Characterization of Lactobacillus acidophilus Isolated from Piglets and Chicken

  • Lactic acid bacteria were isolated from piglets and chicken and characterized. Lactic acid bacteria showing resistance to low pH and bile, adhesion to intestinal epithelium cells, and the inhibition of Escherichia coli and Salmonella spp. were identified as Lactobacillus acidophilus. L. acidophilus PF01 survived for 2 h in MRS broth adjusted to pH 2. L. acidophilus CF07 was less resistant than L. acidophilus PF01 to pH 2, but survived at pH 2.5 for 2 h. Both of isolates were able to grow in MRS broth containing 0.3% (w/v) bile, with L. acidophilus CF07 being more tolerant to bile than L. acidophilus PF01. L. acidophilus PF01 and CF07 adhered specifically to the duodenal and jejunal epithelium cells of piglet, and the cecal and duodenal epithelium cells of chicken, respectively. Both of isolates did not adhere to the epithelium cells of the various animal intestines from which they were isolated. When L. acidophilus was cultured with E. coli and Salmonella spp. in MRS broth, MRS broth containing 2% skim milk powder or modified tryptic soy broth at $37^{\circ}C$, L. acidophilus PF01 and CF07 inhibited the growths of E. coli K88 and K99, and S. enteritidis and S. typhimurium, respectively. Both of isolates were found to possess the essential characteristics of probiotic lactic acid bacteria for piglet and chicken.

  • The influence of nutritional conditions on the histological changes of hepatocyte and intestinal epithelium as well as hepatosmatic index (HSI), protein, RNA and DNA concentrations of liver of Rhynchocypris oxycephalus was tested. Although, the starved group showed higher concentrations of protein, DNA and RNA than the fed group, food deprivation resulted in a decrease in the HSI, hepatocyte nucleus size and nuclear height of the intestinal epithelium. The RNA-DNA ratio appears to be a useful index of nutritional status in R. oxycephalus and may be useful for determining if R. oxycephalus is in a period of rapid or slow growth at the time of sampling. Additionally, the data have been interpreted in detail and some biologically important relationships discussed.

  • The morphological studies on the rectal development in the 60-, 90- and 120-day-old fetuses and neonates of Korean native goats were investigated by light microscopy and scanning electron microscopy. The rectal wall appeared to be differentiated into the epithelium, lamina propria, tunica muscularis and serosa in 60-day-old fetus. The rectal epithelia were stratified in some areas and simple columnar in others at 60- and 90-day-old fetuses but was only observed simple columnar epithelium at 120-day-old fetus. The goblet cells and the intestinal glands appeared in the rectal mucosa at 90-day-old fetus and gradually increase in number during gestation period. The rectal villi appeared at 60-day-old fetus and increased in length and width until 90-day-old fetus but the villi appeared to be shorter and degenerated after 120-day-old fetus. Only the rudiment trace of the villi remained at birth. The tunica muscularis of the rectum were continuously developed during gestation period. The inner circular and outer longitudinal muscle layers of rectum were observed in the 120-day-old fetuses. Under scanning electron microsoopy, the rectal villi appeared at 60-day-old fetus. The pentagonal/hexagonal microridges and numerous microvilli were observed on the luminal surface of rectal wall. Low and thumb-like rectal villi were developed at 90-day-old fetus. The rectal villi became degenerated after 120-day-old fetus. Only the rudiment of villi was remained at birth. Many circular folds were well developed. Opening portions of intestinal glands appeared like slot.

