  • Pancreatic lipase-inhibitory activity of the rhizome of Alpinia officinarum (AO) and its antihyperlipidemic activity were measured. When water extract of AO was stepwise fractionated with organic solvents, ethylacetate fraction exhibited the most potent inhibition. From it, Diarylheptanoid was isolated as an inhibitor of pancreatic lipase and we investigated its in vitro inhibitory effect of lipase activity and in vivo antihyperlipidemic effect. (omitted)

The Stydy on Application to Cosmetics of Phellinus Iimteus

  • Phellinus linteus was artificially cultivated in kangwon province in Korea. The air-dried phellinus linteus was frozen in liquid nitrogen tank and powdered in jar. 10g of the powder was extracted with each 200g of ethanol, methanol, distilled water and 1,3-butylene glycol/distilled water 4 hours under refluxing and then the liquidextract was concentrated under reduced pressure. As a result of analysis by high performance liquid chromatography and thin layer chromarography, many kinds of sugar and flavonoids were detected. Also we knew that phellinus linteus' extract had a strong UV-ray absorption. In the efficacy test for applying to cosmetics, free radical scavenging effect was confirmed. As a result, 2% of sample was the most potent inhibitory effect and the free radical savenging activity, was 0.31%. This is more effective than any other meterial. In the test of antioxidative activity against lipid autoxidation, phellinus linteus' extract had a good effect by 46% while vitamine E was 42.3%. The immunological activity of phellinus linteus was showed through the activation of macrophage cell. Actually, phellinus linteus activated macrophage function of 1.1-1.8 times including nitrite production compared to control. The whitening effect of phellinus linteus was showed through the inhibition of tyrosinase activity, melanin biosynthesis of S. bikiniensis and B-16 melanoma cells. Phellinus linteus' extract was showed strong mushroom tyrosinase inhibitory activity with IC50 value of 0.5% and inhibited melanin biosynthesis with 28mm inhibition zone at 0.005%/paper disc in S. bikinniensis, a bacterium used as an indicator organism in this work. Also it inhibited melanin biosynthesis in B-16 melanoma cells with a minimum inhibitory concentration of 0.134%.

Inhibitory Effects of Scutellaria barbata D. Don on the Cell Proliferation of HeLa cells (반지연(半枝蓮)이 HeLa Cell의 증식억제(增殖抑制)와 사멸(死滅)에 미치는 영향(影響))

  • Purpose : This study was conducted to investigate the inhibitory effects of Scutellaria barbata D. D on on the cell proliferation of HeLa Cells. Methods : Human uterine cervical carcinoma HeLa cells were cultured in the 1%, 5% and 10% concentration of Scutellaria barbata D. D on solution for 24, 48 and 72 hours for the direct inhibitory effects of Scutellaria barbata D. D on. Then we examined the effect of Scutellaria barbata D. D on solution on the cell proliferation inhibition by XTT assay. DNA fragmentation, MAP kinase activity and caspase activity by FACS analysis in HeLa cells. Results : We found that the proliferation of HeLa cells was significantly decreased in Scutellaria barbata D. D on solution containing groups comparing with a control group in a concentration-dependant manner. When HeLa cells were cultivated for 24 hours with 5% Scutellaria barbata D. D on solution containing group, the percentage of HeLa cells with activated caspase was the highest. Scutellaria barbata D. D on solution reduced the MAP kinase activity of HeLa cells comparing with the control group. By the XTT assay, the cell's activity was decreased in 5% and 10% Scutellaria barbata D. D on solution containing groups in 24 and 72 hours cultivation and 10% group in 48 hours. DNA fragmentation and caspase-3 activity of HeLa cells, however, were changed insignificantly. Conclusion : From this study we could suggest that Scutellaria barbata D. D on is available to the inhibition and apoptosis of human cervical carcinoma cell line, HeLa cells in vitro.

