Various bacterial strains were isolated from water in a paddy field, and their seaweed degrading activities were investigated. As the result, 16 strains were obtained from the microflora sample. They were incubated in a liquid medium of sea tangle powder for 3 weeks. Ratios of reduced sugar to total sugar of the liquid medium were measured once a week during the incubation period. Ratio of reduced sugar to total sugar of 30A412 strain was highest. Accordingly, 30A412 strain was incubated in three different liquid media of sodium alginate, sea tangle powder, and sea mustard powder for 3 or 4 weeks. The ratios of reduced sugar to total sugar and cell growth were measured once a week during the incubation period. Ratios of reduced sugar to total sugar and cell growth were increased with the incubation period. Cell growth and degrading ratio were highest in the liquid medium with sea mustard powder.
The current research was focused on the extracellular biosynthesis of bactericidal silver nanoparticles (AgNPs) using cell-free supernatant of a local isolate previously identified as a novel Streptomyces aegyptia NEAE 102. The biosynthesis of silver nanoparticles by Streptomyces aegyptia NEAE 102 was quite fast and required far less time than previously published strains. The produced particles showed a single surface plasmon resonance peak at 400 nm by UV-Vis spectroscopy, which confirmed the presence of AgNPs. Response surface methodology was chosen to evaluate the effects of four process variables ($AgNO_3$ concentration, incubation period, pH levels, and inoculum size) on the biosynthesis of silver nanoparticles by Streptomyces aegyptia NEAE 102. Statistical analysis of the results showed that the linear and quadratic effects of incubation period, initial pH, and inoculum size had a significant effect (p < 0.05) on the biosynthesis of silver nanoparticles by Streptomyces aegyptia NEAE 102. The maximum silver nanoparticles biosynthesis (2.5 OD, at 400 nm ) was achieved in runs number 5 and 14 under the conditions of 1 mM $AgNO_3$ (1-1.5% (v/v)), incubation period (72-96 h), initial pH (9-10), and inoculum size (2-4% (v/v)). An overall 4-fold increase in AgNPs biosynthesis was obtained as compared with that of unoptimized conditions. The biosynthesized silver nanoparticles were characterized using UV-VIS spectrophotometer and Fourier transform infrared spectroscopy analysis, in addition to antimicrobial properties. The biosynthesized AgNPs significantly inhibited the growth of medically important pathogenic gram-positive (Staphylococcus aureus) and gram-negative bacteria (Pseudomonas aeruginosa) and yeast (Candida albicans).
The present study aimed to determine the effects of different storage materials and storage period on some hatchability traits of Japanese quail (Coturnix coturnix japonica) eggs. A total of 32 male and 102 female quail (twelve weeks of ages) were housed in multiple-bird cages. Eggs laid were divided into three groups with respect to the period of storage (I = 1st, 2nd and 3rd days, II = 6th, 7th and 8th days, III = 12th, 13th and 14th days) prior to incubation. A total of 816 eggs was stored in 3 different storage environments during each storage period (B = no use of any storage material, P = use of perlite, H = use of hay) and kept in environmental conditions, where the temperature was $21^{\circ}C$ and relative humidity was 75%, prior to incubation. Statistical analyses were performed after the exclusion of values pertaining to non-fertile eggs (190 eggs) from the data set. The fertility rate of the eggs in the experiment was 76.7%. In the present study, the influence of storage material and different storage periods on egg weight loss were found to be statistically significant (p<0.01). Upon the comparison of hatchability of fertile eggs values, the influence of storage material was determined to be significant (p<0.05), and the influence of storage period was demonstrated to be significant (p<0.01). The storage materials used were determined not to have any influence on early and late embryonic death rates. Perlite was concluded to be safe for use in the storage of hatching eggs. The extension of the storage period (more than 8 days) resulted in decreased hatchability values of fertile eggs in each group.
