Purpose: Necrotizing enterocolitis (NEC) is one of the most serious complications of prematurity. Many risk factors can contribute to the development of NEC. The epidermal growth factor (EGF) plays a major role in intestinal barrier function, increases intestinal enzyme activity, and improves nutrient transport. The aim of this study was to assess the role of epidermal growth factor in the development of NEC in preterm neonates. Methods: In this study, 130 preterm neonates were included and divided into 3 groups, as follows: group 1, 40 preterm neonates with NEC; group 2, 50 preterm neonates with sepsis; and group 3, 40 healthy preterm neonates as controls. The NEC group was then subdivided into medical and surgical NEC subgroups. The serum EGF level was measured using enzyme-linked immunosorbent assay. Results: Serum EGF levels (pg/dL) were significantly lower in the NEC group (median [interquartile range, IQR], 9.6 [2-14]) than in the sepsis (10.1 [8-14]) and control groups (11.2 [8-14], P<0.001), with no significant difference between the sepsis and control groups, and were positively correlated with gestational age (r=0.7, P<0.001). A binary logistic regression test revealed that low EGF levels and gestational ages could significantly predict the development of NEC. The receiver-operating characteristic curve for EGF showed an optimal cutoff value of 8 pg/mL, with 73.3% sensitivity, 98% specificity, and an area under the curve of 0.92. Conclusion: The patients with NEC in this study had significantly lower serum EGF levels (P<0.001), which indicated that EGF could be a reliable marker of NEC in preterm neonates.
Probiotics are microorganisms that can benefit host health when ingested in a live state, and lactic acid bacteria are the most common type. Among fungi, Saccharomyces boulardii (SB) is the only strain known to have a probiotic function with beneficial effects on colitis; however, information on other probiotic yeast strains is limited. Therefore, this study aimed to discover yeast strains expressing intestinal anti-inflammatory activities by exhibiting probiotic properties in dextran sodium sulfate (DSS)-induced colitis mice model. Nuruk (Korean traditional fermentation starter) containing various microbial strains was used as a source for yeast strains, and S. cerevisiae 28-7 (SC28-7) strain was selected with in vitro and in vivo characteristics to enable survival in the intestines. After 14 days of pretreatment with the yeast strains, DSS was co-administered for six days to induce colitis in mice. The results revealed that the disease activity index score was lowered by SC28-7 treatment compared to the DSS group, and the colon length and weight/length ratio were recovered in a pattern similar to that of the normal group. SC28-7 administration significantly reduced the secretion of pro-inflammatory cytokines in the serum and modified the mRNA expression of inflammatory cytokines (interleukin-1β, transforming growth factor-β, and interferon-γ) and proteins involved in gut barrier functions (mucin 2, mucin 3, zonula occludens-1, and occludin) in colon tissues. These results indicate that SC28-7 attenuates DSS-induced colon damage and inflammation, supporting its future use as a probiotic yeast for treating and preventing intestinal inflammatory diseases such as inflammatory bowel disease.
Milk oligosaccharides are the complex mixture of six monosaccharides namely, D-glucose, D-galactose, N-acetyl-glucosamine, N-acetyl-galactosamine, L-fucose, and N-acetyl-neuraminic acid. The mixture is categorized as neutral and acidic classes. Previously, 25 oligosaccharides in bovine milk and 115 oligosaccharides in human milk have been characterized. Because human intestine lacks the enzyme to hydrolyze the oligosaccharide structures, these substances can reach the colon without degradation and are known to have many health beneficial functions. It has been shown that this fraction of carbohydrate can increase the bifidobacterial population in the intestine and colon, resulting in a significant reduction of pathogenic bacteria. The role of milk oligosaccharides as a barrier against pathogens binding to the cell surface has recently been demonstrated. Milk oligosaccharides have the potential to produce immuno-modulation effects. It is also well known that oligosaccharides in milk have a significant influence on intestinal mineral absorption and in the formation of the brain and central nervous system. Due to its structural resemblance, bovine milk is considered to be the most potential source of oligosaccharides to produce the same effect of oligosaccharides present in human milk. This review describes the characteristics and potential health benefits of milk oligosaccharides as well as the prospects of oligosaccharides in bovine milk for use in functional foods.

Obesity and related metabolic diseases are major problems worldwide. Some probiotics are currently considered potential therapeutic strategies for obesity. We aimed to investigate the anti-obesity efficacy of Latilactobacillus sakei WIKIM31 in obese mice induced by a high fat diet. The administration of a high-fat diet with L. sakei WIKIM31 reduced body weight gain, epididymal fat mass, triglyceride and total cholesterol levels in the blood, and remarkably decreased the expression of lipogenesis-related genes in the epididymal adipose tissue and liver. Interestingly, intake of L. sakei WIKIM31 improved gut barrier function by increasing the gene expression of tight junction proteins and suppressing the inflammatory responses. Additionally, L. sakei WIKIM31 enhanced the production of short-chain fatty acids, such as butyrate and propionate, in the intestinal tract. These results showed that L. sakei WIKIM31 can be used as a potential therapeutic probiotic for obesity.