Antimicrobial Activity of Water Soluble Propolis (수용성 프로폴리스의 항균성)

  • In this study, the minimum inhibition concentration(MIC), growth inhibition activity, and colony forming inhibitory activity of water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae and Salmonella enteritidis were tested. The MICs of the water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 312.5 ppm, below 156.3 ppm, 625 ppm, 10,000 ppm, above 10,000 ppm, 10,000 ppm, above 10,000 ppm, above 10,000 ppm, 10,000 ppm, and above 10,000 ppm, respectively. The growth inhibition concentrations against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, and Klebsiella pneumoniae were 156.3 ppm, below 156.3 ppm, 625 ppm, 5,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, and 5,000 ppm, respectively. However, 10,000 ppm did not inhibit the growth of Salmonella enteritidis. Finally, the colony forming inhibitory activities against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 98.0%, 99.8%, 69.8%, 98.1%, 62.0%, 63.1%, 79.5%, 61.9%, 79.6%, and 0.0%, respectively.

P-Glycoprotein Inhibitory Activity of Indonesian Medicinal Plants in Human Breast Cancer Cells

  • In order to examine their effects on the P-glycoprotein (P-gp) activity in human breast cancer cells, MCF-7/ADR, one hundred Indonesian plant extracts were screened. Among them, the five chloroform extracts of Calotropis gigantea, Curcuma aeruginosa, Merremia mammosa, Sindora sumatrana, and Zingiber cassumunar, showed the most potent P-gp inhibitory activity. When each of these extracts was treated together with the anticancer agent, daunomycin, they increased the cytotoxic activity of daunomycin up to $IC_{50}$ values of less than $6.62\;{\mu}M$, which is a value with a positive control, verapamil. Also, other 15 plant extracts exhibited significant P-gp inhibitory activity with $IC_{50}$ values between 6.62 and $13.20\;{\mu}M$. These prospective samples will be subjected to further laboratory phytochemical investigation to find active principles.

  • In our continuing effort to investigate compounds having anti-inflammatory activity from natural products, Ailanthus altissima was examined. Among six compounds isolated from Ailanthus altissima, Luteolin-7-O-glucoside (L7G) along with ethanol extract of Ailnathus altissima (EAa) were chosen to determine their inhibitory activity on secretory recombinant phospholipase $A_2s$ enzyme activity in vitro. As a results, EAa inhibited human recombinant $sPLA_2-V$ ($IC_{50}$ of about 100 ${\mu}g/ml$) and $cPLA_2$, ($IC_{50}$ of about 59 ${\mu}g/ml$), while L7G showed strong inhibitory effect on $sPLA_2-A$, V and $cPLA_2$ with an $IC_{50}$ value of approximately 40 ${\mu}M$, respectively.

Antimicrobial efficacy of endophytic Penicillium purpurogenum ED76 against clinical pathogens and its possible mode of action

  • This study was aimed to evaluate the antimicrobial activity of Penicillium purpurogenum ED76 on several clinically important microorganisms. The endophytic fungus P. purpurogenum ED76 was previously isolated from Swietenia macrophylla leaf. The antimicrobial efficacy of P. purpurogenum ED76 dichloromethane extract was determined via disc diffusion and broth microdilution assay. A kill curve study was conducted and the morphology of extract treated bacterial cells were viewed under scanning electron microscope. The dichloromethane extract showed significant inhibitory activity on 4 test bacteria and 2 test yeasts. The minimal inhibitory concentration of the extract ranged from 125 to $1,000{\mu}g/ml$, which indicates the different susceptibility levels of the test microorganisms to the fungal extract. The kill curve study has revealed a concentration-dependent inhibition for all test microorganisms. With the increase of the extract concentration, the microbial growth was significantly reduced. The scanning electron micrograph of dichloromethane extract-treated Staphylococcus aureus cells showed the total damage of the cells. The cell wall invagination of the bacterial cells also indicates the loss of cellular materials and metabolic activity. The gas chromatography mass spectrometry analysis of the extract also showed that the major compound was stigmasterol, which constitutes 45.30% of the total area. The dichloromethane extract of P. purpurogenum ED76 exhibited significant inhibitory activity on several clinically important bacteria and yeasts. The study proposed a possible mode of action that the extract cause significant damage to the morphology of S. aureus cells.

