  • For the spatiotemporally aligned observation of photothrombosis induction and transient variations of in vivo brain stroke, we developed a novel photothrombosis inducing system compatible to a magnetic resonance imaging (MRI) system using nonmagnetic stereotaxic equipment. From the spatial point of view, the system provides a more reliable level of reproducibility of the photothrombosis in each brain. From the temporal point of view, from T1- and T2-weighted in vivo MR (magnetic resonance) images, the transient variations such as incidence, location, and size of the thrombosis are measured quantitatively. In addition, the final variation is observed in the ex vivo brain by TTC (Triphenyltetrazolium chloride) staining based on histological assay and utilized for the verification of the MR images. From the experimental result of the rat brain, the proposed system shows more reliable characteristics for transient variations of brain strokes.

  • In vivo expression technology (IVET) has been developed to study bacterial gene expression in Salmonella typhimurium during host infection. The expression of selected genes by IVET has been elevated in vivo but not in vitro. The selected genes turned out to be important for bacterial virulence and/or pathogenicity. IVET depends on a synthetic operon with a promoterless transcriptional fusion between a selection marker gene and a reporter gene. The IVET approach has been successfully adapted in other bacterial pathogens and plant-associated bacteria using different selection markers. Pseudomonas putida suppresses citrus root rot caused by Phytophthora parasitica and enhances citrus seedling growth. The WET strategy was adapted based on a transcriptional fusion, pyrBC'-lacZ, in P. putida to study the bacterial traits important far biocontrol activities. Several genes appeared to be induced on P. parasitica hyphae and were found to be related with metabolism and regulation of gene expression. It is likely that the biocontrol strain took a metabolic advantage from the plant pathogenic fungus and then suppressed citrus root rot effectively. The result was parallel with those from the adaptation of IVET in P. fluorescens, a plant growth promoting rhizobacteria (PGPR). Interestingly, genes encoding components for type III secretion system have been identified as rhizosphere-induced genes in the PGPR strain. The type III secretion system may play a certain role during interaction with its counterpart plants. Application of IVET has been demonstrated in a wide range of bacteria. It is an important strategy to genetically understand complicated bacterial traits in the environment.

  • Diseases in the gastro-intestinal track are on an increasing trend. In order to diagnose a patient, the various signals of the digestive organ, such as temperature, pH, and pressure, can offer the helpful information. Among the above mentioned signals, we choose the pressure variation as a monitoring signal. The variation of a pressure signal of the gastro-intestinal track can offer the information of a digestive trouble or some clues of the diseases. In this paper, a pressure monitoring system for the digestive organs of a living pig is presented. This system concept is to transmit the measured biomedical signals from a transmitter in a living pig to wireless receiver that is positioned out of body. The integrated solution includes the following parts: (1) the swallow type pressure capsule, (2) the receiving set consisting of a receiver, decoder box, and PC. The merit of the proposed system if that the monitoring system can supply the precise and repeatable pressure in the gastro-intestinal track. In addition, the design of low power consumption enables it to keep sending reliable signals while the pressure capsule is working in the digestive organ. The subject of the study for the pressure monitoring system is in-vivo experiments for a living pig. We achieved the pressure tracings in digestive organs and verified the validity of system after several in-vivo tests using pressure monitoring system. As a result, we found each organ has its own characterized pressure fluctuation.

  • The chlorophyll fluorescence combined with the O-J-I-P transients were examined in the leaves of the crinum plants (Crinum asiaticum var. japonicum BAK.), in order to satisfy the demand for rapid in vivo measurement of vitality, and to apply easily to approach questions of economical interest concerning the plant vitality. The photosynthetic efficiency, Fv/Fm, of crinum plants dramatically decreased depending on temperature drop in winter. In summer, the Fv/Fm values was lower in day time than at dawn and night, suggesting that photosynthetic efficiency is chronically photoinhibited in day time. In winter, there was no prominent diurnal fluctuations of Fv/Fm values. However, based on the O-J-I-P transient, PI$\_$NO/ and SFI$\_$NO/ dramatically increased at noon in summer, and $\psi$o/(1-$\psi$o) diurnally fluctuated in winter. These results indicated that vitality indexes such as PI$\_$NO/, SFI$\_$NO/ and $\psi$o/(1-$\psi$o) can be used as the indicators for in vivo measurement of environmental stresses.

  • Huperzine A-loaded microspheres composed of poly(D,L-lactide-co-glycolide) were prepared by an O/w emulsion solvent evaporation method. The characterization of the microspheres such as drug loading, size, shape and release profile was described. The in vitro release in the initial 7 days was nearly linear with $10\%$ released per day. Thereafter drug release rate became slow gradually and about $90\%$ drug released at day 21. The in vitro release rate determined by dialysis bag method had a good correlation with the in vivo release rate. Huperzine A aqueous solution was intramuscularly injected (i.m.) at 0.4mg/kg and microspheres were intramuscularly injected at 8.4 mg eq huperzine A/kg in rats. The maxium plasma concentration $(C_{max})$ after i.m. microspheres was only $32\%$ of that after i.m. solution. Drug in plasma could be detectd until day 14 and about $5\%$ of administered dose was residued at the injection site at day 14. The relative bioavailability of huperzine A microspheres over a period of 14 days was $94.7\%$. Inhibition of acyecholinesterase activity (AchE) in rat's cortex, hippocampus and striatum could sustain for about 14 days. In conclusion, huperzine A-loaded microspheres possessed a prolonged and complete drug release with significant inhibition of AchE for 2 weeks in rats.

