Systemic Analysis of Antibacterial and Pharmacological Functions of Anisi Stellati Fructus (대회향의 시스템 약리학적 분석과 항균작용)

  • The purpose of this study was to acquire the active compounds of Anisi stellati fructus (ASF) and to analyze the genes and diseases it targets, focusing on its antibacterial effects using a system pharmacological analysis approach. Active compounds of ASF were obtained through the Traditional Chinese Medicine Systems Pharmacology (TCMSP) Database and Analysis Platform. This contains the pharmacokinetic properties of active compounds and related drug-target-disease networks, which is a breakthrough in silico approach possible at the network level. Gene information of targets was gathered from the UnitProt Database, and gene ontology analysis was performed using the David 6.8 Gene Functional Classification Tool. A total of 201 target genes were collected, which corresponded to the nine screened active compounds, and 47 genes were found to act on biological processes related to antimicrobial activity. The representative active compounds involved in antibacterial action were luteolin, kaempferol, and quercetin. Among their targets, Chemokine ligand2, Interleukin-10, Interleukin-6, and Tumor Necrosis Factor were associated with more than three antimicrobial biological processes. This study has provided accurate evidence while saving time and effort to select future laboratory research materials. The data obtained has provided important data for infection prevention and treatment strategies.

  • Advanced persistent threat (APT) attacks are attacks aimed at a particular entity as a set of latent and persistent computer hacking processes. These APT attacks are usually carried out through various methods, including spam mail and disguised banner advertising. The same name is also used for files, since most of them are distributed via spam mail disguised as invoices, shipment documents, and purchase orders. In addition, such Infostealer attacks were the most frequently discovered malicious code in the first week of February 2021. CDR is a 'Content Disarm & Reconstruction' technology that can prevent the risk of malware infection by removing potential security threats from files and recombining them into safe files. Gartner, a global IT advisory organization, recommends CDR as a solution to attacks in the form of attachments. There is a program using CDR techniques released as open source is called 'Dangerzone'. The program supports the extension of most document files, but does not support the extension of HWP files that are widely used in Korea. In addition, Gmail blocks malicious URLs first, but it does not block malicious URLs in mail systems such as Naver and Daum, so malicious URLs can be easily distributed. Based on this problem, we developed a 'Dangerzone' program that supports the HWP extension to prevent APT attacks, and a Chrome extension that performs URL checking in Naver and Daum mail and blocking banner ads.

Effect of the Application of Hand Washing Education Program for the Children with Intellectual Disability on Hand Washing Frequency and Methods during COVID-19 Pandemic (코로나-19 확산 속에서 손 씻기 교육 프로그램 적용이 지적장애 아동들의 손 씻기 빈도와 방법에 미치는 영향)

  • The purpose of this study is to analyze the effect of the application of hand washing education program for the children with intellectual disability on hand washing frequency and methods during COVID-19 Pandemic. Subjects were 14 children with intellectual disability and they participated in the program using visual video materials. Hand washing education program was consisted of introduction, theory education, and practice and it performed 2 times a week, total 16 sessions during 8 weeks. Hand washing theory education was consisted about the importance and application of hand washing, proper hand washing steps and methods, use of hand washing tools, use of hand sanitizers including characteristics, and prevention of skin damage from hand washing. Hand washing assessment was used by hand washing frequence and methods observation report. Hand washing frequency was measured by the hand washing frequency during the daily routine and it was analyzed by the items divided into day and month units. Hand washing methods was measured by the items divided into use of hand washing tools, time, areas, drying methods after hand washing. As the results, after hand washing education program, the hand washing frequence was increased and hand washing methods were improved. Hand washing is much important as a active method to prevent the infection during the COVID-19 Pandemic. Thus, based on this findings, the application of hand washing education program should be considered to increase the hand washing frequency and improve the hand washing methods of the children with intellectual disability.

Microdroplet Impact Dynamics at Very High Velocity on Face Masks for COVID-19 Protection (코로나-19 보호용 페이스 마스크에서의 액적 고속 충돌 거동)

  • Facial masks have become indispensable in daily life to prevent infection and spread through respiratory droplets in the era of the corona pandemic. To understand how effective two different types of masks (i.e., KF-94 mask and dental mask) are in blocking respiratory droplets, i) we preferentially analyze wettability characteristics (e.g., contact angle and contact angle hysteresis) of filters consisting of each mask, and ii) subsequently observe the dynamic behaviors of microdroplets impacting at high velocities on the filter surfaces. Different wetting properties (i.e., hydrophobicity and hydrophilicity) are found to exhibit depending on the constituent materials and pore sizes of each filter. In addition, the pneumatic conditions for stably and uniformly dispensing microdroplets with a certain volume and impacting behaviors associated with the impacting velocity and filter type change are systematically explored. Three distinctive dynamics (i.e., no penetration, capture, and penetration) after droplet impacting are observed depending on the type of filter constituting the masks and droplet impact velocity. The present experimental results not only provide very useful information in designing of face masks for prevention of transmission of infectious respiratory diseases, but also are helpful for academic researches on droplet impacts on various porous surfaces.

