VERTICAL OZONE DENSITY PROFILING BY UV RADIOMETER ONBOARD KSR-III

  • The UV radiometer payload was launched successfully from the west coastal area of Korea Peninsula aboard KSR-III on 28, Nov 2002. KSR-III was the Korean third generation sounding rocket and was developed as intermediate step to larger space launch vehicle with liquid propulsion engine system. UV radiometer onboard KSR-III consists of UV and visible band optical phototubes to measure the direct solar attenuation during rocket ascending phase. For UV detection, 4 channel of sensors were installed in electronics payload section and each channel has 255, 290, 310nm center wavelengths, respectively. 450nm channel was used as reference for correction of the rocket attitude during the flight. Transmission characteristics of all channels were calibrated precisely prior to the flight test at the Optical Lab. in KARI (Korea Aerospace Research Institute). During a total of 231s flight time, the onboard data telemetered to the ground station in real time. The ozone column density was calculated by this telemetry raw data. From the calculated column density, the vertical ozone profile over Korea Peninsula was obtained with sensor calibration data. Our results had reasonable agreements compared with various observations such as ground Umkhr measurement at Yonsei site, ozonesonde at Pohang site, and satellite measurements of HALOE and POAM. The sensitivity analysis of retrieval algorithm for parameters was performed and it was provided that significant error sources of the retrieval algorithm.

Somatic Hybrids by Electro-Protoplast Fusion between N. tabacum and N. glutinosa (담배(N. tabacum)와 N. glutinosa 종간 원형질체 융합식물의 생성)

  • Protoplasts, isolated from leaf of N. tabacum NR-/SR+ and N. glottnosa were electrofused and divided with a plating efficiency of 30∼35% in AAPI 9M medium. Green callus lines were selected in protoplast-derived colonies on MSNO3 selection medium with 1.2mg/ml streptomycin sulfate on the basis of nitrate reductase proficiency and streptomycin resistance. Four putative hybrid plant lines regenerated from the green callus lines had intermediate morphology between that of parents with respect to floral shape, corolla length and ovate leaf blade. Zymograms of leaf peroxidase and esterase from these putative hybrid plant lines showed isozyme profiles derived from both parents and also, they exhibited additional and lost bands. Cytological analysis of two putative hybrid plant lines gave chromosome counts of 2n=66 in L22 and 2n=54 in L44 which were less than the expected number of N. tabacum(2n=48) and N. glutinosa(2n=24).

Polyploidy and speciation in Korean endemic species of Indigofera grandiflora (Leguminosae) (한반도 고유종 큰꽃땅비싸리의 배수성과 종분화)

  • Chromosome number, morphological variation and RAPD analysis were investigated to study on the speciation of Indigofera in Korea. Chromosome numbers of I. kirilowii (2n=16) and I. koreana (2n=32) are consistent with the previous reports. In this study tetraploid (2n=32) and hexaploid (2n=48) of I. grandiflora are newly observed. Indigofera grandiflora is distributed around Mt. Kaya area together with I. kirilowii and I. koreana. The former species has the larger sizes in plant height, leaves and flowers than the latter two and shows intermediate form between the two species in hairs on leaves and flowers which are one of the most important taxonomic characters in this group. In the RAPD analysis, I. grandiflora is similar to I. koreana than I. kirilowii but RAPD band patterns revealed difference between tetra- and hexaploid of the species. These results suggested that Korean endemic species of I. grandiflora (2n=16, 32, 48) might has multiple origin through polyploidization and/or hybridization between I. kirilowii (2n=16) and I. koreana (2n=32) around Mt. Kaya area where the latter two grow together.

