볏짚의 Nafion 가수분해물에 대한 Pichia stipitis CBS 5776의 에탄올 발효 (Ethanol Fermentation of Nafion-Catalyzed Hydrolyzates of Rice Straw by Pichia stipitis CBS 5776)

  • Pichia stipitis CBS 5776을 이용하여 볏짚 가수분해물의 fermentability를 조사하고 가수분해시 고체 강력산인 Nafion과 재래식 황산에 의한 가수분해물을 비교 검토하였다. 볏짚의 Nafion 가수분해물은 산 가수분해물보다 세포 증식 및 발효에 대한 저해 수준이 낮았으며 3.2 g/l 이상의 Pichia stipitis inoculum을 사용하면 Nafion 가수분해물의 발효에서 에탄올 생산이 가능하였다.

  • Four nonpathogenic isolates of Fusarium oxysporum isolated from spinach showed suppressive effect on the occurrence of the Fusarium wilt of spinach caused by F. oxysporum f. sp. sprinaciae, among which NF01 controlled the disease most effectively. And NF01 was not pathogenic to tomato, cucumber, radish and spinach. This isolate was further tested for the biological control of the disease. The isolate was not inhibitory to the growth of the pathogen on potato sucrose agar medium, however the Fusarium wilt disease occurred less by drenching spore suspension of the nonpathogenic isolate. The control effect of the isolate was higher at lower inoculum level of the pathogen than at the higher inoculum level, and in the pretreatments than the simultaneous treatment of the isolate with the pathogen inoculation. The nit mutants of the isolate were easily formed on chlorate containing media, and was reisolated selectively as nit mutant from infected soil and plants. The reisolation rate of the isolate as opposed to pathogen was high at preinoculated soil and plants relative to the simultaneous inoculation of the isolate with the pathogen.

Morphological Characteristic Regulation of Ligninolytic Enzyme Produced by Trametes polyzona

  • A newly isolated white rot fungal strain KU-RNW027 was identified as Trametes polyzona, based on an analysis of its morphological characteristics and phylogenetic data. Aeration and fungal morphology were important factors which drove strain KU-RNW027 to secrete two different ligninolytic enzymes as manganese peroxidase (MnP) and laccase. Highest activities of MnP and laccase were obtained in a continuous shaking culture at 8 and 47 times higher, respectively, than under static conditions. Strain KU-RNW027 existed as pellets and free form mycelial clumps in submerged cultivation with the pellet form producing more enzymes. Fungal biomass increased with increasing amounts of pellet inoculum while pellet diameter decreased. Strain KU-RNW027 formed terminal chlamydospore-like structures in cultures inoculated with 0.05 g/L as optimal pellet inoculum which resulted in highest enzyme production. Enzyme production efficiency of T. polyzona KU-RNW027 depended on fungal pellet morphology as size, porosity, and formation of chlamydospore-like structures.

Effects of Substrate to Inoculum Ratio on the Biochemical Methane Potential of Piggery Slaughterhouse Wastes

  • The aim of this study was to assess the effect of substrate to inoculum ratio (S/I ratio) on the biochemical methane potential (BMP) and anaerobic biodegradability ($D_{deg}$) of different piggery slaughterhouse wastes, such as piggery blood, intestine residue, and digestive tract content. These wastes were sampled from a piggery slaughterhouse located in Kimje, South Korea. Cumulative methane production curves for the wastes were obtained from the anaerobic batch fermentation having different S/I ratios of 0.1, 0.5, 1.0, and 1.5. BMP and anaerobic biodegradabilities ($D_{deg}$) of the wastes were calculated from cumulative methane production data for the tested conditions. At the lowest S/I ration of 0.1, BMPs of piggery blood, intestine residue, and digestive tract content were determined to be 0.799, 0.848, and $1.076Nm^3kg^{-1}-VS_{added}$, respectively, which were above the theoretical methane potentials of 0.539, 0.644, and $0.517Nm^3kg^{-1}-VS_{added}$ for blood, intestine residue, and digestive tract content, respectively. However, BMPs obtained from the higher S/I ratios of 0.5, 1.0, and 1.5 were within the theoretical range for all three types of waste and were not significantly different for the different S/I ratios tested. Anaerobic biodegradabilities calculated from BMP data showed a similar tendency. These results imply that, for BMP assay in an anaerobic reactor, the S/I ratio of anaerobic reactor should be above 0.1 and the inoculum should be sufficiently stabilized to avoid further degradation during the assay.