Korean food waste, one of the abundantly available but environmentally problematic organic wastes in Korea, was utilized as solid-substrate by fungal strain Aspergillus niger ATcC 6275 for the production of enzymemixture containing amylase, cellulase and xylanase. The enzyme mixture can be used as high value-added animal feed. Solid-state fermentation method yielded a 84-fold enhancement in xylanase activity compared with submerged fermentation method. The effect of incubation period, incubation temperature, pH of medium, moisture content, inoculum size and enrichment of the medium with nitrogen and carbon sources were observed for optimal production of these enzymes The optimal amylase activity of 33.10 U/g, cellulase activity of 24.41 U/g, xylanase activity of 328.84 U/g were obtained at 8 days incubation with 50%(w/w) soy bean flake, with incubation temperature of $25^{\circ}C$, pH of 6.38, optimal moisture content of 55% and with inoculum size of $3.8{\times}10^6$spore/g. Enzyme activities were enhanced when ImM $CaSO_4$, 2% Malt extract and 2% galactose were added as mineral, nitrogen and carbon enrichment respectively.
Bioactivity of the extract from various processed garlic was evaluated based on the inhibition of lipoxygenase(LPO), and the effect of some stabilizers on the bioactivity was investigated. Water, ethanol or chloroform extract of 30 min boiled garlic showed 75%, 76% or 70% inhibition, respectively, compared to extracts of fresh garlic. In pickled garlic, LPO inhibition decreased gradually during storage. Chloroform extract of 40 day-stored pickled garlic inhibited LPO by 77%, and even on 60th day storage it still retained inhibitory effect of 73%, compared to that of fresh garlic. Meanwhile, the powdered (freeze-dried) garlic showed more bioactivity(80%) than the other processed garlics, and moreover, the jrreversible/unstable components seem to be stabilized by freeze-drying. The optimum pH for stabilization of bioactive components in garlic macerate was pH 3 for 48 hr incubation and pH 11 for 6 hr incubation. In addition, the effect of NaCl was not so great, although but maximal stabilization was observed at 150 mM. Stabilizing effect of $\alpha$-tocopherol was markedly great, and at 6mM it showed 308% stabilizing effect after 48hr incubation. More stabilizing effect was observed at lower concentrations of ascorbic acid($\leq$0.6mM) than higher concentrations. The stabilizing effect of soybean oil was found to be remarkable only during initial period(6 hr) of incubation.
Deoxygenation process in which CBOD(carbonaceous BOD) is biochemically oxidized to reduced inorganic compounds is of great significance in the oxygen demand of stream waters. The CBOD decay rate has traditionally been determined in a laboratory by CBOD bottle incubation method. But in this study, CBOD decay rates were obtained by laboratory incubation method and natural waters experiment. Average CBOD decay rate for the Nakdong river(upper zone) in natural waters were 0.553 $day^{-1}$ during April 2005 to January 2007. The values in the middle and down parts of the Nakdong river in natural waters were 0.384 $day^{-1}$ and 0.252 $day^{-1}$ at the same period of time, respectively. Average CBOD decay rates using by incubation method in the upper/middle/down parts of the main stream in the Nakdong river basin was 0.270 $day^{-1}$, and 0.289 $day^{-1}$, and 0.283 $day^{-1}$ during April 2005 to January 2007, respectively.

In order to characterize saikosaponin biosynthesis in Bupleurum falcatum, the expression of five isoprenoid pathway genes and their relationship to saikosaponin accumulation in the hairy roots were analyzed. The hairy roots exhibited a rapid accumulation of saikosaponins when incubated in a root culture medium (3XRCM). Homology-based RT-PCR was used to isolate core fragments of five genes, HMGR, IPPI, FPS, SS, and OSC, from the hairy roots. The deduced amino acid sequences exhibited amino acid identities of more than 85% to previously reported genes. Using the fragments as probes, the expression of these five genes in the hairy roots during incubation in 3XRCM medium was examined. Expression of all five genes in the hairy roots increased soon after incubation. In particular, the SS and OSC genes were coordinately induced at 8 days of incubation, and their expression persisted throughout the incubation period. A quantitative HPLC analysis showed that the saikosaponin content of the hairy root culture also began to increase at 8 days of culture. The correlation between SS transcript level and saikosaponin content in the hairy roots suggests that transcriptional regulation plays a regulatory role in saikosaponin biosynthesis.