Cytochemical and Immunocytochemical Study on the Cellulase Activity in the Digestive Tract of the Land Snail Nesiohelix (동양달팽이 Nesiohelix samarangae 소화관에서의 cellulase 활성에 대한 세포화학적 및 면역세포화학적 연구)

  • In order to observe the anticellulolytic localization in the epithelia of the digestive tract such as esophagus, crop, and intestine of a Korean land snail N. samarangae, a cytochemical method and a immunogold labelling method were applied. For the cytochemical study on the cellulase activity, Benedict reaction method applied. And for the immunocytochemical study, the rabbit serum immunoglobuins (IgG) was obtained from the rabbits injected with cellulase which was extracted from body fluid of the snail. The digestive tract tissues of N. samarangae were fixed with 4% paraformaldehyde and 2% OsO4 and embedded in Lowicryl K4M at -40$^{\circ}C$ under UV light (360 nm). The thin sections were loaded on the nickel grids and stained with the serum IgG and protein A-gold complex (particle size: 10 nm). Observations were undertaken with transmission electron microscope (Jeol, JEM-1010). The epithelium of the digestive tract was consisted of five types of cells. In the cytochemical study, the reaction products were found along the periphery of the vacuoles derived from the Bebedict reaction. In the immunocytochemical study, the protein-A gold particles were selectively labelled in Type 1, Type 3 and Type 4 cells in intestinal tissue. membranes of rER, in the surrounding cytoplasm of the rER and secretory granules, and in the apical cytoplasm of the cells. On the material being secreted from the apical cytoplasm was also labelled with the immunogold particles. The all results obtained throughtout present study suggest that the intestinal epithelium of the snail N. samarangae seretes cellulase as one of digestive enzymes.

Effects of Glucagon-like Peptide-2 on Morphology, Proliferation and Enzyme Activity of Intestinal Enterocyte Cells of Weaned Piglets In vitro

  • This study was conducted according to the single-factor design principle to investigate in vitro the effects of different glucagon-like peptide-2 (GLP-2) concentrations (0, $1{\times}10^{-11}$, $1{\times}10^{-10}$, $1{\times}10^{-9}$, $1{\times}10^{-8}$ and $1{\times}10^{-7}$ mol/L) on the morphology, proliferation and enzyme activity of intestinal enterocyte cells of 28-d-old weaned piglets. These cells were primary cultured in 4 pieces of 24-well cell culture plate. After having been grown for 48 h in culture media with hGLP-2, the ileal enterocyte cells of 28-d-old weaned piglets exhibited the typical characteristics of simple columnar epithelium. Compared with the control groups, the quantities of treated cells significantly increased (p<0.05) and their corresponding absorption values in 540 nm (MTT OD) also significantly increased (p<0.01). Likewise, lactic acid concentration, total protein content and protein retention significantly increased (p<0.05). $Na^{+}$, $K^{+}$-ATP enzyme activity was more active (p<0.05), although the activity of alkaline phosphatase, lactic acid dehydrogenase and creatine phosphokinase in culture media significantly decreased (p<0.01). To summarize, the results indicated that GLP-2 in vitro is capable of promoting the proliferation of intestinal enterocyte cells of 28-d weaned piglets, restraining their apoptosis and maintaining the integrity of their morphology.

Heat shock protein 90β inhibits apoptosis of intestinal epithelial cells induced by hypoxia through stabilizing phosphorylated Akt

  • Intestinal epithelial cell (IEC) apoptosis induced by hypoxia compromise intestinal epithelium barrier function. Both Akt and Hsp90 have cytoprotective function. However, the specific role of Akt and $Hsp90{\beta}$ in IEC apoptosis induced by hypoxia has not been explored. We confirmed that hypoxia-induced apoptosis was reduced by $Hsp90{\beta}$ overexpression but enhanced by decreasing $Hsp90{\beta}$ expression. $Hsp90{\beta}$ overexpression enhanced BAD phosphorylation and thus reduced mitochondrial release of cytochrome C. Reducing $Hsp90{\beta}$ expression had opposite effects. The protective effect of $Hsp90{\beta}$ against apoptosis was negated by LY294002, an Akt inhibitor. Further study showed that Akt phosphorylation was enhanced by $Hsp90{\beta}$, which was not due to the activation of upstream PI3K and PDK1 but because of stabilization of pAkt via direct interaction between $Hsp90{\beta}$ and pAkt. These results demonstrate that $Hsp90{\beta}$ may play a significant role in protecting IECs from hypoxia-induced apoptosis via stabilizing pAkt to phosphorylate BAD and reduce cytochrome C release.

