The present study investigated effects of antifungal and carboxylesterase inoculant on rumen fermentation with different rumen pH. Corn silage was treated without inoculant (CON) and with a mixed Lactobacillus brevis 5M2 and L. buchneri 6M1 (MIX). Rumen fluid was collected from two cannulated Hanwoo heifers before morning feeding (high rumen pH at 6.70) and 3 h after feeding (low rumen pH at 6.20). Dried corn silage was incubated in the rumen buffer (rumen fluid + anaerobic culture medium at 1:2 ratio) for 48 h at 39℃. Eight replications for each treatment were used along with two blanks. Both in a high and a low rumen pH, MIX silages presented higher (p<0.05) the immediately degradable fraction, the potentially degradable fraction, total degradable fraction, and total volatile fatty acid (VFA) than those of CON silages. Incubated corn silages in a low rumen pH presented lower (p<0.05) total degradable fraction, ammonia-N, total VFA (p=0.061), and other VFA profiles except acetate and propionate, than those in a high rumen pH. The present study concluded that application of antifungal and carboxylesterase inoculant on corn silage could improve degradation kinetics and fermentation indices in the rumen with high and low pH conditions.

'Ihis experiment was carried out to select a carrier as rhizobium inoculant for alfalfa. The Rhizobium inoculants were produced in main materials(organic and inorganic materials) and submaterial(Ca$CO_3$, and sucrose), and than the changes of Rhizobium population of inoculants were scrutinized from directly after the production to 12 months at $4^{\circ}$C and $25^{\circ}$C. Directly after the production of inoculants, their Rhizobium numbers were 4-9 $\times$$10^9$/ginoculant in organic materials and 1-8 $\times$$10^8$/g inoculant in inorganic materials. When they were stored at $25^{\circ}$C, most of inoculants was being started in reduction of rhizobium numbers after 2 months and was reduced by tenfold of initial rhizobium number after 4-6 months, but inoculant of bentonite+ vermiculite+sucrose 1% was not reduced by tenfold after 12 months. When they were stored at $4^{\circ}$C, most of inoculants was being started in reduction of rhizobium numbers after 4 months but was not reduced by tenfold of initial rhizobium number after 12 months, and the effect of sucrose addition to bentonite+ vermiculik did nearly not appear. Preservation of Rhizobium inoculants was more effective at 4C than 25 C . 'Ihe Rhizobium inoalant produced in bentonite+vermiculite(l:3 w/w)+ sucrose 1% was not only high Rhizobium population(8.3 $\times$$10^8$/g inoculant) but also survival effect of rhizobium in being stored at $25^{\circ}$C.
The improvement of the fermentation quality of rice straw silage by application of lactic acid bacteria (LAB) and glucose was investigated in this study. Sixteen rice varieties were harvested at the maturity stage and the rice straw was ensiled with LAB inoculant ($1{\times}10^5$ cfu/g of fresh weight) and glucose (2% of fresh weight). Inoculation with LAB improved the fermentation as reflected in reductions in pH, acetic acid (by 3.7 to 78.3%), butyric acid (by -6.0 to 100.0%) and ammonia nitrogen (by 1.0 to 71.7%) concentrations, and increases in lactic acid (by 43.9 to 282.9%) and crude protein concentrations compared with the control. Application of LAB plus glucose was more effective in improving fermentation quality than LAB alone. The variety of rice straw which contained relatively high levels of water soluble carbohydrates (WSC) tended to obtain better fermentation quality. The results suggested that LAB application and selection of rice varieties whose straw contained high levels of WSC were effective in improvement of the fermentation quality of rice straw silage.

The effects of endophytic microbial inoculation and temperature on the phenolic content of tartary buckwheat (TP) sprouts were investigated. TP seeds were inoculated with Herbaspirillum spp. at concentrations (%v/v) of 0 (control), 10, 20, and 40% at 20, 25, and $30^{\circ}C$ in a growth chamber for seven days. It was observed that the phenolic content (PC) including flavonoid, rutin, and tanin increased with an increase in inoculant rate at $20^{\circ}C$, whereas the PC content increased with an increase in temperature regardless of the inoculant rate. Therefore, it is suggested that increasing the inoculant rate is effective at achieving higher phenolic contents when plants are grown at lower temperatures.

