Consensus channelome of dinoflagellates revealed by transcriptomic analysis sheds light on their physiology

  • Ion channels are membrane protein complexes mediating passive ion flux across the cell membranes. Every organism has a certain set of ion channels that define its physiology. Dinoflagellates are ecologically important microorganisms characterized by effective physiological adaptability, which backs up their massive proliferations that often result in harmful blooms (red tides). In this study, we used a bioinformatics approach to identify homologs of known ion channels that belong to 36 ion channel families. We demonstrated that the versatility of the dinoflagellate physiology is underpinned by a high diversity of ion channels including homologs of animal and plant proteins, as well as channels unique to protists. The analysis of 27 transcriptomes allowed reconstructing a consensus ion channel repertoire (channelome) of dinoflagellates including the members of 31 ion channel families: inwardly-rectifying potassium channels, two-pore domain potassium channels, voltage-gated potassium channels (Kv), tandem Kv, cyclic nucleotide-binding domain-containing channels (CNBD), tandem CNBD, eukaryotic ionotropic glutamate receptors, large-conductance calcium-activated potassium channels, intermediate/small-conductance calcium-activated potassium channels, eukaryotic single-domain voltage-gated cation channels, transient receptor potential channels, two-pore domain calcium channels, four-domain voltage-gated cation channels, cation and anion Cys-loop receptors, small-conductivity mechanosensitive channels, large-conductivity mechanosensitive channels, voltage-gated proton channels, inositole-1,4,5-trisphosphate receptors, slow anion channels, aluminum-activated malate transporters and quick anion channels, mitochondrial calcium uniporters, voltage-dependent anion channels, vesicular chloride channels, ionotropic purinergic receptors, animal volage-insensitive cation channels, channelrhodopsins, bestrophins, voltage-gated chloride channels H+/Cl- exchangers, plant calcium-permeable mechanosensitive channels, and trimeric intracellular cation channels. Overall, dinoflagellates represent cells able to respond to physical and chemical stimuli utilizing a wide range of G-protein coupled receptors- and Ca2+-dependent signaling pathways. The applied approach not only shed light on the ion channel set in dinoflagellates, but also provided the information on possible molecular mechanisms underlying vital cellular processes dependent on the ion transport.

이온 크로마토그래피용 모세관 컬럼의 개발 (Development of Capillary Columns for Ion Chromatography)

  • 본 연구에서는 적은 양의 시료를 분석할 수 있는 이온 크로마토그래피용 모세관 컬럼을 개발하였다. 내경이 작은 모세관 컬럼을 사용하는 경우 이동상의 유속은 보통 $5{\sim}15{\mu}L/min$ 정도 되고 컬럼의 길이는 50~150 mm 정도이다. 컬럼은 RSL-300 fused silica 모세관(내경 : 0.53 mm, 외경 : 0.67 mm)과 AG14 column resin(support : polystyrene-divinylbenzene, functional group : aulkyl quatemary ammonium)을 이용하여 만들었다. 또한 이런 모세관 컬럼에 맞는 새로운 전도도 셀과 억제컬럼도 함께 개발하여 사용하였다. 이 모세관 컬럼을 이용하여 실제 음이온들(fluoride, nitrite, nitrate, chlorate)을 분석한 결과 재현성 있고 좋은 크로마토그램을 얻었다.

이온주입에 의한 폴리머의 전기특성 조사 (Electrical properties of polymers by ion implantation)

  • Ion implantation has been shown to significantly alter the surface properties of polymers. Polycarbonate(PC) and Polyimide(PI) were irradiated with 50keV $N^+$, $Xe^+$ ions to the fluences of $1{\times}10^{16}{\sim}3{\times}10^{17}\;cm^2$. The ion beam-induced modification of the electrical conductivity and the related structural features have been studied for polymers. The beam-induced chemical and structural modifications have been investigated by using X-ray Phooelectron Spectroscopy(XPS) and Fourier Transform-Infrared Spectroscopy(FT-IR), while the modification of the electrical properties was followed by performing a complete set of sheet resistance measurements. Samples irradiated at higher fluence showed a good conductivity, with a saturation value of $10^7{\Omega}/sq$. The XPS data demonstrate that the modification of the electrical properties is due to the progressive formation with increasing ion fluence of a dense amorphous carbon network, while PF-IR data reveal that material degradations through bond breaking are the main effects.

산화물유리에서의 $Na^{+}$이온과 $Ag^{+}$이온 교환에 따른 전기전도도 변화에 관한 연구 (Electrical Conductivity in oxide glasses subjected to a sodium silver ion-exchange treatment)

  • $Na_2O$함량 20, 52, 30 mol% 변화를 준 $Na_2O$-CaO-$Al_2O_3$-$B_2O_3$-$SiO_2$계 유리를 제조하여 $AgNO_3$용융염에 각각 0, 24, 36, 48 시간동안 이온교환처리를 행한후 시편의 전기적 특성을 분석하였다. 이온 교환처리된 유리의 저항값과 활성화에너지값이 이온교환처리 되지 않은 모유리보다 낮았다. $Na_2O$의 함량이 증가할수록, 이온교환시간이 증가할수록 전기전도도는 증강하였다. 본 실험에서는 $Na_2O$ 함량 30 mol%의 유리를 48시간 이온교환시킨후의 시료의 전기전도도가 제일 좋았으면 $200^{\circ}C$ 에서의 전기전도도는 $1.78\times$$10^{-4}$S/cm이었다.

