miR-153 Silencing Induces Apoptosis in the MDA-MB-231 Breast Cancer Cell Line

  • MicroRNAs (miRNAs) are small, non-coding RNAs (18-25 nucleotides) that post-transcriptionally modulate gene expression by negatively regulating the stability or translational efficiency of their target mRNAs. In this context, the present study aimed to evaluate the in vitro effects of miR-153 inhibition in the breast carcinoma cell line MDA-MB-231. Forty-eight hours after MDA-MB-231 cells were transfected with the miR-153 inhibitor, an MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was utilized to determine the effects of miR-153 on cell viability. Flow cytometry analysis and assessment of caspase 3/7 activity were adopted to determine whether miR-153 affects the proliferation rates and apoptosis levels of MDA-MB-231 cells. Our results showed that silencing of miR-153 significantly inhibited growth when compared to controls at 48 hours, reducing proliferation by 37.6%, and inducing apoptosis. Further studies are necessary to corroborate our findings and examine the potential use of this microRNA in future diagnostic and therapeutic interventions.

Current Evidence on the Relationship Between Two Polymorphisms in the NBS1 Gene and Breast Cancer Risk: a Meta-analysis

  • Introduction: Published studies on the association between Nijmegen breakage syndrome 1(NBS1) gene polymorphisms and breast cancer risk have been inconclusive, and a meta-analysis was therefore performed for clarification. Methods: Eligible articles were identified by a search of MEDLINE and EMBASE bibliographic databases for the period up to March 2012. The presence of between-study heterogeneity was investigated using the chi-square-based Cochran's Q statistic test. When there was statistical heterogeneity, the random effects model was chosen; otherwise, fixed effects estimates were reported as an alternative approach. Results: A total of 11 eligible articles (14 case-control studies) were identified, nine case-control studies were for the 657del5 mutation (7,534 breast cancer cases, 14,034 controls) and five case-control studies were for the I171V mutation (3,273 breast cancer cases, 4,004 controls). Our analysis results indicated that the 657del5 mutation was associated with breast cancer risk (carriers vs. non-carriers: pooled OR =2.63, 95% CI: 1.76-3.93), whereas the I171V mutation was not (carriers vs. non-carriers: pooled OR =1.52, 95% CI: 0.70-3.28). Conclusion: The present meta-analysis suggests that the 657del5 gene mutation in the NBS1 gene plays a role in breast cancer risk, while the I171V mutation does not exert a significant influence.

Two-Layer Hand-Sewn Esophagojejunostomy in Totally Laparoscopic Total Gastrectomy for Gastric Cancer

  • Purpose: Different esophagojejunostomy (EJ) reconstruction methods are used after totally laparoscopic total gastrectomy (TLTG), and none is considered a standard technique. This report describes a 2-layer hand-sewn EJ technique during TLTG; we also evaluated postoperative morbidity associated with this technique. Materials and Methods: This retrospective cohort study included all consecutive patients who underwent TLTG for gastric cancer (GC) from 2012 to 2016 at 2 affiliated teaching hospitals. All participating surgeons performed standardized intracorporeal 2-layer hand-sewn EJ. Results: We included 51 patients who underwent TLTG for GC and standardized EJ anastomosis. Twenty-seven (53%) were male, and the median age was 60 (36-87) years. The average operative time was $337{\pm}71minutes$ and intraoperative bleeding was $160{\pm}107mL$. There were no open conversions related to EJ. Postoperative morbidity was observed in 9 (17.0%) patients. There was no postoperative mortality. EJ leakage was observed in 2 patients (3.8%) and 1 patient (1.9%) developed EJ stenosis. Patients with leakage were managed non-operatively and the patient with stenosis required endoscopic dilation. The median length of hospital stay was 8 (6-29) days. Conclusions: Two-layer hand-sewn EJ during TLTG for GC is a feasible and safe technique. This method avoids a laparotomy for reconstruction and the disadvantages associated with laparoscopic introduction of mechanical staplers for EJ, and provides an alternative for alimentary tract reconstruction after TLTG.