Critical Review and Alternatives to the Decriminalization of Tattooing (문신시술의 비범죄화에 대한 비판적 검토와 대안)

  • South Korean law strictly prohibits engagement in medical activities by non-medical practitioners. In the country, tattooing is classified as a medical practice, and non-medical practitioners who engage in it are penalized because they are unauthorized to carry out this procedure. In reality, however, people rarely seek tattooing services from medical personnel. Arguing that their freedom of job selection is violated, non-medical personnel who make a living as tattoo artists reject the characterization of the procedure as a form of medical treatment and demand the decriminalization of tattooing by non-medical practitioners. Nevertheless, tattooing can cause health- and hygiene-related dangers when it is not performed by medical professionals because it involves penetration into the skin using needles. Hence, stringent management is necessary for infection prevention. The gap between reality and the law gives rise to the need for proactive thinking about the institutionalization of tattoo practice by non-medical personnel. Policymakers should reflect on the fact that only minimal tattooing services are currently performed by medical staff while also accounting for health and safety. On this basis, this study examined tattoo-related legislation in South Korea to determine whether the procedure corresponds to medical practice and identify ways to solve problems that occur from the perspective of health care. As a response that promotes safety and reflects reality, this research proposed a three-phase approach.

20 (S)-ginsenoside Rh2 inhibits colorectal cancer cell growth by suppressing the Axl signaling pathway in vitro and in vivo

  • Background: Colorectal cancer (CRC) has a high morbidity and mortality worldwide. 20 (S)-ginsenoside Rh2 (G-Rh2) is a natural compound extracted from ginseng, which exhibits anticancer effects in many cancer types. In this study, we demonstrated the effect and underlying molecular mechanism of G-Rh2 in CRC cells in vitro and in vivo. Methods: Cell proliferation, migration, invasion, apoptosis, cell cycle, and western blot assays were performed to evaluate the effect of G-Rh2 on CRC cells. In vitro pull-down assay was used to verify the interaction between G-Rh2 and Axl. Transfection and infection experiments were used to explore the function of Axl in CRC cells. CRC xenograft models were used to further investigate the effect of Axl knockdown and G-Rh2 on tumor growth in vivo. Results: G-Rh2 significantly inhibited proliferation, migration, and invasion, and induced apoptosis and G0/G1 phase cell cycle arrest in CRC cell lines. G-Rh2 directly binds to Axl and inhibits the Axl signaling pathway in CRC cells. Knockdown of Axl suppressed the growth, migration and invasion ability of CRC cells in vitro and xenograft tumor growth in vivo, whereas overexpression of Axl promoted the growth, migration, and invasion ability of CRC cells. Moreover, G-Rh2 significantly suppressed CRC xenograft tumor growth by inhibiting Axl signaling with no obvious toxicity to nude mice. Conclusion: Our results indicate that G-Rh2 exerts anticancer activity in vitro and in vivo by suppressing the Axl signaling pathway. G-Rh2 is a promising candidate for CRC prevention and treatment.

Discharge Analysis of Chungcheongbuk-do Residents using National Hospital Discharge In-depth Injury Survey in the Recent 5 Years (퇴원손상심층조사 자료를 이용한 최근 5년간의 충청북도 거주민의 퇴원 분석)

  • This study was performed to generate basic data to establish a health promotion plan for residents of Chungcheongbuk-do by identifying characteristics of discharged patients residing in the Chungcheongbuk-do area from an In-Depth Post-Discharge Injury Survey reported by the Korea Centers for Disease Control and Prevention(KCDCP). The Report provided data on demographic characteristics, medical institution use characteristics, medical use characteristics, and disease characteristics of patients discharged from medical institutions with 100 or more beds from 2013 to 2017. The total number of Chungcheongbuk-do residents who were admitted and discharged from 2013 to 2017 was estimated to be 1,656,590, and the discharge rate was 21,089, which was higher than the national average of 13,882 in 2016. The regions where the discharge rate increased during this period include Goesan, Yeongdong, Boeun, Okcheon, Jeungpyeong, and Eumseong-gun, which are mainly rural areas. Among the patients hospitalized and discharged from hospitals outside the Chungcheongbuk-do area, the discharge rate of patients who used hospitals in Incheon/Gyeonggi areas and Daejeon/Chungnam areas increased slightly. Among the malignant tumor patients, the number of lung cancer(included trachea & bronchial cancer) patients was the highest. In addition, the discharge rate was highest for patients with respiratory diseases. This study suggests that efforts need to be made to lower the discharge rate for infection, circulatory disease, genitourinary disease, and musculoskeletal disorder patients

