Gallic Acid Inhibits STAT3 Phosphorylation and Alleviates DDS-induced Colitis via Regulating Cytokine Production

  • Signal transducer and activator of transcription 3 (STAT3) is associated with various human diseases, such as cancer, auto-immune disease, and intestinal inflammation. The limited and inadequate effect of standard approaches for treating inflammatory bowel disease (IBD) has prompted to develop alternative anti-colitis agents through inhibition of STAT3. Here, we show that gallic acid (GA), a 3,4,5-trihydroxybenzoic acid, markedly reduced phosphorylation of STAT3. Among the derivatives of benzoic acids, GA showed significant inhibition on STAT3 phosphorylation. In addition, GA ameliorated the dextran sodium sulfate (DSS)-induced acute colitis as determined by the measurement of symptomatic and histological indices. The suppression of DSS-induced acute colitis by GA treatment may be related to the regulation of cytokines and growth factors. Furthermore, GA inhibited phosphorylation of STAT3 in the colon tissue of DSS-treated mice. These findings may be useful in comprehending the molecular action of GA on STAT3 phosphorylation and provide novel insights into the potential application of GA in the treatment of STAT3-related inflammatory disease, such as IBD.

The produced mealworm meal through organic wastes as a sustainable protein source for weanling pigs

  • This study was designed to evaluate the influence of dietary supplementation of Tenebrio molitor larvae (TM) as an alternative source of protein to fish meal on growth response, retention of nutrient, hematological factors, immune system, and intestinal integrity in weaned pigs. A total of 180 weanling pigs (initial bodyweight of 6.27 ± 0.15 kg; 21 d) were distributed based on the bodyweight between 3 treatments with 6 replicates in each treatment including 10 piglets per replicate. The diets corresponding to treatments consisted of a fish meal diet (CON), a fish meal-dried mealworm diet (TM1, 50% replacement of fish meal with TM meal), and a dried mealworm diet (TM2, 100% replacement of fish meal with TM meal). This study was performed in two phases as phase 1 (d 0 to 14) and phase 2 (d 15 to 28). The pigs in the TM2 treatment showed a greater gain to feed ratio compared with the TM1 pigs in phase 1. Throughout the experimental period, the average daily gain (ADG) of the TM1 pigs was significantly greater than that of the TM2 treatment. The IgG concentration was increased in the TM1 and TM2 treatments compared with the CON pigs in phase 1. In conclusion, the supplementation of TM meal (50% replacement of fish meal) did not show any adverse effects in the performance of weanling pigs, however, 100% replacement of fish meal with TM meal is not recommended.

  • 가장 대중적(大衆的)으로 이용(利用)되고 있는 식품(食品)인 라면, 식빵 건조인절미를 쥐의 기초사료(基礎飼料)와 여러 가지로 혼합(混合)하여 10개군(個群)의 쥐에 대(對)해 사육(飼育)실험하여 생리적(生理的) 변화(變化)를 조사(調査)한 바 다음과 같았다. 1. 체중증가율(體重增加率)은 라면과 기초사료를 1 : 4 또는 1 : 2로 혼합(混合)한 경우가 가장 우수(優秀)하였으며 대조구(對照區)를 능가하였고 라면, 식빵, 인절미 등을 단용(單用)한 경우는 약간 저조(低調)하였다. 2. 사육시험중(飼育試驗中)의 장내세균(腸內細菌) flora의 변동(變動), 간기능(肝機能) 그리고 신장(腎臟)의 기능(機能)은 모두 정상적(正常的)이었으나, albumin globulin의 비율(比率)은 전(全) 시험구(試驗區)에서 대조구(對照區)와 공(共)히 약간 낮은 치(値)를 보였다. 3. 간(肝), 신장(腎臟)의 조직도 전시험구(全試驗區)에서 대조구(對照區)와 공(共)히 전시험기간중(全試驗其間中) 이상(異常)이 없었다.

  • Kefir exhibits antimicrobial activity in vitro against gram-positive and gram negative bacteria, as well as some fungi. The ability of LAB to inhibit the growth of closely related bacteria is well known. This inhibition of pathogenic and spoilage microbes may be due to the production of organic acids, hydrogen peroxide, acetaldehyde, diacetyl, carbon dioxide, or bacteriocins. Lactobacilli are the major contributors to acid production and, hence, a determining factor in the flavor development in kefir. Lactic acid, proteolytic activity, and acetaldehyde are the essential flavor compounds in kefir. Both acid and bacteriocins contribute to the antimicrobial activity of kefir and kefir grains. Kefir is rich in proteins, calcium, vitamin $B_{12}$, niacin, and folic acid. Many studies have investigated the benefits of consuming kefir and have shown that it is a natural probiotic, which when consumed regularly, can help relieve intestinal disorders, promote bowel movement, reduce flatulence, and improve the overall health of the digestive system. Tibetan kefir, which is different from traditional kefir, is produced in China. It has been reported to exhibit antimicrobial activity that is nearly identical to that of traditional kefir. Kefir production is considered a rapidly growing food industry in China.

