Polyhydroxyamides(PHAs) having poly(ethylene glycol)methyl ether (MPEG) and/or dimethylphenoxy pendant groups were synthesized by solution polycondensation at low temperature. The inherent viscosities of the PHAs measured at $35^{\circ}C$ in DMAC or DMAc/LiCl solution were in the range of $0.51\~2.31dL/g$. This precursor polymers were studied by FT-IR, $1H-NMR$, DSC, and TGA. Solubility of the precursors with higher MPEG unit was increased, especially the polymer having MPEG $(M_n=1100)$ was soluble or partially soluble in ethanol, methanol, and water as well as aprotic solvents, but the PBOs were nearly insoluble in a variety of solvents. PHAs were converted to polybenzoxazoles (PBOs) by thermal cyclization reaction with heat of endotherm. In case of the precursors having MPEG nit, the precursor polymers with a higher $M_n$ were fully cyclized at a lower temperature than one with a lower $M_n$.
Objective: The present study was to investigate the extraction conditions of dietary fiber from dried cassava pulp (DCP) and cassava distiller's dried grains (CDG) under different NaOH concentrations, and the Fourier transform infrared (FTIR) was used to determine the dietary fiber components. Methods: The dried samples (DCP and CDG) were treated with various concentrations of NaOH at levels of 2%, 4%, 6%, and 8% using a completely randomized design with 4 replications of each. After extraction, the residual DCP and CDG dietary fiber were dried in a hot air oven at 55℃ to 60℃. Finally, the oven dried extracted dietary fiber was powdered to a particle size of 1 mm. Both extracted dietary fibers were analyzed for their chemical composition and determined by FTIR. Results: The DCP and CDG treated with NaOH linearly or quadratically or cubically (p<0.05) increased the total dietary fiber (TDF) and insoluble fiber (IDF). The optimal conditions for extracting dietary fiber from DCP and CDG were under treatment with 6% and 4% NaOH, respectively, as these conditions yielded the highest TDF and IDF contents. These results were associated with the FTIR spectra integration for a semi-quantitative analysis, which obtained the highest cellulose content in dietary fiber extracted from DCP and CDG with 6% and 4% NaOH solution, respectively. The principal component analysis illustrated clear separation of spectral distribution in cassava pulp extracted dietary fiber (DFCP) and cassava distiller's dried grains extracted dietary fiber (DFCDG) when treated with 6% and 4% NaOH, respectively. Conclusion: The optimal conditions for the extraction of dietary fiber from DCP and CDG were treatment with 6% and 4% NaOH solution, respectively. In addition, FTIR spectroscopy proved itself to be a powerful tool for fiber identification.
Effects of different processes or texture of cucumber pickles fermented in 7% NaCl solution were studied. Cucumbers were treated with different processes: control; preheating in 7% NaCl, 0.04M $CaCl_2$ solution ($60^{\circ}C$, 1hr); combination of preheating and post microwave heating (5 min, midium high level, 2450 Mhz); preheating in the solution which was adjusted to pH 8.5. In each process the pH and total acidity of brine solution, total pectin contents in alcohol insoluble solids (AIS), hot water soluble pectin (HWSP) contents and firmness of cucumber pickles were determined at 0, 5, 10, 15 and 25 days of storage periods. Total pectin contents and the firmness of pickles were decreased during 25 days of storage. Control group showed the lowest value and post microwave heating group marked the highest value in total pectin contents and firmness retention of cucumber pickles. The results of this study indicated that total pectin content seems to correlate with the firmness of cucumber pickles. However, HWSP content did not show any correlation with firmness of cucumber pickles. Post microwave heating method could be used to retain firmness of fermented cucumber pickles.
