Production and Secretion of Human Interleukin-18 in Transgenic Tobacco Cell Suspension Culture

  • Interleukin-18 (IL-18), otherwise known as interferon-gamma-inducing factor (IGIF), is one of several well characterized and important cytokines that contribute to host defenses. The complementary DNA (cDNA) of mature human interleukin-18 gene (hIL-18) was fused with the signal peptide of the rice amylase 1A gene (Ramy1A) and introduced into the plant expression vector under the control of a duplicated CaMV 35S promoter. The recombinant plasmid was transformed into tobacco (Nicotiana tabacum L. cv Havana) using the Agrobacterium-mediated transformation method. The integration of the hlL-18 gene into the genome of transgenic tobacco plants was confirmed by polymerase chain reaction (PCR) amplification and its expression was observed in the suspension cells that were derived from the transgenic plant callus by using Northern blot analysis. The hlL-18 protein was detected in the extracts of the transgenic callus and in the medium of the transgenic tobacco suspension culture by using immunoblot analysis. Based upon enzyme-linked immunosorbant assay (ELISA) results, the expression level of the hlL-18 protein approximated $166{\mu}g/L$ in the suspension culture medium. Bioassay results from the induction of $interferon-{\gamma}$ from a KG-1 cell line indicated that the hlL-18 secreted into the suspension culture medium was bioactive.

  • 논리적으로 동일한 종류에 속하여 서로 유사한 구조를 가지는 많은 XML 문서들이 서로 다른 종류로 분류되어 서로 다른 문서형 정의(DTD)를 가지게 되는 경우가 많다. 이로 인하여 XML 문서를 저장하기 위한 데이타베이스의 스키마가 서로 다르게 되고, 동일한 데이타베이스에 저장되어야 하는 XML 문서들이 서로 다른 데이타베이스에 저장되는 문제점이 발생하게 된다. 이러한 문제점을 해결하기 위하여 본 논문에서는 유한 오토마타와 트리구조를 이용하여 유사한 구조를 가지는 XML 문서들의 DTD를 통합하는 알고리즘을 제안한다. 유한 오토마타는 DTD의 반복연산자나 연결자를 표현하기에 적합하고 표현 방법이 단순하므로 DTD 통합 알고리즘의 복잡도를 감소시킬 수 있다. 또한 제안한 알고리즘의 효과성을 검증하기 위하여 국내 학회 논문지의 논문 DTD를 통합하는데 본 논문에서 제안한 알고리즘을 적용한다.

XML 기반의 통합 임상정보를 효율적으로 저장하기 위한 XML 압축 기법에 대한 연구 (A Study on XML Compress method for efficient integration and storing of XML-based Clinical Information)

  • 임상정보 문서는 환자 진료기록뿐만 아니라 처방전, 개인적 유전자정보를 가지고 있다. 이러한 임상 정보 문서는 병원 시스템들간에 교환 및 공유함으로써 양질의 의료서비스를 제공할 수 있다. 이와 관련하여 임상정보의 통합을 위한 기존의 연구들은 각각 HL7 메시지를 XML 문서로 변환하고 XML 기반의 CDA 를 관계형 데이터베이스에 저장하는 연구가 진행되었다. 그러나 관계형 데이터베이스는 문서의 데이터 별 테이블 단위로 생성, 저장된다. 그러나 HL7 과 CDA 는 문서 중심의 XML 문서이기 때문에 관계형 데이터베이스에 저장 시 문서 별 많은 변이가 존재하여 테이블 증가를 갖는다. 따라서 비정규적인 구조에 적합한 데이터베이스를 선택하기 위해 XML 전용 데이터베이스와 관계형 데이터베이스 비교하고 효율적 저장을 위해 압축기법을 제시한다. 압축기법을 적용한 임상 정보 데이터베이스는 대용량 임상정보 문서의 크기를 압축함으로써 문서의 크기를 줄임으로써 데이터베이스의 효율적 저장을 향상시킨다.

대학 캠퍼스 건물 내부휴게공간의 공간적 특성에 관한 연구 (A Study on Spatial Characteristics of Indoor Rest Zones in the Campus Buildings)

  • This study investigates the physical characteristics of indoor rest zones using the Space Syntax Method. A conceptual framework is based on analysis of design works presented in competitions(544 rest zones of 24 buildings on 7 campuses). The analysis on the unit type of rest zones showed that they were mostly available as open type, followed by semi-open type and outdoor rest places. On the other hand, by classifying the shape of whole building into traffic lines and nodal points, it was found that the buildings with 3 or more traffic lines have lower availability than others, because the buildings required more nodal points along a little higher complexity and zoning formed at increased nodal points between building masses. The analysis on the location of rest zones on traffic lines showed that hall type was most excellent in the level of availability. Finally, the analysis on the floor height showed that hall-like open rest places and outdoor places like rooftop garden functioned as a factor to enhance the integration of different spaces. In conclusion, this research will help the follow-up studies make spatial creation planning that may enhance the spatial efficiency and availability of Indoor rest zones on the campus.

  • The Optical Art is based on the principle of visual perception of the illusionary effects which induce psychological responses. It has influenced greatly on the Texile Design in that unique iJlusionary creativity of pattern simulates the visual sense of special movement; the dynamic psylosophy of vitalism. The Optical pattern has become a highly valued item due to its innovative effect in aesthetic direction. According to Vitor Vasarely the pioneer in this area, the integration and the inseparability of form and color which he calls 'Plastic Unity' provides the basis for the composition of infinite variety. The composition of infinite variety. The composition reveals the complex interaction between the space and form relating to order, repetition, combination and permutation. It is not simple to create optical patterns due to the extreme complexity composed by the multi-dimension and the infusion of form and color giving immensely varied movement. The purposes of this study are as follows; 1) to classify the complex processes of optical pattern on the basis of formative method. 2) to develop creative ideas for progressive contemporary textile design In this study, the analysis of applied methods is concentrated, which is based 1) on the gradual modification and on the transformation of the basic plastic elements which depend on thle direction of visual points involVing contradictory perspectives 2) on the composition varied special situations by repeating, overlapping and converging a series of idetUical units or by means of irrdiation, radiation and etc.

