Variation in the number of nucleoli and incomplete homogenization of 18S ribosomal DNA sequences in leaf cells of the cultivated Oriental ginseng (Panax ginseng Meyer)

  • Background: Wild ginseng, Panax ginseng Meyer, is an endangered species of medicinal plants. In the present study, we analyzed variations within the ribosomal DNA (rDNA) cluster to gain insight into the genetic diversity of the Oriental ginseng, P. ginseng, at artificial plant cultivation. Methods: The roots of wild P. ginseng plants were sampled from a nonprotected natural population of the Russian Far East. The slides were prepared from leaf tissues using the squash technique for cytogenetic analysis. The 18S rDNA sequences were cloned and sequenced. The distribution of nucleotide diversity, recombination events, and interspecific phylogenies for the total 18S rDNA sequence data set was also examined. Results: In mesophyll cells, mononucleolar nuclei were estimated to be dominant (75.7%), while the remaining nuclei contained two to four nucleoli. Among the analyzed 18S rDNA clones, 20% were identical to the 18S rDNA sequence of P. ginseng from Japan, and other clones differed in one to six substitutions. The nucleotide polymorphism was more expressed at the positions 440-640 bp, and distributed in variable regions, expansion segments, and conservative elements of core structure. The phylogenetic analysis confirmed conspecificity of ginseng plants cultivated in different regions, with two fixed mutations between P. ginseng and other species. Conclusion: This study identified the evidences of the intragenomic nucleotide polymorphism in the 18S rDNA sequences of P. ginseng. These data suggest that, in cultivated plants, the observed genome instability may influence the synthesis of biologically active compounds, which are widely used in traditional medicine.

The influence of subjective awareness of dental health status on the use of dental hygienic devices (주관적인 구강건강상태 인식이 구강관리용품의 인지 및 사용에 미치는 영향)

  • Objectives: The purpose of the study is to investigate the influencing factors of subjective awareness of dental health status on the use of dental hygienic devices. Methods: A self-reported questionnaire was completed by 320 patients who visited dental clinics in Seoul and Gyeonggi-do from May to June 2014. Except incomplete answers, 284 copies were analyzed by SPSS WIN 18.0 program. The questionnaire was adapted from Park & Youn, and Chae. The questionnaire consisted of five questions of the general characteristics of the subjects, one question of subjective dental health status, eight questions of awareness of dental hygienic devices, and eight questions of use of dental hygienic devices. Results: Although the awareness of self dental health state was shown to statistically significantly affect the awareness of dental hygiene devices(p<0.001) which is the dependent variable, it was shown to not affect the use. Conclusions: Based on such results, to improve and maintain the dental health, an individual's subjective dental health state should be identified and provide accurate and objective information on the dental hygiene devices that are suitable to individual dental state, as well as make the patient's use of dental hygiene devices increase through continuous boosting of motivation.

The Impact of Poverty on Self-Rated Health in Philippines: A Mediated Moderation Model of Health Behaviors and Family and Friend Support (필리핀 지역주민의 빈곤이 주관적 건강상태에 미치는 영향: 건강행태와 가족 및 친구 지지의 매개된 조절효과 분석)

  • Background: Socio-economical status, represented by poverty, is a potent factor in predicting health status, because preventable illness and death occurs due to poverty and socio-economical situation. This study aims to provide information towards on the correlation between poverty and self-rated health in consideration of elements of health behaviors and family and friend support in Philippines. Methods: Data was collected on 15th to 28th of February 2011 by using structured questionnaire through interview method. Study area is Antipolo and Tondo in Manila, Philippines. Sample size was 1,100 but only 658 cases was analyzed due to incomplete questionnaire. Results: Results show that the poverty has direct negative influence on self-rated health, while the presence of family and friend support decreased negative influence. And through the analysis of mediated moderation model, similarly poverty has also indirect negative effect on self-rated health by health behaviors and family and friend support. Especially, regular exercise was found to be a major variable that mediates poverty and self-rated health. Conclusion: In conclusion, to improve the self-rated health state of respondents need to provide emotional support especially from family and friends. Also regular exercise should be encouraged by raising health awareness through continuous health education and promotion for the residents in order to develop health behaviors.

