Effects of Ultraviolet Blood Irradiation in a Type 1 Diabetic Rabbit (제1형 당뇨토끼에서 자외선 혈액 조사 효과)

  • The purpose of this study is to evaluate the effects of ultraviolet blood irradiation on the blood when a low dose of ultraviolet (UV) C is directly irradiated to the blood in a diabetic rabbit model and to evaluate the effects on treatment for diabetes. This study results indicate that the reduced body weight is increased and blood glucose levels are significantly reduced after the UBI treatment is performed when compared to those prior to the UBI treatment. In addition, $HCO_3{^-}$ levels and blood pH were elevated and lowered, respectively. When the UBI treatment is performed in a diabetic rabbit model, in this result indicate that blood glucose levels are reduced.

Induction of micronuclei in human, rabbit and dog lymphocytes irradiated in vitro with gamma radiation (사람, 토끼 및 개 유래 말초혈액 림프구의 미소핵을 이용한 방사선 피폭의 생물학적 선량측정)

  • The frequencies of gamma-ray-induced micronuclei (MN) in cytokinesis-blocked (CB) lymphocytes at several doses were measured in three donors of three species (human, rabbit, dog). Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was : human : $y=0.1184D+0.01867D^2+0.01$, rabbit : $y=0.0387D+0.00528D^2+0.01$ (y = number of MN/CB cells and D = irradiation dose in Gy). The relative sensitivity of rabbit lymphocytes compared with human lymphocytes was estimated by best fitting linear-quadratic model based on the radiation-induced MN data over the range from 0 Gy to 4 Gy. In the case of MN frequency with 0.2, the relative sensitivities of rabbit lymphocytes was 0.39. These data indicate that the induction of MN in rabbit CB cells following irradiation was much less sensitive to the MN induction effects of gamma-irradiation than those from human. The MN assay with dog lymphocytes was very difficult and time-consumed because the dog PHA-stimulated lymphocytes yielded cultures with very low level of CB cells formation in the condition of this experiment. Our in vitro radiobiological study confirmed that the cytogenetic response obtained in blood from rabbit can be utilized for application in environmental studies.

RABBIT HEATING BY MICROWAVE EXPOSURE AT VARIOUS AMBIENT TEMPERATURES

  • The potential ability of environmental temperature to enhance the effect of microwave radiation (7 GHz) was experimentally studied for rabbit heating after simultaneous application of both agents. The tested ambient temperatures (30 and $38^{\circ}C$) didn't exert a considerable influence upon rabbit heat homeostasis after the used duration of exposure (3 hours and 15 minutes, correspondingly). The synergistic interaction of microwave irradiation and ambient temperature was demonstrated for rabbit heating. Power flux density of microwave irradiation was shown to be a determinant of the synergistic interaction effectiveness. For the fixed ambient temperature ($30^{\circ}C$), the synergism was shown to be observed only within a definite power flux density ($0-100\;mW{\cdot}cm^{-2}$), inside of which there was an optimal intensity ($20\;mW{\cdot}cm^{-2}$), which maximized the synergistic effect. Any deviation of the power flux density from the optimal value resulted in a reduction of the synergy. It is concluded that any assessment of the health or environmental risks should take into account the synergistic interaction between ambient temperature and microwave radiation.

  • Despite a number of recent studies on appendix its function appears to remain unknown. The present studies were undertaken in order to extend and confirm the previous studies concerning the role of appendix in immune response. An early hemagglutinin response of mercaptoethanol sensitive antibody(IgM antibody) in rabbit injected intravenously(i.v.) with 200mcg of bovine gamma globulin(BGG) was abolished by lethal whole body irradiation(900 r), but preserved in animals whose appendix and bone marrow were shielded during irradiation. Late formation of mercaptoethanol resistant antibody(IgG antibody) and the development of memory in bone marrow shielded animals were not affected by irradiation of the appendix. Formation of either IgM or IgG antibody to sheep red blood cells(SRBC) injected i.v. as determined by direct plaque forming cell(DPFC) technique in spleen were effectively abolished by appendectomy, thymectomy, or both followed by irradiation. When bone marrow was shielded in combination with autologous appendix reconstitution, DPFC response was about 5 times greater than the sum of two. Lysed appendix cells failed to restore the response. Lethally irradiated rabbits restored with combination of autologous appendix and thymus cells showed DPFC responses which were essentially normal. Three pools of appendix were obtained by manual separation technique and were stimulated with soluble concanavalin A(Con A), phytohemagglutinin-P(PHA) and pokeweed mitogen(PWM). Rabbit appendix cells responded to Con A, PHA and PWM. Cells of thymus dependent area(TDA) of the appendix were relatively enriched in their response to T cell mitogens compared to dome and follicle cells. The PHA/Con A responsive ratio of appenix TDA subpopulation was high, indicating that Con A responsive cells have a wider distribution among appendix. This finding showed that interfollicular area of the appendix is thymus-dependent. The present studies confirmed other evidence that the rabbit appendix cells itself are unable to form antibody and T lymphocytes in appendix TDA may be heterogenous, and that the appendix cells are synergistic with either bone marrow or thymus cells in the early hemagglutinin on splenic antibody response to BGG or SRBC.

