Analysis of Optimum Condition for Alcoholic Drink Production Using Onion Extract. (양파즙을 사용한 알코올 음료 제조를 위한 최적조건 검토)

  • Onions are considered to be a promising source of the alcoholic drink because they are rich in sugar, amino acids and various nutrients. To isolate strains of Saccharomyces cerevisiae producing ethanol of higher concentration, 19 strains were subjected to screening. Among them, the strain producing the highest concentration of ethanol was OJ-8 strain. Onion's oder was effectively removed by treatment for 30 min with 10% (w/v) charcoal against medium and then heat treatment of onion extract for 40 min at $100^{\circ}C$. The optimum conditions for alcoholic fermentation was investigated in medium containing the onion extract. The optimal conditions for ethanol production was obtained by standing culture for 5 days at $25^{\circ}C$ with 5% inoculum volume.

Three Different Viruses Isolated from Typical Weed Plants that Grown Adjacent to Common Crop Fields

  • Weeds are widely grown in the field and are infected by many viruses. A survey was conducted to identify viruses infecting weeds in Korea. Virus-infected weed samples including Rorippa indica (L.) Hiern, R. islandica (Oed.) Bord, Crepidiastrum denticulatum (Houtt.) Pak & Kawanno, Achyranthes japonica (Miq.) Nakai, and Chrysanthemum boreale (Makino) Makino were collected in Kyonggi Province. These weeds were grown in the greenhouse and were isolated on 10 test plants. Several virus isolates were isolated fron infected tissues and were further studied by host range assay, serological test, electron microscopy (EM), reverse transcription-polymerase chain reaction (RT-PCR) and sequencing. Each isolated virus strain was mechanically transmitted to weeds and various hosts including Nicotiana spp., Brassica spp., Vigna unguiculata, Capsicum annuum, and Cucumis sativus and showed systemic mosaic, vein clearing, necrosis, mottle, malformation, chlorosis, and/or death of host plants in some cases. Each virus was then purified using infected leaves and observed by EM. From these results three viruses were isolated and identified as Turnip mosaic virus (TuMV), Broad bean wilt virus (BBWV), and Cucumber mosaic virus (CMV). RT-PCR using virus-specific oligonucleotide primers and the cloning were conducted to determine the nucleotide sequences of coat proteins of the three viruses their amino acid sequence were deduced. The amino acid sequence homologies were about 92.7 to 99.7%, 96.2 to 97.7%, and 93.9 to 98.6% to other reported TuMV, BBWV, and CMV strains, respectively. These results suggest that many weeds may serve as primary inoculum source of diseases caused by TuMV, BBWV, CMV and that the management of these viral diseases can be achieved through weed control.

Effect of Temperature on Removal of Ammonia in the Ceramic Biofilter Inoculated with Earthworm Casts (지렁이 분변토를 접종한 세라믹 바이오필터의 암모니아 제거에 미치는 온도의 영향)

  • Removal of ammonia using the porous ceramic biofilter inoculated with earthworm casts was characterized. By assuming a plug air flow in the biofilter and applying the Michaelis-Menten equation, the maximum removal rate of $NH_3$ was $280.7g-N{\cdot}m^{-3}{\cdot}h^{-1}$($18.0g-N{\cdot}kg^{-1}{\cdot}d^{-1}$) at $30^{\circ}C$. $NH_3$ removal rate was increased as temperature increases from $15^{\circ}C$ to $35^{\circ}C$. The maximum removal rate was $285.8g-N{\cdot}m^{-3}{\cdot}h^{-1}$($18.8g-N{\cdot}kg^{-1}{\cdot}d^{-1}$) at $35^{\circ}C$. At $15^{\circ}C$, the $NH_3$ removal rate was $122.8g-N{\cdot}m^{-3}{\cdot}h^{-1}$($8.1g-N{\cdot}kg^{-1}{\cdot}d^{-1}$). When 210 ppm $NH_3$ was supplied to the biofilter at space velocity of $220h^{-1}$, the removal efficiency of $NH_3$ at 15, 25, 30 and $35^{\circ}C$ was 80, 90, 95, and 96%, respectively. The removal rate of the ceramic biofilter was 3 to 15 times higher than other biofilters comparing the removal efficiency of $NH_3$ per unit volume of carrier. This result indicates that earthworm casts and porous ceramics are very good inoculum source and carrier, respectively, for the $NH_3$-degrading biofilter.

