Oxidative Potential of Some Endophytic Fungi Using 1-Indanone as Substrate

  • The oxidative potential of the fungus Penicillium brasilianum, a strain isolated as endophytic from a Meliaceae plant (Melia azedarach), was investigated using 1-indanone as substrate to track the production of monooxygenases. The fungus produced the dihydrocoumarin from 1-indanone with the classical Baeyer-Villiger reaction regiochemistry, and (-)-(R)-3-hydroxy-1-indanone with 78% ee. Minor compounds that had resulted from lipase and SAM activities were also detected. The biotransformation procedures were also applied using a collection of Penicillium and Aspergillus fungi obtained from M. azedarach and Murraya paniculata. The results showed that Baeyer-Villiger were mostly active in fungi isolated from M. azedarach. Almost all fungi tested produced 3-hydroxy-1-indanone.

Spectrophotometric Determination of Platinum (IV) with 2-Oximino-1-indanone (2-Oximino-1-indanone을 이용한 Pt(IV) ion의 정량에 관한 연구)

  • A method is described for the spectrophotometric determination of platinum (IV) with 2-oximino-1-indanone based on solvent extraction of Pt:2-oximino-1-indanone complex. The 2-oximino-1-indanone reacted with Pt(IV) to form a dark-orange complex which shows a characterisic maximum absorption at 342nm. The optimum PH for the platinum extraction lies between 5.4~8.0. Beer's law obeys up to 0.98-16.3ppm of platinum (IV) and the molar absorption coefficient is $1.06{\times}10^{-4}L.mol^{-1}.cm^{-1}$. The relative standard deviation of the method was $\times2.1%$. The composition of the complex is estimated to be Pt : In= 1 : 1, by the mole ratio method and ion exchange resin experiment. The optimum condition for the determination of platinum has been studied in detail. The 2-oximin-1-indanone is found to be a selectivereagent for the determination of platinum, since the synthesixed 2-oximino-1-indanone did not react with other metals such as cobalt, cadmium, copper, manganese nickel, iron, lead and zinc, to form the complex. In this studies, we have also clarified Sindhwani and Singh's spectrophotometric determination data of various metals with acenaphthenequinone monooxime (Talanta 20,248, 1973), whose results were not correct.

Beckmann Rearrangements of 1-Indanone Oxime Derivatives Using Aluminum Chloride and Mechanistic Considerations

  • Hydrocarbostyril, which is a key intermediate in our new synthetic route to 6-nitroquipazine, can be prepared from 1-indanone oxime by Beckmann rearrangement. We have optimized the reaction by using a Lewis acid, aluminum chloride,in the yield of 91% instead of common acids such as polyphosphoric acid,and sulfuric acid used in conventional Beckmann rearrangement (20% in the literature, 10% in our experiment).The optimized condition is established by usingthree equivalents of aluminum chloride in CH2Cl2 at -40 $^{\circ}C$ - room tempera-turefor 40 min. We have applied this condition to other 1-indanone derivatives, such as 4-methyl-, 4-methoxy-, 4-nitro and 6-nitro-1-indanones. The mechanism ofthis BR has been proposed on the basis of the effect of tem-perature and substituenton product ratio, with the aid of PM3 calculation for a model system.

  • 1-Amino-5,6-dimethoxyindan hydrochloride was synthesized from 3- (3,4-dirnethoxyphenyl)propionic acid by intramolecular Friedel-Crafts acylation, oximation with hydroxylamine, and reduction with an overall yield of 74%. 2-Amino-5,6-dimethoxyindan hydrochloride was synthesized from 3-(3,4-dirnethoxyphenyl)propionic acid by intramolecular Friedel-Crafts acylation, oximation with isoamylnitrite, reduction in NaOH and reaction with HCI to form 5,6-dimethoxy-2-indanone, which was reacted with hydroxylamine and reduced with an overall yield of 42%. 5,6-Dimethoxyindan-1,2-dione-2-oxime, which was catalytically hydrogenated to afford cis-, and trans-1-amino-5,6-dimethoxyindan-1-ol as 3 : 1 ratio. This mixture was treated with Li and reacted with chloroacetyl chloride. Cis isomer was acylated and cyclized to synthesize rir -( ${\pm}$ )-7,8-dimethoxy-4,4a,5,9b-tetrahydroindeno[1,2-b][1,4]oxazin-3(2H)-one, but trans isomer was just acylated to form amide.

  • The title compound, ${\alpha},{\alpha}$-dichloro-o-methylacetophenone, was prepared and its photochemical behavior was investigated. Addition of an extra chlorine at alpha position to the carbonyl showed many different features from photochemical reactivities of mono chloro analogue, 2,5-dimethylphenacyl chloride. In benzene, a rearrangement product with a formal 1,5-Cl shift and a reduction product were formed beside indanone. In methanol, solvolysis and cyclization of a common dienol intermediate occurred at comparable reaction rates.
