Mitochondria play key roles in the production of cell's energy. Their dominant function is the synthesis of adenosine 5'-triphosphate (ATP) from adenosine diphosphate (ADP) and phosphate (Pi) through the oxidative phosphorylation. Evaluation of drug-induced mitochondrial toxicity has become increasingly important since mitochondrial dysfunction has recently been implicated in numerous diseases including cancer and diabetes mellitus. Mitochondrial functions have been monitored via oxygen consumption, mitochondrial membrane potential, and more importantly via ATP synthesis since ATP synthesis is the most essential function of mitochondria. Various analytical methods have been employed to investigate ATP synthesis in mitochondria, including high performance liquid chromatography (HPLC), bioluminescence technique, and pH measurement. However, most of these methods are based on destructive analysis or indirect monitoring through the enzymatic reaction. Infrared absorption spectroscopy (IRAS) is one of the useful techniques for real-time, label-free, and direct monitoring of biological reactions [1,2]. However, the strong water absorption requires very short path length in the order of several micrometers. Transmission measurements with thin path length are not suitable for mitochondrial assays because solution handlings necessary for evaluating mitochondrial toxicity, such as rapid mixing of drugs and oxygen supply, are difficult in such a narrow space. On the other hand, IRAS in the multiple internal reflection (MIR) geometry provides an ideal optical configuration to combine solution handling and aqueous-phase measurement. We have recently reportedon a real-time monitoring of drug-induced necrotic and apoptotic cell death using MIR-IRAS [3,4]. Clear discrimination between viable and damaged cells has been demonstrated, showing a promise as a label-free and real-time detection for cell-based assays. In the present study, we have applied our MIR-IRAS system to mitochondria-based assays by monitoring ATP synthesis in isolated mitochondria from rat livers. Mitochondrial ATP synthesis and hydrolysis were in situ monitored with MIR-IRAS, while dissolved oxygen level and solution pH were simultaneously monitored with O2 and pH electrodes, respectively. It is demonstrated that ATP synthesis and hydrolysis can be monitored by the IR spectral changes in phosphate groups in adenine nucleotides and MIR-IRAS is useful for evaluating time-dependent drug effects of mitochondrial toxicants.
This report does studied for making the method of conserving bracket murals in Daeungjeon of Jikjisa Temple, through the scientific way. Results of evaluated the conservation status at the braket mural paintings, most serious damage is structural damage like cracks, breakage, and delamination. After optical investigation, a characteristic point wasn't found such as underdrawing or traces of a coat of paint. The ultrasonic examination speed by each wall painting was measured from about 195.8 m/s to 392.7 m/s, according to the location of the surface, and it was able to compare the surface properties according to the location. In Infrared-thermal image measurement shows that wall layer separation and paint layer delamination are closely detected, therefore it was able to judge of damage on the objective way. Material analysis revealed that the walls were made by sand and weathering soil. The wall layer combined sand with less than fine sand size by nearly 5:5, and the finishing layer was found to have mixed medium sand and fine sand at approximately 6:4 rates. However, In case of finishing layer, mixing ratios of sizes less than very fine sand were found to be significantly lower than wall. Therefore, it is estimated that the plysical damage such as the separation between the layers of the walls created in the braket mural paintings, is continuously caused by changes in the internal stresses and volume ratio caused by the density differences between the wall and the finishing layers.
According to recent ground collapse occurrence, ground subsidence is increasing every year in downtown area, which is a social problem. The purpose of this study is to investigate the relationship between ground water level lowering and ground collapse through laboratory model experiments. After mixing 1:1 granite weathered soil with sand, sandy soil was formed as a relative density of 30%, 50%, and 80%. And then the changes of soil discharge with change of groundwater level were compared. The physical property of material of which particle distribution were well graded with maximu dry unit weight of $1.94kg/cm^3$ and internal friction angle of 37degrees. Ground water levels were measured at 10 cm, 20 cm, and 30 cm from the bottom. As a result, the experiment shows that the higher the groundwater level works the higher the discharge velocity and the magnitude of underground cavity also increases with elapsed time. Finally, the cumulative quantity of soil discharge occurred up to 30 kg at the elapsed time, 35 minutes. It was also confirmed that the range of ground collapse increased due to soil discharge with ground water level lowering.