리팜피신과 오플로삭신에 내성인 Enterococcus faecalis 균주의 개발 (Development of Enterococcus faecalis Strains Resistant to Rifampicin and Ofloxacin)

  • The preparation of Enterococcus faecalis RSI is used as a therapeutics for human intestinal disorders. However, the microbe in this preparation is usually very sensitive to rifampicin and fluoroquinolones. If this preparation is taken with rifampicin or fluoroquinolones, its therapeutic effect can not be expected. E. faecalis RFR11, containing resistance to rifampicin was obtained by MNNG mutation method. Serial passage of E. faecalis RFR11 produced E. feacalis OFR16 on agar with 2-fold minimal inhibitory concentration of ofloxacin produced. E. feacalis OFR16 was resistant to fluoroquinolones up to 8-256 fold higher than that for the original strain. E. faecalis OFR16 also exhibited identical characteristics with the parent strain when they were tested for lactic acid formation and growth inhibition of E. coli MB4-5737 and Shigella sonnei MB4-10411. From in vitro test, it was identified that rifampicin and ofloxacin is not inactivated by certain factors of E. faecalis OFR16. Conclusively. E. faecalis OFR16, rifampicin and fluoroquinolones resistant mutant, is an efficient strain that has insensitivity against rifampicin and fluoroquinolones and original biochemical characteristics of the parent strain.

국내 시판용 Frozen Yogurt의 병원성 미생물 검출 및 미생물학적 품질 평가에 관한 연구 (Detection of E. coli O157:H7 and Listeria monocytogenes, and Appraisal for Microbiological Qualities in the Commercial Frozen Yogurt Products in Korea)

  • Recently the high outbreaks of intestinal disease caused by the consumption of frozen dairy foods containing pathogenic bacteria has generated considerable interest in the causative agent such as Listeria monocytogenes and E. coli O157:H7. This study was carried out to detect the above pathogens and compare the microbiological qualities of three commercial forzen yogurt products. The main results obtained were as follows. L. monocytogenes coliforms and E. coli O157:H7 were not detected in the total of seven frozen yogurt samples. For microbiological qualities the viable lactic counts of products manufactured by FA company were about 2.9$\times$108 -1.6$\times$109cfu/ml 1.7$\times$106 cfu/ml for FB's and 1.2$\times$106 cfu/ml for FC's The PH values of FA's FB's and FC's products was in the range of pH 4.1~5.3 and the values of FA's were 4.1~4.6 compared by the pH 5.2~5.3 of FB's and FC's products. During refrigeration of the test samples the survival rates of L. monocytogenes spiked into thawed frozen yogurt sample(FA's FB,s and FC's) were 0.55% 15.61% and 16.89% respectively. On the other hand E. coli O157:H7 and L. monocytogenes were 12.4% and 25.0% for FA's 10.8% and 20.8% for FB's and 10.26% and 22.7% for FC's under 37$^{\circ}C$ storage, As the results described above each frozen yogurt products were different in microbiological qualities. The survival rates of pathogens spiked into the samples increased with the pH of the products. This indicates that the pH or any other factors pre-sumable supressed the growth of E. coli O157:H7 and L. monocytogenes in frozen yogurt products.

가금티브스균 Salmonella gallinarum의 생육을 저해하는 길항미생물의 선발 및 동정 (Isolation, Identification and Cultural Condition of the Antagonistic Microorganism Against Salmonella gallinarum Causing Fowl Typhoid)

  • 가금티프스는 가금류에 Salmonella gallinarum이 원인균이 되어 발병하는 양계산업에 막대한 지장을 주는 질병이다. 가금티프스를 억제하기 위한 생균제 개발을 위한 목적으로써 가금티프스 원인균인 Salmonella gallinarum의 생육을 저해시킬 수 있는 길항균주를 토종닭의 내장으로부터 분리하여 생육특성과 길항물질 생산성을 조사하고, 이 균주를 분류학적으로 동정하였다. 분리된 길항균주는 Bacillus amyloliquefaciens와 98% 상동성을 나타내어 최종적으로 Bacillus amyloliquefaciens Y3로 명명하였다. 0.3% maltose, 0.2% $NH_4Cl,\; 37^{\circ}C$ pH 7에서 균생육 및 길항물질의 생산능이 가장 우수하였으나 장내 담즙에 대한 내성은 크게 나쁘지 않을 것으로 확인되어졌다. 생산된 길항물질을 추정하여 본 결과 분자량이 10,000보다 작은 저분자물질이었으며 $80^{\circ}C$에서 20분간 열처리한 후에도 80%의 활성을 유지하는 내열성 물질임을 확인할 수 있었다. 향후 선발되어진 Bacillus amyloliquefaciens Y3가 생산해내는 길항물질에 대한 연구와 개량을 통하고 장내 정착성 실험을 거쳐 우수균주로 확인되면 양계산업에 사용될 우수한 생균제로 개발이 가능할 것이라 생각된다.