  • To adopt the development in the medical scenario IoT developed towards the advancement with the processing of a large amount of medical data defined as an Internet of Medical Things (IoMT). The vast range of collected medical data is stored in the cloud in the structured manner to process the collected healthcare data. However, it is difficult to handle the huge volume of the healthcare data so it is necessary to develop an appropriate scheme for the healthcare structured data. In this paper, a machine learning mode for processing the structured heath care data collected from the IoMT is suggested. To process the vast range of healthcare data, this paper proposed an MTGPLSTM model for the processing of the medical data. The proposed model integrates the linear regression model for the processing of healthcare information. With the developed model outlier model is implemented based on the FinTech model for the evaluation and prediction of the COVID-19 healthcare dataset collected from the IoMT. The proposed MTGPLSTM model comprises of the regression model to predict and evaluate the planning scheme for the prevention of the infection spreading. The developed model performance is evaluated based on the consideration of the different classifiers such as LR, SVR, RFR, LSTM and the proposed MTGPLSTM model and the different size of data as 1GB, 2GB and 3GB is mainly concerned. The comparative analysis expressed that the proposed MTGPLSTM model achieves ~4% reduced MAPE and RMSE value for the worldwide data; in case of china minimal MAPE value of 0.97 is achieved which is ~ 6% minimal than the existing classifier leads.

Epidemiological Aspects and Trends of the Extensive Eradication Programs for Bovine Brucellosis Outbreaks-Associated Human Brucellosis in Korea, 2002~2013

  • Over the last decade, human brucellosis (HB) has become a principal zoonosis in Korea. The objective of this study was observing the epidemiological trends of the extensive eradication programs for bovine brucellosis (BB) outbreaks-associated HB in Korea from 2002 to 2013. The raw data analysis in this study was obtained from the website of brucellosis outbreaks by Korea Center for Disease Control and Prevention (KCDC) and Animal and Plant Quarantine Agency (QIA), Korea, 2002~2013. The number of cases of BB outbreaks-associated HB in Korea was increased after 2003 and peaked in 2006 before decreasing thereafter. There were a total of 89,240 infected BB in domestic cattle with a cumulative incidence rate (CIR) of 611.8 per 100,000 cattle; during the same period there were a total of 703 cases of HB with a CIR of 0.12 per 100,000 persons. Moreover, the correlation coefficient of brucellosis outbreaks between cattle and human was highly significant (r=+0.985). The attack ratio of HB was 7.88 per 1,000 BB cases. All of the control measures of HB were applied in the extensive eradication programs for brucellosis outbreaks of infection source in domestic animals and contributed significantly to the reduction in the outbreaks of HB in Korea.

Sustained release of alginate hydrogel containing antimicrobial peptide Chol-37(F34-R) in vitro and its effect on wound healing in murine model of Pseudomonas aeruginosa infection

  • Background: Antibiotic resistance is a significant public health concern around the globe. Antimicrobial peptides exhibit broad-spectrum and efficient antibacterial activity with an added advantage of low drug resistance. The higher water content and 3D network structure of the hydrogels are beneficial for maintaining antimicrobial peptide activity and help to prevent degradation. The antimicrobial peptide released from hydrogels also hasten the local wound healing by promoting epithelial tissue regeneration and granulation tissue formation. Objective: This study aimed at developing sodium alginate based hydrogel loaded with a novel antimicrobial peptide Chol-37(F34-R) and to investigate the characteristics in vitro and in vivo as an alternative antibacterial wound dressing to treat infectious wounds. Methods: Hydrogels were developed and optimized by varying the concentrations of crosslinkers and subjected to various characterization tests like cross-sectional morphology, swelling index, percent water contents, water retention ratio, drug release and antibacterial activity in vitro, and Pseudomonas aeruginosa infected wound mice model in vivo. Results: The results indicated that the hydrogel C proved superior in terms of cross-sectional morphology having uniformly sized interconnected pores, a good swelling index, with the capacity to retain a higher quantity of water. Furthermore, the optimized hydrogel has been found to exert a significant antimicrobial activity against bacteria and was also found to prevent bacterial infiltration into the wound site due to forming an impermeable barrier between the wound bed and external environment. The optimized hydrogel was found to significantly hasten skin regeneration in animal models when compared to other treatments in addition to strong inhibitory effect on the release of pro-inflammatory cytokines (interleukin-1β and tumor necrosis factor-α). Conclusions: Our results suggest that sodium alginate -based hydrogels loaded with Chol-37(F34-R) hold the potential to be used as an alternative to conventional antibiotics in treating infectious skin wounds.