This study is to form the self assembly of cubic crystalline phase to penetrate into the skin epidermis. The various performance synthesized diglyceryl phytylacetate (DGPA) having hydroxyl group (-OH) and 4 methyl chains with phytyl group was carried out as an amphoteric lipid such as emulsifying power, self assembly. Emulsifying activity of DGPA was very stabilized on only 1% of small content, it could make a W/O emulsion containing high internal phase incorporated with water. Cubic liquid crystal structure with DGPA on three-phase diagram was formed, when mixed DGPA, dimethicone (2CS), and water. Through three-phase diagram forming the cubic liquid crystal area, hexagonal structure zone, and mixing water phase and hexagonal structure area, reversed micelle area were respectively certified. Its structure was proved by the SAXS (small angled x-ray scattering) analysis. As an application, formation of cubosome containing 10% of magnesium ascorbylphosphate and 5% of pyridoxine tris-hexyldecanoate was encapsulated. Occlusive effect of cubosome had above 1.7 times better than reversed micelle. From using poloxamer of dispersing agent, phase structure recovered from W/O emulsion to cubic liquid crystal phase when storage in $33^{\circ}C$ incubator. Therefore, our this study is expected to be as epidermal-dermal skin absorbers in skin care cosmetics and pharmaceuticals industries as raw materials to form a cubic crystal phase through a more in-depth research to DGPA having amphoteric lipid property.

First of all, the multi-story Housing applying resident's participation in europe was classified by the menu selection method, the two-step supply method and the cooperative method. And then I analyzed flexible unit plan of cases for deriving the planning methode and the characteristics of flexibility. First, I analyzed the area and form of the unit plan, structure and Installation, fixed and variable elements to derive the planning method. The area of units are distributed from a minimum of $35m^2$ to a maximum of $150m^2$, and many of the unit planes have a narrow front and a deep depth. The structure is a long-span wall-structure or a skeleton structure, and is designed without any columns and bearing walls in the interior space for flexibility in spatial composition. The vertical shafts are located in the center of the unit in a box-form or in the corner at the unit dividing wall for free placement of interior wall. Fixed elements are framework and facility systems. Most of the future residents in the two-steps supply method and the cooperative method were able to freely design the internal space within the zoning concept proposed by the architect and change the location of the facade element within module system proposed by the architect. Second, the characteristics of the flexibility applied to the unit plan were divided in integrated flexibility, functional flexibility, construction flexibility, and supply flexibility. The integrated flexibility enables residents to give the variable space combination based on the complex structure of the inner space for providing various living experiences. Regarding functional flexibility, the three-dimensional spatial structure with neutral space has multi-functionality according to the needs of residents and easily accepts mixing of hybrid programs such as work and residence. Constructive flexibility allows residents to create identity by freely planning interior space and changing the size or location of facade components in a determined system of architects. Finally, various types of size and space composition are proposed and realized in the whole building applying menu selection method, so that flexibility in the offer can accommodate and integrate various types of living.

In order to increase the electrochemical performance of thermal battery anode, LIFT anode having the same weight but a larger lithium content in electrodes was fabricated by mixing lithium, iron and titanium. By applying these electrodes, a single cell and a thermal battery were prepared, and the effect of LIFT anode on electrochemical performance was evaluated. The LIFT-applied single cell presented a better cell performance than LIFe-applied single cell at 500℃ and 550℃. The discharge performance of LIFT-applied single cell, which included the operating time (787s), specific capacity (1,683 Asg-1), and electrode utilization (80.7%), was improved collectively compared to the LIFe applied single cell (736s, 1,245 As g-1, and 74.6%) at 500℃. As the discharge progressed, the internal resistance of LIFT anode decreased, because the lithium migration path was formed due to the presence of large titanium particles among iron particles. These results were analyzed in terms of the microstructure of electrode using SEM. Energy density of LIFT-applied single cell also increased by 10% to 142.1 Wh kg-1 compared to that of LIFe-applied single cell (127.4 Wh kg-1). In addition, the LIFT-applied single cell presented a stable discharge performance for 6,500s without a short circuit which could occur by molten lithium under an open circuit voltage condition with a high pressure (4 kgf cm-2). As observed in the high temperature thermal battery performance tests, the voltage and specific capacity of LIFT-applied thermal battery are superior to those of LIFe-applied thermal batteries, indicating that the energy density of LIFT-applied thermal batteries should remarkably increase.

Objective : In this study, we investigated the effect of SAL5(mixing extracts of Schisandra chinensis Baillon, Artemisia capillaris Thunb., and Aloe vera Linne) on chronic ethanol-induced fatty liver model. Methods : Sprague-Dawley male rats were fed Liber-DeCarli (normal), ethanol liquid diet (control), SAL5 (200 mg/kg). We administrated the SAL5 on chronic ethanol-induced fatty liver model for 5 weeks. We measured alkaline phosphtase (ALP), alanine transminase (ALT), aspartate transminase (AST) and ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG), superoxide dismutase (SOD), catalase, glutathione (GSH) and malondialdehyde (MDA) level in liver. Liver histopathology was examined by Hematoxylin-eosin and Oil red O staining of the fixed liver tissues. Real-time PCR was performed to measure the mRNA expression of inflammatory cytokines and MMP-2, MMP-9. Results : SAL5 administration resulted in significantly decreased liver marker enzymes activities of alanine transminase (ALT), ${\gamma}-glutamyl$ transpeptase (${\gamma}-GTP$) in serum and triglyceride (TG) activities in liver. The control group decreased the activities of superoxide dismutase (SOD), catalase (CAT) with the reduced level of glutathione (GSH) in liver. On the other hand, SAL5 group increased the activities of SOD, CAT and the level of GSH. SAL5 delayed the development of an alcoholic fatty liver by reversing fat accumulation in the liver, as evidenced in histological observations. The gene expression of mRNA were significantly decreased at the $IL-1{\beta}$, $TNF-{\alpha}$, NOS-II and MMP-2 by SAL5. Conclusions : These results indicate that SAL5 might have protective effect chronic ethanol-induced fatty liver models.
