A Routine System for Generation of Fertile Transgenic Rice Plants Using Biolistic Method

  • A routine system based on particle bombardment of embryogenic callus for recovery of fertile transgenic rice (Oryza sativa L.) plants was developed. Embryogenic callus was established within 2-3 months from calli derived from mature seeds of Korean rice cultivar, Nagdongbyeo. The callus was bombarded with the plasmid pRQ6 containing the $\beta$-glucuronidase gene (gusA) and hygromycin phosphotransferase gene (hph, conferring resistance to hygromycin B), both driven by CaMV 35S promoter. Placement of cells on an osmoticum-containing medium (0.2 M sorbitol and 0.2 M mannitol) 4 hrs prior to and 16 hrs after bombardment resulted in a statistically significant increase with 3.2-fold in transient expression frequency gusA. In five independent experiments, the average frequency of transformation showing GUS activities was $8.86\%$. A large number of morphologically normal, fertile transgenic rice plants were obtained. Integration of foreign gene into the genome of $R_0$ transgenic plants was confirmed by Southern blot analysis. GUS and HPT were detected in $R_1$ progeny and Mendelian segregation of these genes was observed in $R_1$ progeny.

  • In the near future, the voltage of power system for passenger vehicle will be changed to 42V from existing 14V./ Because of increasing power and voltage ratings used in the vehicle the motor drive system has high switching dv/dt and it generates electromagnetic interference (EMI) To solve these problems multi-level inverter system may be used The feature of multi-level inverter is the output voltage to be synthesized from several levels of voltage Because of this feature high switching dv/dt and EMI can be reduced in the multi-level inverter system But as the number of level is increased manufacturing cost is getting expensive and system size is getting large. Because of these disadvantages the application of multi-level inverter has been restricted only to high power drives. The method to reduce manufacturing cost and system size is to integrate circuit of multi-level inverter into a few chips But isolated power supply and signal isolation circuit using transformer or opto-coupler for drive circuit are obstacles to implement the integrated circuit (IC) In this paper a drive circuit of 4-level inverter suitable for integration to hybrid or one chip is proposed In the proposed drive circuit DC link voltage is used directly as the power source of each gate drive circuit NPN transistors and PNP transistors are used to isolate to transfer the control signals. So the proposed drive circuit needs no transformers and opto-couplers for electrical isolation of drive circuit and is constructed only using components to be implemented on a silicon wafer With th e proposed drive circuit 4- level inverter system will be possible to be implemented through integrated circuit technology Using the proposed drive circuit 4- level inverter system is constructed and the validity and characteristics of the proposed drive circuit are proved through the experiments.

Sparkover Voltage Estimation of Standard Sphere Gaps for Negative Polarity by Calculation of Ionization Index

  • The field utilization factor (equation omitted) (the mean electric field / the maximum electric field) of standard sphere gaps was calculated by the charge simulation method, taking into account the ground plane and shanks. n changes mainly with g/r and slightly with 1$_1$, 1$_2$ and 1, where D=2r is the sphere diameter, g is the gap length, 1$_1$ and 1$_2$, respectively, are the lengths of the upper and lower shank, and t is the shank diameter. Generally, (equation omitted) increases as 1$_1$,1$_2$ and t each becomes larger. IEC standard 60052(2002) limits t$\leq$0.2D 1$_1$$\geq$1D and prescribes A=1$_2$+D+g where A is the height of the spark point on the upper sphere. Therefore, (equation omitted) is the largest when A=9D and the smallest when A=3D. The simple equation of a straight line, (equation omitted)=1- (g/3r), can generally be used as a representative value of (equation omitted) for a wide variety of sphere diameters that are permitted by the IEC standard. The maximum electric field E$_{m}$ at sparkover of standard air gaps has also been calculated by the relation E$_{m}$=V/(equation omitted)g). E$_{m}$ describes a U-curve for g/r, up to the sphere diameter of 1 m. Moreover, for 1.5-m and 2-m diameters and especially .for negative polarity, sparkover voltages have been calculated by integration of the ionization index.index.

  • As the need for stable and high speed wireless Internet service Brows, the wireless LAN service provider hurries to preempt wireless LAN service market. IAPP(InterAccess Point protocol) is defined to be able to provide a secure handoff mechanism of wireless LAN terminal information between AP(Access Point)s, and the related IEEE standard is IEEE 802.11f. For the secure handoff of wireless LAN terminal, it is necessary to transfer terminal's authentication & accounting information securely from old AP to new AP IEEE 802.11f recommends RADIUS server as IAPP server which authenticates AP and provides information for secure channel between APs. This paper proposes IAPP server using Diameter protocol to overcome the limit of RADIUS sewer, and describes about the interaction between server components and integration method with the current IAPP client system.

  • The rigorous validation of the accuracy of metamodels is an important topic in research on metamodel techniques. Although a leave-k-out cross-validation technique involves a considerably high computational cost, it cannot be used to measure the fidelity of metamodels. Recently, the mean$_0$ validation technique has been proposed to quantitatively determine the accuracy of metamodels. However, the use of mean$_0$ validation criterion may lead to premature termination of a sampling process even if the kriging model is inaccurate. In this study, we propose a new validation technique based on the mean and variance of the response evaluated when sequential sampling method, such as maximum entropy sampling, is used. The proposed validation technique is more efficient and accurate than the leave-k-out cross-validation technique, because instead of performing numerical integration, the kriging model is explicitly integrated to accurately evaluate the mean and variance of the response evaluated. The error in the proposed validation technique resembles a root mean squared error, thus it can be used to determine a stop criterion for sequential sampling of metamodels.