Studies on the Normal Frequency and Effects of X-irradiation on Sex Chromatin of Neutrophil Leukocyte in the Rabbit (가토의 호중성백혈구 성염색질의 정상빈도와 X선조사에 대하여)

  • The frequency of sex chromatin in neutrophil leukocyte of 14 female and 4 male normal, adult rabbits was examined and compared to those of single total X-ray irradiation. 1. The average frequency of of drumstck was 9 % in female and 0.1% in male, and that of sessile nodule was 3.35% in female and 0.9% in male , in normal condition rabbit . These results fairly determined sex ratio by sex chromatin in neutrophil leukocyte. 2. The drumstick frequency reduces to half after X-irradiaton but did not show any regular tendency or variation in morphologicla form, indicating the stability of chromatin to X-irradiation. 4. The chromatin satellite was found with a reasonable frequency in the some kinds of irradiated cells, polymorphonuclear neutrophil (above 7 lobes 0 and megakarycyte.

CHARACTERISTICS OF THE PNEUMATIC TRANSFER SYSTEM AND THE IRRADIATION HOLE AT THE HANARO RESEARCH REACTOR

  • This paper describes the results of an irradiation test and the specifications of the pneumatic transfer system (PTS) in the NAA #3 irradiation hole at the HANARO research reactor, which was reinstalled after some modifications of the operation mode at the end of 2004. The outer and inner diameters of the PE transfer tube are 34.1 and 27.5 mm, respectively. PE rabbit was used for sample irradiation. The $N_2$ gas pressure of the PTS lines was adjusted to 0.75 bar. The average sending time to the reactor was $8.5{\pm}0.3$ s and the average receiving time back to the receiver was $3.2{\pm}0.2$ s. The internal and external temperature of the irradiation tube was measured in a range of 50 to $80^{\circ}C$ for a 40 s to 80 s irradiation time, respectively. The optimum irradiation time was estimated to be less than 80 s. The thermal, epithermal and fast neutron flux at 30 MW thermal power were $1.42{\pm}0.01{\times}10^{14},\;1.51{\pm}0.04{\times}10^{13}$ and $9.48{\pm}0.69{\times}10^{11} n{\cdot}cm^{-2}{\codt}s^{1-}$, respectively. The cadmium ratio was approximately 9.40. The data obtained will be applied to supplement user information and for reactor management.

CONTRIBUTION OF HANARO IRRADIATION TECHNOLOGIES TO NATIONAL NUCLEAR R&D

  • HANARO is a multipurpose research reactor located at the Korea Atomic Energy Research Institute (KAERI). Since the commencement of its operation in 1995, various neutron irradiation facilities, such as rabbit irradiation facilities, fuel test loop (FTL) facilities, capsule irradiation facilities, and neutron transmutation doping (NTD) facilities, have been developed and actively utilized for various nuclear material irradiation tests requested by users from research institutes, universities, and industries. Most irradiation tests have been related to national R&D relevant to present nuclear power reactors such as the ageing management and safety evaluation of the components. Based on the accumulated experience as well as the sophisticated requirements of users, HANARO has recently supported national R&D projects relevant to new nuclear systems including the System-integrated Modular Advanced Reactor (SMART), research reactors, and future nuclear systems. This paper documents the current state and utilization of irradiation facilities in HANARO, and summarizes ongoing research efforts to deploy advanced irradiation technology.

