66 days and the enhancement ratio ranged from 1.37 to 2.23. The most effective combination was when cis-DDP was given 4 hours before irradiation. Tumor growth delay for microscopic tumor after 5 Gy of radiation and 5 mg/kg of cis-DDP ranged from 3.55 to 11.98 days with enhancement ratio from 2.05 to 6.92. Microscopic tumors showed response significantly greater than additive in every time interval and the most effective treatments were when cis-DDP was given 2 and 1 hour before irradiation. In in vitro experiment, the surviving fraction after 6 Gy of radiation and 1 hour exposure to 4 ${\mu}M$ cis-DDP fluctuated as a function of time between treatments, but the difference between maximum and minimum surviving fractions was very small. According to the above results the sequence and time interval between irradiation and chemotherapy is very critical especially for the management of microscopic tumors as in the case of postoperative adjuvant treatment.

Optimization of Iso-flavonoids Extraction Process from Kudge Using Ultrasonic Irradiation Energy (초음파에너지를 이용한 칡으로부터 이소플라보노이드의 추출공정 최적화)

  • In this study, we used the ultrasonic extraction process as a method to extract antioxidant substances from kudzu, and measured the content of iso-flavonoids puerarin, daidzein, daidzin contained in kudzu. The response surface methodology which is a statistical analysis method for optimizing the extraction amount of iso-flavonoids from the kudzu and the process condition for maximizing the yield was applied. It is the final objective of this study to effectively derive the condition of the process that matches the target response with a minimum number of experiments and analyze the effect of each process condition on the response. In the response surface methodology, the central composite design was applied and the optimum condition was analyzed, and the three independent variables were set to ultrasonic irradiation time, volume ratio of ethanol/ultrapure water, ultrasonic irradiation power. Using the response surface methodology, the optimum conditions with the maximum extraction yield and the content of iso-flavonoids were evaluated as ultrasonic irradiation time (24.75 min), ethanol / ultrapure water volume ratio (39.75 vol%), ultrasonic irradiation power (592.36 W). The overall satisfaction level appears as high as 0.8938, which is recognized at a significance level within 5%. As a result of analyzing the optimization process, it was confirmed that the ultrasonic irradiation time is the factor that most affects the responses.

Effects of External Whole-Body gamma-Irradiation on Blood Picture and Growth in Young Rabbits (Cobalt-60 gamma 선(線) 외부전신조사(外部全身照射)가 어린 토끼의 혈액상(血液像)과 성장률(成長率)에 미치는 영향(影響))

  • In the event a population of animals is exposed to ionizing radiation, proper disposition of animals will minimize loss to the farmer and protect the public from unwholesome meat and dairy products. Clinical response is an important factor in considering salvage of animals for food following exposure to ionizing radiation whether the dose is lethal or not. It is the purpose of this report to present the discussions of blood picture and growth of y ung growing rabbits exposed to cobalt-60 gamma-irradiation. The experimental animals were Hyrnalayan rabbits of 65 days old. The body weight for all animals of the time of irradiation was approximately 450g. The 96 rabbits used in the experiment were allotted t groups of eight. Rabbits in one group served as controls and the others were exposed to single doses of 100, 200, 300, 400, 500, 600 and 700 Roentgens (R). The exposure does rate averaged 15 R per minute. Central blood was obtained by heart puncture. Erythrocyte and leukocyte enumerations and, determinations of hemoglobin in blood and hematocrit value were made in the usual manner. Erythrocyte counts, concentrations of hemoglobin in blood, and hematocrit readings dropped to the lowest value between 1 and 4 weeks postirradiation, returning towards the control values. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. There was a progressive decrease with increasing radiation does in the above values. Mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration were not affected by the irradiation. After irradiation of each does the numbers of total leukocyte and the absolute numbers of various types of white cell began to decrease rapidly, and reached a minimum after 2 weeks, returning towards normal levels. But recovery was not complete at the time of the last determination on 6 weeks postirradiation. The reductions in total leukocyte and differential counts as the radiation does increases were apparent. Relative neutrophilia was observed in each irradiated group because of more expense of lymphocytes after irradiation. Growth rate of the rabbits was not affected by 100 R of whloe-body irradiation, while rabbits irradiated with 200, 300, 400 and 500 R showed marked depression of growth rate. As the radiation does increases, a depression of growth rate was apparent. The rabbits exposed to 600 and 700 R ceased from growing and recovery was not recognized during the experimental period.

