PSL, TL, ESR 및 GC/MS 분석을 통한 감마선 조사된 유지종실류의 검지 특성 연구 (Detection Characteristics of Gamma-Irradiated Seeds by using PSL, TL, ESR and GC/MS)

  • 본 연구는 비허용 품목인 유지종실류 5종(달맞이꽃씨, 홍화씨, 유채씨, 해바라기씨 및 아마씨)을 선정하여 광자극발광법(PSL), 열발광법(TL), 전자스핀공명법(ESR) 및 기체크로마토그래프/질량분석법(GC/MS)을 활용하여 검지 특성 연구를 수행하였다. PSL 및 TL 측정 결과, 5종 모두 적용 가능성이 높은 것으로 판단하였다. ESR 분석결과는 5종 모두 조사시료에서 조사유래의 특이 ESR signal이 관찰되지 않아 적용 가능성이 낮다고 판단하였다. GC/MS 분석 결과, 홍화씨, 유채씨, 해바라기씨 및 아마씨의 경우에는 조사 유래의 hydrocarbon류가 검출되어 적용 가능성이 높은 것으로 판단하였다. 특히, oleic acid에서 유도된 8-heptadecene ($C_{17:1}$), 1,7-hexadecadiene ($C_{16:2}$)이 조사 여부를 확인하는 화학적 마커로 활용 가능성이 높았다. 달맞이꽃씨 경우에는 oleic acid에서 유도된 hydrocarbon류가 검출되지 않아 적용 가능성이 낮다고 판단하였다. 또한, 달맞이꽃씨, 홍화씨 및 해바라기씨에서 감마리놀렌산의 주요 원료인 linoleic acid에서 유도된 1,7,10-hexadecatriene ($C_{16:3}$)과 6,9-heptadecadiene ($C_{16:3}$)이 검출되어 조사여부를 확인하는 화학적 검지 마커로 활용 가능성이 높다고 판단하였다. 본 연구 결과를 통해 유지종실류 5종에 대한 적용 가능성 및 다중분석 체계(PSL-TL-GC/MS)를 확립하였다.

Changes of the Binding Abilities of Immunoglobulin G and E on Gamma-Irradiated Ovalbumin by Proteolytic Enzymes

  • This study evaluated the binding abilities of rabbit anti-ovalbumin (OVA) immunoglobulin G (IgG) and egg-allergic patient IgE on gamma-irradiated OVA during proteolysis using pepsin and trypsin. The concentrations of both the intact and the irradiated OVAs decreased during proteolysis when detected with IgG However, when detected by patient IgE the concentration of the intact OVA decreased up to 30 min after the trypsin treatment and increased thereafter. Irradiated OVA detected by patient IgE showed a lower initial concentration (0.16%) than that of the intact OVA, and this reduced concentration was maintained stably. The results indicate that irradiation, rather than enzymatic treatment, could reduce the binding of the irradiated and enzyme-treated OVA. Therefore, gamma irradiation has potential as an effective method to reduce OVA-induced allergy and may enhance the safety of egg-allergic individuals.

Effects of Irradiation Temperature on the Sensory Quality Improvement of Gamma-irradiated Ganjang-gejang, Korean Traditional Marinated Raw Crab Portunus trituberculatus in Soybean Sauce

  • This study was conducted to confirm quality properties of sterilized Ganjang-gejang (marinated crab Portunus trituberculatus) with Korean soy sauce using by gamma irradiation and to improve quality of sterilized Ganjang-gejang. The Ganjang-gejang was irradiated at dose of 3, 6, 9, 12, and 15 kGy by gamma irradiation and there was evaluated in microbiological, physicochemical, and sensory properties. Total aerobic bacteria and fungi contents of non-irradiated samples were about 6 and 4 log CFU/g level, respectively. Gamma-irradiated samples at above 9 kGy did not contain aerobic bacteria or fungi at detection limit less than 2 log CFU/g, but sensory scores were significantly decreased depending on the irradiation dose. To improve the sensory qualities of gamma-irradiated Ganjang-gejang, the temperature was adjusted during sample irradiation. When samples were irradiated under freezing temperatures, especially on dry ice, the TBARS and the deterioration of sensory qualities of Ganjan-Gejang were reduced. Different odor patterns were observed among samples, as observed using electronic nose analysis system. The results of this study indicated that treatment with irradiation under low temperatures may help to preparing high-quality Ganjang-gejang.