  • In this study, polyurethane acrylate macromer was synthesized and it was used in a gel polymer electrolyte, and then its electrochemical performances were evaluated. LiCoO$_2$/GPE/MCF cells were also prepared and their performances depending on discharge currents and temperatures were evaluated. ionic conductivity of the gel polymer electrolyte with PUA at room temperature and -20$^{\circ}C$ was ca. 4.5 x 10$\^$-3/ S/cm and 1.7${\times}$10$\^$-3/ S/cm, respectively. GPE was stable electrochemically up to 4.5 V vs. Li/Li$\^$+/. LiCoO$_2$/GPE/MCF cell showed a good high-rate and a low-temperature performance.

Polarization Behavior of Li4Ti5O12 Negative Electrode for Lithiumion Batteries

  • $Li_4Ti_5O_{12}$ is prepared through a solid-state reaction between $Li_2CO_3$ and anatase $TiO_2$ for applications in lithium-ion batteries. The rate capability is measured and the electrode polarization is analyzed through the galvanostatic intermittent titration technique (GITT). The rate characteristics and electrode polarization are highly sensitive to the amount of carbon loading. Polarization of the $Li_4Ti_5O_{12}$ electrode continuously increases as the reaction proceeds in both the charge and discharge processes. This relation indicates that both electron conduction and lithium diffusion are significant factors in the polarization of the electrode. The transition metal (Cu, Ni, Fe) ion added during the synthesis of $Li_4Ti_5O_{12}$ for improving the electrical conductivity also greatly enhances the rate capability.

폴리비닐알콜을 이용한 직접메탄을 연료전지용 이온교환막 제조에 관한 연구 (Studies on the Preparation of the Poly(vinyl alcohol) ion Exchange Membranes for Direct Methanol Fuel cell)

  • 본 연구는 직접메탄을 연료전지(Direct Methanol Fuel Cell, DMFC)용 전해질 막으로 이용되는 양이온교환막의 개발에 관한 것이다. 투과증발공정에서 메탄을 Barrier로 잘 알려져 있는 Poly(vinyl alcohol)을 Base polymer로 사용하고 양이온 교환기가 포함되어 있는 Poly(acrylic acid)를 가교제로 사용하여 가교제의 함량변화에 따른 메탄을 투과도(Methanol permeability), 이온전도도(Ion conductivity), 이온교환용량(Ion exchange capacity), 함수율(Water content), 고정이온농도(Fixed ion concentration)를 통해 막 특성을 측정하였다. 메탄올 투과도와 이온전도도는 가교제인 PAA함량이 증가함에 따라 감소하다가 15%이상에서는 증가하는 경향을 보였다. 이것은 가교제의 함량증가로 인한 가교의 영향과 가교제에 포함되어 있는 친수성기의 도입으로 이와 같은 결과가 나타난다고 예상된다. 실험결과를 통해 DMFC에 적용가능성이 있는 막은 $25^{\circ}C,\; 50^{\circ}C$에서의 메탄을 투과도가 $6.49{\times}10^{-8}/cm^2/s,\; 2.85{\times}10^{-7}/cm^2/s$, $25^{\circ}C,\; 50^{\circ}C$에서의 이온전도도가 $2.66{\times}10^{-3}\;S/cm,\; 9.16{\times}10^{-3}\;S/cm,$ 이온교환용량이 1.32 meq/g membrane, 함수율이 0.25 g $H_2O$/g membrane, 고정이온농도가 5.25 meq/g $H_2O$인 PVA/PAA-$160^{\circ}C$ 15% 막으로 예측된다.

Oxygen Permeability, Electronic and ionic Conductivities and Defect Chemistry of Ceria-Zirconia-Calcia

  • The total conductivity and oxygen permeation in (Ce1-xZrxO2)0.9(CaO)0.1 solid solutions were measure das a function of temperature and oxygen partial pressure. Empirically, σ at given x and T was expressed essentially by σ=σo2+σeo Po2-1/4, where σo2 and σeo are constant. Applying a standard defect model in which major defects are Cace", Cece' and Vo in ideal solution, we can assign σo2 as the oxide ion conductivity decreases while the electronic conductivity increases with the increase in Zr content. Using the oxide ion and electronic conductivities thus determined, the oxygen permeation flux was calculated for respective Po2 and T conditions at which the measurements were made. The calculated values were found to agree with the observed ones.

  • The Velocity Autocorrelation Function (VAF) of the sodium ions is calculated for a range of temperature from 250K to 1000K and converted into the linear ac-conductivity and ac-susceptibility response via Fourier transformation. A peak is found in the conductivity around $6{\times}10^{12}$ Hz that has some of the character of a Poley absorption. Here it is shown to be due to an harmonically coupled site vibrations of the sodium atoms, which extend only over a limited range. At frequencies below the peak the conductivity tends towards a constant i.e. dc value corresponding to a constant flow of ions through the simulation cell. At high temperatures the conductivity due to this ion transport process behaves like a metal with an insulator to metal transition occurring around a specific temperature.

Polysulfone/SPEEK 블랜드 고분자 전해질 막 제조 및 특성 연구 (Preparation and Their Characterization of Blended Polymer Electrolyte Membranes of Polysulfone and Sulfonated Poly(ether ether ketone))

  • Poly(ether ether ketone)(PEEK)를 황산을 사용하여 설폰화시킨 후 폴리설폰과 다양한 조성으로 혼합하여 블렌드 고분자 전해질 막을 제조하였고 조성의 변화에 따른 메탄을 투과도, 수소이온전도도, 그리고 이온교환용량의 변화를 측정하였다. 폴리설폰의 경우 이온전도도는 낮은 반면에 메탄올에 대한 저항은 우수하였다. 그러나, 설폰화된 PEEK의 양이 증가함에 따라 메탄을 투과도와 수소이온전도도가 함께 증가하였다. 이온전도도와 메탄을 투과도의 비로부터 폴리설폰의 양이 20%일 때 가장 좋은 선택성을 가지는 것을 알 수 있었다.