One of the most powerful health management practices is the use of antibiotics, but their use is being restricted because of health safety issues. The swine industry has been looking for various alternatives to antibiotics and increasingly considers the use of dietary factors like feed ingredients, feed additives, feed formulation practices, or feeding methods, instead of using antibiotics to improve pig health and performance. Among other alternatives to antibiotics, spray-dried plasma may be a candidate. Spray-dried plasma is a blood product that provides bioavailable nutrients and physiologically active components such as immunoglobulins, glycoproteins, growth factors, peptides, etc. It is an excellent protein source with balanced and highly digestible amino acids. Several beneficial physiological activities depend on components of spray-dried plasma, such as immune competence (antibacterial activity), modulation of microbiota and/or immune system, integrity of intestinal barrier function, etc. These beneficial effects can contribute to improvement of pig performance and health by modulation of microbiota in the digestive tract and/or immune system. Therefore, it is suggested that spray-dried plasma has great potential as an antibiotics alternative.
장 질환은 점막 세포의 파괴로부터 진행되어 장 상피세포벽의 기능상실, 비정상적인 장벽 활동을 야기하며, 이는 또 다시 염증반응의 가속화를 야기하는데[1], 여러 연구에도 불구하고 염증성 장질환에 대한 병변은 뚜렷이 밝혀진 바가 없다. 지난 수년간 병리학적 기전에 근거한 염증성 장질환에 대해 여러 연구가 이루어져 왔으며, 현재 만성적인 염증성 장 질환의 경우 여러 종류의 혈중 사이토카인 증가로 인한 과도한 세포 면역반응과 관련이 있을 것으로 추정되고 있다. 이러한 이유로 비정상적인 면역반응을 유도하는 전염증성 사이토카인인 TNF-${\alpha}$와 같은 특정 사이토카인의 발현을 전사단계에서부터 선택적으로 제거하는 방법을 통해 염증성 질환의 예방 및 치료에 접근하고자 하는 시도가 기대를 모으고 있다. 향후 면역반응 조절을 통한 염증성 장 질환 연구는 장 질환 자체의 세부적인 치료법에 대한 발전뿐만 아니라 염증과 관련된 여러 질병들의 병리학적 증상에 대해서도 새로운 접근법을 제시할 수 있을 것이다. Immunex(Enbrel), J&J/Centocor(Remicade)와 같은 사이토카인 억제제-쥐에서 유도된 단일클론 항체-의 경우 여러 연구를 통해 염증성 장질환 환자들의 증상을 완화하는 것으로 밝혀졌으나 면역과 관련된 부작용을 동시에 갖고 있으며, 비용적인 문제와 주사제를 이용해야 하는 치료방법의 제한점을 가지고 있다. 이러한 이유로 환자 모두가 사이토카인 억제 약물을 통한 치료를 받는 것이 현실적으로 어려운 상황이다. 본 연구는 양(羊)의 초유(初乳)에서 생성된 TNF-${\alpha}$ 항체의 TNF-${\alpha}$의 활성을 억제를 통한 염증반응 완화능을 세포단계의 생물검정과 장 염증이 유도된 동물모텔을 통해 검증하였다. 양(羊)의 초유(初乳)에서 생성된 TNF-${\alpha}$ 항체의 사이토카인 발현 억제능을 살펴보기 위하여 세포 생물검정을 수행하였다. 항체는 1 : 10,000 희석배율에서 TNF-${\alpha}$의 활성을 완전히 억제하였으며 동일한 양의 다른 양유(羊乳)를 이용한 실험에서도 억제능의 정도에 일부 차이를 보였으나 대조군에 비하여 모든 실험군에서 TNF-${\alpha}$의 활성이 억제었다. 동물실험 1의 경우 초기 시범 실험을 통해 염증유도 물질인 PAF(Platelet activating factor)와 LPS(Lipopolysaccharides)의 투여량을 설정하였으나, 본 실험 중 과도한 장내 염증반응으로 인해 출혈을 일으키거나 폐사하였으며, 동물실험 2에서는 TNBS(Trinitrobenzenesulphonic acid)를 이용한 대장염 유도를 시도하였으나, 실험군의 50% 정도만이 대장염으로 인한 전형적인 체중 감소와 일반적인 병리학 증세를 보였다. 이상의 결과로 미루어 면역반응을 통해 생성된 양(羊)의 초유(初乳)에 함유된 TNF-${\alpha}$ 항체는 WEHI-13 VAR 세포의 TNF-${\alpha}$ 활성을 유의적으로 억제함을 확인할 수 있었다. 동물실험 1의 경우, 예상되는 TNF-${\alpha}$ 항체의 염증반응 억제 효과보다 유도된 염증반응의 정도가 강하였고, 실험 2의 경우, 대장염 유도에 대한 실험동물간의 민감성 차이를 나타내었다. 향후 항TNF-${\alpha}$ IgA 치료법을 통한 염증 유래 장질환 연구를 위해서 적합한 실험동물 모델의 개발이 필요하며 더 많은 항체 개발과 함께 수반되는 전임상 및 임상실험 역시 이루어져야 할 것으로 사료된다.