  • Morphometric, band-pattern and electrophoretic analysis on Cobitis taenia complex were performed to investigate the morphological and genetic differentiation and to clarify their taxonomic status. Intermediate types of band-pattern (C and D type) were more frequently expressed than that of types of C. t. taenia(type A) and C. t. lutheri (type B). Sexual dimorphism of band-pattern was observed not only in C. t. taenia(type A) and C. t. lutheri(type B). Sexual dimorphism of band-pattern was observed not only in C. t. lutheri but also in C. t. taenia and C. t. striata as well. Discriminant function analysis based on 19 morphological characters shows no significant differences among C. taenia complex. The degree of genic variation of C. t. striata was higher ( =1.48, P=31.2%, HD=0.009) than those of C. t. striata was higher( =1.48, P=31.2%, HD=0.082 and HG=0.009) than those of C. t. lutheri ( =1.37, P=2.7%, HD=0.058 and HG=0.065). The average genetic similarities between C. t. lutheri and C. t. taenia-C. t. striata were S=0.62 and S=0.66 respectively and these values indicate that C. t. tanenia has evolved specific level of differentiation. C. t. striata and C. t. lutheri show subspecifc level of close genetic similarity (S=0.82). Based on the divergent time estimate (Nei, 1975) it is assumed that C. t. tanenia was branched off from the other subspecies about two million years before present (MYBP) and C. t. striata and C. t. lutheri were differentiated about 0.6 MYBP. The use of C. sinesis an the scientific name for the Korean C. t. taenia, proposed by Kim and Lee (1988) seems incorrect since they are quite different in the structure of lamina circularis (Vladycov, 1935), the external morphology and distribution (Cheng and Zheng, 1987) and the chromosome number(Yu et al., 1989). Kim and Lee(1988) also argued that C.t. striata and C. t. lutheri should be treated as distinct species but the present study and other reports (Kim and Lee, 1984; Kim and Yang, 1993) do not support it. We conclude that C. t. taenia is a good species and C. t. striata and C. t. lutheri are subspecific status. Their scientific names should be revised in the future.

  • A low power single-chip CMOS receiver for 60 GHz mobile application are proposed in this paper. The single-chip receiver consists of a 4-stage current re-use LNA with under 4 dB NF, Cgs compensating resistive mixer with -9.4 dB conversion gain, Ka-band low phase noise VCO with -113 dBc/Hz phase noise at 1 MHz offset from 26.89 GHz, high-suppression frequency doubler with -0.45 dB conversion gain, and 2-stage current re-use drive amplifier. The size of the fabricated receiver using a standard 0.13 ${\mu}m$ CMOS technology is 2.67 mm$\times$0.75 mm including probing pads. An RF bandwidth is 6.2 GHz, from 55 to 61.2 GHz and an LO tuning range is 7.14 GHz, from 48.45 GHz to 55.59 GHz. The If bandwidth is 5.25 GHz(4.75~10 GHz) The conversion gain and input P1 dB are -9.5 dB and -12.5 dBm, respectively, at RF frequency of 59 GHz. The proposed single-chip receiver describes very good noise performances and linearity with very low DC power consumption of only 21.9 mW.

Comparison of Cognitive Loads between Koreans and Foreigners in the Reading Process

  • Objective: This study aims to measure cognitive load levels by analyzing the EEG of Koreans and foreigners, when they read a Korean text with care selected by level from the grammar and vocabulary aspects, and compare the cognitive load levels through quantitative values. The study results can be utilized as basic data for more scientific approach, when Korean texts or books are developed, and an evaluation method is built, when the foreigners encounter them for learning or an assignment. Background: Based on 2014, the number of the foreign students studying in Korea was 84,801, and they increase annually. Most of them are from Asian region, and they come to Korea to enter a university or a graduate school in Korea. Because those foreign students aim to learn within Universities in Korea, they receive Korean education from their preparation for study in Korea. To enter a university in Korea, they must acquire grade 4 or higher level in the Test of Proficiency in Korean (TOPIK), or they need to complete a certain educational program at each university's affiliated language institution. In such a program, the learners of the Korean language receive Korean education based on texts, except speaking domain, and the comprehension of texts can determine their academic achievements in studying after they enter their desired schools (Jeon, 2004). However, many foreigners, who finish a language course for the short-term, and need to start university study, cannot properly catch up with university classes requiring expertise with the vocabulary and grammar levels learned during the language course. Therefore, reading education, centered on a strategy to understand university textbooks regarded as top level reading texts to the foreigners, is necessary (Kim and Shin, 2015). This study carried out an experiment from a perspective that quantitative data on the readers of the main player of reading education and teaching materials need to be secured to back up the need for reading education for university study learners, and scientifically approach educational design. Namely, this study grasped the difficulty level of reading through the measurement of cognitive loads indicated in the reading activity of each text by dividing the difficulty of a teaching material (book) into eight levels, and the main player of reading into Koreans and foreigners. Method: To identify cognitive loads indicated upon reading Korean texts with care by Koreans and foreigners, this study recruited 16 participants (eight Koreans and eight foreigners). The foreigners were limited to the language course students studying the intermediate level Korean course at university-affiliated language institutions within Seoul Metropolitan Area. To identify cognitive load, as they read a text by level selected from the Korean books (difficulty: eight levels) published by King Sejong Institute (Sejonghakdang.org), the EEG sensor was attached to the frontal love (Fz) and occipital lobe (Oz). After the experiment, this study carried out a questionnaire survey to measure subjective evaluation, and identified the comprehension and difficulty on grammar and words. To find out the effects on schema that may affect text comprehension, this study controlled the Korean texts, and measured EEG and subjective satisfaction. Results: To identify brain's cognitive load, beta band was extracted. As a result, interactions (Fz: p =0.48; Oz: p =0.00) were revealed according to Koreans and foreigners, and difficulty of the text. The cognitive loads of Koreans, the readers whose mother tongue is Korean, were lower in reading Korean texts than those of the foreigners, and the foreigners' cognitive loads became higher gradually according to the difficulty of the texts. From the text four, which is intermediate level in difficulty, remarkable differences started to appear in comparison of the Koreans and foreigners in the beginner's level text. In the subjective evaluation, interactions were revealed according to the Koreans and foreigners and text difficulty (p =0.00), and satisfaction was lower, as the difficulty of the text became higher. Conclusion: When there was background knowledge in reading, namely schema was formed, the comprehension and satisfaction of the texts were higher, although higher levels of vocabulary and grammar were included in the texts than those of the readers. In the case of a text in which the difficulty of grammar was felt high in the subjective evaluation, foreigners' cognitive loads were also high, which shows the result of the loads' going up higher in proportion to the increase of difficulty. This means that the grammar factor functions as a stress factor to the foreigners' reading comprehension. Application: This study quantitatively evaluated the cognitive loads of Koreans and foreigners through EEG, based on readers and the text difficulty, when they read Korean texts. The results of this study can be used for making Korean teaching materials or Korean education content and topic selection for foreigners. If research scope is expanded to reading process using an eye-tracker, the reading education program and evaluation method for foreigners can be developed on the basis of quantitative values.

