Influences of Sulfur Dioxide on the Growth of Ornamental Trees (아황산가스가 조경수목의 생육에 미치는 영향)

  • This study was to investigate ornamental trees injuries at the Tobong ornamental tree farm, which is located in Uichongpu City, Kyonggido. The injury was caused by $SO_2$ gas from the smoke of the vicinal factories. The results obtained are as follows: 1 The total concentration of sulfur in leaves was increased with incerased degree of injury The most severe injuries were found at the distance between 70-120m from the western boundary of the Tobong ornamental 4ree farm with a highest concentration of total sulfur at $0.5-0.6\%$. This means the injuries caused by the acute injury and the temperature changes. 2. Significant differences were found between normal and injured portion of the same trees in Juniperus chinensis. 3. Anatomical study of the leaf tissues, showed the plasmolysis occured both in spongy and epidermal cells, leading to shrinkage and destruction. An intercellular space and stomatal periphery became wider.

Effect of Diclazuril on the Bursa of Fabricius Morphology and SIgA Expression in Chickens Infected with Eimeria tenella

  • The effects of diclazuril on the bursa of Fabricius (BF) structure and secretory IgA (SIgA) expression in chickens infected with Eimeria tenella were examined. The morphology of the BF was observed by hematoxylin and eosin staining, while ultrastructural changes were monitored by transmission electron microscopy. E. tenella infection caused the BF cell volumes to decrease, irregularly arranged, as well as, enlargement of the intercellular space. Diclazuril treatment alleviated the physical signs of damages associated with E. tenella infection. The SIgA expression in BF was analyzed by immunohistochemistry technique. The SIgA expression increased significantly by 350.4% (P<0.01) after E. tenella infection compared to the normal control group. With the treatment of diclazuril, the SIgA was relatively fewer in the cortex, and the expression level was significantly decreased by 46.7% (P<0.01) compared with the infected and untreated group. In conclusion, E. tenella infection in chickens induced obvious harmful changes in BF morphological structure and stimulated the expression of SIgA in the BF. Diclazuril treatment effectively alleviated the morphological changes. This result demonstrates a method to develop an immunological strategy in coccidiosis control.

An Acidic PATHOGENESIS-RELATED1 Gene of Oryza grandiglumis is Involved in Disease Resistance Response Against Bacterial Infection

  • Wild rice, Oryza grandiglumis shows hyper-resistance response to pathogen infection. In order to identify genes necessary for defense response in plants, we have carried out a subtractive hybridization coupled with a cDNA macroarray. An acidic PATHOGENESIS-RELATED1 (PR1) gene of the wild rice is highly identical to the acidic PR1 genes of different plant species. The OgPR1a cDNA has an apparent single open reading frame with a predicted molecular mass 40,621 Da and an isoelectic point of 5.14. Both in silico analysis and a transient expression assay in onion epidermal cells revealed that the OgPR1a protein could be localized in intercellular space in plants. The OgPR1a mRNA was strongly transcribed by the exogenous treatment with ethylene and jasmonic acid as well as protein phosphatase inhibitors. Additionally, ectopic expression of the OgPR1a conferred disease resistance on Arabidopsis to the bacterial and fungal infections.

Systemic Induction of the Small Antibacterial Compound in the Leaf Exudate During Benzothiadiazole-elicited Systemic Acquired Resistance in Pepper

  • Plants protect themselves from diverse potential pathogens by induction of the immune systems such as systemic acquired resistance (SAR). Most bacterial plant pathogens thrive in the intercellular space (apoplast) of plant tissues and cause symptoms. The apoplastic leaf exudate (LE) is believed to contain nutrients to provide food resource for phytopathogenic bacteria to survive and to bring harmful phytocompounds to protect plants against bacterial pathogens. In this study, we employed the pepper-Xanthomonas axonopodis system to assess whether apoplastic fluid from LE in pepper affects the fitness of X. axonopodis during the induction of SAR. The LE was extracted from pepper leaves 7 days after soil drench-application of a chemical trigger, benzothiadiazole (BTH). Elicitation of plant immunity was confirmed by significant up-regulation of four genes, CaPR1, CaPR4, CaPR9, and CaCHI2, by BTH treatment. Bacterial fitness was evaluated by measuring growth rate during cultivation with LE from BTH- or water-treated leaves. LE from BTH-treatment significantly inhibited bacterial growth when compared to that from the water-treated control. The antibacterial activity of LE from BTH-treated samples was not affected by heating at $100^{\circ}C$ for 30 min. Although the antibacterial molecules were not precisely identified, the data suggest that small (less than 5 kDa), heat-stable compound(s) that are present in BTH-induced LE directly attenuate bacterial growth during the elicitation of plant immunity.