Grandmothers' Experience of Child Care with Grandchildren: A Grounded Theory Approach (손자녀를 돌보는 외할머니의 양육경험: 근거이론적 접근)

  • Purpose: This study examined grandmothers' experience of child care with grandchildren to understand how it affects their stage of life based on the methodology of grounded theory. Methods: This study used grounded theory, a method of qualitative study, as the theoretical foundation. The subjects of this study were 10 grandmothers who have reared their grandchildren for at least 6 months and volunteered to participate in the study after listening to an explanation. Resources were collected through in-depth interviews and the interviews were recorded and dictated. Results: The core category of participants in this study was "solidifying the family relationship". There were five steps in the process of "solidifying the family relationship". The first step of process was "affection motion" of rearing to help their daughters. The second step was "conflict" due to physical constraint and burden of child care. The third step was "acceptance" based on compensation and support and the fourth step was "development" in search of their ego. The last step was "integration" of family relationship. Conclusion: This study provided basic data for appropriate nursing intervention to grandmother to care for their grandchildren according to the five steps of grandmother's child care experience process.

Development of a simulation method for the subsea production system

  • The failure of a subsea production plant could induce fatal hazards and enormous loss to human lives, environments, and properties. Thus, for securing integrated design safety, core source technologies include subsea system integration that has high safety and reliability and a technique for the subsea flow assurance of subsea production plant and subsea pipeline network fluids. The evaluation of subsea flow assurance needs to be performed considering the performance of a subsea production plant, reservoir production characteristics, and the flow characteristics of multiphase fluids. A subsea production plant is installed in the deep sea, and thus is exposed to a high-pressure/ low-temperature environment. Accordingly, hydrates could be formed inside a subsea production plant or within a subsea pipeline network. These hydrates could induce serious damages by blocking the flow of subsea fluids. In this study, a simulation technology, which can visualize the system configuration of subsea production processes and can simulate stable flow of fluids, was introduced. Most existing subsea simulations have performed the analysis of dynamic behaviors for the installation of subsea facilities or the flow analysis of multiphase flow within pipes. The above studies occupy extensive research areas of the subsea field. In this study, with the goal of simulating the configuration of an entire deep sea production system compared to existing studies, a DES-based simulation technology, which can logically simulate oil production processes in the deep sea, was analyzed, and an implementation example of a simplified case was introduced.

Stability of the Divergent Barotropic Rossby-Haurwitz Wave (발산 순압 로스비-하우어비츠 파동의 안정성)

  • Stability of the barotropic Rossby-Haurwitz wave is investigated using the numerical models on the global domain. The Rossby-Haurwitz wave under investigation is composed of the basic zonal flow of super-rotation and a finite amplitude spherical harmonic wave. The Rossby-Haurwitz wave is given as either steady or unsteady wave by adjusting the strength of the super-rotating zonal flow. Stability as well as the growth rate of the wave in the numerical simulation is determined by comparing the perturbation amplitude at two different time stages. Unstable modes of the Rossby-Haurwitz wave exhibited a horizontal structure composing of various zonal-wavenumber components. The vorticity perturbation for some modes showed a discontinuity around the area of weak flow, which was found robust regardless of the horizontal resolution of the model. Fourier finite element model was shown to generate the unstable mode in earlier stage of the time integration due to less accuracy compared to the spherical harmonic spectral model. Taking the overall accuracy of the models into consideration, the time by which the unstable mode begin to dominate over the spherical harmonic wave was estimated.

Tethers tension force effect in the response of a squared tension leg platform subjected to ocean waves

  • The tension leg platform (TLP) is one of the compliant structures which are generally used for deep water oil exploration. With respect to the horizontal degrees of freedom, it behaves like a floating structure moored by vertical tethers which are pretension due to the excess buoyancy of the platform, whereas with respect to the vertical degrees of freedom, it is stiff and resembles a fixed structure and is not allowed to float freely. In the current study, a numerical study for square TLP using modified Morison equation was carried out in the time domain with water particle kinematics using Airy's linear wave theory to investigate the effect of changing the tether tension force on the stiffness matrix of TLP's, the dynamic behavior of TLP's; and on the fatigue stresses in the cables. The effect was investigated for different parameters of the hydrodynamic forces such as wave periods, and wave heights. The numerical study takes into consideration the effect of coupling between various degrees of freedom. The stiffness of the TLP was derived from a combination of hydrostatic restoring forces and restoring forces due to cables. Nonlinear equation was solved using Newmark's beta integration method. Only uni-directional waves in the surge direction was considered in the analysis. It was found that for short wave periods (i.e., 10 sec.), the surge response consisted of small amplitude oscillations about a displaced position that is significantly dependent on tether tension force, wave height; whereas for longer wave periods, the surge response showed high amplitude oscillations that is significantly dependent on wave height, and that special attention should be given to tethers fatigue because of their high tensile static and dynamic stress.

The Establishment of BPR for National Spatial Data Infrastructure Quality Management System (국가공간정보통합체계 품질관리시스템 구축을 위한 BPR 수립)

  • In Korea, National spatial data infrastructure has implemented in order to integrated manage and share the national spatial information producted by public agencies and local governments. The necessities of systematic quality management are raised, because information, which is generated by different agencies, is integrative managed by national level. In this paper, the establishment of BPR(Business Process Reengineering) for national spatial data infrastructure quality management system. Quality management business is defined as quality management object definition, quality measuring, evaluation and analysis, and quality enhancement process. Next, activities for each process are designed. For the quality management business, business rule(BR) is required for determining error. We derive the BR for six objects(legal-dong, railway boundary, railway centerline, road boundary, road centerline, building) among the basic spatial information. Other information's BR can be generated by using the derivation method described in this paper. Based on the BPR of this paper and derived BR, national spatial data infrastructure quality management system can be implemented in the future.