담배세균성마름병[립고병(立枯病)]에 대한 담배품종의 저항성 검정법 (Screening Procedure of Tobacco Cultivars for Resistant to Bacterial Wilt Caused by Ralstonia solanacearum)

  • Bacterial wilt caused by Ralstonia solanacearum has become a severe problem on tobacco in Korea. No effective single control measure is available at present time. One of the most potential way for controlling the bacterial wilt on tobacco is growing tobacco cultivars resistant to the bacterial wilt. In this study, optimal conditions for screening tobacco cultivars resistant to the bacterial wilt were examined to provide reproducible and efficient methods in growth chamber testing and field experiments for evaluating plant disease resistance. For this, already-known inoculation methods, inoculum densities, and incubation temperature, and plant growth stages at the time of inoculation were compared using tobacco cultivars resistant (Nicotiana tabacum cv, NC95), moderately resistant (N. tabacum cv. SPG70), and susceptible (N. tabacum BY4) to the bacterial disease. It was determined that root-dipping of tobacco seedlings at six true leaf stage into the bacterial suspension with inoculum level of $10^8$ colony-forming units (CFU)/ml for 20 min before transplanting was simple and most efficient in testing for resistance to the bacterial wilt of tobacco caused by R. solanacearum, for which disease incidences and severities were examined at 2 weeks of plant growth after inoculation at $20{\sim}25^{\circ}C$ in a growth chamber. These experimental conditions could discriminate one tobacco cultivar from the others by disease severity better than any other experimental conditions. In field testing, the optimum time for examining the disease occurrence was late June through early July. These results can be applied to establishing a technical manual for the screening of resistant tobacco cultivars against the bacterial wilt caused by R. solanacearum.

Production of Biomass and Bioactive Compounds from Cell Suspension Cultures of Eurycoma longifolia in Balloon Type Bubble Bioreactors

  • Eurycoma longifolia is an important rare medicinal plant that contains valuable bioactive compounds. In the present study, cell suspension culture of E. longifolia was established for the production of biomass and phenolic compounds. Various medium parameters, such as concentration of auxin, salt strength of the medium, and sucrose and nitrogen concentrations, were optimized for the production of biomass at the flask-scale level. Full strength Murashige and Skoog (MS) medium supplemented with $3.0mg{\cdot}L^{-1}$ naphthaleneacetic acid (NAA), 3% (w/v) sucrose, 0:60 $NH{_4}^+:NO{_3}^-$ was found suitable for biomass accumulation. Based on the optimized flask-scale parameters, cell suspension cultures were established in balloon-type bubble bioreactors, and bioprocess parameters such as inoculum density and aeration rate were optimized. Inoculum density of $50g{\cdot}L^{-1}$ and increasing aeration rate from 0.05 to 0.3 vvm, with increases every 7 days, were suitable for the accumulation of both biomass and phenolic compounds. With the optimized conditions, $14.70g{\cdot}L^{-1}$ dry biomass, $10.33mg{\cdot}g^{-1}$ DW of phenolics and $3.89mg{\cdot}g^{-1}$ DW of flavonoids could be achieved. Phenolics isolated from the cell biomass showed optimal free radical scavenging activity.

근류균 접종제 개발을 위한 우수 증량제의 선발 및 생산 최적조건 (Establishment of some Conditions for the Development of Legume Inoculant)

  • Rhizobium균을 이용하여 근류균 접종제를 생산하기 위한 기초연구로 우수한 증량제(carrier material)를 선발하고, 근류균의 접종량, carrier의 숙성 및 보존온도, 멸균조건등 근류균 접종제 생산을 위한 각종 조건을 확립하기 위하여 실험한 결과는 다음과 같다. 1. 근류균 접종제 생산을 위한 carrier material source로는 퍼라이트 및 토탄이 우수한 것으로 나타났다. 2. 근류균의 접종량(inoculum size)에 따른 경시적 생존율은 접종초기 $10^4cells/g$ 이하의 균주를 접종하여도 1~2주후에는 $5{\times}10^8cells/g$로 증가하여 $10^8cells/g$ 접종구와 비슷한 균수를 나타내었다. 3. Carrier의 숙성 및 보존온도에 따른 근류균의 경시적 생존율은 특별한 보온이나 냉장시설없이 실온($20^{\circ}C$) 보존에서도 $10^9cells/g$의 높은 균수가 12주째까지 유지되었다. 4. Carrier의 멸균방법에 따른 근류균의 경시적 생존율은 가압습열멸균 처리구에서 생존율이 가장 양호하였으나 각 처리에 따른 큰 차이는 볼 수 없었고, 특히 퍼라이트의 경우에는 멸균처리없이도 12주째 까지 $10^9cells/g$ 이상의 높은 균수를 유지하였다.