Biological Analysis of Enzymatic Extracts from Sargassum fulvellum Using Polysaccharide Degrading Enzyme (Polysaccharide Degrading Enzyme을 이용한 참모자반 효소분해 추출물의 생리활성 연구)

  • SC092 strain, producing a polysaccharide degrading enzyme, was isolated from the seawater. This strain was identified as Microbulbifer sp. using the comparative sequence analysis against known 16S rRNA sequence. A polysaccharide degrading enzyme from this strain was used to acquire the enzymatic extracts of Sargassum fulvellum. DPPH radical scavenging and SOD activity of the enzyme extracts of S. fulvellum were about 61.9% and 82.9% at 2 mg/mL, respectively. Nitrite scavenging activities was 52.5% at 2 mg/mL on pH 1.2. In addition, ${\alpha}$-glucosidase inhibitory activity was also increased in a dose-dependent manner and was about 52.7% at 2 mg/mL. To determine the influence of enzyme extracts of S. fulvellum on alcohol metabolism, the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. ADH and ALDH activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}$-glucosidase inhibitory activity of enzyme extracts of S. fulvellum was remarkably increased in a dose-dependent manner and was about 52.7% at 2 mg/mL. These results indicate alcoholizing and ${\alpha}$-glucosidase inhibitory activities can be enhanced by the enzymatic extracts of S. fulvellum.

3D-QSAR of Angiotensin-Converting Enzyme Inhibitors: Functional Group Interaction Energy Descriptors for Quantitative Structure-Activity Relationships Study of ACE Inhibitors

  • A new set of functional group interaction energy descriptors relevant to the ACE (Angiotensin-Converting Enzyme) inhibitory peptide, QSAR (Quantitative Structure Activity Relationships), is presented. The functional group interaction energies approximate the charged interactions and distances between functional groups in molecules. The effective energies of the computationally derived geometries are useful parameters for deriving 3D-QSAR models, especially in the absence of experimentally known active site conformation. ACE is a regulatory zinc protease in the renin-angiotensin system. Therapeutic inhibition of this enzyme has proven to be a very effective treatment for the management of hypertension. The non bond interaction energy values among functional groups of six-feature of ACE inhibitory peptides were used as descriptor terms and analyzed for multivariate correlation with ACE inhibition activity. The functional group interaction energy descriptors used in the regression analysis were obtained by a series of inhibitor structures derived from molecular mechanics and semi-empirical calculations. The descriptors calculated using electrostatic and steric fields from the precisely defined functional group were sufficient to explain the biological activity of inhibitor. Application of the descriptors to the inhibition of ACE indicates that the derived QSAR has good predicting ability and provides insight into the mechanism of enzyme inhibition. The method, functional group interaction energy analysis, is expected to be applicable to predict enzyme inhibitory activity of the rationally designed inhibitors.

  • Minimum inhibition concentration(MIC), growth inhibition activity, and colony forming inhibitory activity of grapefruit seed extract against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Escherichia coli, Salmonella enterifidis and Serratia marcescens were tested. MIC of grapefruit seed extract against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Escherichia coli, Salmonella enteritidis and Serratia marcescens was 12.5, 12.5, 12.5, 50, 50, 100ppm, respectively. Growth inhibition concentration of grapefruit seed extract against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Escherichia coli, Salmonella enteritidis and Serratia marcescens was below 1.0, 6.25, below 1.0, 6.25, 25, 25ppm, respectively. Colony forming inhibitory activity of grapefruit seed extract against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Escherichia coli, Salmonella enteritidis and Serratia marcescens was 93.9, 94.0, 99.9, 4.4, 82.7, 86.4%, respectively. Colony forming inhibitory activities of grapefruit seed extract against Gram positive bacteria were higher than that against Gram negative bacteria.