사료급여별 산란계의 체 성장 및 체 조성 특성 비교

  • 산란계의 성장과정 중의 체 조성과 기관의 발달이 어떻게 이루어지고 성장제한에 따라 차이가 어떻게 나타나는가를 구명하기 위하여 갈색 산란계 1,080수를 공시하여 70주령시까지 체 조성 및 기관의 발육특성을 조사하였다 산란 개시지점까지의 체 조성은 동일 체중에서 상이하였는데, 특히 지방함량이 제한급이구가 유의적으로 낮았다. 산란개시 이후 체 조성을 처리별 차이가 없이 비슷하게 나타났다. 번식기관은 1,000 g 이후 서서히 발달하여 산란개시 시기인 1,400 g 근처에서 급격히 증가하였는데 발육은 대조구, T2, T1의 순으로 나타났으며 산란개시 시점과 일치하였다. 소화기관 및 간장의 비율은 제한급이구가 1,000g 시까지지 높았으며 이후 전 기간 일정하게 유지되었다. 이상의 결과에서 제한급이는 육성기체 조성에 영향을 미치고 이러한 영향은 산란개시 시기와 관계가 있었다.

버섯 섭취와 장내 미생물 균총의 변화 (Changes in gut microbiota with mushroom consumption)

  • 버섯의 섭취는 버섯의 유용 성분에 의해 면역체계 및 장내미생물균총의 변화를 유발한다. 그중 많이 알려진 β-glucan 구조 기반 물질들은 염증매개물질의 분비는 억제하고 대식세포의 활성은 유발하여 면역력의 증진을 도와인체의 면역기능을 증진시킨다. 직접적인 유용 성분 이외의 다른 물질은 장내 미생물균총에 의해 단쇄지방산으로 변화되며, 변화된 단쇄지방산은 면역 및 다양한 질병의 완화를 유발하게 된다. 버섯이 포함하고 있는 성분은 β-glucan 외에도 장내미생물균총을 변화하게 유도하는 prebiotic로써의 역할을 수행하게 된다. 변화된 장내미생물균총은 외부로부터 들어오는 다양한 감염성 균의 감염을 막으며, 면역체계의 균형 유지를 도우며, 질병을 예방하게 된다. 국내 섭취 빈도가 높은 새송이, 표고, 느타리, 팽이, 목이, 양송이 버섯의 유용 성분의 탐색, 장내미생물균총의 변화 및 섭취 시 인체의 단쇄지방산 변화 연구는 추후 버섯의 유용 성분, 장내미생물 및 질병과의 인과관계를 풀어내는데 기여할 것으로 여겨진다.

Antimicrobial activity of fermented Maillard reaction products, novel milk-derived material, made by whey protein and Lactobacillus rhamnosus and Lactobacillus gasseri on Clostridium perfringens

  • Objective: The objective of this study was to evaluate the antimicrobial effects of fermented Maillard reaction products made by milk proteins (FMRPs) on Clostridium perfringens (C. perfringens), and to elucidate antimicrobial modes of FMRPs on the bacteria, using physiological and morphological analyses. Methods: Antimicrobial effects of FMRPs (whey protein plus galactose fermented by Lactobacillus rhamnosus [L. rhamnosus] 4B15 [Gal-4B15] or Lactobacillus gasseri 4M13 [Gal-4M13], and whey protein plus glucose fermented by L. rhamnosus 4B15 [Glc-4B15] or L. gasseri 4M13 [Glc-4M13]) on C. perfringens were tested by examining growth responses of the pathogen. Iron chelation activity analysis, propidium iodide uptake assay, and morphological analysis with field emission scanning electron microscope (FE-SEM) were conducted to elucidate the modes of antimicrobial activities of FMRPs. Results: When C. perfringens were exposed to the FMRPs, C. perfringens cell counts were decreased (p<0.05) by the all tested FMRPs; iron chelation activities by FMRPs, except for Glc-4M13. Propidium iodide uptake assay indicate that bacterial cellular damage increased in all FMRPs-treated C. perfringens, and it was observed by FE-SEM. Conclusion: These results indicate that the FMRPs can destroy C. perfringens by iron chelation and cell membrane damage. Thus, it could be used in dairy products, and controlling intestinal C. perfringens.