  • The ultrastructure and silk formational process of the flagelliform glands, one of the capture thread producing organs in the orb web spider (Nephila clavata L. Koch), have been studied with electron microscope. A pair of flagelliform glands consist of three distinct regions: excretory duct, storage sac and convoluted tail. This gland is connected to the large spinning tube (spigot) of the posterior spinnerets with two pairs of aggregate glands, and showed a characteristic spigot structure called "triad". The excretory duct consists of three long segments surrounded by the same sheath of connective tissue. The distal duct near the spinning tube bears the electron lucent subcuticles which have the functions of water removal and orientation of the silk fiber, whereas the proximal duct near the sac contains the cuticule precursor transported through the intercellular space. Cytoplasms of the sac and tail regions have well developed rough endoplasmic reticulum and contain the round secretory granules commonly, but the electron densities and internal textures of the granules do not coincided. Adjacent epithelial cells are connected with the specialized septate junctions, and secretory granules accumulated at the apical pole are released to the lumen by the apocrine secretion.secretion.

Effect of simulated Acid rain on Foliar Structural of Changes of Ginkgo biloba and Pinus thunbergii (은행나무와 곰솔에 처리된 인공산성비에 의한 잎의 형태변화)

  • Visible injury symptoms such as necrosis, chlorosis and premature senscence in the leaves of Ginkgo bloba and Pinus thunbergii treated with acid rain of pH 3.2 or below were observed. The epicuticular wax erosions were observed by SEM after exposure to acid rain of pH 2.4 and 3.2 in G. biloba and pH 4.0 below in P. thunbergii. The adaxial epidennal cells and sponge parenchyma cells were compressed, and those were distorted in the leaves of G. biloba treated with simulated acid rain of pH 3.2 or below. However, vascular tissue was intact. With increase of acidity, mesophyll cells were smaller than those of control while intercellular space in mesophyU was increased. In P. thunbergii, sponge parenchyma cells and vascular tissue except epidennis were distorted after exposure to acid rain of pH 2.4. The size change of stomata in foliar injury was not observed, but the stomatal index and size of stomatal aperture in leaves treated with acid rain increased. The stomata of injured leaf were opened in both species examined.amined.

AN ELECTRON MICROSCOPIC STUDIES OF RADIATION EFFECTS ON THE ENAMEL DEVELOPMENT OF THE RAT MOLAR (방사선조사가 백서구치법랑질발육에 미치는 영향에 관한 전자현미경적 연구)

  • Mandibular first molars of the rats were undertaken to observe the radiosensitivity of amelogenesis. Twenty four Sprague-Dawley rats received 396cGy radiation with the MK Cell irradiator using Cs-137, and twenty four rats served as control. They were devided into two groups; Group 1 which received radiation at the 14th day after gestation and group 2 which received radiation at the 19th day after gestation. Light Microscopy and Transmitted Electron Microscopy investigation was undertaken in the group 1 at the 15th, 18th, 20th, 22nd(2 days after birth), and 25th day(5 days after birth) after gestation, and in the group 2 at the 21th(birth day), 22nd(2 days after birth), and 25th(5 days after birth) day after gestation. The following histopathologic findings were obtained. 1. Compared with control group, experimental group showed a delayed formation of enamel and dentin due to slow rate of differentiation of undifferentiated mesenchymal cells. 2. In the experimental groups, the arrangement of the inner enamel epithelium was irregular and there were many vacuoles in the cytoplasm. There were dilated rER and mitochondria, increase of the intercellular space, and loss of the cellular polarity. 3. In the group 1, early enamel without Tomes' process, and early organic matrix was observed at the 25th day after gestation. 4. In the group 2, histopathologic changes were similar to those of the group 1, but the degree of changes was more severe than that of the group 1.