Investigation on spanwise coherence of buffeting forces acting on bridges with bluff body decks

  • In the traditional buffeting response analysis method, the spanwise incomplete correlation of buffeting forces is always assumed to be same as that of the incident wind turbulence and the action of the signature turbulence is ignored. In this paper, three typical bridge decks usually adopted in the real bridge engineering, a single flat box deck, a central slotted box deck and a two-separated paralleled box deck, were employed as the investigated objects. The wind induced pressure on these bridge decks were measured via a series of wind tunnel pressure tests of the sectional models. The influences of the wind speed in the tests, the angle of attack, the turbulence intensity and the characteristic distance were taken into account and discussed. The spanwise root coherence of buffeting forces was also compared with that of the incidence turbulence. The signature turbulence effect on the spanwise root coherence function was decomposed and explained by a new empirical method with a double-variable model. Finally, the formula of a sum of rational fractions that accounted for the signature turbulence effect was proposed in order to fit the results of the spanwise root coherence function. The results show that, the spanwise root coherence of the drag force agrees with that of incidence turbulence in some range of the reduced frequency but disagree in the mostly reduced frequency. The spanwise root coherence of the lift force and the torsional moment is much larger than that of the incidence turbulence. The influences of the wind speed and the angle of attack are slight, and they can be ignored in the wind tunnel test. The spanwise coherence function often involves several narrow peaks due to the signature turbulence effect in the high reduced frequency zone. The spanwise coherence function is related to the spanwise separation distance and the spanwise integral length scales, and the signature turbulence effect is related to the deck-width-related reduced frequency.

Development of Dual Sensor for Prognosticating Fatigue Failure of Mechanical Structures (구조물의 피로파괴 예지를 위한 이중센서 개발)

  • Because of the inherent uncertainties caused by the manufacturing process variations, future loading conditions, and incomplete damage models, the lifetimes of mechanical structures under field conditions are significantly different from the results obtained in the laboratories. In this study, a dual sensor was developed to prognosticate the fatigue failure of structures under these uncertain conditions, and its effectiveness was demonstrated on a rectangular columnar structure under repeated uni-axial loading. The dual sensor is a slightly weaker structure embedded in the target structure, so that failure occurs in the sensor earlier than in the target structure. From the signal differences in the strain gauges in the embedded dual sensor, it is possible to differentiate between the normal status and warning status, even under variable loads.

Seoul some dental hygienist in the factors affecting job satisfaction study (서울시 일부 치과위생사의 직무만족에 영향을 미치는 요인)

  • Purpose : First, this study is to look into difference between dental hygienists' demographic characteristics and their job satisfaction by examining their demographic characteristics and job satisfaction. Second, this study is to analyze demographic factors of dental hygienists which influence their job satisfaction. Method : This study was made through mail survey with questionnaires for dental hygienists who are personally performing dental hygienist duties at dental clinics in Seoul between June 19 and July 24, 2008 by looking into factors influencing dental hygienists' job satisfaction in order to provide them as background data to enhance their job satisfaction. Total 180 questionnaires were distributed, and among these, 156 questionnaires excluding 24 questions of incomplete responses were statistically analyzed with SPSS 12.0. Result : First, the total mean of dental hygienists' job satisfaction was found to be 2.99 points out of 5 points, and that of their autonomy to be the highest as 3.26, and that of their salary to be the lowest as 2.55. Second, for relation between general characteristics and job satisfaction of dental hygienists, significant difference was found just in job experience(p<0.05), current position(p<0.05), average salary(p<0.001), and the expected period of engagement in dental hygienist duties(p<0.01). Third, descriptive power on demographic characteristics, which were used as an independent variable influencing their job satisfaction, was found to be 17.7%, and among the said factors, average salary had a statistically significant effect on their job satisfaction(p<0.01). Conclusion : The point of salary factor was lowest among 5 factors composing dental hygienists' job satisfaction, and as a result of multiple regression analysis, a factor influencing job satisfaction was average salary. Therefore salary system is required to be improved in order to increase their job satisfaction, and further studies on factors influencing their job satisfaction are also required to be made.