The Effect of Bacteriocin Produced by Lactobacillus plantarum on the Growth of Listeria monocytogenes

  • The inhibitory effect of Lactobacillus plantarum (Lb. plantarum) which is bacteriocin­producing strain against the growth of Listeria monocytogenes (L. monocytogenes) was examined in trypticase soy broth (TSB). TSB was inoculated with 104 cells/me L. monocytogenes and then with different numbers $(10^6\;10^4\;and\;10^2\;cells/ml)$ of Lb. plantarum. The mixed cultures were incubated at 37, 25 and $4^{\circ}C$. The most effective inhibition of was found at $37^{\circ}C$ and a less inhibition at $25^{\circ}C$. However, there was no significant change in the cell numbers of both L. monocytogenes and Lb. plantarum at $4^{\circ}C$. At same incubation temperature, the higher initial inoculum level of Lb. plantarum, the better inhibitory effect against L. monocytogenes. In addition, TSB was inoculated with L. monocytogenes at different initial inoculum levels of $10^6,\;10^4$ and $10^2$ cells/me and then supplemented with 0, 30, 60 and 100 AU/ml of bacteriocin produced by Lb. plantarum. The mixed cultures were incubated at 37, 25 and $4^{\circ}C$. L. monocytogenes of three different initial inoculum levels began to be inhibited in the presence of more than 60 AU/ml of bacteriocin at $37^{\circ}C$. In TSB containing more than 60 AU/me of bacteriocin and incubated at $25^{\circ}C$, L. monocytogenes decreased by 2 log-units during the period of 12 hrs incubation and thereafter remained steady. At $4^{\circ}C$, L. monocytogenes decreased by 1.5 log-units in the presence of 60 AU/ml bacteriocin during the period of 4 days incubation and dropped to the non-detectable level in TSB with 100 AU/ml bacteriocin.

  • 감자 괴경에 타박상을 입혔거나 세균현탁액을 주입했을 때 낮은 water potential(-6.46 bar) 보다 높은 water potential (-6.06 bar)에서 더 쉽게 부패를 초래하였으며, 높은 water potential을 가진 괴경과 낮은 water potential을 가진 괴경 사이에 있어서의 부패의 진전에는 큰 차이를 발견할 수 있었으며, 현탁액의 농도의 종류에 따른 괴경의 연부병의 이병정도는 높은 water potential에서 높았으며, 세균 현탁액 농도의 $ED_{50}$은 높은 water potential에서 $10^{8.5}\;cells/ml$이며 낮은 water potential에 서는 $100^{9.8}\;cells/ml$이었으며, 높은 water potential과 낮은 water potential $ED_{50}$에서는 작은 차이를 인정할 수 있었다. 이 시험에서 감자는 상처나 타박상이 나지 않도록 다루어야 하며 수확후 충분히 건조하여야 오랜 운반과 저장중에 연부병 발생을 크게 줄일 수 있다는 것을 나타낸다.

감나무 탄저병균(Collectotrichum gloeosporioides)의 월동 전염원 조사 (Survey of Overwintering Inoculum Potential of Anthracnose of Sweet Persimmon Caused by Colletotrichum gloeosporioides)

  • 2006년에서 2007년까지 2년간 자연상태에서 월동후 식물체 부위별 단감 탄저병의 병원균 분리비율을 조사하기 위해 병든 가지의 병반과 눈, 죽은 가지, 잎자루, 잎, 건전한 가지와 눈, 월동한 과실 등에서 조사를 하였다. 4월에 조사한 결과 병든 가지의 병반에서 93.3% 분리되었으며, 병든 가지 눈에서 46.7% 분리되었으며, 죽은 가지에서 36.7% 병원균이 분리되었다. 잎자루에서 23.3% 분리되었으며, 잎에서 16.7% 분리가 되었으며, 낙과된 과실에서 6.7% 분리되었지만 건전한 가지와 눈에서는 병원균이 분리되지 않았다. 그러나 생육중에 가지, 잎, 잎자루, 과실에 발생한 병든 부위에서는 병원균이 100%분리되었다.