The purpose of this study was to find out required time for recovery of nutritional disorder by Fe-DTPA treatment in induced Fe-deficient tomato cultivars and to select stable Fe-chelate in high pH of nutrient solution. The pH levels of nutrient solution were amended with 6.0, 7.0, and 8.0. Then Fe-EDTA (Ethylenediaminetetraacetic acid, ferric-sodium salt), Fe-DTPA (Sodium ferric diethylenetriamine pentaacetate), and Fe-EDDHA (Ethylenediamine-N,N-bis (2-hydroxyphenylacetic acid) ferric-sodium salt)) were treated as Fe $2.0mg\;L^{-1}$ concentration. The Fe-DTPA and Fe-EDDHA were stable in the nutrient solution of pH 6.0~8.0 but Fe-EDTA in nutrient solution of pH 8.0 was to become insoluble by 25%. The Fe $2.0mg\;L^{-1}$ as Fe-DTPA was treated for recovery of Fe deficient tomato seedlings. In case of Redyoyo and Supersunroad cultivars, total chlorophyll and Fe contents of leaves were recovered as much as those of normal leaves in 5 days. The Rafito cultivar for complete recovery was taken 7 days. When Fe $2.0mg\;L^{-1}$ as Fe-DTPA was supplied to Fe-deficient tomato seedlings, in geotype, heme oxigenase recovered as much as normal leaves in 24 hours in the Rafito and Redyoyo. However, it was not remarkable difference by elapsed time in the Supersunroad.
In this study, poly(DL-lactide-co-glycolide) nanoparticles loaded with water-insoluble vitamins such as vitamin A (Retinol) and vitamin E acetate were prepared by the emulsification diffusion method. Polymer solution was prepared by the two water-miscible organic solvent, such as ethanol and acetone. Because of its biocompatible property, polyethyleneglycol-polypropyleneglycol diblock copolymer was used as surfactant and stabilizer. The influence of some preparative variables on the nanoparticle formation and on the loading efficiency of active agents, such as the type and concentration of stabilizing agent, the stirring methods, the water/oil phase ratio and the polymer concentration were investigated in order to control and optimize the process. After preparation of nanoparticles loaded with active agent, particle size and distribution were evaluated by the light scattering particle analyzer. The loading efficiency of active agents was evaluated by the UV-visible spectroscopy. As the results, particle size were 50-200 nm and dispersibility was monodisperse. The optimum loading efficiency of active agents was observed 50-60%. It was found that the appropriate of selections of binary solvent mixtures and polymeric concentrations in both organic and aqueous phases could provide good yield and favorable physical properties of PLGA nanoparticles.
Action of a cyclodextrin glycosyltransferase (CGTase) produced from alkalophilic Bacillus sp. No.4 was studied in a solution containing starch and sucrose to prepare glycosyl sucrose syrup with good sweetness and antidecaying properties of teeth. In the initial stage of the reaction the CGTase produced cyclodextrin, however, the cyclodextrin disappeared and glycosyl sucrose was formed with the lapse of reaction time. The best proportion of sucrose to starch for prodution of glycosyl sucrose was about 1 : 1. The optimum pH and temperature of the coupling reaction was pH 6.0 and $60^{\circ}C$, respectively. Main composition of glycosyl sucrose syrup prepared with 20% starch and 20% sucrose was sucrose 18%, glucosyl sucrose ($G_{2}F$) 15.3% and maltosyl sucorse ($G_{3}F$) 11.3%. And glucose, maltose and maltotriose were produced very little. Smaller amounts of acid and insoluble glucan were formed in the syrup by Streptococcus mtans OMZ176 than in the sucrose. Therefore, the prepared glycosyl sucrose sucrose syrup is expected to prevent teeth from decaying.
In order to effectively utilize marine animal skin wastes in marine processing manufacture, conger eel skin, file fish skin and arrow squid skin as raw material of edible gelatin were screened. Conger eel skin was the highest in the collagen content, followed by Ole fish skin and arrow squid skin, in the order named. In the fish skins, the soluble and insoluble collagens occupied $67.4%{\sim}72.3%\;and\;27.7{\sim}32.6%$, respectively, and in the arrow squid skin, 30.4ft and 69.6ft, respectively. No difference in the amino acid composition between soluble and insoluble collagens was detected. Collagen from the marine animal skin catched in coasted and offshore water in Korea consisted ${\alpha}$ chain and ${\beta}$ chain, and ${\alpha}$ chain were hetero type. The sum of proline and hydroxyproline contents in conger eel skin collagen was higher than that in the other skin collagens, while was lower than that pork skin collagen. Conger eel skin collagen exhibited a higher denaturation temperature in solution and a higher degree of proline hydroxylation, compared with skin collagen of the respective species. The physical properties such as gel strength, melting point and gelling point of conger eel skin gelatin were superior to those of file fish skin and arrow squid skin gelatins.