The Effect of Root Exudate and Chemotaxis on Host Recognition in Soybean-Bradyrhizobium Symbiosis (대두(大豆)-근류균(根瘤菌) 공생(共生)에서 뿌리분비물(分泌物)과 화학주성(化學走性)이 숙주인식(宿主認識)에 미치는 영향(影響))

  • This study was carried out to research the effect of the chemotaxis of Bradyrhizobium japonicum KCTC 2422 and its mutant toward soybean root exudate and to elucidate the effect of the lectin of host specificity (Host Recognition) in soybean-Bradyrhizobium symbiosis. The results obtained were as follows: The homogeneities of the purified lectins from soybean and pea seed was ascertained chromatographically and electrophoretically. Gel electrophoresis of soybean lectin in the presence of sodium dodecyl sulfate appeared a single protein band, whereas pea lectin appeared two protein bands. Soybean lectin from 2 cultivars formed immunoprecipitin arcs at same position with anti-soybean lectin rabbit IgG, but pea lectin did not form immunoprecipitin lines with anti-soybean lectin rabbit IgG. Chemotactic responses of KCTC 2422, LPN-100 and LCR-101 toward proline in capillary assays were 3.1, 1.3 and 1.0-fold above background, respectively. The chemotactic responses of KCTC 2422, LPN-100, and LCR-101 toward Paldal crude root exudate in capillary assays were 3.5, 1.4 and 1.4-fold above background, respectively. The present work shows that B. japonicum and its mutants are capable of very different responses toward root exudate fraction. The chemotactic responses of KCTC 2422 was most with neutral fraction, least with anionic fraction and intermediate with cationic fraction. The nitrogenase activity of soybean nodule was shown in 15days after inoculation with LCR-101. However, we couldn't find out the nodules when soybean was inoculated with LPN-100. From these result we can suppose that the chemotaxis of Bradyrhizobium plays inportant the role of forming the nodule (host recognition) in the soybean-B. japonicum symbiosis.