  • 각종 가공식품 원료로 사용되고 있는 건조 채소류 중 표고버섯, 시금치, 무청, 미나리, 무, 호박 등 6종에 대하여 1, 4, 7 kGy의 선량으로 감마선을 조사하고, photostimulated luminescence kGy와 thermoluminescence (TL) 등 물리적 분석방법을 이용하여 감마선 조사 여부를 확인하였다. PSL 측정에서 비조사 표고버섯, 시금치, 무청, 미나리, 무 및 호박에서는 각각 $194.0{\pm}223.4,\;695.5{\pm}26.2,\;598.0{\pm}4.2,\;266.5{\pm}9.2,\;503.0{\pm}190.9$$270.5{\pm}245.4$ 수준의 photon count가 측정되어 비조사 시료의 범위(700 이하의 값)에 속하였으며, 1 kGy 이상 조사군에서는 모두 방사선 조사시료의 범위인 5000 이상의 photon count를 나타내었다. 또한 6종의 시료에 대해 TL분석을 실시한 결과, 비조사군의 TL ratio$(TL_1/TL_2)$는 0.021 이하를, 1 kGy 이상 조사군에서는 0.653 이상으로 나타나 본 연구에 사용된 6종의 건조 채소류는 PSL과 TL분석에 의해 조사여부의 검지가 가능한 것으로 확인되었다.

방사선조사식품에 대한 지식, 태도 및 행위 측정도구 개발 (Development of a Tool to Measure Knowledge, Attitude and Behavior towards Irradiated Food)

  • 방사선조사식품을 바로 알리고 선택할 수 있는 행위를 향상시키는 개입전략의 근거를 도출하는데 활용하기 위해 교육학적 접근에서 방사선조사식품에 대한 지식, 태도 및 행위를 측정하는 도구를 개발하고자 하였다. 방사선조사식품에 대한 지식은 20개 문항에 대해 3개의 요인이 추출되었다. 요인1은 '방사선조사식품의 원리', 요인2는 '방사선조사식품의 영향', 요인3은 '방사선조사식품의 현황'관련으로 각각 명명 하였다. 각 요인의 분산 설명력은 요인1이 43.1%, 요인2가 12.0%, 요인3이 9.9%로서 총 65.0%가 설명되었다. 방사선조사식품에 대한 태도는 4개 문항에 대해 1개의 요인이 추출되었고, 측정척도의 요인분석 결과 '방사선조사식품에 대한 태도'로 명명하였으며 요인의 분산 설명력은 71.1%로 설명되었다. 방사선조사식품에 대한 행위는 5개 문항에 대해 1개의 요인이 추출되었고, 요인의 분산설명력은 57.6%로 설명되었다.

곡류의 감마선 조사 검지를 위한 DEFT/APC 방법의 이용 (Detection of Gamma-Irradiated Grains by Using DEFT/APC Method)

  • For the screening of gamma-irradiated grains, domestic rice, glutinous rice, barley, and wheat were irradiated with 0.1, 0.3, 0.5, 0.7, and 1.0 kGy and screened using the DEFT/APC (Direct Epifluorescent Filter Technique/Aerobic Plate Count) method. The log DEFT/APT units increased with the dose increment in all samples, whereas the log APC unit decreased gradually. For rice, barley, and wheat, unirradiated and irradiated samples with below 0.3 kGy had 2.0 or lower logarithmic units, whereas those with 0.5 kGy or higher had 2.0 or higher logarithmic units. For glutinous rice, the sample irradiated with 0.5 kGy showed 1.92 logarithmic unit and those with 0.7 kGy or higher had 2.0 or higher logarithmic units. These results suggest that if the grains show 2.0 or higher logarithmic units, they could be assumed to have been irradiated at a dose level of at least 0.5 kGy. In conclusion, grains could be easily screened through the DEFT/APC method.

ESR Spectroscopy에 의한 감마선 조사된 향신료의 확인 (Detection of Gamma-Irradiated Spices by ESR Spectroscopy)

  • Electron spin resonance(ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity of irradiated spices and to identify the stability of radicals after storage. Red, white and black peppers, and garlic powders were irradiated with doses of 0, 1, 5, 10, 20 and 30 kGy at room temperature using a Co-60 irradiator. Triplet ESR signals were observed in irradiated pepper powders, while singlet ESR signals were observed in irradiated garlic powders. Those characteristic signals were not detected in non-irradiated samples. The strength of ESR signals linearly increased with the applied doses(1~3 kGy). Highly positive correlation coefficients ($R^2$=0.9757~0.9933) were obtained between the irradiation doses and the corresponding ESR signal intensities. The signal intensities of irradiated samples were stable even after 97 days of storage at room temperature.