Sensory Characteristics of Citrus Vinegar fermented by Gluconacetobacter hansenii CV1 (제주 감귤식초 발효균주 선발)

  • Citrus juice, a concentrate manufactured by the Jeju Provincial Corporation, was converted into vinegar orderly by alcohol and acetate fermentation. The juice with a 6-fold dilution by distilled water was used as the sole nutrient source throughout the experiments. The diluted juice contained 12.96Brix of total sugar, $0.632\%$ of total acid and $20.23{\mu}g/ml$ of hesperidin. Naringin was not detected from the juice. Citrus wine having $5.6\~6.3\%$ alcohol was produced from the diluted juice after 3 days of fermentation at $28^{\circ}C$. A kind of citrus-malomelo-yeast CMY-28 was used for the wine fermentation. The wine was successfully fermented for 8 days at $30^{\circ}C$ after inoculation of seed vinegar which contained active cells of acid producing bacteria CV1. The inoculum size of the seed vinegar was controlled to $10\%$(v/v) of the citrus wine. The wine was converted into vinegar by the fermentation process. Citrus vinegar, the final fermentation product, was colored with very thin, radish-yellow and was transparent. It's acidity ranged between $5.8\~6.2\%$ of that of acetic acid. The vinegar attained the best score by sensory test among several natural fruit vinegars. It was clear from the results that high quality citrus vinegar could be produced from concentrated citrus juice. However, the fermentation conditions should be improved to reduce the amount of reducing alcohol.

Bacterial Community Profiling during the Manufacturing Process of Traditional Soybean Paste by Pyrosequencing Method (Pyrosequencing을 이용한 전통된장 제조과정 중 세균군집구조의 분석)

  • In order to evaluate the diversity and change of bacterial population during the manufacturing process of traditional soybean paste (doenjang), bacterial communities were analyzed using 16S rRNA gene-based pyrosequencing. In rice straw, the most important inoculum source for fermentation, the bacterial sequences with a relative abundance greater than 1% were assigned to four phyla, Proteobacteria (71%), Actinobacteria (20.6%), Bacteroidetes (4.2%), and Firmicutes (1.3%). Unlike bacterial community composition of rice straw, a different pattern of bacterial population in meju was observed with predominantly high abundance (99.1%) of Firmicutes. Phylum composition in young doenjang was almost same as that of meju. Major genera in young doenjang were Bacillus (81.3%), Clostridium (6.9%) and Enterococcus (6.3%) and the predominant species among bacterial population was B. amyloliquefaciens (63.6%). Abundance of the phylum Firmicutes in mature doenjang was 99.98%, which was even higher value than those in meju and young doenjang. Predominant species in mature doenjang were B. amyloliquefaciens (67.3%), B. atrophaeus (12.7%), B. methylotrophicus (6.5%), B. mojavensis (3.2%), and B. subtilis. (2.5%), which were also identified as major species of the microbial flora in meju. These results suggested that rice straw was a primary source for supplement of Bacillus species in manufacturing the traditional doenjang and that some species of Bacillus strains were mainly involved in the fermentation process of traditional doenjang.

