The survival of Colletotrichum acutatum was investigated in soil, infected fruits, and infected fruit debris incorporated into soil at several temperatures with different soil moisture levels. Samples were examined at 2-week intervals for 18 weeks to determine the survival of the pathogen based on the number of colony forming unit (CFU) of C. acutatum recovered on a semi-selective medium. C. acutatum conidia survived in both sterile and non-sterile soil at 4 and $10^{\circ}C$ for 18 weeks. If infected pepper fruits were completely dried, C. acutatum survived for 18 weeks at temperature from 4 to $20^{\circ}C$. Soil temperature and moisture affected the survival of C. acutatum in infected fruit debris incorporated into soil after air-drying. The effect of soil moisture on survival was weaker at low temperatures than at high temperatures. For up to 16 weeks, conidia were recovered from fruit debris in soil that had been kept at 4 to $20^{\circ}C$ and below 6% soil moisture. Conidia were recovered from fields until approximately 6 months after pepper fruits were harvested. Using PCR with species-specific primers and a pathogenicity test, we identified conidia recovered from soil and infected fruit from both the laboratory and field as C. acutatum and as the primary inoculum causing pepper anthracnose.

This experiment was carried out to investigate the primary inoculum of strawberry anthracnose in nursery field. The pathogen, Colletotrichum gloeosporioides was not detected in soil and weeds of nursery field but symptom of anthracnose was developed in mother plants collected from market after incubation in humid chamber, The symptom of anthracnose was expressed in the strawberry plant that reserved for 17 days in field after inoculation by spore suspension but was not observed there after. When inoculated leaves were observed by SEM, only appressoria were observed 7 days after inoculation. So, it is guessed that dissemination of Colletotrichum sp. into nursery held will be by contamination of mother plants, and diagnosis by naked eyes may be impossible because symptom will be not developed if environment is to be adequate to penetration and in case of imperfect penetration after germination, the pathogen remains appressorium to achieve penetration.
We investigated turfgrass diseases and inoculum density at nationwide turfgrass cultivation sites in year of 2013. Occurrences of large patch and rust disease appeared in September. Brown patch recorded in September to October at Namhea and Pythium blight disease occurred outbreaks in early July at Namhea site. Some sites in Namhea damaged 3% area of total cultivation field by Sclerotinia homoeocarp. In Daepyeong (Gyeongnam), Fairy ring and large patch were recorded. Severe takeall and fairy ring have been observed in Gochang-si. Multi-site in Cheongju-si, brown patch was observed in pandemic level. Interesting enough, a cool-season turfgrass cultivate sites in Pyeongtaek-si brown patch, leaf blast, summer patch, and Curvularia leaf spot were investigated during the surveys period. Inoculum densities (Rhizoctonia spp.) at turfgrass cultivations sites were increased as cumulatively in all survey sites. The investigation result indicated that the disease occurrence and pathogens are similar as diseases in golf courses.
In order to illustrate the role of conidia of Mycosphaerella nawae as a secondary inoculum in nature, pseudothecial development on persimmon leaves was investigated microscopically. The fungal ascospores have been believed as the primary or only inoculum source in nature, however, pseudothecia were readily formed on persimmon leaves infected naturally and artificially by conidia. The pseudothecia of M. nawae were found to form in the tissues of infected leaves while the leaves were still hanging on the trees. The size of pseudothecia were approximately 51.0~122.4$\times$51.0~112.2 ${\mu}{\textrm}{m}$ (82.8 $\times$72.5 ${\mu}{\textrm}{m}$in average), the shapes were spherical, ovoid or occidental pear type. The sizes of asci were approximately 30.6~61.2$\times$8.2~10.2 ${\mu}{\textrm}{m}$(46.6$\times$9.4 ${\mu}{\textrm}{m}$ in average) and the shapes were cylinder or banana. The ascospores were mostly spindle type, and the sizes were 10.2~12.2$\times$3.1~4.1 ${\mu}{\textrm}{m}$ (11.4$\times$3.2 ${\mu}{\textrm}{m}$ in average)-like. The pseudothecial formation was initiated before defoliation and morphological characteristics of the pseudothecia, ascus and ascospores on the infected leaves were fully illustrated in this study. Results indicated that conidia of M. nawae induce circular leaf spot of persimmon as much as ascospores, and might play an important role of the disease epidemics in nature.