In this paper, the recycling of waste Ni-MH battery by-products for electric vehicle is studied. Although rare earths elements still exist in waste Ni-MH battery by-products, they are not valuable as materials in the form of by-products (such as an insoluble substance). This study investigates the recovering of rare earth oxide for solvent extraction A/O ratio, substitution reaction, and reaction temperature, and scrubbing of the rare earth elements for high purity separation. The by-product (in the form of rare earth elements insoluble powder) is converted into hydroxide form using 30% sodium hydroxide solution. The remaining impurities are purified using the difference in solubility of oxalic acid. Subsequently, Yttrium is isolated by means of D2EHPA (Di-[2-ethylhexyl] phosphoric acid). After cerium is separated using potassium permanganate, lanthanum and neodymium are separated using PC88A (2-ethylhexylphosphonic acid mono-2-ethylhexyl ester) and it is calcinated at a temperature of 800 ℃. As a result of the physical and chemical measurement of the calcined lanthanum and neodymium powder, it is confirmed that the powder is a microsized porous powder in an oxide form of 99.9% or more. Rare earth oxides are recovered from Ni-MH battery by-products through two solvent extraction processes and one oxidation process. This study has regenerated lanthanum and neodymium oxide as a useful material.
We investigated the effects of a mutan (water-insoluble ${\alpha}$-glucans) isolated from Streptococcus mutans on the healing of bone defect in rat. Sprague-Dawley rats were divided into control (saline-treated), lipopolysaccharide (LPS)-treated, and mutan-treated groups (n=6 per group). Experimental bone defects were surgically created with round fissure bur at the buccal surface of the left mandibular. The control groups was administered with saline solution (0.1 ml/100 g), while the LPS and mutan group was given LPS and mutan (1 mg/kg body weight) three times weekly. After 4 weeks the rats were sacrificed, the healing of bone defect was assessed by bone mineral density (BMD) and micro-computed tomography (${\mu}CT$) examination. Percent bone volume (bone volume/tissue volume [BV/TV]), trabecular thickness (Tb.Th), and trabecular number (Tb.N) parameters of ${\mu}CT$ showed higher values in control group than LPS and mutan group. Bone surface/volume ratio (BS/BV), trabecular bone pattern factor (Tb.Pf), and structure model index parameters of ${\mu}CT$ showed higher values in LPS group than mutan group. BMD values of mutan treated-alveolar bones were significantly lower for than that of the LPS group. Therefore, we suggest that mutan, water-insoluble ${\alpha}$-glucans from S. mutans may be induce the induction of periodontal diseases.
The effect of sludge digestion on the leaching efficiency of heavy metals from sludge by bioleaching with Thiobacillus thiooxidans MET was investigated. The used sludges were a non- and anaerobically digested. The leaching efficiency of heavy metals was strongly dependent on the pH of the sludge solution rather than the sludge condition and stolid concentration. The lower the pH the more heavy metal was leached from 3.0 of pH. The sequent orders of leaching heavy metals were Zn, Cu, and Cr. Although the buffering capacity of non-digested sludge was similar to anaerobically digested sludge, the pH decrease rate of the anaerobically digested sludge solution was faster than that of the non-digested sludge solution due to the higher sulfur oxidation rate of T. thiooxidans MET in the anaerobically digested sludge. The amount of leached heavy metals from the anaerobically digested sludge showed higher than that of non-digested sludge at the same pH value. This result might be caused by the difference of the insoluble metal types, which were contained in the sludge. An increase in sludge solids concentration decreased the leaching efficiency of heavy metals in the range of solids concentration 10 g/L to 70 g/L. The optimum ratio of S° to sludge stolid was 0.1 in both the sludge. The bioleaching process of heavy metals with T. thiooxidans MET showed the disinfecting effect over 90% as well as the reduction effect in sludge weight of 20%.