Homozygous Missense Epithelial Cell Adhesion Molecule Variant in a Patient with Congenital Tufting Enteropathy and Literature Review

  • Congenital diarrheal disorders (CDDs) with genetic etiology are uncommon hereditary intestinal diseases characterized by chronic, life-threatening, intractable watery diarrhea that starts in infancy. CDDs can be mechanistically divided into osmotic and secretory diarrhea. Congenital tufting enteropathy (CTE), also known as intestinal epithelial dysplasia, is a type of secretory CDD. CTE is a rare autosomal recessive enteropathy that presents with intractable neonatal-onset diarrhea, intestinal failure, severe malnutrition, and parenteral nutrition dependence. Villous atrophy of the intestinal epithelium, crypt hyperplasia, and irregularity of surface enterocytes are the specific pathological findings of CTE. The small intestine and occasionally the colonic mucosa include focal epithelial tufts. In 2008, Sivagnanam et al. discovered that mutations in the epithelial cell adhesion molecule (EpCAM, MIM# 185535) were the genetic cause of CTE (MIM# 613217). More than a hundred mutations have been reported to date. Furthermore, mutations in the serine peptidase inhibitor Kunitz type 2 (SPINT2, MIM# 605124) have been linked to syndromic CTE. In this study, we report the case of a 17-month-old male infant with congenital diarrhea. Despite extensive etiological workup, no etiology could be established before admission to our center. The patient died 15 hours after being admitted to our center in a metabolically decompensated state, probably due to a delay in admission and diagnosis. Molecular autopsy with exome sequencing revealed a previously reported homozygous missense variant, c.757G>A, in EpCAM, which was confirmed by histopathological examination.

  • Gastrointestinal cyst found within the oral cavity has been reported as a rare occurrence with less than 40 cases. The tongue is the preferred site with almost 60% of cysts occurring within oral cavity. The cyst usually shows an asymptomatic swelling in the floor of the mouth, about 30% of affected individuals have symptoms related to difficulties with feeding, swallowing and respiration. The histopathologic features of the cyst vary with respect to the type of epithelium lining; gastric, squamous, intestinal and respiratory epithelium. The pathogenesis is not known, but it may be related to entrapment of undifferentiated endoderm within oral cavity during early fetal development. In this article, we report a case of a gastrointestinal cyst of the tongue in a 16-month-old girl.

  • The insect midgut epithelium is generally lined with a unique chitin and protein structure, the peritrophic membrane (PM), which facilitates food digestion and protects the gut epithelium. We used gel electrophoresis and mass spectrometry to identify the extracted proteins from the silkworm PM to obtain an in-depth understanding of the biological function of the silkworm PM components. A total of 305 proteins, with molecular weights ranging from 8.02 kDa to 788.52 kDa and the isoelectric points ranging from 3.39 to 12.91, were successfully identified. We also found several major classes of PM proteins, i.e. PM chitin-binding protein, invertebrate intestinal mucin, and chitin deacetylase. The protein profile provides a basis for further study of the physiological events in the PM of Bombyx mori.

  • Vaccine is one of the best known and most successful applications of immunological theory to human health and it protects human life through inducing the immune response in systemic compartment. However, when we consider the fact that mucosal epithelium is exposed to diverse foreign materials including viruses, bacteria, and food antigens and protects body from entry of unwanted materials using layer of tightly joined epithelial cells, establishing the immunological barrier on the lining of mucosal surfaces is believed to be an effective strategy to protect body from unwanted antigens. Unfortunately, however, oral mucosal site, which is considered as the best target to induce mucosal immune response due to application convenience, is prone to induce immune tolerance rather than immune stimulation. Since intestinal epithelium is tightly organized, a prerequisite for successful mucosal vaccination is delivery of antigen to mucosal immune induction site including a complex system of highly specialized cells such as M cells. Consequently, development of efficient mucosal adjuvant capable of introducing antigens to mucosal immune induction site and overcome oral tolerance is an important subject in oral vaccine development. In this review, various approaches on the development of oral mucosal adjuvants being suggested for effective oral mucosal immune induction.