Preliminary Study of Semi-continuous Liquid Recirculating Anaerobic Digestion for Source Separated Food Waste (음식물류 폐기물 처리를 위한 준 회분식 액순환 건식 혐기성 소화법에 대한 기초연구)

  • In this study, the experiment was carried out to produce methane by applying Semi-Continuous Leachate Recirculation Anaerobic Digestion System fed with source separated food waste from school cafeteria. There were two systems and each system consisted of a bioreactor and a liquid tank. Each bioreactor had a screen near the bottom of the reactor. 2.5L of separated liquid was transferred to the liquid tank for 30min each day by using a tubing pump and the liquid from the liquid tank was pumped to the bioreactor at the upper of the bioreactor as soon as the transfer was ended. Through this circulation, the liquid having high concentration of VFAs was supplied to the top of bioreactor. At the beginning of the experiment, food waste/inoculum anaerobic sludge volume ratio was 2:8 that is 9g VS/L of OLR(Organic Loading Rate). Feeding was conducted every two weeks. Experimental results showed that the contents of moisture, combustible matter, ash were 65.91%, 32.73%, and 1.36%, respectively. Two different food waste loading were studied. The average organic loading rates were 3.51g VS/d for System A and 3.86g VS/d for System B, respectively. The average produced methane based on food waste fed to bioreactor were observed as $6.30m^3CH_4/kgVS{\cdot}d$ for system A and $4.94m^3CH_4/kgVS{\cdot}d$ for System B, respectively.

Effect of NO3- and NH4+ Concentrations on Root Growth and Eleutherosides Accumulation in adventitious root Culture of Eleutherococcus senticosus (가시오갈피의 부정근 배양시 부정근의 생육과 eleutheroside류의 함량에 미치는 NO3-와 NH4+ 비율 및 농도의 영향)

  • This study was carried out to investigate the effect of $NO_3{^-}$ and $NH_4{^+}$ on the adventitious root growth and eleuthroside synthesis of Eleutherococcus senticosus during 3 L-bioreactor culture. The change of medium component ratio was also measured during culture. The fresh weignt of adventitious root reached to the greatest level of 24.4g FW/L in the presence of 50 mM $NO_3{^-}$ and 10 mM $NH_4{^+}$, representing 3.4-fold increase compared to the 60 mM $NH_4{^+}$. However, as the increase of the portion of $NH_4{^+}$, the root growth was decreased. Maximum eleutheroside B and E1 production were $249{\mu}g/g$ and $43{\mu}g/g$, respectively, with 30 mM total nitrogen source. The maximum production of eleutheroside E were $788{\mu}g/g$ with 120 mM total nitrogen source. The greatest weight of adventitious root increased up to 6.2 fold of inoculum size within 9 weeks. The change of pH was influenced from 4.81 to 6.35 and the amounts of $NH_4{^+}$ and $K^+$ were decreased during culture periods. From these results we suggest, need further study of various treatment to increase the growth of biomass and the accumulation of useful secondary metabilites.

Distribution and Favorable Conditions for Mycelial Growth of Cordyceps pruinosa in Korea (한국에 자생하는 붉은자루동충하초(Cordyceps pruinosa)의 분포와 균사생장에 적합한 조건)

  • Cordyceps pruinosa grows upon dead pupae of Lepidoptera and produces one or $3{\sim}4$ club-shaped stromata per host. The stromata have distinct club-shaped head and long stalk. The length of stromata varies from $1{\sim}3\;cm$. Apical head consists of densely crowded semi-immersed perithecia, which are $360{\sim}400\;{\times}\;180{\sim}200\;{\mu}m$ in size. Asci are $150\;{\mu}m$ in length and $2.8{\sim}3\;{\mu}m$ in diameter. Ascospores, which are $124{\sim}141\;{\mu}m$ in length, have thin thread-like structures in the middle with part-spores attached on both sides. Each ascospore does not separate into part-spores after dispersal, but each part-spore germinates and together develops a colony. The imperfect form produces phialides of $15{\sim}24\;{\times}\;2{\sim}3\;{\mu}m$ size, with spherical or spindle shaped conidia of $4{\sim}6\;{\times}\;1.8{\sim}2.4\;{\mu}m$ size, The anamorph was identified as Mariannaea elegans Samson. YMA and SDAY agar media with pH 7 was produced abundant mycelial growth with high density. Best mycelial growth was observed when dextrin was used as a carbon source. Lactose, saccharose and sucrose also produced high mycelial growth. Peptone, yeast extract and tryptone produced abundant mycelial growth, when used as nitrogen sources. Highest mycelial growth and density was observed when C/N ratio was 1 : 1 at the concentration of 12.5 g/l each. $KH_2PO_4$ was the best mineral source for mycelial growth. Highest mycelial dry wt. was produced in YM and SDAY broths. Optimum inoculum for 100 ml of liquid broth was 6 mycelial discs. Similarly, optimum liquid culture period was 7 days.