Water Absorption, Cooking Properties and Cell Structure of Gamma Irradiated Soybeans (감마선조사 대두의 수분 흡수와 조리특성 및 세포구조)

  • Gamma irradiation was applied to soybean(Glycine max.), Hwangkeum, at dose levels of 0, 5, 10 and 20 kGy to improve the physical properties of soybeans. The time to reach a fixed moisture content was reduced depending on the increment of soaking temperatures and applied irradiation dose levels. Irradiation at 5~20 kGy resulted in reduction in soaking time of the soybeans by about 3~6 hrs at soaking temperature of $20^{\circ}C.$ The degree of cooking of soybeans in boiling water was determined by measuring the maximum cutting force of cotyledon. The cutting force to reach complete cooking was about 145g/g. Irradiation at 5~20 kGy resulted in a reduction of cooking time of soybeans by 55~75% as compared to the nonirradiated soybean. In electron microscopic observation of seed coat inner, the parenchyma of nonirradiated soybean showed tight fibrillar structure, whereas that of irradiated soybeans showed loosened and deformed structure. The microstructure of compressed cells and cotyledon epidermis was also deformed by gamma irradiation. In subcellular structure of cotyledon, the roundness of protein body was deformed and changed to spike shape at 20 kGy. Also, the size of lipid body decreased as the irradiation dose levels increased.

Effects of Gamma-irradiation on Anti-diabetic and Cytotoxic Activities of Heat-treated Mistletoe (Viscum album) (감마선 조사가 열처리 겨우살이의 항당뇨 및 세포독성에 미치는 영향)

  • Mistletoe (Viscum album) has been widely used as a functional food material for various therapeutic purposes from ancient time. In this study, we examined anti-diabetic and cytotoxic activities of heated-treated mistletoe and the effects of gamma-irradiation on its activities. Heat-treated mistletoe extract was prepared by heating during different time (3, 6, 9 and 12 h) and gamma-irradiated with different doses of 0, 10, 30, 50, 70 and 100 kGy. Heat-treated mistletoe extracts showed a concentration-dependent cytotoxicity on rat insulinoma RINm5F cells and the effect was gradually decreased as heating time increased up to 12 h. 12 h heat-treated extract was no cytotoxic. Gamma-irradiation enhanced the reduction of heat-treated mistletoe-induced cytotoxicity and the decreasing effect was an irradiating dose-dependent. In particular, all of 70 kGy irradiated and heat-treated mistletoe extracts did not showed the cytotoxicity and the effect was comparable to 12 h heat-treated mistletoe extract. Among those extracts, 3 h heat-treated mistletoe extract gradually increased the insulin secreting activity by gamma-irradiation and the effect was the best at 70 kGy, whereas 12 heat-treated extract was no effect. On the test of ${\alpha}$-glucosidase inhibitory activity, 3 h heat-treated mistletoe extract showed the concentration dependent effects and gamma-irradiation induced more activity at 70 kGy, compared to non-irradiated 3 h and 12 h heated mistletoe extracts. These results suggest that the combination of heat treatment and gamma-irradiation might be more effective than only heat-treatment for improving the anti-diabetic activity of mistletoe extract and reducing its cytotoxicity.

Effects of Ultrasonic Irradiation on Physical Properties of Silica/PEG Hybrids

  • The effect of ultrasonic radiation is reported for silica-poly(ethylene glycol) system prepared without the solvent using sol-gel processing by varying various parameters such as ultrasonic irradiation time, PEG content and HCl/TEOS molar ratio. The property of sonogel is compared with classic gel which has been prepared with ethanol as a solvent by traditional sol-gel processing. SEM, BET, DTA-TGA, density and Vickers hardness measurements are carried out for analyzing the samples. The gelation time is found strongly dependent on radiation time, PEG content and pH value, and has been discussed on the basis of existing theories. The $SiO_2-10$ & 20 wt% PEG sonogel exhibited superior optical, physical and gel properties as compared to the classic gel, hence, found suitable for device applications. The ultrasonic radiation increased the density and surface area, and also reduced the pore size which is well supported by the shift in the peak of DTA curve. The DTA thermogram was found similar to that of pure silica gel.