The widespread use of antimicrobials causes antibiotic resistance in bacteria. The use of butyric acid and its derivatives is an alternative tactic. This review summarizes the literature on the role of butyric acid in the body and provides further prospects for the clinical use of its derivatives and delivery methods to the animal body. Thus far, there is evidence confirming the vital role of butyric acid in the body and the effectiveness of its derivatives when used as animal medicines and growth stimulants. Butyric acid salts stimulate immunomodulatory activity by reducing microbial colonization of the intestine and suppressing inflammation. Extraintestinal effects occur against the background of hemoglobinopathy, hypercholesterolemia, insulin resistance, and cerebral ischemia. Butyric acid derivatives inhibit histone deacetylase. Aberrant histone deacetylase activity is associated with the development of certain types of cancer in humans. Feed additives containing butyric acid salts or tributyrin are used widely in animal husbandry. They improve the functional status of the intestine and accelerate animal growth and development. On the other hand, high concentrations of butyric acid stimulate the apoptosis of epithelial cells and disrupt the intestinal barrier function. This review highlights the biological activity and the mechanism of action of butyric acid, its salts, and esters, revealing their role in the treatment of various animal and human diseases. This paper also discussed the possibility of using butyric acid and its derivatives as surface modifiers of enterosorbents to obtain new drugs with bifunctional action.
전염증성 사이토카인의 분비를 조절하는 전사인자인 NF-${\kappa}B$는 염증성 장질환 환자의 대장 점막에서 발현이 증가되어 있다고 보고하고 있으며 이를 억제하여 대장의 염증을 억제하려는 연구가 진행 중이다. 루테올린은 다양한 한약제에 포함되어 있는 플라보노이드 중 하나로 항염증 및 항산화작용이 있다고 알려져 있으며 LPS 자극된 대식세포에서 TNF-${\alpha}$ 분비의 억제 뿐만 아니라 전사인자인 NF-kB의 발현을 억제하여 항염증작용을 하는 것으로 알려져 있다. 본 연구에서는 DSS을 이용한 염증성 장질환 모델에서 루테올린의 장염의 치료효과를 보고자 하였다. C57BL/6 NF-${\kappa}B^{EGFP}$ 쥐에게 2.5% DSS를 투여하여 장염을 유발하였으며 치료군(n=6)에서는 매일 루테올린(1 mg/kg, vol 0.1 ml)을 비위관을 통해 경구투여 하였으며, 비치료군(n=6)에서는 매일 같은 양의 vehicle(vol 0.1 ml)를 투여하여 정상대조군(n=6)과 비교하였다. 실험 기간 동안 질병활성도를 기록하였으며, 투약 6일 후 모든 쥐를 희생하여 대장을 분리하여 길이를 측정하고, 조직검사를 시행하였다. 또한 대장점막조직을 배양하여 m IL-12 p40의 분비를 측정하였고, 공초점 형광현미경하에서 EGFP 발현의 정도를 관찰하였다. 질병의 활성도의 관찰에서 치료군에서 2.7점, 비치료군에서 2.5점으로 양군 사이에 유의한 차이가 없어 루테올린에 대한 장염예방효과는 없었다. 또한 치료군과 비치료군에서 대장점막의 m IL-12 p40의 분비 각각 $535.2{\pm}198.2pg/ml$, $412.5{\pm}48.2pg/ml$ 로 양군 사이의 차이는 없었다. 흥미롭게도 EGFP 발현은 치료군에서 오히려 높게 나타났으며, 공초점 형광현미경관찰에서 대부분의 고유근층하 대식세포에서 증가되어 있음을 알 수 있었다. 이상의 결과로 루테올린의 경구 투여는 DSS 유발 염증성 장질환 모델에서 대장 점막 염증을 억제하는 대체 요법으로써의 치료효과는 관찰할 수 없었으며, 향후 항염증작용이 있다고 알려져있는 플라보노이드 물질의 개발에 신중함이 있어야 할 것으로 사료된다.