The Incidence and Distribution of Viral Diseases in Pepper by Cultivation Types (시설 및 노지재배 고추의 바이러스병 발생과 분포)

  • In the year of 2002 annual nationwide survey of virus diseases occurring in the pepper fields and greenhouses in Korea, the distribution and the incidence of viral diseases was investigated. The pepper samples from both greenhouses (155 samples) and open fields (227 samples) were collected and further analyzed to detect eleven different viruses by RT-PCR. The results indicate that no sample collected from both greenhouse and open field seems to be infected by TMV, RMV, PVY, AMV, and TSWV. On the other hand, CMV, BBWV2, PepMoV, PMMoV, TMGMV and ToMV are readily identified from greenhouse and open field samples by RT-PCR. The infection rates of the collected samples between greenhouse and open field are largely different. Comparing with 10% of virus-infected pepper samples grown in greenhouse, approximately one third of pepper samples collected from open field are infected. The mixed-infection rates in the virus-infected greenhouse and open field samples are 16% and 61%, respectively. The dominant virus occuring in greenhouse is PMMoV, indicating that virus-infected seed stocks and infected plant debris in the growing area may be important sources of inocula. On the other hand, both CMV and BBWV2 are dominant viruses in open field. This may indicate that the migration of viruliferous insect vectors into pepper fields may be the most important source of inoculum. Also, the survey shows that BBWV2 is newly immerging virus to be controlled in Korea. The discrepancies on the distribution and the occurrence of viral diseases between field and greenhouse may provide a fact that the accumulation and distribution of inoculum by successive cultivation and the migration of viruliferous vectors into growing areas are likely to be important factors to determine the incidence of viral diseases. Therefore, the further studies on epidemiology and the consideration of new breeding program of pepper are essential to minimize virus diseases.

Effect of Nitrogen on Eleutheroside production and Adventitious Root Growth in Eleutherococcus koreanum Nakai Bioreactor Cultures (생물반응기를 이용한 섬오갈피나무의 부정근 배양시 질소농도 및 NH4+와 NO3- 비율이 부정근의 생육과 eleutherosides 함량에 미치는 영향)

  • This study was carried out to investigate the effect of ${NO_3}^-$ and ${NH_4}^+$ on the adventitious root growth and eleuthroside synthesis of Eleutherococcus koreanum in 5 L-bioreactor culture. The change in the medium components was also measured during culture. The fresh weignt of adventitious root reached to the highest level of 30.8 g FW/L in the presence of both 50 mM ${NO_3}^-$ and 10 mM $NH_4^+$, representing 3.6-fold increase compared to the 60 mM ${NH_4}^+$ alone. However, as the increase of the portion of ${NH_4}^+$, the root growth was decreased. However, the maximum eleutheroside B, E and E1 contents were $57.3{\mu}g/g$ DW, $188.4{\mu}g/g$ DW and $47.3{\mu}g/g$ DW, with 30 mM, 60 mM and 15 mM total nitrogen source, respectively. Fresh weight of adventitious root increased up to 6.8-fold of inoculum size within 9 weeks. The amounts of ${NH_4}^+$, $K^+$, ${NO_3}^-$ and ${PO_4}^-$ were decreased during culture periods. Based on these results, we suggest that various further studies are required to increase the biomass and the useful secondary metabilites.