감마선 조사선량에 따른 버찌(Fruit of Prunus serrulata L. var. spontanea Max. wils.) 에탄올 추출물의 항산화활성 (Antioxidative Properties of Ethanolic Extracts from Flowering Cherry (Prunus serrulata L. var. spontanea Max. wils.) Fruit with Various Doses of Gamma Radiation)

  • 본 연구는 폐자원으로 버려지는 버찌의 활용 증대방안 모색을 위한 기초자료 마련을 위해 버찌의 에탄올 추출물에 0~20 kGy 선량으로 감마선 조사를 실시하고, 이에 따른 항산화활성 변화를 측정하였다. 총 폴리페놀 함량 측정결과, 10 kGy 조사구에서 30.35 mg/g으로 가장 높은 값을 나타내었으나 감마선 조사에 따른 일정한 증가나 감소의 경향은 나타내지 않았다. DPPH radical 소거능 측정결과 10 kGy 조사구에서 327.52 ppm으로 가장 낮은 $IC_{50}$값을 나타내 가장 높은 활성을 나타내었으며 대체적으로 비조사구보다는 조사구에서 높은 활성을 나타내었다. 환원력 측정 결과, 2.5 mg/mL 이하 농도에서 비조사구에 비해 조사구에서 높은 활성을 나타내었고 20 kGy 조사구에서 대체적으로 높은 환원력을 나타내었다. 아질산염 소거능을 측정한 결과 pH 4.2 이하의 pH에서 조사구 및 비조사구 모두 73~94% 이내의 높은 소거능을 나타내 pH 의존적인 아질산염 소거능을 보였으나 조사에 따른 활성 증가는 나타내지 않았다. FRAP value를 측정결과 3 kGy 조사구에서 0.38 mM로 가장 높은 활성을 나타내었다. 결론적으로 버찌 에탄올 추출물에 대한 감마선 조사는 일부 선량에서 추출물의 항산화 활성 증가를 나타내며, 사용목적에 따른 적정 선량의 감마선 조사는 생리활성물질의 증가를 유도하여 산업적인 면에서 효율적으로 이용될 수 있을 것으로 사료된다.

  • Structural modification of corn starch by gamma irradiation was evaluated for under dry conditions at varied intensities from 0 to 40 kGy. Under scanning electron microscopy, the granule shape of corn starch was not significantly affected by the irradiation up to 40 kGy. In addition, X-ray diffraction and melting patterns of the irradiated starches were similar to those of the native starch, indicating that crystalline regions in the starch granules were not changed by irradiation. However, the pattern of gel permeation column chromatography showed a significant increase in partial hydrolysis of gamma irradiated starch samples. The degree of polymerization and the paste viscosity of irradiated starch samples dose-dependently decreased significantly with irradiation, and increased solubility and clarity were observed in the irradiated starch solution. In addition, the degree of retrogradation decreased as irradiation dose increased. Irradiation of corn starch has advantages over the ordinary acid or the enzyme hydrolysis modification methods. It does not affect the granular shape and crystalline phase of starch during hydrolysis, and the process can be carried out in dry state.

The Microwave-Assisted Extraction of Fats from Irradiated Meat Products for the Detection of Radiation-Induced Hydrocarbons

  • Hydrocarbons have been successfully used as a chemical marker in order to identify irradiated from non-irradiated foods. The method for determining hydrocarbons consists of extraction of fats, followed by separation of hydrocarbons by florisil column chromatography, and then identification of hydrocarbons by GC/MS. Currently, solvent extraction method for fats has certain limitations with regard to extraction time and solvent consumption. Commercial hams and sausage were irradiated at 0 and 5 kGy, and the efficiency of microwave-assisted extraction (MAE) and conventional solvent extraction (CSE) methods on the extraction of radiation-induced hydrocarbons from the meat products was compared. Significant levels of hydrocarbons, mainly composed of 1,7-hexadecadien, 1,7,10-hexadecatriene, and 6,9-heptadecadiene, were detected in the extracts from irradiated hams and sausages by both CSE and MAE methods. Both methods were acceptable in extracting hydrocarbons from samples, but MAE method required apparently reduced amounts of solvent from 150 (CSE) to 50 mL and reduced extraction time from 23 (CSE) to 5 min.
