개 외이염에 대한 Enrofloxacin과 Silver Sulfadiazine 국소제제의 치료효과 (Efficacy of Enrofloxacin and Silver Sulfadiaznine Topical Otic Suspension for the Treatment of Canine Otitis Externa)

  • 본 연구는 개 외이염에 대한 enrofloxacin-silver sulfadiazine (Baytril$^{(R)}$ otic, Bayer, USA)의 치료효과를 평가하는 것이다. 24두의 외이염 이환견을 대상으로 처치군에는 바이트릴 오틱을, 대조군에는 증류수를 이도내 7일간 국소투약한 후 7일후에 임상증상, 미생물 및 효모균수를 통해 효과를 비교하였다. 또한 외이염에서 분리된 세균 및 효모균에 대한 바이트릴 오틱의 최소억제농도를 산출하였다. 주요 분리균으로는 Staphylococcus pseudintermedius, Pseudomonas aeruginosa, Proteus spp and Malassezia pachydermatis.이었다. 임상증상의 경감은 81%, 미생물수는 87% 감소하였다. 바이트릴 오틱은 분리된 균에 대한 살균효과가 충분하였고 국소투약에 대한 부작용은 관찰되지 않았다. 따라서 바이트릴 오틱은 개의 외이염 감염에 대한 효과적이며 안전한 제제로 판단된다.

  • Background. 5-(and-6)-carboxy-2',7'-dichlorofluorescein diacetate, succinimidyl ester mixed (CFSE) is the fluorescent labelling agent of living cells and used to trace the cells in vivo after transplatnation of various cells. The CFSE labelled cells can maintain fluorescence for up to 7 days after labelling. The MC3T3-E1 cell line (MC3T3) has been used for many studies about osteoblast, which is well known as a mouse preosteoblast. So the CFSE would be used to trace the transplanted MC3T3. However there are few reports about CFSE labelling of MC3T3. This study is aimed to know about adequate concenturation and incubation time of CFSE to MC3T3. Materials and methods. The MC3T3 was incubated in a humidified atmosphere of 95% air with 5% $CO_2$ at $37^{\circ}C$ using ${\alpha}$-minimal essential medium (${alpha}$-MEM) containing10% FBS and gentamycin. Ten mM CFSE solution in dimethylsulphoxide (DMSO: 1%) was diluted with phosphate buffered saline (PBS) and final concentration of culture medium was, respectively, 5, 10, 15, 20, 25 and 30 ${{\mu}M$. Then the MC3T3 was incubated with CFSE in a humidified atmosphere of 95% air with 5% $CO_2$ at $37^{\circ}C$ for 5, 10, 15, 20, 25, 30, 35, 40 and 45 minutes in each concentration. The fluorescence of CFSE labelled cells was analysed with a inverted fluorescence microscope. The duration of cell labelling was also studied. Trypan blue dye exclusion test was done for cell viability. Results. For concentration between 5 and 10 ${\mu}M$, CFSE did not significantly label the MC3T3 in vitro. The destruction of MC3T3 was observed at the concentration of 20 ${\mu}M$. In the concentration of 15 ${\mu}M$, the best labelling was obtained at an incubation period between 15 and 30 minutes. The MC3T3 labelled with an incubation period of 15 minutes at 15 ${\mu}M$ was still fluorescent 7 days after CFSE labelling. The mean cell viability was 95.93%. Conclusion. These results suggests an incubation period of 15 minutes at 15 ${\mu}M$ of CFSE provides best labelling of MC3T3 in vitro.

오매(烏梅)의 다성분동시분석 및 마우스를 이용한 경구 단회투여 급성독성시험 (Simultaneous determination and acute toxicity study of Fructus mume extracts in ICR mice)

  • Objectives : Previous studies have shown that Fructus mume (FM) has anti-platelet effects. The present study was performed to determine the acute oral toxicity and quality control of a crude extract of FM in ICR mice. Methods : We investigated the in vivo single dose acute toxicity of FM 95% ethanol extract. This test was orally administered once by gavage to 20 male and 20 female mice at dose levels of 0 (control group), 1250, 2500 and 5000mg/kg body weight, respectively. Mortalities, clinical findings, autopsy findings and body weight changes were monitored daily for the 14 days following the administration. HPLC analysis was performed for the simultaneous determination of ursolic acid and p-hydroxycinnamic acid in FM. Reverse-phase chromatography using a C18 column and photodiode array detection at 211 nm was used for quantification of the two maker components. The mobile phase for gradient elution consists of water and acetonitrile. Results : We observed survival rates, general toxicity, change of body weight, and autopsy. The mice did not die after single oral administration of maximum dose of FM. Autopsy of animal revealed no abnormal gross finding. Therefore, $LD_{50}$ value of FM for ICR mice was more than 5000mg/kg on oral route. The HPLC analysis showed that ursolic acid and p-hydroxycinnamic acid amounts to 9.75- and 0.12% in the extract with the retention times of 47.99- and 15.38 minutes, respectively. Conclusions : These results suggest that no toxic dose level of FM in mice is considered to be more than 5000mg/kg. Consequently, it was concluded that FM have no effect on acute toxicity and side effect in ICR mice. For the quality control of FM extract, simultaneous determination of ursolic acid and p-hydroxycinnamic acid was established.