Changes of Activities of Rabbit Pulmonary Surfactant Incubated at $37^{\circ}C$, and effect of X-Irradiation in Vitro ($37^{\circ}C$에서 incubate 한 가토(家兎) 폐포표면활성물질(肺胞表面活性物質)의 활성도(活性度)의 변화(變化)와 in vitro X 선조사(線照射)의 영향(影響))

  • In an attempt to understand the possible effects of temperature and X-irradiation on the activities of surfactant in rabbits, the pulmonary surfactant from the rabbit was subjected to the incubation at $37^{\circ}C$ and X-irradiation with 900r in vitro, and activities of surfactant were measured at 10, 30, 60, and 90 minutes. Tension-area diagram of the lung extract was recorded automatically by the modified Langmuir-wilhelmy balance with a synchronized recording system designed in this Department. A comparison was made with the normal and the following results were obtained. 1) The maximal surface tension, minimal surface tension, width of the tension area diagram at the surface area of 40% and stability index of the normal rabbit lung extract were $31.6{\pm}3.11\;dynes/cm,\;8.2{\pm}0.56\;dynes/cm,\;21.4{\pm}4.40\;dynes/cm\;and\;1.12{\pm}0.22$,respectively. 2) In the $37^{\circ}C$ incubation group, maximal surface tension was similar to the normal value, while minimal surface tension was significantly lower and stability infer was markedly higher than the normal. 3) In the group where X-irradiation of 900r in vitro was applied, maximal surface tension did not differ greatly with the normal or the $37^{\circ}C$ incubation group. The minimal surface tension was significantly lower than the normal but comparing with the $37^{\circ}C$ incubation group, some decrease in minimal surface tension was noted. The width of the tension·area diagram at 40% and stability index in the irradiated group were significantly higher than the normal but a tendency of increase was noted comparing with the $37^{\circ}C$ incubation group.

HISTOMORPHOMETRIC STUDY ON THE INFLUENCE OF STEROID TOPICAL IRRIGATION AND IMPLANT SURFACE ON BONE HEALING IN THE IRRADIATED RABBIT TIBIA (방사선 조사 후 매식한 임프란트의 표면 종류와 스테로이드 관주에 따른 골 치유 효과에 대한 조직 형태학적 연구)

  • The purpose of this study was to evaluate and compare the influence of Steroid topical irrigation and implant surface on bone healing in the irradiated rabbit tibia. Implant to bone contact surface ratio and the pattern of bone healing around hydroxyapatite(HA) coated implant and pure titanium (Ti) implant which were inserted into the irradiated rabbit tibia were compared. 16 Korean house mature male rabbits were used as experimental animal. Each rabbit received 15 Gy of irradiation. 4 weeks after the irradiation, two holes were prepared in the irradiated tibia of each rabbits, where two surface type of implants were inserted :1) HA coated type and 2) pure Ti type. Right before placing implants, one group of rabbit received steroid irrigation and the other group did saline. After the irrigation, two implants of HA coated type and pure Ti type were inserted into the tibia of each rabbits. Each rabbit were sacrificed at 2nd, 4th, and 8th week after the implantation and the specimens were observed by the light microscope. The pattern of bone healing and histomorphometric analysis of the implant-bone interface were done. The results were as follows. 1. All implants inserted into the irradiated tibia of rabbit did not show any sign of clinical mobility and the bone around implants inserted into the irradiated tibia of rabbit did not show any resorption. 2. The bone to implant contact surface ratio around HA coated implants that received steroid irrigation got more bone to implant contact surface ratio than that of the saline irrigation. This result showed statistically significant(p<0.05). There was no statistically significant difference in 8th week group. 3. Though there was no statistically significant difference HA coated implants had more bone to implant contact surface ratio than pure Ti implant in 2nd and 4th groups, and there was no difference in 8th week group. 4. All implants inserted into the irradiated tibia of rabbit had exhibited successful osseointegraion.

Pathologic Changes in the Rabbit lung Following Single Dose Irradiation (방사선 조사후 관찰한 가토 폐의 병리학적 변화)

  • The damage which radiation produces in tissues such as the lungs can be discussed at the molecular, biophysical, cellular, and organ levels. The cellular effects of irradiating the lungs are related to the histologic and clinical sequelae. In the present study the right lung of rabbits were exposed to single dose of 20 Gy of X-irradiation. Animals from each group were sacrificed monthly for 6 months postexposure. Sections of lung were examined by light microscopy(LM) and by transmission electron microscopy(TEM). Multiple exudative lesions were seen at 2 months after the 20Gy irradiation,and they progressed to a proliferative and then reparative fibrotic lesion by 6 months. Changes in epithelial lining of lung components, particulary the presence of type II pneumocytes were found by both LM and TEM. Capillary endothelial damages were less pronounced. The possible implication of cellular components in radiation pneumonitis and fibrosis is discussed.