Catalyst poisoning by ionomers in membrane electrode assemblies of alkaline membrane fuel cells has been reported recently. We tried to improve the membrane electrode assembly's performance by controlling the solvent's ratio during electrode manufacturing. 4 Different mixing ratios of N-Methyl-2-pyrrolidone (NMP) and ethylene glycol (EG) gave four different cathode electrodes with platinum and Fuma-Tech ionomers. The electrode with higher EG improved polarization performance by about 36% compared to the NMP-based commercial ionomer. The dependence of the ionomer's dispersibility on the solvent seems responsible for the difference, which means that the non-uniform distribution of ionomers improves the performance of the electrode. High-frequency resistance, internal resistance corrected polarization curve, Tafel slope, mass activity, and impedance spectroscopy characterized the electrode. We can find that the existence of poor solvent improves cathode electrode performance. It seems to be the result of reduced poisoning of the catalyst according to the particle size distribution of the ionomer.
From two viewpoints the writer investigated the beauty of space by overall arrangement and composition of a picture in Oriental painting. In particular, she examined the expanded representation and significance of space use which had not properly been recognized in the field of a colored picture contrary to a painting in India ink. She studied that the boundary for the representation and appreciation of space was unlimited to one field of painting by studying and analyzing it in connection with other fields of art which had something in common with it in techniques or languages of representation. The writer considered the aspects of similar forms and spirits as the methods of understanding and representing the essence of an object in creating a work. She generally considered the aspect of perfecting knowledge by studying the principle of an actual thing for the representation of revealed forms corresponding to the aspect of similar forms, and tried to reach the stage of 'materialization' united with the spirit of the subject of creation for the formless forms corresponding to the aspect of representing an artist's inner world as well as the external shapes of things. She tried to reach the stage of spiritual cultivation in pursuit of the boundary between 'mental vanity' and 'sitting quietly and attaining the state of perfect selflessness', which were presented by Chuang-tzu, to express the spirituality internal to it. She recognized that the projection of the cultivation on a work could convey internal essence as well as external forms to a picture. It was because the image of the form represented in a picture was based on the aesthetic experience got from realty. In the concept of space and a method of representing it, she explored and analyzed the basic concept of space, arranged the concept of space shown in Oriental ideas dividing it into the concepts of space in Confucianism, Taoism and the Zen sect. What she felt acutely through this study was that she should establish the identity of her work by succeeding to, changing and re-creating tradition based on the historical heritage left by successive excellent painters and theorists. Putting together all these things showed that establishing the identity in the world of work pursued and oriented by her required searching the direction in future works by mixing tradition with modern times in a creative way, which is just the purpose of study in this thesis.
The mixing of rice and brown rice produced in different years is banned in Korea by the grain management act. However, there has been no reported method for discriminating the production year of rice. The objective of this study was to develop a method for discriminating the production year of rice and brown rice based on their phospholipids content. One hundred rice samples and 130 brown rice samples produced between 2012 and 2015 were collected. Twelve phosphatidylcholine components were analyzed by liquid chromatography-tandem mass spectrometry. Phosphatidylcholine was used as an internal standard to calculate the peak intensity of the samples. A statistical analysis of the results showed that the centroid distance between the stale and new rice was 4.16 and the classification ratio was 97%. To verify the calculated discriminant, 61 and 40 rice samples were collected. The accuracy of discrimination was 82% by primary verification and 80% by secondary verification. The statistical analysis of brown rice showed that the centroid distance between the stale and new brown rice was 3.14 and the classification ratio was 96%. To verify the calculated discriminant, 10 samples of new rice and 30 samples of stale rice were collected and the accuracy of discrimination was 93%. The accuracy of discrimination for rice stored at room temperature was 57.9-92.1% and that for rice stored at a low temperature was 86.8-94.7%, depending on the storage period. For brown rice, the detection accuracy was 94.7-100% at room temperature and 92.1-100% at a low temperature, depending on the storage period. The accuracy of discrimination for rice was affected by the storage temperature and time, while that for brown rice was more than 92% regardless of the storage conditions. These results suggest that the developed discriminant analysis method could be utilized to determine the production year of rice and brown rice.