  • Background: Artesunate, extracted from Artemisia annua, has been proven to have anti-cancer potential. Allicin, diallyl thiosulfinate, the main biologically active compound derived from garlic, is also of interest in cancer treatment research. This object of this report was to document synergistic effects of artesunate combined with allicin on osteosarcoma cell lines in vitro and in vivo. Methods: After treatment with artesunate and allicin at various concentrations, the viability of osteosarcoma cells was analyzed by MTT method, with assessment of invasion and motility, colony formation and apoptosis. Western Blotting was performed to determine the expression of caspase-3/9, and activity was also detected after drug treatment. Moreover, in a nude mouse model established with orthotopic xenograft tumors, tumor weight and volume were monitored after drug administration via the intraperitoneal (i.p.) route. Results: The viability of osteosarcoma cells in the combination group was significantly decreased in a concentration and time dependent manner; moreover, invasion, motility and colony formation ability were significantly suppressed and the apoptotic rate was significantly increased through caspase-3/9 expression and activity enhancement in the combination group. Furthermore, suppression of tumor growth was evident in vivo. Conclusion: Our results indicated that artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis.

  • Body water compartments in vivo were determined in Holstein cattle with age ranging from 5 to 521 days to obtain an equation to estimate volumes of body water. Live weight ranged from 47 to 480 kg. Compartments were determined as antipyrine space for total body water (TBW), thiocyanate space for extracellular water (ECW) and Evans blue dye space for plasma water (PW). Body water compartments expressed as a percentage of live weight decreased as age in days increased and significantly correlated with age in days. Regression analyses revealed that prediction equations had low accuracy. Regression equations of body water compartments on live weight (WT, kg) were useful for the prediction of body fluid with a high accuracy. Live weight significantly regressed on age in days (Day), which was inferred to be utilized for estimation of standardized live weight in case animals were emaciated by certain causes such as severe diarrhea or dehydration. In conclusion, following equations were presented to estimate body water compartments of cattle in vivo : TBW in liters = $0.556({\pm}0.007)WT+10$, r = 0.993, $SE{\pm}0.7$ ECW in liters = $0.321({\pm}0.008)WT+10$, r = 0.978, $SE{\pm}0.8$ PW in liters = $0.0502({\pm}0.0012)WT+1.6$, r = 0.0983, $SE{\pm}0.1$ WT (kg) = $0.772({\pm}0.018)Day+24$, r = 0.982, $SE{\pm}2.3$.

  • A method of detecting lead was developed using square wave anodic stripping voltammetry (SWASV) with DNA-carbon nanotube paste electrode (CNTPE). The results indicated a sensitive oxidation peak current of lead on the DNA-CNTPE. The curves were obtained within a concentration range of 50 $ngL^{-1}-20mgL^{-1}$ with preconcentration time of 100, 200, and 400 sec at the concentration of $mgL^{-1}$, ${\mu}gL^{-1}$, and $ngL^{-1}$, respectively. The observed relative standard deviation was 0.101% (n = 12) in the lead concentration of 30.0 ${\mu}gL^{-1}$ under optimum conditions. The low detection limit (S/N) was pegged at 8 $ngL^{-1}$ ($2.6{\times}10^{-8}M$). Results showed that the developed method can be used in real-time assay in vivo without requiring any pretreatment and pharmaceutical samples, and food samples, as well as other materials requiring water source contamination analyses.

  • The aim of this study is to evaluate the effects essential oil from Lavendular officinalis on the production of UVB-irradiated-induced nitric oxide(NO), in vivo and in vitro. NO is a recently discovered mediator of cell communication involved in a variety of physiological and pathophysiological processes. This enzyme is present in various tissues including smooth muscle cells and macrophages and take part in several immunopathological process. In vitro, the cytotoxicity and cell viability of aroma oil was evaluated by the MTT assay in the concentration of 0.01, 0.05, 0.1%. And, the effect of aroma oil was investigated to production of NO in human fibroblast cells line CCD-986sk ($2{\times}10^5$ cell/well) after UVB-irradiation with aroma oil (0.01, 0.1, and 1%). The result showed that aroma oil did not affected the production of NO. In vivo, it was investigated to production of NO after UVB- irradiation with aroma oil. The experimental groups were divided into four groups. Aroma oil was stimulated the production of NO by itself. As the results, all of the in vitro and in vivo, aroma oil were affected production of NO by dependent the concentration-manners.