Purpose: Methane production potential in aerobic digestion was assessed according to feed to inoculum (F/I) ratio for food waste only, and mixing ratio of two materials for food waste and swine manure to give a basic data for the design of anaerobic digestion system. Methods: Anaerbic digestion test was performed using a lab scale batch reactor at $35^{\circ}C$ for six different feed to inoculum (F/I) ratios (0.50, 0.72, 1.14, 1.50, 2.14 and 3.41), three food waste to swine manure ratios (100:0, 60:40 and 40:60) with two different loading concentrations (10g VS/L and 30g VS/L). Results: For food waste only, the highest biogas yield of 1008 mL/gVS was obtained at 0.50 of F/I. For the co-digestion of food waste and swine manure mixture, the highest biogas yield of 1148 mL/gVS was obtained at a mixing ratio of 40:60 with loading concentration of 10g VS/L. Conclusions: F/I ratio for the food waste only, mixing ratio of food waste and swine manure, and co-substrate loading rate affected the biogas production rate. For the low loading rate, there was not so much difference according to the mixing ratio of food waste and swine manure, but for the high loading rate higher biogas yield was acquired for the co-digestion of food waste and swine manure than for the food waste alone (mixing ratio, 100:0).

The efficiency of the OECD MITI(I) test which is designated as a standard method for measuring the biodegradability of chemicals by the Korean Ministry of Environment was evaluated and compared with those of two other OECD ready biodegradability tests: the manometric respirometry test and the closed bottle test. All the tests were applicable to a wide variety of organic chemicals and there were no significant differences in biodegradation rates of eight test chemicals and a reference chemical with three methods. However, the MITI(I) test had unnecessary expense and complexity in preparation of a mixed inoculum. Decrease in bacterial diversity during pre-conditioning of the inoculum for 1 month was observed and therefore the mixed inoculum did not ensure the presence of a variety of species in the test. Based on the simplicity, cost-effectiveness and reproducibility of the test procedure, it is considered that the manometric respirometry test is the most adequate method which can replace the MITI(I) test method.
The current research was focused on the extracellular biosynthesis of bactericidal silver nanoparticles (AgNPs) using cell-free supernatant of a local isolate previously identified as a novel Streptomyces aegyptia NEAE 102. The biosynthesis of silver nanoparticles by Streptomyces aegyptia NEAE 102 was quite fast and required far less time than previously published strains. The produced particles showed a single surface plasmon resonance peak at 400 nm by UV-Vis spectroscopy, which confirmed the presence of AgNPs. Response surface methodology was chosen to evaluate the effects of four process variables ($AgNO_3$ concentration, incubation period, pH levels, and inoculum size) on the biosynthesis of silver nanoparticles by Streptomyces aegyptia NEAE 102. Statistical analysis of the results showed that the linear and quadratic effects of incubation period, initial pH, and inoculum size had a significant effect (p < 0.05) on the biosynthesis of silver nanoparticles by Streptomyces aegyptia NEAE 102. The maximum silver nanoparticles biosynthesis (2.5 OD, at 400 nm ) was achieved in runs number 5 and 14 under the conditions of 1 mM $AgNO_3$ (1-1.5% (v/v)), incubation period (72-96 h), initial pH (9-10), and inoculum size (2-4% (v/v)). An overall 4-fold increase in AgNPs biosynthesis was obtained as compared with that of unoptimized conditions. The biosynthesized silver nanoparticles were characterized using UV-VIS spectrophotometer and Fourier transform infrared spectroscopy analysis, in addition to antimicrobial properties. The biosynthesized AgNPs significantly inhibited the growth of medically important pathogenic gram-positive (Staphylococcus aureus) and gram-negative bacteria (Pseudomonas aeruginosa) and yeast (Candida albicans).
