Metabolism of Pyrimidine Deoxyribonucleosides and Heat-resistivity of CdR-aminohydrolase in the Mouse Small Intestine (생쥐 小腸에서의 Pyrimidine Deoxyribonucleoside 代謝와 CdR-aminohydrolase의 熱抵抗性)

  • The metabolism of CdR-2-$^14 C$ and UdR-2-$^14 C$ in mouse small intestine has been studied in connection with the effect of heat treatment on the enzymes concerned in vitro. CdR-2-$^14 C$ is deaminated reaidly by CdR-aminohydrolase at nucleoside level and then degraded into U by the action of nucleosidase which is quite resistant to cleave N-pentose bond of cytosine nucleosides, CdR and CR. High inactivation temperature of $80^\\circC$ was observed for CdR-aminohydrolase, while nucleosidase has an inactivation temperature of $60^\\circC$. CdR-aminohydrolases in various tissues of mouse were inactivated at $80^\\circC$, but not one in tissues of rabbit. It might be assumed that there are correlations between order specificity and inactivation temperature of the enzyme. A physiological significance of the appearance of CdR-aminohydrolase in differentiated tissues of mammals possibly be regarded as a main function in catabolic pathways.

Analysis of Gene Expression Responses to a Salmonella Infection in Rugao Chicken Intestine Using GeneChips

  • Poultry products are an important source of Salmonella enterica. An effective way to reduce food poisoning due to Salmonella would be to breed chickens more resistant to infection. Unfortunately host responses to Salmonella are complex with many factors involved. To learn more about responses to Salmonella in young chickens of 2 wk old, a cDNA Microarray containing 13,319 probes was performed to compare gene expression profiles between two chicken groups under control and Salmonella infected conditions. Newly hatched chickens were orally infected with S. enterica serovar Enteritidis. Since the intestine is one of the important barriers the bacteria encounter after oral inoculation, intestine gene expression was investigated at 2 wk old. There were 588 differentially expressed genes detected, of which 276 were known genes, and of the total number 266 were up-regulated and 322 were down-regulated. Differences in gene expression between the two chicken groups were found in control as well as Salmonella infected conditions indicating a difference in the intestine development between the two chicken groups which might be linked to the difference in Salmonella susceptibility. The differential expressions of 4 genes were confirmed by quantitative real-time PCR and the results indicated that the expression changes of these genes were generally consistent with the results of GeneChips. The findings in this study have lead to the identification of novel genes and possible cellular pathways, which are host dependent.

Effects of the Oral Administration of a Probiotic Combination on the Expression of Cytokine and the Histopathology of the Large Intestine in an Animal Model of Enteritis

  • It is known that lactic-acid bacteria (LAB) helps keeping the intestine healthy and to enhance its immunologic competence. In addition, it is known to control the composition of the enterobacteria and the intestinal inflammatory reaction by inducing immunological enhancement. This study was performed, in a mouse model, to test the treatment and preventive effects of LAB of inflammatory bowel disease (IBD), which was induced by a blend of LAB-administering trinitrobenzene sulfonic acid (TNBS). To obtain the animal model of IBD, 2% TNBS was rectally administered once to a five-week-old male Balbc/J mouse. A probiotic combination was administered to the prevention group five times a week for eight weeks before the inducement of enteritis, and the mixture was administered to the treatment group five times a week, after the administration of TNBS. The changes in the levels of the cytokines of the lymph nodes and the tissue of the large intestine were observed, both with the naked eye and with a microscope. The observation showed that the levels of inflammatory cells, infiltration, and necrosis were much lower in the LAB-administered groups than in that of the control group. In addition, the inflammatory cytokines (e.g., TNF-${\alpha}$, IL-17A) decreased in the lymph nodes and the tissues of the large intestine. The results indicated that the administration of the combination to the animal model suppressed the inflammatory cytokines in the large intestine and in the lymph nodes, which in turn suppressed the progression of colitis.

Effects of Nutritional Level on Digestive Enzyme Activities in the Pancreas and Small Intestine of Calves Slaughtered at Same Body Weight

  • Six Holstein heifer calves weaned at 45 days-of-age were randomly allocated into high daily gain (1.1 kg/d, HDG) and low daily gain (0.56 kg/d, LDG) groups, and were slaughtered at 170 kg of live weight. Energy intake level in the feeding period was 2.4 $\times$ maintenance in 105 days for HDG and 1.4 $\times$ maintenance in 216 days for LDG calves. Total length of the small intestine was identical between groups, but both weights of the pancreas and of the small intestinal mucosa were greater (p < 0.01) for HDG calves. Alpha-amylase, lipase, proteinase, and trypsin activities of the whole pancreas were higher (p < 0.05) in HDG calves. Disaccharidase activity of the whole small intestinal mucosa was also higher (p < 0.10) for HDG than for LDG calves. However, the enzymatic activities, expressed as per gram or per protein of the pancreas and the small intestinal mucosa, were not affected (p > 0.10) by the plane of nutrition. These results suggest that the digestive enzyme activity in the small intestine varies primarily with the weight of tissues synthesizing the enzyme.