  • This study is an analyze for Casa del Fascio, of Giuseppe Terragni, an important architect of fascism period of Italy. Based on the spot reinvestigation of first copies and unpublished data in a archive for a month of 2002. August, Terragni, Como, to compromise with the limit of studies up to the present. General's from early scheme to final constructed scheme, finally offered standard way to understand Terragni's scheme. Result of analyze are to two conclusions. 1) It has direct bearings between physical environment of site which was to build from original scheme and final decided external form. Especially there is direct relation between facade and composition of Duomo Cathedral, in the center of Como. 2) Although the image for scheme by architect represented as a physical form which was impossible to analyze the data as development sequence from archive, it influences on the first image, which had changed many times, exists as important element to final constructed scheme.

  • Immunotherapy is an effective treatment option for gynecological malignancies. Radiologists dealing with gynecological patients undergoing treatment with immune checkpoint inhibitors should be aware of unconventional immune-related imaging features for the evaluation of tumor response and immune-related adverse events. In this paper, immune checkpoint inhibitors used for gynecological malignancies and their mechanisms of action are briefly presented. In the second part, patterns of pseudoprogression are illustrated, and different forms of immune-related adverse events are discussed.

  • Baird-Parker agar with egg yolk/tellurite emulsion (BPA) is widely accepted as a medium for the enu-meration of Staphylococcus aureus in foods. Howerver, it is not vompletely selective and colonies of other genera of species could be similar to those of Staphylococcus aureus. Moreover, the strains of Staphylococcus aureus that are lecithinase negatrive could go unnoticed. Both facts could affect the counts. The aim of this study was to dtermine whether the enumeration of the colonies with the typical morphol-ogy of Staphylococcus aureus on BPA is sufficien to quantity this species in poultry meat. Forty chiken carcasses were tested for Staphylococcus aureus by surface plating using BPA, Results indicate that the predictiv value of the morphoogy of the colonies of BPA is 85.71% and 68.42% for typical and atypical colonies of Staphylococcus aureus, respectively. However, Staphylococcus aureus counts (after identification) and counts of typical colonies did not show any significant differences(P>0.05) and are significantly(P<0.001) correlated (r=0.996).These results suggest that , for screening purposes. enumeration of Staphylococcus aureus from poultry meat does not require any identification of stains. resulting in a saving of time and money.

Gigantomastia as a Cause of Pulmonary Hypertension

  • Reduction mammaplasty is the gold standard treatment for gigantomastia. We report one female patient with juvenile gigantomastia associated with severe pulmonary hypertension where her pulmonary pressure decreased significantly after the surgery, improving her quality of life. A 22-year-old female patient with gigantomastia since 10 years old, tricuspid regurgitation, and pulmonary thromboembolism antecedent was admitted to the emergency department. Her oxygen saturation was 89%. Acute heart failure management was initiated. An echocardiogram reported left ventricle ejection fraction (LVEF) of 70% with severe right heart dilation, contractile dysfunction, and arterial pulmonary pressure (PASP) of 110 mm Hg. A multidisciplinary team considered gigantomastia could generate a restrictive pattern, so a Thorek reduction mammoplasty with Wise pattern was performed. Presurgical measurements were: sternal notch to nipple-areola complex, right 59 cm, left 56 cm. Three days after surgery, the patient could breathe without oxygen support. In the outpatient follow-up, patient referred reduction of her respiratory symptoms and marked improvement in her quality of life. Six months after surgery, a control echocardiogram showed a LVEF of 62% and PASP of 85 mm Hg. Pulmonary hypertension may be present in patients with gigantomastia. Reduction mammoplasty may be a feasible alternative to improve the cardiac signs and symptoms in patients with medical refractory management.