Sclerotium rolfsii causing southern blight on numerous vegetable and fruit crops was isolated from stems of chili peppers showing wilting symptoms. The pathogen was identified by morphological observation and DNA sequencing analysis of ITS region. To select an effective fungicide for control of southern blight, we investigated the inhibition efficacy of thirty fungicides included in nine groups of fungicides with different mechanisms of action. A fungal growth inhibition assay was conducted through an agar dilution method by using mycelial discs and sclerotia of the pathogen as inoculum, respectively. When mycelial discs were used as an inoculum, several fungicides showed good inhibitory activity against the mycelial growth of S. rolfsii 12-6. All DMI fungicides tested had a good inhibition except for prochloraz which had low inhibitory effect. All strobilurin fungicides tested except for kresoxim-methyl and all SDHI fungicides tested except for boscalid and fluopyram, had a good inhibition. Also, fludioxonil, a protective fungicide and fluazinam had a good inhibitory effect. Interestingly, when sclerotia were used as an inoculum, inhibition efficacy was increased for fluopyram, a SDHI fungicide, and for some protective fungicides such as propineb, chlorothalonil, dithianon, and folpet. All the fungicides selected in this study should be tested in the field for their control activities against stem rot for practical use in chili pepper cultivation.
Two isolates of Tricholoma matsutake T-008 and T-034, preserved in Entomopathogenic Fungal Culture Collection (EFCC) of Korea, were used in the present study. The isolates had 100% Bootstrap homology with Tricholoma matsutake U62964 and T. matsutake AB188557 and AF309538 preserved in Gene Bank of NCBI. Mycelial growth of T. matsutake was highest in TMM and MYA at $25^{\circ}C$. The highest dry wt. of mycelium was obtained after 65 days of culture, when 6 mycelial discs were inoculated in 100 ml of broth in 250 ml shaking flask. Mycelial mats were observed in clumped condition at the inoculation sites of pine forest after two weeks of inoculation. After 5 months of inoculation, mycelia mats were observed growing inside soil and walls of a few inoculation sites, while mycelial mats growth up to $5{\sim}8$ cm were observed in the roots of pine tree after 6 months. The survival rate of the inoculum was about 40% of the total inoculation sites. The survival rate was found below 20% when the mycelium was inoculated in the summer. The reasons for low survival rates of the mycelium were mainly due to dry season and the soil-borne small animals such as earthworm and mole. After one year of inoculation, no external difference was observed between the artificially inoculated mycelia and the naturally existing mycelia of T. matsutake. The present study showed that fruiting bodies of T. matsutake could be produced by artificial inoculation under the appropriate environmental conditions.
A disease forecast model for bacterial grain rot (BGR) of rice, which is caused by Burkholderia glumae, was developed in this study. The model, which was named 'BGRcast', determined daily conduciveness of weather conditions to epidemic development of BGR and forecasted risk of BGR development. All data that were used to develop and validate the BGRcast model were collected from field observations on disease incidence at Naju, Korea during 1998-2004 and 2010. In this study, we have proposed the environmental conduciveness as a measure of conduciveness of weather conditions for population growth of B. glumae and panicle infection in the field. The BGRcast calculated daily environmental conduciveness, $C_i$, based on daily minimum temperature and daily average relative humidity. With regard to the developmental stages of rice plants, the epidemic development of BGR was divided into three phases, i.e., lag, inoculum build-up and infection phases. Daily average of $C_i$ was calculated for the inoculum build-up phase ($C_{inf}$) and the infection phase ($C_{inc}$). The $C_{inc}$ and $C_{inf}$ were considered environmental conduciveness for the periods of inoculum build-up in association with rice plants and panicle infection during the heading stage, respectively. The BGRcast model was able to forecast actual occurrence of BGR at the probability of 71.4% and its false alarm ratio was 47.6%. With the thresholds of $C_{inc}=0.3$ and $C_{inf}=0.5$, the model was able to provide advisories that could be used to make decisions on whether to spray bactericide at the preand post-heading stage.