Studies on characteristics and pathogenicity of Yersinia sp. Isolated from cultured olive flounder, Paralichthys olivaceus (양식넙치에서 분리한 Yersinia sp.의 일부 특성과 병원성에 관한 연구)

  • Characteristics and pathogenicity of Yersinia sp. strain isolated from diseased cultured olive flounder, Paralichthys olivaceous were studied. The causative organisms were identified as Yersinia sp. by biochemical and biophysical characteristics. The strain was named KF-1, and it showed the optimal growth rate at pH 9.0 and 1% NaCl. Total cell protein peptide bands of the Yersinia sp. KF-1 were between 14.4-100.6 Kd in molecular weight by the electrophoretic analysis, and a total of 28 bands appeared. The band found at 32.8 Kd in molecular weight was the major one of electrophoretic phase. In the pathogenicity test of the isolate to the flounder injecting with $1.0{\times}10^7$ cfu/fish 9 died out of 10 within 60 hrs, and in the group with $1.0{\times}10^7$ cfu/fish 5 died within 60 hrs. Thus the $LD_{50}$ was presumed to be $1.0{\times}10^7$ cfu/fish. In the drug sensitivity test KF-1 strain was sensitive to chloramphenicol, gentamycin, kanamycin, streptomycin and pefloxacin, and intermediate to erythromycin, and resistant to ampicillin, cephalothin, sulfamethoxazole/trimethoprim, tetracycline and vancomycin.

  • Weathering of biotite shows a biotite-vermiculite-kaolinite sequence at the early stage, but presents biotite-kaolinite sequence without a significant intermediate phase (vermiculite) at the late stage from the weathering profile of the granitic gneiss. Secondary 1:1 phyllosilicates are kaolinite and halloysite which show different weathering textures originated by a different formation mechanism. Kaolinitization began from the edges of biotite and propagated toward the interior of grain along a multilayered front. $10 \AA$ layers of biotite are interleaving with $7\AA$ layers of kaolinite and c-axis of two phases is consistent. Kaolinite pseudomorph of biotite is isovolumetric, compared to the biotite boundary and includes many band-like porosities parallel to the cleavage. Platy kaolinite formed by 1:1 layer fur layer replacement of biotite. Halloysitization proceeded outward from the grain edges which were foliated as fine flakes and bent at the right angle for cleavage Halloysites were extensively fanning out and greatly increased the volume of grain. This indicated that halloysite tubes were formed by epitaxial overgrowth on the surface of biotite with import of Si and Al from the external solution by dissolution of plagioclase. These halloysites have abnormally high Fe content ( ~11%).

Effects of Ethephon and Gibberellin on Sex Expression, and Subsequent Changes in Protein Contents, Peroxidase Activities, and Isoperoxidase Isoperoxidase Patterns of Cucumis sativus L.   • This experiment was conducted to study sex expression and subsequent changes in protein and peroxidase after three cultivars of cucumber plants were treated with ethephon and gibberellin. The three cultivars of cucumber used in this study included 'Sayeup' (monoecious type), 'Sinrokdadaki' (gynoecious type), and 'Seonghowon' (intermediate type). The ethephon at 250 ppm and gibberellin at 100 ppm were treated at the 2-leaf and 4-leaf stages, and subsequent sex expression and changes in protein contents, peroxidase activities and isoperoxidase banding patterns by disc polyacrylamide gel electrophoresis were studied. The results can be summarized as follows: 1. Ethephon treatment slightly increased number of pistillate flowers and significantly decreased number of staminate flowers in the three cultivars, while gibberellin treatment significantly increased number of staminate flowers in both gynoecious 'Sinrokdadaki' and intermediate 'Seonghowon' and did not increase number of staminate flowers in monecious 'Sayeup' 2. There were some differences among three cultivars in protein contents, protein banding and isoperoxidase banding patterns of seeds and germinating seeds. However, it was not obvious to differentiate monoecious from gynoecious cultlvars by these characters. 3. Protein contents in the leaves. and stem apex after ethephon and gibberellin treatment increased gradually at the 2-leaf stage, but decreased at the 4-leaf stage. Protein contents in stem apex at the 4-leaf stage without treatment were much higher in 'Sinrokdadaki' and 'Seonghowon' than in monoecious 'Sayeup'. Protein contents in the stern apex at the 4-leaf stage were increased in the ethephon-treated monoecious 'Sayeup' and decreased in 'Sinrokdadaki' and 'Seonghowon' compared with untreated plants. 4. Peroxidase activities in the leaves and stem apex gradually decreased at the 2- leaf stage, but increased at the 4-leaf stage. Peroxidase activities in stern apex at the 4-leaf stage were significantly increased by ethephon treatment. 5. The number of protein bands in the three cuitivars after treatment gradually decreased in leaves and stem apex at the 2-leaf stage, but increased in the 4-leaf stage. The protein banding pat terns of stern apex of the ethephon-treated monoecious 'Sayeup' at the 4-leaf stage were gradually shifted to the banding.