Correlation between EGFR Gene Mutations and Lung Cancer: a Hospital-Based Study

  • Epidermal growth factor receptor (EGFR) is one of the targeted molecular markers in many cancers including lung malignancies. Gefitinib and erlotinib are two available therapeutics that act as specific inhibitors of tyrosine kinase (TK) domains. We performed a case-control study with formalin-fixed paraffin-embedded tissue blocks (FFPE) from tissue biopsies of 167 non-small cell lung carcinoma (NSCLC) patients and 167 healthy controls. The tissue biopsies were studied for mutations in exons 18-21 of the EGFR gene. This study was performed using PCR followed by DNA sequencing. We identified 63 mutations in 33 men and 30 women. Mutations were detected in exon 19 (delE746-A750, delE746-T751, delL747-E749, delL747-P753, delL747-T751) in 32 patients, exon 20 (S786I, T790M) in 16, and exon 21 (L858R) in 15. No mutations were observed in exon 18. The 63 patients with EFGR mutations were considered for upfront therapy with oral tyrosine kinase inhibitor (TKI) drugs and have responded well to therapy over the last 15 months. The control patients had no mutations in any of the exons studied. The advent of EGFR TKI therapy has provided a powerful new treatment modality for patients diagnosed with NSCLC. The study emphasizes the frequency of EGFR mutations in NSCLC patients and its role as an important predictive marker for response to oral TKI in the south Indian population.

The Dynamics of Local Politics in the Philippines: A Case Study on the PBMA in Surigao del Norte Province (필리핀 지방정치의 역동성: 북부 수리가오 주의 토착 기독교 종교단체 (PBMA) 사례 연구)

  • It is generally acknowledged that Philippines politics has achieved a basic soil for democracy at the macro-level by the two incidents of 'People Power' (1986 & 2001). However, in spite of such an achievement, Philippine politics at the micro-level, does not enjoy the same reputation. Institutionalized 'Bossism' or 'patron/clientelism' political culture has been attributed to make such a gap. This paper aims to bridge the gap by a longitudinal observation on local politics of a particular province in the Philippines. Surigao del Norte is the province picked up for the study. The reason why this particular province attracts special concerns is that there appears to come a very dynamic local politics, led by an indigenous Christian organization, the PBMA (Philippines Benevolent Missionary Association), which was founded in the mid 1960s. With the coming of the organization in the province, the political power of the traditional families has been narrowed. On the other hand, new families, not only the leader family of the PBMA but also the family allied with the PBMA, has expanded political influence. Following steps are adopted to understand the meaning of such a change. First, institutional frame of Philippine politics and elections will be reviewed. Second, demographic changes will be investigated on Surigao del Norte province to find out how popular the PBMA organization is among the people. Third, local election outcomes will be examined to describe the changing nature of political landscape of the province. This will reveal how the relationship between the old and new families has been developed in the province. Lastly, based on the facts discovered by a longitudinal observation, an attempt to evaluate theories on Philippines politics will be made Conclusions are as follows. To understand the dynamics of Philippine politics, 'expectations from the below,' or, 'passions of the people,' suggested by Ileto, needs more attention. Furthermore, 'mutual accommodation' between domination and resistance, coined by Abinales, also demands more appreciation. The case of local politics in the Surigao del Norte, with the coming of the PBMA, is sure to show a concrete example of changing politics in the 'changeless land.'

Isolation and Characterization of the Smallest Bacteriophage P4 Derivatives Packaged into P4-Size Head in Bacteriophage P2-P4 System

  • Bacteriophage P4, a satellite phage of coliphage P2, is a very useful experimental tool for the study of viral capsid assembly and cos-cleavage. For an in vitro cos-cleavage reaction study of the P2-P4 system, new shortened and selectable markers containing P4 derivative plasm ids were designed as a substrate molecules. They were constructed by swapping the non-essential segment of P4 DNA for either the kanamycin resistance (kmr) gene or the ampicillin resistance (apr) gene. The size of the genomes of the resulting markers were 82% (P4 ash8 delRI:: kmr) and 79% (P4 ash8 delRI:: apr) of the wild type P4 genome. To determine the lower limit of genome size that could be packaged into the small P4-size bead, these shortened P4 plasmids were converted to phage particles with infection of the helper phage P2. The conversion of plasmid P4 derivatives to bacteriophage particles was verified by the heat stability test and the burst size determination experiment. CsCl buoyant equilibrium density gradient experiments confirmed not only the genome size of the viable phage form of shortened P4 derivatives, but also their packaging into the small P4-size head. P4 ash8 delRI:: apr turned out to be the smallest P4 genome that can be packaged into P4-sized head.