  • The appearance of some gastrointestinal endocrine cells in the Meckel's diverticulum (MD) of the bean goose, Anser fabalis Latham was observed using specific antisera against serotonin, gastrin, cholecystokinin (CCK)-8, glucagon, secretin, somatostatin and human pancreatic polypeptide (HPP) with the peroxidase antiperoxidase (PAP) method. Among these specific antisera, serotonin-, gastrin-, CCK-8-, somatostatin- and HPP-immunoreactive cells were demonstrated in this study. Serotonin-, gastrin- and somatostatin-immunoreactive cells were detected at moderate frequency and CCK-8- and HPP-immunoreactive cells was rare and low frequencies, respectively. These immunoreactive cells were located in the superficial epithelium, intestinal crvpt and intestinal glands with spherical or spindle shaped cells having long cytoplasmic processes (open typed-cell). Mucosal layer of MD was composed of simple columnar epithelium and numerous intestinal glands. In addition, numerous lymphatic tissues were also demonstrated. In conclusion, histological profiles of MD were similar to any parts of the large intestine, especially the cecum, but the appearance, distribution and relative frequency of gastrointestinal endocrine cells were similar to those of upper parts of the small intestine. Although the exact digestive functions were unknown, the finding that the appearance, distribution and relative frequency of gastrointestinal endocrine cells in MD is similar to small intestine may be considered as distinct evidence that this organ may have some digestive functions.

Systemic Aspergillosis associated with bovine viral diarrhea virus infection in Korean native calves (한우송아지의 BVD바이러스 감염과 Aspergillus fumigatus의 복합감염)

  • Systemic aspergillosis were outbroken of 17 Korean native calves at livestock farm in Kyunggi province. Two 3 months old calves were died after having diarrhea, coughing, dyspnea, nasal discharge, and abdominal breathing. These calves were diagnosed of aspergillosis as multifocal to coalescing pyogranulomatous inflammation with intralesional aspergillus infiltration at liver, kidney, spleen, heart, lymph nodes, lungs, intestine and cerebrum. Aspergillus fumigatus was cultured from the affected tissues. And the small intestine was positive against bovine viral diarrhea (BVD) virus on FA test. We considered that the secondary fungal infections killed the calves after BVD virus infection primarily.

Biological Activities of Acidic Extracts of the Starfish Asterina pectinifera (별 불가사리(Asterina pectinifera) 조직별 초산추출물의 생리활성 탐색)

  • The present study was performed to examine the contraction and relaxation responses of the smooth muscles, and search for antimicrobial and antioxidant activities in the tissues, of the starfish Asterina pectinifera. Frozen samples were extracted with distilled water containing 1% acetic acid. Extracts from all tissues showed potent antimicrobial activity against Escherichia coli D31. Relatively high levels of antimicrobial activity were also detected in the body extracts. Liver, tube feet, and body extracts caused contraction responses in the dorsal retractor muscles (DRM) of the starfish. In contrast, all tissues examined exhibited contractile activity in the esophagus of squid Todarodes pacificus. In addition, liver and gonad extracts caused contraction responses upon application to the intestine of the puffer fish Takifugu pardalis. Relaxation effects on the DRM of starfish were identified in most of the extracts, while no relaxant activity was detected in body extracts. Extracts from all tissues examined also exhibited antioxidant activities. The results of this study suggest that starfish are a potential source of novel bioactive compounds.