Co-administration of alcohol and combination antiretroviral therapy (cART) in male Sprague Dawley rats: a study on testicular morphology, oxidative and cytokines perturbations

  • Alcohol consumption alongside combination antiretroviral therapy (cART) has attracted research interest, especially because of increasing male infertility. This study investigated the combined effects of alcohol and cART on testicular morphology, biomarkers of oxidative stress, inflammation, and apoptosis. Rats, weighing 330-370 g, were divided into four groups of six animals each; control, alcohol treated (A), cART, and alcohol plus cART treated (A+cART). Following 90 days treatment period, animals were euthanized, testis extracted, and routinely processed for histology and immunohistochemical analysis. Significantly decreased epithelial area fraction, increased luminal and connective tissue area fractions, and reduction of epithelial height and spermatocyte number, were recorded in the treated groups compared to control. Extensive seminiferous epithelial lesions including widened intercellular space, karyolysis, and sloughing of germinal epithelium were recorded in all the treated groups. Furthermore, upregulation of inducible nitric oxide synthase and 8-hydroxydeoxyguanosine, interleukin-6, and caspase 3 recorded in treated animals, was more significant in A+cART group. Also, the levels of interleukin-1β and tumor necrosis factor-α were more elevated in A and cART treated groups than in A+cART, while MDA was significantly elevated in cART and A+cART treated groups compared to control group. Altogether, the results indicate testicular toxicity of the treatments. It is concluded that consuming alcohol or cART induces oxidative stress, inflammation, and apoptosis in testis of rats, which lead to testicular structural and functional derangements, which are exacerbated when alcohol and cART are consumed concurrently. The result will invaluably assist clinicians in management of reproductive dysfunctions in male HIV/AIDS-alcoholic patients on cART.

Morphological and Physiological Characteristics of Acclimated Liriodendron tulipifera Tree Produced by Several Types of In Vitro Germination Culture (백합나무 체세포배 기내발아 방식에 따른 순화묘의 형태적 및 생리적 특성)

  • The acclimatization of in vitro propagated plants is an important step to produce vigorous plants for clonal forestry and in vitro micro-environment may affect the growth in ex vitro condition. To monitor in vitro environmental effects on the growth in ex vitro condition, several culture systems such semi-solid medium(SS), temporary immersion bioreactor(TIB) and continuous immersion bioreactor(CIB) culture types were tested to compare for the growth of acclimated plants of Liriodendron tulipifera. Results suggested that morphological characters, stomatal conductance, evapotranspiration and chlorophyll contents of acclimated plants were affected by the different of in vitro culture conditions. CIB type of culture was resulted to the lowest value in the biomass of acclimated plants. Net photosynthsis rate of CIB was the same level as those of SS and TIB. However, stomatal conductance, evapotranspiration and $CO_2$ partial pressure in the intercellular air space were lower than those of SS and TIB. The amounts of chlorophyll a, b and carotenoids were also lower than those of the other two culture systems. TIB, showing a little lower or higher value than SS in many growth character, is recommended rather than CIB to produce healthy yellow poplar plants in ex situ condition.

  • In Korea, a study on the anatomical features of pitch pine (pinus rigida Miller) branch wood through photo-microscopical method was reported in 1972 by Lee. Therefore, as a further study of Lee's on the anatomical features in branch wood of pinus rigida miller that grows in Korea, compression wood, opposite wood, and side wood were selected and treated for the purpose of comparing their structures revealed on cross and radial surface through scanning electron microscope in this study. The obtained results in this study were summarized as follows; 1. The trachied transition from earlywood to late wood is very gradual and the tracheids are nearly regular in both arrangement and size in compression wood but this transition in opposite wood and side wood is abrupt and the tracheids in opposite wood and side wood are less regular than those in compression wood. Also, the annual ring width of opposite wood is narrower than that of compression wood or side wood and the rays revealed on cross surface of side wood are more distinct than compression wood and opposite wood rays. 2. The tracheids of compression wood show roundish trends especially in earlywood but those of opposite wood and side wood show some angular trends. And intercellular space, helical cavity, and spiral check are present in both earlywood and latewood of compression wood but not present in opposite wood and side wood irrespective of earlywood and latewood. 3. The wall thickness of latewood tracheid is similar to that of earlywood tracheid in compression wood whereas the wall thickness of latewood tracheid is by far thicker than that of earlywood tracheid in opposite wood and side wood and the S3 layer of secondary wall is lack in compression wood tracheid unlike opposite wood and side wood tracheid. 4. The tracheids in compression wood are often distorted at their tips unlike those in opposite wood and side wood and the bordered pit in compression wood tracheid is located at the bottom of helical groove unlike that in opposite wood and side wood tracheid. 5. The bordered pits in radial wall of opposite wood and side wood tracheids are oval in shape but those of compression wood tracheids show some modified oval shape. 6. In earlywood of side wood, the small apertures of cross-field pits are roundish triangle to rectangle and the large one are fenestriform through the coalition of two small ones. However, the small apertures of cross-field pits are upright oval and the large ones are procumbent oval shape in earlywood of opposite wood and the apertures of cross-field pits in compression wood are tilted bifacial convex lens shape in earlywood and slit in late wood because of the border on tracheid side.