Objective: This study was conducted to confirm the effects of new inoculants producing-antifungal or esterase substances on rye silage and its rumen fermentation indices by comparing wild with mutated types. Methods: Rye harvested at dough stage was ensiled into 3 L mini bucket silo (1 kg) for 90 d in triplicate following: distilled water at 20 μL/g (CON); Lactobacillus brevis 100D8 (AT) and its inactivation of antifungal genes (AT-m) at 1.2×105 cfu/g, respectively; and Leuconostoc holzapfelii 5H4 (FD) and its inactivation of esterase genes (FD-est) at 1.0×105 cfu/g, respectively. After silo opened, silage was sub-sampled for the analysis of ensiling quality and its rumen fermentation indices. Results: Among the wild type inoculants (CON vs AT vs FD), FD inoculant had higher (p<0.05) in vitro digestibilities of dry matter and neutral detergent fiber, the total degradable fraction, and total volatile fatty acid in rumen, while AT inoculant had higher (p<0.05) lactate, acetate, and lactic acid bacteria in silage. Silage pH and the potentially degradable fraction in rumen increased (p<0.05) by inactivation of antifungal activity (AT vs AT-m), but lactate, acetate, and lactic acid bacteria of silage decreased (p<0.05). In silage, acetate increased (p<0.05) by inactivation of esterase activity (FD vs FD-est) with decreases (p<0.05) of pH, ammonia-N, lactate, and yeast. Moreover, inactivation of esterase activity clearly decreased (p<0.05) in vitro digestibilities of dry matter and neutral detergent fiber, the total degradable fraction, and total volatile fatty acid in the rumen. Conclusion: This study concluded that FD inoculant confirmed esterase activity on rye silage harvested at dough stage, while AT inoculant could not be confirmed with antifungal activity due to the absence of mold in all silages.
For the purpose of developing a high quality agricultural microbial inoculant, methods and materials for improving encapsulation were investigated. Preparation of capsule was conducted by improving extrusion system with micro-nozzle and peristaltic pump. The sodium alginate was selected because of its cheapness, stability of cells, and gel formation ability. The yields, physical properties and gel formation abilities of extractable alginate from sea tangle were investigated by hot water extractable and alkali soluble methods. The extraction yields of hot water extractable alginate (HWEA) and alkali soluble alginate (ASA) from sea tangle were 8 and 20%, respectively. The HWEA was almost not viscous even in 1.5% of the sample solution, whereas the ASA was very highly viscous in above 3% sample solution. The gel formation ability of each samples varied from 1.5% to 5% and the ASA showed a good gel formation ability at 3% solution as commercial alginate (CA). The soil microbial inoculant, Bacillus thuringiensis, Bacillus subtilis, Lactobacillus plantarum and Geotrichum candidum encapsulated sodium alginate with starch and zeolite for stabilizer. The survivability of encapsulated soil microbial inoculant using alginate without stabilizer appeared to be 66, 52, 70 and 50%, respectively. Inclusion of starch and zeolite with alginate bead increased viabilities in Bacillus sp. and Geotrichum candidum by 81-83% and 89%, respectively.
The effect of sulfur content and inoculant on solidification microstructure and mechanical properties of gray cast iron have been investigated. The main results obtained are as follows, 1. In the FeSi3Ca1Ba inoculated irons, with the variation of sulfur content, low sulfur levels (${\sim}0.03%$) yield low chill depth, high tensile strength, good wear resistance and type A graphite with a pearlite matrix. High sulfur levels( >0.08%) provide high chill depth, low mechanical proper ties and type D graphite with small amount ferrite. 2. In case of inoculant variation with normal FeSi, FeSi3Ca1Ba, 30CaSi and uninoculation at 0.03%S level, lower chill depth and higher tensile strength was obtained in the order ; 30CaSi, FeSi3Ca1Ba normal FeSi, uninoculation.
The abilities and fading behaviors of 5 different inoculants were evaluated by the thermal analyses of the melts which were held at constant temperature after addition in induction furnace. The silicon content was preserved at the high holding temperature of 1,450${\sim}$1,500$^{\circ}C$, which reduced by oxidation at the low one. The inoculation temperature should be higher to obtain better and reproducible result. The inoculation performance of 1%Ba-Fe-Si was the best on the bases of effect and fading behavior.