Identification of DEA Determinant Input-Output Variables : an Illustration for Evaluating the Efficiency of Government-Sponsored R&D Projects (DEA 효율성을 결정하는 입력-출력변수 식별 : 정부지원 R&D 과제 효율성 평가를 위한 실례)

  • In this study, determinant input-output variables are identified for calculating Data Envelopment Analysis (DEA) efficiency scores relating to evaluating the efficiency of government-sponsored research and development (R&D) projects. In particular, this study proposes a systematic framework of design and analysis of experiments, called "all possible DEAs", for pinpointing DEA determinant input-output variables. In addition to correlation analyses, two modified measures of time series analysis are developed in order to check the similarities between a DEA complete data structure (CDS) versus the rest of incomplete data structures (IDSs). In this empirical analysis, a few DEA determinant input-output variables are found to be associated with a typical public R&D performance evaluation logic model, especially oriented to a mid- and long-term performance perspective. Among four variables, only two determinants are identified : "R&D manpower" ($x_2$) and "Sales revenue" ($y_1$). However, it should be pointed out that the input variable "R&D funds" ($x_1$) is insignificant for calculating DEA efficiency score even if it is a critical input for measuring efficiency of a government-sonsored R&D project from a practical point of view a priori. In this context, if practitioners' top priority is to see the efficiency between "R&D funds" ($x_1$) and "Sales revenue" ($y_1$), the DEA efficiency score cannot properly meet their expectations. Therefore, meticulous attention is required when using the DEA application for public R&D performance evaluation, considering that discrepancies can occur between practitioners' expectations and DEA efficiency scores.

  • Central cord syndrome (CCS) is the most common incomplete spinal cord injury (SCI) syndrome, occured by cervical hyperextension injury. It is characterized by disproportionately greater motor impairment in the upper extremities compared to the lower extremities, bladder dysfunction, and variable degrees of sensory loss below the level of injury. Recently 36 year-old male was admitted with upper motor neuron weakness, sensory loss in the upper extremities and stiff neck. We report the improvement of his symptoms through both western medical treatment and oriental medical treatment.

Assessing the Willingness-to-Pay of a Scuba Diving Management in a Soft Coral Marine Protected Area (다이버를 대상으로 한 연산호(軟珊瑚) 서식 해양보호구역 다이빙 관리제도의 지불의사액 분석)

  • A marine protected area (MPA) is a spatially designated section of the ocean where the biodiversity is high. Munseom MPA has a unique underwater landscape including soft coral, subtropical fish, and benthos, attracting many scuba divers. This paper intends to evaluate the willingness-to-pay (WTP) for the scuba diving management policy in the Munseom MPA. As a stated preference valuation method, a choice experiment was applied to assess divers' preferences with economic values for the policy. The attributes used in the study included zoning (temporal closure), conservation education, daily permit, soft coral area, and diving fee. We collected 333 questionnaires through online and field surveys and used 296 after excluding incomplete responses for the final data analysis. Study results show that a six-month closure, a decrease in soft coral habitat, and the diving fee were statistically significant. A positive coefficient with regard to the six-month closure variable indicates that divers preferred the policy, and the marginal willingness to pay (MWTP) was estimated to be KRW 8,637 per person per dive. The aggregate WTP of KRW 233 million was obtained by multiplying the MWTP by the number of visiting divers per year. When comparing the levels of recreation specialization, more skilled divers preferred the management option. The results of this study will help predict the priority of management measures and the levels of acceptability among divers when diving management is implemented in Korea in the future. This study suggests that marine environmental policies need to consider divers as one of the important stakeholders for marine conservation.

A Novel SLC25A15 Mmutation Causing Hyperornithinemia-Hyperammonemia-Homocitrullinuria Syndrome (Hyperornithinemia-hyperammonemia-homocitrullinuria 증후군을 유발하는 SLC25A15 유전자의 새로운 변이)

  • Hyperornithinemia-hyperammonemia-homocitrullinuria syndrome (HHH syndrome) is a neurometabolic disorder with highly variable clinical severity ranging from mild learning disability to severe encephalopathy. Diagnosis of HHH syndrome can easily be delayed or misdiagnosed due to insidious symptoms and incomplete biochemical findings, in that case, genetic testing should be considered to confirm the diagnosis. HHH syndrome is caused by biallelic mutations of SLC25A15, which is involved in the urea cycle and the ornithine transport into mitochondria. Here we report a boy with spastic paraplegia and asymptomatic younger sister who have compound heterozygous mutations of c.535C>T (p.R179*) and c.116C>A (p.T39K) in the SLC25A15 gene. We identified that p.T39K mutation is a novel pathogenic mutation causing HHH syndrome and that p.R179*, which is prevalent in Japanese and Middle Eastern heritage, is also found in the Korean population.