This study was conducted to compare in vitro rumen fermentation characteristics among corn grains imported from America, Brazil, Argentina and Ukraine A and Ukraine B. Two Holstein steers, each surgically fitted with a ruminal cannula, consuming total mixed ration were used as rumen fluid donors. In vitro rumen fermentation experiments were performed in a completely random design which included a control (no corn) and treatments with 3.0 g of corn from different geographical origins, i.e., America, Brazil, Argentina, and Ukraine A and Ukraine B, respectively. Ruminal pH, ammonia-N, volatile fatty acid (VFA) and total gas production were measured at 0, 1, 3, 6, 12, 24 and 48 h post-incubation, respectively. No differences (p > 0.05) in mean ruminal pH appeared among the treated groups, however, ruminal pH patterns differed; i.e. corn treated groups had dramatically lower pH compared with control during the entire incubation period. Similarly, no different patterns between the groups in ammonia-N (p > 0.05) appeared until 6 h post-incubation. Unexpectedly, higher ammonia-N concentration for control than that for the corn treated groups appeared after 12 h post-incubation despite that for all groups increased. Total VFA was similar between the groups until 6 h post-incubation, but VFA after 12 h post-incubation was different (p < 0.05), i.e. VFA for corn from Argentina, Ukraine A, Ukraine B, and Brazil were comparatively higher than for America. Overall, data in this study showed that the corns of different origins may have different feed values to ruminants despite having similar chemical compositions.
We investigated fermentation characteristics and the amount of exopolysaccharide (EPS) produced, and the correlation between EPS and rheological properties in stirred yogurt fermented with Streptococcus thermophilus St-Body 1. The changes of pH and lactic acid concentrations of yogurt showed different patterns according to fermentation temperatures. About 20 to 40% of lactose was utilized during 6 hrs incubation. The higher number of lactic acid bacteria was obtained at 31℃ incubation temperature rather than other incubation temperatures. The higher amount of EPS was produced at 12 hr or 24 hr rather than other incubation period at the same fermentation temperatures. The viscosity continuously increased during the fermentation period, however the syneresis of yogurt was shown the lowest value at 37℃ for 30 hrs incubation. In physical properties, the hardness and adhesiveness increased continuously with decreasing cohesiveness and elasticity as incubation process was extended. The stirred yogurt fermented with S. thermophilus St-Body 1 at 37℃ for 24 hr resulted in the highest score at each sensory evaluation category.
Cultivation characteristics of 12 strains of cauliflower mushroom (Sparassis crispa) collected in Korea were investigated by growing the mushroom on sawdust medium of Larix kaempferi. As cultivation characteristics, incubation period for full growth of mycelium in a cultivation bottle, cultivation time period taken for first harvest, and mushroom color and yield were examined. S. crispa KFRI 723 showed the shortest for incubation period with 59 days while S. crispa KFRI 746 showed the longest with 94 days. The earliest mushroom harvesting was achieved by 29 days from S. crispa KFRI 746 and the latest was by 63 days from S. crispa KFRI 691. The colors of fruit body of the tested strains can be divided into three groups; S. crispa KFRI 700 was white, S. crispa KFRI 747 was yellow brown, and the others were light yellowish. KFRI 700 yielded the most as 163 g from 380 g sawdust media, while KFRI 746 and KFRI 747 were the lowest with 58 g and 35 g, respectively. As results of cultivation characteristics of 12 strains of cauliflower mushroom, we consider that three strains (KFRI 700, 723 and 724) of S. crispa are suitable for sawdust cultivation on L. kaempferi in the aspects of mycelial growth period, harvesting period and mushroom production, respectively.