Evaluating sulfoxaflor residues in pig tissues using animal modeling

  • Maximum residue limits (MRL) for pesticides in feed have been set to protect public health and produce safe livestock products. In vivo experiments to establish MRL are essential, as livestock are commonly used to obtain reliable in vivo quantitative information. Here, we aimed to evaluate whether small laboratory animals can replace or reduce monogastric livestock in experiments to quantify pesticide residues in vivo after oral consumption through feed. First, 24 pigs and rats were randomly assigned to four groups and fed 0, 3, 9, or 30 mg/kg of sulfoxaflor. After four weeks, serum, muscle, fat, liver, kidney, and small intestine samples were collected, and sulfoxaflor residues were analyzed using liquid chromatography - tandem mass spectrometry. Sulfoxaflor residues in pig tissues were significantly correlated with those in rat tissues. Model equations were formulated based on the residual sulfoxaflor amount in pig and rat tissues. The calculated and measured sulfoxaflor residues in pigs and rats showed more than 90% similarity. Sulfoxaflor did not affect body weight gain, feed intake, or the feed conversion ratio. Therefore, we concluded that pesticide residue quantification in vivo to establish MRL could be performed using small laboratory animals instead of livestock animals. This would contribute to obtaining in vivo pesticide residue information and reducing large-scale livestock animal experiments.

A Study on the Effect of Sangbaegpitang & Supungsungiwhan on the Glucose Metabolism of db/db Mice (상백피탕(桑白皮湯)과 수풍순기환(搜風順氣丸)이 db/db Mice의 당대사(糖代謝)에 미치는 영향(影響))

  • In this study, body weight levels of glucose, insulin and triglyceride in blood and glucosidase activity of the small intestine were investigated to determine the effect of Sangbaegpitang and Supungsungiwhan on the glucose metabolism of db/db mice. The GLUT4 mRNA of muscle tissue and the Acetyl CoA Carboxylase and the activation rate of GLUT2 mRNA of liver tissue were measured by the reverse transcription-polymerase chain reaction method and by the vitro transcription. The results were obtained as follows: 1. In the Sangbaegpitang administration group, (1) The level of triglyceride was decreased significantly and the glucosidase activity of the small intestine was inhibited remarkably, (2) The amounts of the GLUT4 mRNA in muscle tissue and Acetyl CoA Carboxylase mRNA in liver tissue were increased significantly. (3) Though glucose level in both fasting and non-fasting, were decreased and the insulin level in blood was increased, the results showed no statistical significance. 2. In the Supungsungiwhan administration group, (1) The levels of glucose and triglyceride were decreased significantly in the blood of non-fasting animals. (2) The glucosidase activity of small intestine was inhibited markedly and the amounts of GLUT4 mRNA of muscle tissue and GLUT2 mRNA of liver tissue were increased significantly. (3) The glucose levels in the fasting group were reduced, while insulin level was increased but showed no statistical significance, Based on the above results, our conclusions are as follows: Sangbaegpitang & Supungsungiwhan are thought to be capable of inhibiting the activity glucosidase, the enzyme which influences carbohydrate metabolism in the small intestine of db/db mice(the experimental diabetic model) and delaying the absorption of carbohydrate, thus proving effective on inhibiting the increase of non-fasting glucose level effectively. Futhermore Sangbaegpitang and Supungsungiwhan are though: to be capable of preventing the composition of free fatty acids by restoring the production of GLUT4 mRNA of muscle tissues and GLUT2 mRNA of liver tissues. Those results suggests that above prescriptions can be applied to non-insulin dependent diabetes mellitus in order to improve insulin resistance.

Cadmium Elimination in Tissue of Olive Flounder, Paralichthys olivaceus after Long-Term Exposure (카드뮴 장기노출 후 넙치, Paralichthys olivaceus의 기관에 따른 카드뮴의 배출)

  • Experiments were carried out to investigate the elimination of cadmium in tissues (gill, intestine, liver, kidney and muscle) of flounder, Paralichthys olivaceus, after sub-lethal Cd exposure (5, 50, 100 ${\mu}g/L$). During the depuration phase, Cd concentration in the gill decreased immediately following the end of the exposure period. The elimination rates at the end of depuration periods were $18\%$ for 50 ${\mu}g/L$ exposure and $70\%$ for 100 ${\mu}g/L$ exposure. Intestine showed fastest elimination rates of Cd at all concentration. At the end of the depuration period, the Cd concentration was similar to that in the control, Cd elimination in liver significantly decreased after 10 days of depuration period. After 20 days of depuration, the elimination rate was $66.20\%$ in the fish exposed to 50 ${\mu}g/L$ and $86.22\%$ in the fish exposed to 100${\mu}g/L$. The order of cadmium elimination in tissues were decreased intestine>liver${\geq}$gill>>kidney during depuration periods. In this study the gill, intestine and liver showed faster elimination rate of Cd in 50, 100 ${\mu}g/L$-Cd exposure concentration. After the end of the Cd exposure, the Cd concentration in the kidney slowly decreased or remained constant and the Cd concentration in the muscle slowly increased or remained constant.