IMACIS-1을 이용한 위장관 종양의 방사면역신티그램 (Radioimmunoscintigraphy Using IMACIS-1 in Gastrointestinal Cancer)

  • Most of the diagnostic methods currently used for the detection of neoplastic masses provide indirect evidence. To obtain greater specificity in the interpretation of neoplasias by in vivo methods, the immunological approach appears to be most promising. Two problems that interfered with progress in this field were the lack of tumor specific antigen and the lack of well-defined and reproducible antibodies. To improve the sensitivity and specificity of radioimmunoscintigraphy as a technique for tumor localization, the use of monoclonal antibodies, fragments of antibodies and single photon emission computerized tomography (SPECT) are reasonable. The obvious advantages of monoclonal antibodies are their homogeneity, their specificity for the immunizing antigen and the reaction with a single determinant-thus no large immunecomplexes with antigen are formed. Monoclonal antibody technique has recently provided an opportunity to reevaluate the role of nuclear medicine for the diagnosis of malignant diseases by using the immunological approach. Out first results by means of radioimmunoscintigraphy of CEA and CA 19-9 producing tumors using a cocktail of fragments F $(ab')_2$, of mocolonal antibodies to CA 19-9 and CEA labeled with $^{131}I$ (IMACIS-1) are reported. The aims of this investigation was to evaluate the role of immunoscintigraphy in patients with colorectal and other cancers for diagnosis of local recurrences and metastasis. This report contains results of the first 8 colorectal and pancreas cancer patients with the elevation of the level of serum CEA and/or CA 19-9. IMACIS-1 was injected intravenously during 30 minutes in 100 ml saline solution after skin test. Planar scintigrams were recorded 3, 5 and 7 days after the injection of the IMACIS-1. Anterior, lateral and posterior views of the liver as well as anterior and posterior views of the pelvis were obtained in each patients as an $^{131}I-antibody$ image. We were able to localize exactly the malignant process with the double-nuclide double-compound $^{99m}Tc\;^{131}I$ (Tc+l) scintigrams. In Tc & I double-nuclide scintigraphy, computer subtraction display provided more clear localization of the tumor. We compared the results of radioimmunoscintigraphy with CT, ultrasonograms, conventional scintigrams. The results were as follows: 1) The sensitivity and specificity of radioimmunoscintigraphy using the fragments $F(ab')_2$ of the cocktails of CEA and CA 19-9 monoclonal antibodies were 80% and 100% respectively. 2) Tumor detection rate was not proportionated to the level of serum tumor markets. 3) Second tracer technique was essential for tumor localization as an anatomic landmark using double-nuclide scintigraphy. 4) A slow infusion of the antibodies was necessary to prevent the formation of large immune complexes. 5) Tumor/non-tumor radioactivity was most elevated at 7 days delayed imaging. 6) Using planar scintigraphic technique of $^{131}I$ labeled monoclonal antibodies are possible for imaging most of the tumors.

Efficient Anti-Tumor Immunotherapy Using Tumor Epitope-Coated Biodegradable Nanoparticles Combined With Polyinosinic-Polycytidylic Acid and an Anti-PD1 Monoclonal Antibody

  • Vaccination with tumor peptide epitopes associated with MHC class I molecules is an attractive approach directed at inducing tumor-specific CTLs. However, challenges remain in improving the therapeutic efficacy of peptide epitope vaccines, including the low immunogenicity of peptide epitopes and insufficient stimulation of innate immune components in vivo. To overcome this, we aimed to develop and test an innovative strategy that elicits potent CTL responses against tumor epitopes. The essential feature of this strategy is vaccination using tumor epitope-loaded nanoparticles (NPs) in combination with polyinosinic-polycytidylic acid (poly-IC) and anti-PD1 mAb. Carboxylated NPs were prepared using poly(lactic-co-glycolic acid) and poly(ethylene/maleic anhydride), covalently conjugated with anti-H-2Kb mAbs, and then attached to H-2Kb molecules isolated from the tumor mass (H-2b). Native peptides associated with the H-2Kb molecules of H-2Kb-attached NPs were exchanged with tumor peptide epitopes. Tumor peptide epitope-loaded NPs efficiently induced tumor-specific CTLs when used to immunize tumor-bearing mice as well as normal mice. This activity of the NPs significantly was increased when co-administered with poly-IC. Accordingly, the NPs exerted significant anti-tumor effects in mice implanted with EG7-OVA thymoma or B16-F10 melanoma, and the anti-tumor activity of the NPs was significantly increased when applied in combination with poly-IC. The most potent anti-tumor activity was observed when the NPs were co-administered with both poly-IC and anti-PD1 mAb. Immunization with tumor epitope-loaded NPs in combination with poly-IC and anti-PD1 mAb in tumor-bearing mice can be a powerful means to induce tumor-specific CTLs with therapeutic anti-tumor activity.