XML의 주체 기반 암호화를 이용한 콘텐츠 패키지의 접근 제어 (Access Control of Content Package by Using XML Subject-based Encryption)

  • XML 문서가 웹 문서의 표준으로 자리 잡음에 따라 많은 정보들이 XML 문서 형식으로 표현되면서 XML 문서의 보안에 관한 요구도 커지고 있다. 현재까지의 XML 보안에 관한 연구는 암호화와 전자 서명 같은 통신상의 보안에 관한 연구가 중심이 되었으나 XML 문서가 방대해지고 복잡해짐에 따라 XML 문서에 대한 통신상의 보안뿐 아니라 관리적인 보안이 필요하게 되었다. 하지만 XML 문서의 암호화는 단순한 통신상의 보안만을 제공할 뿐 관리적 보안 요소인 다양한 사용자와 다양한 접근 권한 정책을 반영하지 못하고 있다. 본 논문에서는 XML 문서의 보안을 위해 주체별 권한 정책을 반영한 접근 제어를 보장하는 주체 기반 XML문서의 암호화 기법을 제안한다. 접근 제어를 보장하는 암호화를 수행함으로써 다양한 사용자의 다양한 권한 정책을 반영할 수 있다.

PLC를 이용한 온실의 환경제어 (Control of Environments in Greenhouse Using Programmable Logic Controller)

  • This study was carried out to develop the control system with PLC and its operating software and to investigate its control ability of greenhouse environments. Two experimental greenhouses were controlled by PLC and ON/OFF controller, respectively. In greenhouse controlled by PLC, target values of air temperature, relative humidity and $CO_2$ concentration were automatically changed. In warm-water heating, the variation of air temperature was reduced to $\pm$ $0.6^{\circ}C$ by the method of proportional-integration(PI) control with an inverter. In ventilation, the variation of air temperature was reduced, since windows open and close with multistage by mutual relation formula among the target, indoor, and outdoor temperature. Relative humidity at daytime was maintained with range of 35% to 55% by PLC controlled fogger. $CO_2$ concentration was automatically controlled from 300 to 800 $\mu$molㆍ$mol^{-1}$ according to amount of solar radiation. The suppling amount and frequency of nutrient solution were controlled by total integrated solar radiation. Difference in the yield of cucumber in the greenhouse controlled by PLC and by ON/OFF controller was not significant at the 5% level.

데오도라이트를 이용한 위성체 얼라인먼트 측정에 관한 연구 (A Study of Spacecraft Alignment Measurement with Theodolite)

  • 위성체 정렬은 위성체 조립 및 시험과정에서 중요한 부분이다. 인공위성이 우주궤도상에서 성공적인 임무를 수행하기 위해서는 자세제어 및 탑재체용 부분품들에 대하여 측정허용오차 $0.1^{\circ}{\sim}0.7^{\circ}$의 정밀하고 정확한 측정이 요구되며 정렬된 상태에서의 위성체 좌표계의 정확한 방향좌표를 측정하여 지상에 위치한 위성체 관제부에서 위성체의 자세제어 등에 사용하도록 제공하게 된다. 본 논문에서는 자동시준에 의한 위성체 정렬 측정 이론에 대하여 기술하고 데오도라이트를 사용하여 위성체 정렬을 측정할 수 있는 측정방법 및 그 측정 결과에 대하여 고찰해 보고자 한다.

블루오션 전략을 적용한 요구공학 프로세스 (Requirements Engineering Process Applying the Blue Ocean Strategy)

  • According to the change of customer needs, integration with other systems, and the advent of new technology, it is inevitable for the existing system requirements to be requested to redesign. However, there are actually no methodologies and processes to be able to reorganize the existing requirements or integrate a number of requirements different from each other. The traditional requirements processes do not work when solving such problems. Thus, since the existing approaches to develop requirements are not helpful, many traditional requirement engineering processes can not help falling in Red Ocean. In this paper, in order to solve such the problems, we propose a novel requirements engineering process applying Blue ocean strategy which is used to open a non-competition market not discovered yet. Blue Ocean approach is a novel effective method to elicit and build up new requirements from customers. In order to accomplish the strategy, this paper presents a framework and processes to discover new perspectives and analyze the corresponding requirements. In addition, some pieces of case study are presented to introduce how our approach is applied to practical system developments and the evaluation analysis for how valid it is.

평면 다물체 동역학 해석에서 GPU 병렬 프로그래밍의 계산효과 (Calculation Effect of GPU Parallel Programing for Planar Multibody System Dynamics)

  • In this paper, the equations of motions for planar multibody dynamics are established for considering the parallel programming based on GPU. Cartesian coordinates are used to formulate the equations of motion and implicit integration method called HHT-alpha is employed. Open chain multibody system is considered for computer simulation. CUDA toolkit is employed for establishing the GPU parallel programming. The exactness of the analysis is verified from the comparison with ADAMS. The results from parallel computing based on GPU are compared with the results from the sequential programming based on CPU in terms of calculation time. The multiple pendulum with bodies and joints is employed for the computer simulation. In the pendulum system that has 290 bodies, the parallel program indicates an improved efficiency of about 25.5 second(15.5% improvement). It is noted that the larger the size of system is, the time efficiency is better.