고려 홍삼이 대뇌 백질 미세구조 및 인지기능에 미치는 효과 : 침입 오류를 중심으로 (Effects of Korean Red Ginseng on White Matter Microstructure and Cognitive Functions : A Focus on Intrusion Errors)

  • Objectives Although ginseng has been reported to protect neuronal cells and improve various cognitive functions, relationship between ginseng supplementation and response inhibition, one of the important cognitive domains has not been explored. In addition, effects of ginseng on in vivo human brain have not been investigated using the diffusion tensor imaging (DTI). The purpose of the current study is to investigate changes in intrusion errors and white matter microstructure after Korean Red Ginseng supplementation using standardized neuropsychological tests and DTI. Methods Fifty-one healthy participants were randomly allocated to the Korean Red Ginseng (n = 26) or placebo (n = 25) groups for 8 weeks. The California Verbal Learning Test was used to assess the number of intrusion errors. Intelligence quotient (IQ) was measured with the Korean Wechsler Adult Intelligence Scale. Depressive and anxiety symptoms were evaluated using Hamilton Depression Rating Scale, Hamilton Anxiety Rating Scale, and Hopkins Symptom Checklist-25. The fractional anisotropy (FA) was measured from the brain DTI data. Results After the 8-week intervention, Korean Red Ginseng supplementation significantly reduced intrusion errors after adjusting age, sex, IQ, and baseline score of the intrusion errors (p for interaction = 0.005). Change in FA values in the left anterior corona radiata was greater in the Korean Red Ginseng group compared to the placebo group (t = 4.29, p = 0.04). Conclusions Korean Red Ginseng supplementation may be efficacious for improving response inhibition and white matter microstructure integrity in the prefrontal cortex.

X-선 조사식품 4종의 유전독성학적 안전성 평가 (Genotoxicological Safety Evaluation of X-ray Irradiated Four Foods)

  • 본 연구는 방사선 조사식품에 대한 소비자들의 수용성을 증대하고, 감마선조사의 대체기술로 X-선 조사의 상용화를 확대할 목적으로 30 kGy X-선 조사식품 4종(닭고기, 전란분말, 건파, 후추)의 유전독성학적 안전성 평가를 실행하였다. Salmonella Typhimurium TA98, TA100, TA1535 및 TA1537에 대한 X-선 조사식품 4종의 복귀변이 집락 수를 조사한 결과, 대사 활성계 도입 및 부재 시 모두 시험 적용 농도인 $40{\sim}5,000{\mu}g/plate$의 범위에서 복귀변이 집락 수의 농도 의존적인 증가 혹은 감소를 보이지 않았다. 그리고 포유류 배양세포를 이용한 염색체 이상 시험에서도 X-선 조사식품 4종은 $625{\sim}5,000{\mu}g/mL$의 시험 적용 농도에서 염색체 이상 유발능이 5% 미만이어서 염색체 이상을 유발하지 않는 것으로 나타났다. 또한 설치류 망상적혈구를 이용하여 X-선 조사식품 4종의 소핵 형성 시험을 수행한 결과 시험 적용 농도인 250~2,000 mg/kg의 범위에서 소핵을 가진 망상적혈구의 출현율이 음성대조군과 유의한 차이를 나타내지 않아 소핵을 유발하지 않음을 확인하였다. 이상의 결과를 바탕으로 위해 분석 결과 30 kGy X-선 조사식품 4종(닭고기, 전란분말, 건파, 후추)은 본 시험조건에서 유전 독성이 없는 것으로 나타났다.

Arsenic Toxicity on Duck Spermatozoa and the Ameliorating Effect of L-Ascorbic Acid

  • The objectives of this study were to understand the possible mechanism of duck sperm toxicity induced by arsenic exposure in vivo, and to investigate the roles of the antioxidant L-ascorbic acid in ameliorating the arsenic-induced sperm impairment. To test the acute toxicity, the percentages of mortality of mature drakes treated with different concentrations of trivalent sodium arsenite, As (III), and pentavalent sodium arsenate, As (V) were measured. The LD50 value of As (III) for mature drakes was $4.89{\pm}1.49$ ppm. Although As (V) didn't cause any deaths even at a concentration of 40 ppm, the chronic toxicity of As (V) on sperm quality was shown by a decreased fertilization rate. When the concentrations of As (V) were above 0.4 ppm, fertilization rates were lower than those of 0.04 ppm and control. Drakes treated with 40 ppm of As (V) had the highest malondialdehyde (MDA) level in the testis tissue, $3.100{\pm}0.218{\mu}mole/g$ testis. This showed that 40 ppm of As (V) significantly induced lipid peroxidation in testis tissue. For the 1.2 ppm As (III) treatment, several significant effects were observed: (1) sperm motility was decreased most dramatically by $52.0{\pm}9.1$% after three days of incubation; (2) fertilization rate of artificially inseminated semen was the lowest, $26.4{\pm}15.4$; (3) the MDA concentration in testis tissue, $7.846{\pm}0.246{\mu}mole/g$ testis, was significantly higher than the others (p<0.05); (4) the sperm number, $1.17{\pm}0.40({\times}10^9)$, was significantly lower than with the 60 ppb and control treatments (p<0.05); (5) a black appearance and soft texture was observed in the testis tissue. The antioxidant L-ascorbic acid administered along with 1.2 ppm As (III) decreased the toxicity of arsenic. The ameliorating effects included: improved sperm motility, increased sperm number and fertilization rate, and decreased MDA concentration in the testis tissue. This study suggests that the toxicity of the trivalent arsenic on sperm quality is partly from free radicals generated by its metabolic pathway, and the antioxidant ascorbic acid ameliorates arsenic-caused sperm impairment.

잔디 피시움마름병(Pythium blight)의 생물학적 방제를 위한 길항 미생물의 선발과 효력 검정 (Development of Antagonistic Microorganism for Biological Control of Pythium Blight of Turfgrass)

  • Pythium spp. 균에 의해 발생되는 피시움마름병(Pythium blight)은 골프장 퍼팅 그린에서 문제가 되는 대표적 병해이다. 본 연구에서는 Pythium aphanidermatum를 피시움마름병 병원균으로 하여 생물학적 방제법에 이용할 수 있는 길항 미생물을 1차 선발하였고, 선발된 길항 미생물에 대해 폿트 수준의 생물 검정 실험을 실시하였다. 두 실험을 동해 선발된 미생물 중에서 Relative Performance Indies 값을 기준으로 가장 우수한 것을 선별하여 피시움마름병에 대한 길항 미생물로 최종 선발하였고, 동정한 결과 Bacillus subtilis로 밝혀졌다. 최종 선발된 미생물은 B. subtilis GB-0365라 명명하였고, 화학 농약에 대한 내성 실험과 혼용 처리에 따른 효과를 검정하여 화학 농약과 시간의 차이를 두고 교호 살포할 경우 방제효과가 보다 높게 나타났다. 골프장의 간이 묘포장에서 자연 발생된 피시움마름병에 대한 B. subtilis GB-0365의 방제효과를 검증한 결과, 무처리 대비 방제가는 56.4%, 화학 농약 대비 방제가는 60.9%로 우수한 방제 효과을 가진 것으로 판명되었다.

스쿠티카충 Miamiensis avidus에 대한 넙치의 면역반응 (Immune Response of Olive flounder, Paralichthys olivaceus against Miamiensis avidus (Ciliophora: Scuticociliatida))

  • Miamiensis avidus는 넙치에 폐사를 일으키는 스쿠티카 섬모충이다. M. avidus에 대한 넙치의 면역반응을 측정하기 위하여 FKC (formalin killed cell)를 마리당 2.6 × 106 cells을 복강주사하고, FKC를 초음파처리한 것을 2.4 × 106 cells/㎖로 14.9 cm (26.8 g)의 넙치에 2주 간격으로 두 번 면역하였다. 면역한 넙치의 항체는 충체의 운동성둔화 (immobilization)와 응집 (agglutination)을 유발하였다. 면역한 그룹은 면역하지 않은 그룹보다 높은 ELISA항체가를 나타내었다. 또한, 복강주사로 면역한 넙치에서 유의적으로 높은 phagocytic ratio, phagocytic index와 주화활성을 나타내었다. FKC의 면역방어효과를 검증하기 위하여 8.6 cm (6.3 g)의 넙치에 마리당 1.43 × 105 cells을 복강주사면역하고 2.2 ×105 cells/㎖의 농도로 침지면역했다. 2주 간격으로 두 번 면역하고 2.0 × 104과 2.0 × 103 cells/㎖ 농도로 침지 공격 감염시킨 후 폐사율을 관찰하였다. 감염시킨 3주 후 폐사율은 복강주사구에서 감소하였고 침지면역구는 폐사가 지연되었다. 위의 결과로부터 M. avidus의 불활화백신을 넙치에 면역하면 충에 대한 특이면역이 증강되며 증강된 면역반응이 방어력을 제공했을 것으로 생각되었다.
